        Original Articles : Effect of aldosterone on the amplification of oncolytic vaccinia virus in human cancer Lines

        Background/Aims: JX-594 is an oncolytic virus derived from the Wyeth vaccinia strain that causes replication-dependent cytolysis and antitumor immunity. Starting with a cross-examination of clinical-trial samples from advanced hepatocellular carcinoma patients having high levels of aldosterone and virus amplification in JX-594 treatment, we investigated the association between virus amplification and aldosterone in human cancer cell lines. Methods: Cell proliferation was determined by a cell-counting-kit-based colorimetric assay, and vaccinia virus quantitation was performed by quantitative polymerase chain reaction (qPCR) and a viral plaque assay. Also, the intracellular pH was measured using a pH-sensitive dye. Results: Simultaneous treatment with JX-594 and aldosterone significantly increased viral replication in A2780, PC-3, and HepG2 cell lines, but not in U2OS cell lines. Furthermore, the aldosterone treatment time altered the JX-594 replication according to the cell line. The JX-594 replication peaked after 48 and 24 hours of treatment in PC-3 and HepG2 cells, respectively. qPCR showed that JX-594 entry across the plasma membrane was increased, however, the changes are not significant by the treatment. This was inhibited by treatment with spironolactone (an aldosterone-receptor inhibitor). JX-594 entry was significantly decreased by treatment with EIPA [5-(N-ethyl-N-isopropyl)amiloride; a Na+/H+-exchange inhibitor], but aldosterone significantly restored JX-594 entry even in the presence of EIPA. Intracellular alkalization was observed after aldosterone treatment but was acidified by EIPA treatment. Conclusions: Aldosterone stimulates JX-594 amplification via increased virus entry by affecting the H+ gradient. (Korean J Hepatol 2011;17:213-219)

        Designing Co7Fe3@TiO2 Core–Shell Nanospheres for Electromagnetic Wave Absorption in S and C Bands

        Ferromagnetic-dielectric nanocomposites have attracted extensive interests for its high electromagnetic wave absorption(EMA) performance due to the synergetic effects between different components. Herein, we report the design of core–shellstructured Co7Fe3@TiO2 composite particles, which are subsequently annealed in H2/Ar atmosphere to further improve itsEMA performance. The introducing of TiO2dielectric shell together with hydrogen annealing contributes greatly to theelectromagnetic properties due to the increased conductivity and enhanced ferromagnetic resonance. Excellent EMA performanceis achieved in S (2–4 GHz) and C (4–8 GHz) bands in coatings using Co7Fe3@TiO2 as absorbents. Apart from thehigh EMA efficiency, the location of EMA band can be tailored in a wide range through regulating the coating thickness. Specifically, an effective absorption band of 2.0 GHz in C band at a thickness of 2.62 mm, and an effective absorption bandof 1.7 GHz are achieved in S band at thicknesses of 4.0–5.0 mm. The excellent electromagnetic properties are ascribed tothe effective complementary between dielectric loss and ferromagnetic loss.

        Aeromicrobium endophyticum sp. nov., an endophytic actinobacterium isolated from reed (Phragmites australis)

        A Gram-staining-positive, motile and short-rod-shaped actinobacterium designated 9W16Y-2T was isolated from surface- sterilized leaves of reed (Phragmites australis) collected from Taklamakan Desert in Xinjiang Uygur Autonomous Region, China. Colonies were pale greenish yellow, circular, smooth, and convex. The 16S rRNA gene sequence of strain 9W16Y-2T exhibited highest sequence similarities with Aeromicrobium camelliae CGMCC 1.12942T (99.0%) and Aeromicrobium erythreum NRRL B-3381T (97.2%). Phylogenetic analyses based on 16S rRNA gene sequences and single-copy phylogenetic marker genes (pMGs) showed that strain 9W16Y- 2T belonged to the genus Aeromicrobium and formed a monophyletic clade with Aeromicrobium camelliae CGMCC 1.12942T. Furthermore, average nucleotide identity (ANI) and DNA-DNA hybridization (DDH) clearly separated strain 9W16Y-2T from the other species of the genus Aeromicrobium with values below the thresholds for species delineation. The G+C content of the genomic DNA is 68.9 mol%. The diagnostic diamino acid of the cell-wall peptidoglycan was LLdiaminopimelic acid. The predominant menaquinone was MK-9(H4). The major fatty acids (> 10% of the total fatty acids) were C18:0 10-methyl (TBSA) (28.2%), C16:0 (21.0%), C16:0 2-OH (20.8%) and C18:1 ω9c (12.8%). The polar lipid profile comprised diphosphatidylglycerol, phosphatidylglycerol, phosphatidylcholine, phosphatidylinositol, an unidentified aminophospholipid and an unidentified lipid. Based on the phylogenic, phenotypic and chemotaxonomic features, strain 9W16Y-2T represents a novel species of the genus Aeromicrobium, for which the name Aeromicrobium endophyticum sp. nov. is proposed. The type strain is 9W16Y-2T (= CGMCC 1.13876T = JCM 33141T).

        Soft-template-carbonization route to highly textured mesoporous carbon–TiO<sub>2</sub> inverse opals for efficient photocatalytic and photoelectrochemical applications

        <P>Hierarchically organized mesoporous carbon–TiO<SUB>2</SUB> inverse opal nanostructures were synthesized by complementary colloid and block copolymer (BCP) self-assembly, where the triblock copolymer P123 acts simultaneously as the template and the carbon source. Highly ordered mesoporous inverse opal nanostructures with a nano-textured surface morphology and multiple-length scale nanopores provide increased light-activated surface area and scattering effects, leading to enhanced photoabsorption efficiency and the transport of matter. UV-vis absorption, X-ray photoelectron spectroscopy and Mott–Schottky measurement studies show that incorporation of carbon moieties into TiO<SUB>2</SUB><I>via</I> direct conversion of BCPs creates a new energy level above the valence band of TiO<SUB>2</SUB>, resulting in an effective decrease in the band gap.         Screening of Candida utilis and medium optimization for co-production of S-adenosylmethionine and glutathione

        An effective S-adenosylmethionine and glutathione enriching yeast mutant of Candida utilis CCTCC M 209298 was first screened from plates containing 0.5 g/L of DL-ethionine by complex mutagenesis with UV and γ-ray in this study. Medium components optimization for enhanced co-production of S-adenosylmethionine and glutathione by C. utilis CCTCC M 209298 was further carried out using response surface methodology. The significant factors influencing S-adenosylmethionine and glutathione co-production were selected by Plackett-Burman design as sucrose, KH2PO4 and L-methionine, and Box-Behnken design was applied for further optimization studies. Based on these approaches, the optimized concentrations on medium components for higher co-production of S-adenosylmethionine and glutathione were sucrose 35.4 g/L, (NH4)2SO4 10 g/L, KH2PO4 12.3 g/L, MgSO4·7H2O, 0.05 g/L, CaCl20.05 g/L and L-methionine 4.6 g/L. The medium optimization by response surface methodology led to a total production of 589.3 mg/L on S-adenosylmethionine and glutathione, which was 2.4-fold increased compared with the medium without optimization.

        ORiginal Article : Association of IL28B Genotypes and Baseline Serum Interferon-γ-Inducible-Protein-10 Levels with Treatment Response in Hepatitis C Virus Patients in China

        Background/Aims: Several studies have demonstrated that serum interferon-γ-inducible-protein-10 (IP-10) levels at baseline and single nucleotide polymorphisms (SNPs) near the IL28B gene were associated with viral response and treatment outcomes. Our purpose was to assess the combination of pretreatment IP-10 levels with IL28B SNPs as predictors of treatment response to pegylated interferon α-2a plus ribavirin in patients infected with genotype 1 hepatitis C virus in China. Methods: Seventy-two patients with chronic hepatitis C without fibrosis/cirrhosis were enrolled in the study. The virologic parameters and baseline serum IP-10 levels were determined. IL-28B genotypes were determined by sequencing. Results: In this cohort, serum baseline IP-10 levels lower than 426.7 pg/mL could predict rapid virological response/ sustained virological response (SVR). Patients carrying favorable IL28B SNP genotypes had higher SVRs than did those carrying unfavorable variants (IL28B rs12979860, p=0.002; IL28B rs8099917, p=0.020). Combining both baseline IP- 10 and IL28B SNPs could improve the prediction of SVR in favorable allele carriers of IL28B, rs12979860 CC and rs8099917 TT. Serum baseline IP-10 levels and IL28B genotypes were independent predictors of SVR. Conclusions: Our study shows that the combination of baseline serum IP-10 levels and the determination of IL28B SNPs increase the predictability of SVR rates in this cohort.         A novel strategy on the high-cell-density cultivation of Candida utilis for the enhanced production of glutathione

        Efficient glutathione production by high-cell-density cultivation of Candida utilis was investigated. A series of batch glutathione fermentations were carried out and the optimal initial glucose concentration was found to be about 26 g/L. Then, fed-batch fermentation under diverse feeding strategies was used to enhance glutathione production with a total glucose concentration of 150 g/L. Constant glucose feeding strategy cannot meet the requirement of cells at the late period of feeding, while exponential glucose feeding strategy cannot satisfy the needs of cells at the beginning of feeding. Based on the results above, a polynomial glucose feeding strategy was developed to provide enough glucose for cells along with the cultivation, under which both the cell and glutathione productivity were satisfactorily improved. Furthermore, fed-batch fermentation under this strategy with a total glucose concentration of 200 g/L was successfully performed, the dry cell weight and glutathione concentration reached 91.2 g/L and 825 mg/L, respectively.

        中国香港地区CISG适用问题研究

        United Nations Convention on Contracts for International Sale of Goods (CISG) is considered as a core international trade law that may apply to a contract for international sales of goods by most countries in the world. CISG was legislated by United Nations Commission on International Trade Law in 1980 and China was one of the earliest nations that adopted CISG. Although Constitution of the People’s Republic of China states that Chinese territory covers the Chinese mainland, Hong Kong, Macao and Taiwan, there is still a controversy as to whether CISG could be regarded as of applicable law in the Chinese mainland and other territorial units including Hong Kong. This is due to the different legal systems that each region is currently adopting. In this study, we will focus on the pros and cons of application of CISG to Hong Kong region. Even after Hong Kong’s return to Chinese rule, the central government of China still have not made any declaration regarding the application of CISG to Hong Kong for the reason that Hong Kong was once a British colony and United Kingdom was not of a contracting state of CISG. Until now Hong Kong’s application of CISG is instigating many arguments among states in the world. According to the cases of each country, civil law countries such as France claims that CISG should not be adopted by Hong Kong. In contrast, in the cases in common law countries such as United States of America states that CISG must be applied to Hong Kong. As such, there is no universal consensus with regard to this issue. This study will first summarize the positions of each state toward this issue and then analyze how the Chinese government must conduct for the greatest effect. This study will be conducted based on the assumption that the Chinese government is likely to declare that CISG is applicable to Hong Kong. However, it does not mean that this research will completely exclude the possibility that the Chinese government will continuously keep silence. As a participant in international trade, we are obligated to pronounce our opinions for this issue of the application of CISG to Hong Kong before the Chinese government finally makes a statement to this problem.
