Analysis of hematological changes in normal and diarrhea calves

In order to effective treats a sick calf, it is necessary to obtain accurate information about the patient’s condition. However, the standard references for Korean cattle claves are not well known. Therefore, this study aims to present useful clinical values by analyzing normal blood and diarrhea blood of Hanwoo calf. Recently, with the advent of a portable blood analyzer, it is possible to immediately analyze the patient’s condition and severity in the field, not in the laboratory, and to calculate a suitable dosage for supporting fluid therapy. Therefore, in this study, the distribution of red blood cells (RBC) and white blood cells (WBC) were analyzed in normal and diarrhea Hanwoo calves. As a result, Hematocrit (HCT) levels increase significantly between 1 and 20 days in diarrhea positive calves. Changes in leukocyte composition had similar growth patterns in normal and diarrhea calves. As it grew, the proportion of neutrophils decreased and lymphocytes increased. However, the number of WBCs increased from 1 to 10 and 21 to 30 days in diarrhea positive calves, which is closely related to the increase in neutrophils. Therefore, those data can be used for diagnosis and treatment of diarrhea calf.

Comparison of polymerase chain reaction for antigen receptor gene rearrangement and flow cytometric analysis for the diagnosis of canine Lymphoma

( Ru Hui Song ),( Do Hyeon Yu ),( Jun Hwan Kim ),( Hyun Seok Lee ),( Da Mi Lee ),( Chul Park ),( Il Jung Yu ),( Jin Ho Park ) 한국가축위생학회 2011 韓國家畜衛生學會誌 Vol.34 No.3

Lymphoma is the most common hematopoietic malignancy in dogs. Diagnosis of lymphoma is classically performed by morphological assessment and immunohistochemistry. But some cases in the early stage are difficult to distinguish and need more objective and accurate methods. So, Polymerase chain reaction (PCR) for antigen receptor rearrangements (PARR) and flow cytometric immunophenotype of lymphoma have been developed continuously. In this study, we performed these two methods to classify lymphoma type in 3 cases. According to PARR analysis, B cell origin lymphoma was diagnosed in two of three cases by testing PBMC and lymph node. All fine needle aspiration (FNA) samples of lymph nodes had high expression of CD21 on >88% of total cell population and PBMC samples also showed high expression of CD21 on >30% of total lymphocytes in those two cases, while the expression of CD3, CD4 and CD8 was absent. These results suggest that concurrent use of PARR and flow cytometric immunophenotype is more effective and valuable tool for the diagnosis and monitoring of canine lymphoma patients.

Hyperinsulinism in a dog with beta-cell neoplasia (insulinoma)

        Yu, Do-Hyeon,Lee, Jong-Hyun,Song, Ru-Hui,Noh, Dong-Ho,Li, Ying-Hua,Lee, Mi-Jin,Cho, Ara,Kim, Bumseok,Park, Jinho The Korean Society of Veterinary Science 2009 大韓獸醫學會誌 Vol.49 No.4

A six-year-old female cocker spaniel presented with recurring episodes of pelvic limb weakness and intermittent seizures. Laboratory analysis revealed marked hypoglycemia and an elevated serum insulin concentration. A pancreatic beta-cell tumor at stage III ($T_1N_1M_1$) was diagnosed based on serial blood glucose and insulin measurements along with diagnostic imaging. The patient survived for 140 days after diagnosis with medical management, including frequent feeding and prednisolone therapy. On necropsy, necrosis and masses in the peripancreatic omentum and liver were found; pancreatic beta-cell neoplasia with metastasis to the liver was confirmed by histopathologic examination. This case reports hyper-insulinism in a dog presenting with hypoglycemic seizures.

Hemophilia B (factor IX deficiency) in a Labrador retriever dog

( Jun Hwan Kim ),( Dong Ho Noh ),( Ru Hui Song ),( Da Mi Lee ),( Ho Seong Cho ),( Do Hyeon Yu ),( Jin Ho Park ),( Chul Park ) 한국가축위생학회 2011 韓國家畜衛生學會誌 Vol.34 No.2

A 3-month-old intact male, Labrador retriever was presented with the history of coagulopathy and anemia. The results of initial screening tests of the hemostatic system yielded a tentative diagnosis of hemophilia. Activated partial thromboplastin time (APTT) was distinctly prolonged (106 seconds) and prothrombin time (PT) was not detected due to markedly prolonged test time. Whole blood transfusions (20 me l/kg body weight) were carried out prior to assays of coagulation factor. After transfusion, the patient recovered well and hemorrhage ceased. Blood samples were assessed for coagulation factor activity. The patient showed markedly low factor IX coagulation activity (5%, reference range: 7~140%) and was diagnosed with hemophilia B. After recovery, the patient was discharged from the hospital. However, 4 months later the patient was re-hospitalized for recurrence of the initial symptoms. The owner did not want to pursue further treatment and the patient died of respiratory distress two days later.

Ethyl Pyruvate Downregulates Tumor Necrosis Factor Alpha and Interleukin (IL)-6 and Upregulates IL-10 in Lipopolysaccharide-Stimulated Canine Peripheral Blood Mononuclear Cells

YU, Do-Hyeon,NOH, Dong-Ho,SONG, Ru-Hui,PARK, Jinho Japanese Society of Veterinary Science 2010 The Journal of veterinary medical science Vol.72 No.10

We investigated the anti-inflammatory effects of ethyl pyruvate (EP) on LPS-stimulated canine PBMCs in vitro. We found that EP treatment inhibited the mRNA expressions of proinflammatory cytokines (TNF-α and IL-6), but induced mRNA expression of anti-inflammatory cytokines (IL-10). ELISA measurements revealed that EP also effectively downregulates the LPS-induced increase in proinflammatory cytokine release, while upregulating anti-inflammatory cytokine release. These data indicate that EP could be an effective anti-inflammatory agent in dogs.

      • High-mobility group box 1 as a surrogate prognostic marker in dogs with systemic inflammatory response syndrome : HMGB1 in dogs with SIRS

        Yu, Do-Hyeon,Nho, Dong-Ho,Song, Ru-Hui,Kim, Sue-Hee,Lee, Mi-Jin,Nemzek, Jean A.,Park, Jinho Wiley (Blackwell Publishing) 2010 Journal of veterinary emergency and critical care Vol.20 No.3

OBJECTIVE: To evaluate various surrogate markers associated with the inflammatory and counter-inflammatory responses with respect to mortality in dogs with systemic inflammatory response syndrome (SIRS). DESIGN: Prospective observational study. SETTING: Veterinary Teaching Hospital. ANIMALS: Twenty-eight dogs with naturally occurring diseases and SIRS from January 2007 to May 2009. INTERVENTIONS: Upon admission to the veterinary hospital, history and baseline data from the physical examination, including parameters previously defined for meeting SIRS criteria, were documented. Heparinized blood samples were collected and plasma cytokines interleukin-6 (IL-6), IL-10, and high-mobility group box 1 (HMGB1) were measured by sandwich ELISA. MEASUREMENTS AND MAIN RESULTS: In nonsurvivors, median plasma HMGB1 concentrations (0.718 microg/L, interquartile range [IQR]; 0.300-1.626 microg/L) and the ratio of HMGB1 to IL-10 (2.236, IQR; 0.972-5.367) were significantly increased as compared with those found in survivors (0.300 microg/L, IQR; 0.300-0.312 microg/L for HMGB1; 1.017, IQR; 0.862-1.126 for the ratio of HMGB1 to IL-10, P=0.007 and 0.024, respectively). Plasma IL-6, IL-10, and the ratio of IL-6 to IL-10 were not significantly different between groups. Among the parameters studied, HMGB1 and the ratio of HMGB1 to IL-10 performed the best in discriminating outcome in dogs with SIRS according to receiver operator characteristic curve analysis. CONCLUSIONS: Increases in plasma HMGB1 concentration and the ratio of HMGB1 to IL-10 may predict poorer outcomes in dogs with SIRS. The approach described may lead to reliable prognostic biomarkers and new therapeutic concepts in the study of SIRS in dogs.

      • ppGalNAc T1 as a Potential Novel Marker for Human Bladder Cancer

        Ding, Ming-Xia,Wang, Hai-Feng,Wang, Jian-Song,Zhan, Hui,Zuo, Yi-Gang,Yang, De-Lin,Liu, Jing-Yu,Wang, Wei,Ke, Chang-Xing,Yan, Ru-Ping Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.11

Objectives: To investigate the effect of glycopeptide-preferring polypeptide GalNAc transferase 1 (ppGalNAc T1 ) targeted RNA interference (RNAi) on the growth and migration of human bladder carcinoma EJ cells in vitro and in vivo. Methods: DNA microarray assays were performed to determine ppGalNAc Ts(ppGalNAc T1-9) expression in human bladder cancer and normal bladder tissues. We transfected the EJ bladder cancer cell line with well-designed ppGalNAc T1 siRNA. Boyden chamber and Wound healing assays were used to investigate changes of shppGalNAc T1-EJ cell migration. Proliferation of shppGalNAc T1-EJ cells in vitro was assessed using [3H]-thymidine incorporation assay and soft agar colony formation assays. Subcutaneous bladder tumors in BALB/c nude mice were induced by inoculation of shppGalNAc T1-EJ cells and after inoculation diameters of tumors were measured every 5 days to determine gross tumor volumes. Results: ppGalNAc T1 mRNA in bladder cancer tissues was 11.2-fold higher than in normal bladder tissues. When ppGalNAc T1 expression in EJ cells was knocked down through transfection by pSUPER-shppGalNAc T1 vector, markedly reduced incorporation of [3H]-thymidine into DNA of EJ cells was observed at all time points compared with the empty vector transfected control cells. However, ppGalNAc T1 knockdown did not significantly inhibited cell migration (only 12.3%). Silenced ppGalNAc T1 expression significantly inhibited subcutaneous tumor growth compared with the control groups injected with empty vector transfected control cells. At the end of observation course (40 days), the inhibitory rate of cancerous growth for ppGalNAc T1 knockdown was 52.5%. Conclusion: ppGalNAc T1 might be a potential novel marker for human bladder cancer. Although ppGalNAc T1 knockdown caused no remarkable change in cell migration, silenced expression significantly inhibited proliferation and tumor growth of the bladder cancer EJ cell line.

Rotavirus-mediated alteration of gut microbiota and its correlation with physiological characteristics in neonatal calves

        Ja-Young Jang,Suhee Kim,Min-Sung Kwon,이지은,Do-HyeonYu,Ru-hui Song,Hak-Jong Choi,Jin-Ho Park 한국미생물학회 2019 The journal of microbiology Vol.57 No.2

Diarrhea is a fatal disease to neonatal calves, and rotavirus is the main pathogen associated with neonatal calf diarrhea. Although previous studies have reported that the gut microbiota is changed in calves during diarrhea, less is known about whether rotavirus infection alters the structure of the gut microbiota. Here, we characterized fecal microbial communities and identified possible relationships between the gut microbiota profiles and physiological parameters. Five fecal specimens of rotavirus-infected calves from 1 to 30 days after birth and five fecal specimens of age-matched healthy calves were used for the microbial community analysis using the Illumina MiSeq sequencer. Rotavirus infection was associated with reduced rotavirus infection significantly reduced the richness and diversity of the bacterial community. Weighted unique fraction metric analysis exhibited significant differences in community membership and structure between healthy and rotavirus-infected calves. Based on relative abundance analysis and linear discriminant analysis effect size, we found that the representative genera from Lactobacillus, Subdoligranulum, Blautia, and Bacteroides were closely related to healthy calves, while the genera Escherichia and Clostridium were closely affiliated to rotavirus-infected calves. Furthermore, canonical correlation analysis and Pearson correlation coefficient results revealed that the increased relative abundances of Lactobacillus, Subdoligranulum, and Bacteroides were correlated with normal levels of physiological characteristics such as white blood cells, blood urea nitrogen, serum amyloid protein A, and glucose concentration in serum. These results suggest that rotavirus infection alters the structure of the gut microbiota, correlating changes in physiological
