        The main goals of the study were to investigate the sediment quality and trace metal distribution in sediment of Kwangyang Bay, south coastal of Korea. Surface sediment sampling were performed in November, 2000 and February and March, 2001. The concentration of H₂S, IL and COD in sediment were measured by a manual of the Department of Marine Environment of Korea. The extracted trace metals Fe, Cu, Cd, Pb, and Zn in sediments mean analyzed by the flame atomic absorption spectrophotometer with an instrument of model Shimadzu AA-6701F. H₂S, IL and COD in the surface sediment were 1.3 mg/g, 5.9 % and 15.0 mg/g, respectively. The mean content of Fe, Cu, Cd, Pb and Zn were 1523 mg/kg, 4.799 mg/kg, 1.321 mg/kg 9.285 mg/kg and 9.353 mg/kg, respectively. All of these measured trace metals level were over the other reported trace metal level in sediment of coastal area. According to histograms, the content of H₂S, IL and COD and trace metals in sediment of this study were affected by artificial activities rather than natural process.

        Metacercariocidal effects and infection status of Gymnophalloides seoi in oysters

        We studied the infection rate of and various metacercariocidal approaches to controlling Gymnophalloides seoi for prevention of human infection in cultured and natural oysters in Korea. The selected survey areas were Aphae-do (Shinan-gun, Jeollanam-do), which is an endemic area for G. seoi, and Tongyeong (Geonsangnam-do), which is the main production area of oysters in Korea. In the Tongyeong area, the metacercariae of G. seoi were not detected in cultured oysters (0/201) or wild oysters (0/134). Seventy-two G. seoi metacercariae were observed in 33 of 265 natural oysters collected from Aphae-do; however, metacercariae were not detected in the cultured oysters (0/1101) purchased from the Daejeon Fish Market. To investigate the viability of G. seoi metacercariae, various metacercariocidal treatments were used with 3.5% saline and oyster juice used as positive controls. The metacercariae survived for 75.4 h in 3.5% saline and 112.6 h in oyster juice. After the metacercariocidal treatment, G. seoi metacercariae were survived for 13.29 min in tap water, < 20 sec in 4.3% vinegar, no effect in a rinse of the whole oyster body in 70°C water for 1 sec, but 1 sec in a rinse of the whole oyster body in 90°C water for 1 sec. The greatest metacercariocidal effect on G. seoi was from rinsing oysters in 90°C water followed by those from treatment with 20% ethyl alcohol, 4.3% vinegar, and tap water. However, we suggest that the most actual prevention to G. seoi human infection is rinsing the oysters with tap water for at least 30 min.

        Kinetics of HMGB1 level changes in a canine endotoxemia model

        In this study, we investigated the kinetics of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-6 and high mobility group box 1 (HMGB1) concentrations in a 48-h model of canine endotoxemia by lipopolysaccharide (LPS) injection. Four healthy beagles were slowly administered 1 mg/kg of LPS diluted in normal saline, while two others were administered normal saline as controls. Blood collection was performed at 0 h (baseline), 1 h and 3 h (for TNF-${\alpha}$), 6 h, 12 h, 24 h and 48 h of the experiment, and cytokine levels were determined using the sandwich ELISA method. Early increments of TNF-${\alpha}$ and IL-6 were observed (< 3 h), but HMGB1 levels increased the most at 12 h of the experiment and gradually decreased until 48 h. During the whole experiment, IL-6 and HMGB1 were sustained over 12 h of LPS injection, whereas TNF-${\alpha}$ decreased within 6 h of LPS injection. Taken together, canine HMGB1 levels increase relatively late (< 12 h) and sustained longer than TNF-${\alpha}$ and IL-6 in response to endotoxin. This is the first study to evaluate canine HMGB1 cytokine from endotoxemia in dogs.

        Lymphadenopathy Associated with Disseminated Aspergillosis in a Jindo Dog in Korea

        A 2-year-old outdoor, spayed, female Korean Jindo dog was referred for thoracic and abdominal masses. Diagnostic imaging revealed generalized lymphadenopathy and a significant amount of pleural effusion. The cytological evaluation of the pleural effusion and fine-needle aspiration of the mediastinal lymph nodes confirmed the presence of fungal hyphae. The histopathological examination of the mediastinal lymph nodes stained with Grocott’s methenam- ine silver revealed fungal mycelia, and Aspergillus terreus was identified based on fungal DNA sequencing. Thus, the dog was diagnosed with disseminated canine aspergillosis caused by A. terreus. This case report describes a rare case of disseminated canine aspergillosis caused by A. terreus in a Korean Jindo dog.

        Hyperinsulinism in a dog with beta-cell neoplasia (insulinoma)

        A six-year-old female cocker spaniel presented with recurring episodes of pelvic limb weakness and intermittent seizures. Laboratory analysis revealed marked hypoglycemia and an elevated serum insulin concentration. A pancreatic beta-cell tumor at stage III ($T_1N_1M_1$) was diagnosed based on serial blood glucose and insulin measurements along with diagnostic imaging. The patient survived for 140 days after diagnosis with medical management, including frequent feeding and prednisolone therapy. On necropsy, necrosis and masses in the peripancreatic omentum and liver were found; pancreatic beta-cell neoplasia with metastasis to the liver was confirmed by histopathologic examination. This case reports hyper-insulinism in a dog presenting with hypoglycemic seizures.

        Dysmyelopoiesis in a cat with immune-mediated hemolytic anemia

        A 3-year-old spayed female Persian feline with non regenerative anemia showed persistent autoagglutination in EDTA anticoagulated blood. Primary immune-mediated hemolytic anemia (IMHA) was suspected and the underlying causes for IMHA were excluded by radiologic, sonographic, serologic and molecular studies. Cytologic examination of the bone marrow revealed that dysmyelopoiesis and dysplastic changes were prominent in the erythroid cells. These changes included asynchronous maturation of the nucleus and cytoplasm, binucleation, trinucleation, fragmented or lobulated nuclei and multilineages. Mild dysgranulopoiesis and dysmegakaryocytopoiesis were also detected including pseudo Pelger-Huet anomalies, giant band neutrophils, asynchronous maturation of the nucleus and cytoplasm in granulopoiesis and large hypolobulated forms as well as dwarf megakaryocytes in megakaryocytopoiesis. Myelodysplastic syndrome and congenital dysmyelopoiesis was ruled out by the low number of blast cells. Finally, secondary dysmyelopoiesis associated with IMHA was diagnosed and immunosuppressive treatment was successfully responsive.

        Ethyl Pyruvate Downregulates Tumor Necrosis Factor Alpha and Interleukin (IL)-6 and Upregulates IL-10 in Lipopolysaccharide-Stimulated Canine Peripheral Blood Mononuclear Cells

        <P>We investigated the anti-inflammatory effects of ethyl pyruvate (EP) on LPS-stimulated canine PBMCs <I>in vitro</I>. We found that EP treatment inhibited the mRNA expressions of proinflammatory cytokines (TNF-α and IL-6), but induced mRNA expression of anti-inflammatory cytokines (IL-10). ELISA measurements revealed that EP also effectively downregulates the LPS-induced increase in proinflammatory cytokine release, while upregulating anti-inflammatory cytokine release. These data indicate that EP could be an effective anti-inflammatory agent in dogs.</P>

        In vitro Conidial Germination and Mycelial Growth of Fusarium oxysporum f. sp. fragariae Coordinated by Hydrogen Peroxide-and Nitric Oxide-signalling

        Chemicals related to hydrogen peroxide (H<sub>2</sub>O<sub>2</sub>) and nitric oxide (NO) generations were exogenously applied to Fusarium oxysporum f. sp. fragariae (Fof ) causing Fusarium wilt disease in strawberry plants, and regulations of in vitro conidial germination and mycelial growth of the fungus by the chemical treatments were evaluated. H<sub>2</sub>O<sub>2</sub> drastically reduced the conidial germination of Fof in a dose-dependent manner, and treatment with 3-amino-1,2,4-triazole (3-AT) catalase inhibitor also led to dose-dependent inhibition of conidial germination but relatively moderately. Gradual decreases in mycelial growth of Fof were found by high concentrations of H<sub>2</sub>O<sub>2</sub>, whilst exogenous 3-AT slightly increased the mycelial growth. Increasing sodium nitroprusside (SNP) NO donor, N<sup>G</sup>-nitro-l-arginine methyl ester (L-NAME) NO synthase (NOS)-inhibitor and tungstate nitrate reductase (NR) inhibitor led to dose-dependent reductions in conidial germination of Fof in quite different levels. SNP conversely increased the mycelial growth but increasing L-NAME moderately decreased the mycelial growth. Tungstate strongly enhanced mycelial growth. Differentially regulated in vitro mycelial growths of Fof were demonstrated by SNP, L-NAME and tungstate with or without H<sub>2</sub>O<sub>2</sub> supplement. Superoxide anion production was also regulated during the mycelial growth of Fof by nitric oxide. These results show that H<sub>2</sub>O<sub>2</sub> and NO-associated enzymes can be suggested as fungal growth regulators of Fof as well as eco-friendly disease-managing agents in strawberry production fields.

      • High-mobility group box 1 as a surrogate prognostic marker in dogs with systemic inflammatory response syndrome : HMGB1 in dogs with SIRS

        <P>OBJECTIVE: To evaluate various surrogate markers associated with the inflammatory and counter-inflammatory responses with respect to mortality in dogs with systemic inflammatory response syndrome (SIRS). DESIGN: Prospective observational study. SETTING: Veterinary Teaching Hospital. ANIMALS: Twenty-eight dogs with naturally occurring diseases and SIRS from January 2007 to May 2009. INTERVENTIONS: Upon admission to the veterinary hospital, history and baseline data from the physical examination, including parameters previously defined for meeting SIRS criteria, were documented. Heparinized blood samples were collected and plasma cytokines interleukin-6 (IL-6), IL-10, and high-mobility group box 1 (HMGB1) were measured by sandwich ELISA. MEASUREMENTS AND MAIN RESULTS: In nonsurvivors, median plasma HMGB1 concentrations (0.718 microg/L, interquartile range [IQR]; 0.300-1.626 microg/L) and the ratio of HMGB1 to IL-10 (2.236, IQR; 0.972-5.367) were significantly increased as compared with those found in survivors (0.300 microg/L, IQR; 0.300-0.312 microg/L for HMGB1; 1.017, IQR; 0.862-1.126 for the ratio of HMGB1 to IL-10, P=0.007 and 0.024, respectively). Plasma IL-6, IL-10, and the ratio of IL-6 to IL-10 were not significantly different between groups. Among the parameters studied, HMGB1 and the ratio of HMGB1 to IL-10 performed the best in discriminating outcome in dogs with SIRS according to receiver operator characteristic curve analysis. CONCLUSIONS: Increases in plasma HMGB1 concentration and the ratio of HMGB1 to IL-10 may predict poorer outcomes in dogs with SIRS. The approach described may lead to reliable prognostic biomarkers and new therapeutic concepts in the study of SIRS in dogs.</P>
