        Low Molecular Weight Polyethylenimine-Mitochondrial Leader Peptide Conjugate for DNA Delivery to Mitochondria

        It has been found that a number of diseases are associated with mutations in the mitochondrial DNA. Therapeutic gene delivery to mitochondria has been suggested as a clinical option for these diseases. In this study, we developed a gene carrier to mitochondria by the conjugation of mitochondrial leader peptide (LP) to polyethylenimine (PEI). Mitochondrial LP conjugated PEI (PEI-LP) was synthesized with low molecular weight PEI (2,000 Da, PEI2K). Gel retardation assay showed that PEI2K-LP formed complexes at a 1.0/1 weight ratio. In addition, PEI2K-LP protected DNA from the enzymatic degradation for at least 60 min, while naked DNA was completely degraded within 20 min. PEI2K-LP was compared with LP conjugated high molecular weight PEI (25,000 Da, PEI25K) in terms of toxicity and delivery efficiency. MTT assay showed that PEI2K-LP had much lower cytotoxicity than PEI25K-LP to 293 cells. In addition, cell-free DNA delivery assay showed that PEI2K-LP delivered more DNA to mitochondria at a 1.8/1 weight ratio than naked DNA or PEI. This result suggests that PEI2K-LP may be useful for the development of mitochondrial gene therapy system with lower cytotoxicity.

        공공정보자원의 상업화 모델 개발 및 적용방안

        본 논문의 목적은 공공정보자원의 활용도 제고 및 지식정보산업 육성을 통한 국가경쟁력 강화의 기반으로서, 공공정보자원의 상업화 모델을 개발하고 이를 적용하기 위한 주요 이슈를 점검하고 정책적 시사점을 제공하는 것이다. 이를 위하여 미국∙EU∙영국∙캐나다의 공공정보자원 활용 제고를 위한 상업화 정책과 상업화 모델로서 자금조달(Funding Model), 비용회수(Cost Recovery Model), 수익창출(Revenue Generation) 모델을 검토하였다. 이를 토대로 공공정보자원의 생명주기와 가치사슬 모델을 활용하여 ⅰ) 공공기관 직접 상업화, ⅱ) 민간계약 상업화, ⅲ) 민∙관 공동생산 상업화, ⅳ) 민간완전위탁 상업화라는 네가지 모델을 개발하고 장단점을 살펴보았다. 그리고 이러한 모델을 우리나라의 공공정보자원 상업화에 적용하기 위해서 선결되어야 할 주요 이슈로서 공공정보 상업화의 비전과 목표, 상업화의 대상 및 범위, 상업화의 주체, 공공정보 저작권, 요금부과 방식과 이윤배분∙활용 및 예산지원, 관련 법제도 정비 등을 검토하였다. This study formulates a strategy for commercial utilization of the public sector information(PSI) to promote efficient provision and effective utilization of information. Models for commercialization of the PSI are suggested in consideration of the life cycle perspectives and the commercial arrangements of the PSI. As to the life cycle perspectives, three stages of commercialization - Funding, Cost Recovery, and Revenue Generation - are proposed based on the analysis of cost structure and the economic viability of the public sector information. This paper also considers various types of commercialization scheme and suggests four types of business arrangement for commercialization of the PSI as a sustainable mechanism. Reviewing the literature on the major countries and organizations relevant to commercialization of the PSI, we examine each of four types of commercialization arrangement in the context of an information resource life cycle along a value chain of the PSI. The study also identifies key issues such as copyright and suggests relevant policies for PSI commercialization such as pricing scheme, related legal and institutional requirements. The suggestions in this study can be the foundation for further detailed researches and practical policies regarding the PSI commercialization.

        감자 Polyphenol Oxidase의 열안정성

        감자로부터 polyphenol oxidase를 추출하여 열안정성에 영향을 주는 요인을 식품의 제조공정에 유용한 정보를 제공하기 위해 연구를 하였다. 감자의 PPO는 pH 7.0에서 가장 안정하였고, 이 효소는 80℃에서 1분간의 열처리로 효소 활성이 70% 저해되었다. 감자 PPO의 열안정성에 대한 z-value는 12.17±0.58℃이고, 이 효소의 열안정성은 sodium chloride의 첨가로 감소되었으며, 2-mercaptoethanol과 dithiothreitol과 같은 환원제의 첨가로 역시 효소활성이 저해되었다. Factors affecting thermostability of polyphenol oxidase (PPO) from potato were studied for the purpose of providing useful information for food processing operations. The enzyme was most stable at pH 7.0 and it was inhibited to 70% after heat treatment at 80℃ for 1 min. The z-value for the thermal inactivation of the PPO was 12.17±0.58℃. The thermostability of the enzyme was reduced by addition of sodium chloride. And the activity was inhibited by addition of reducing reagents such as 2-mercaptoethanol and dithiothreitol.

        치면열구전색제의 조건에 따른 법랑질 결합강도에 관한 연구

        In this study, the shear bond strengths and accompanying elastic modulus ratio, displacement at bond failure and work up to fracture of pit and fissure sealants to toothe enamel were analyzed by the following variables, those were acid etching, moisture content of the surface of enamel, dentin bonding agent(species and method), thermocycling, saliva contamination. One kind of chemically cured resin based pit and fissure sealant, two kinds of light cured resin based pit and fissure sealants and one kind of glass ionomer based pit and fissure sealant, and two kinds of dentin bonding agents were studied. The buccal surface of extracted premolars were ground flat with # 600 silicone carbide papers, and the ground surface was treated depending on the experimental conditions. After each treatment, each pit and fissure sealant was filled into plastic tube of 3mm diameter, which were laid on the surface of ground enamel surface, and were cured. Sealant bonded teeth specimens were immersed in 37℃ distilled water form 24 hours. The shear bond strength of sealant to enamel surface was measured with Universal testing machine with the cross head speed of 1mm/min. From the obtained load-deflection curves, modulus ratio, bond strength, displacement at bond failure and work up to fracture were analyzed. From this experiment, the following results were obtained : 1. Comparing the results of standard condition(i. e. acid etching, dry surface, no dentin bonding agent and 300 thermocyling), the elastic modulus ratio of chemically cured resinbased pit and fissure sealant was significantly higher than those of other materials(P<0.05), and the bond strength, displacement at bond failure and work up to fracture of glass ionomer based pit and fissure sealant were significantly lower than those of other materials(P<0.05). 2. The results of ANOVA with the variables of type of material, acid etching, moisture condition of the enamel surface, dentin bonding agents were as follows ; 1) variables showed significant main effect on the shear bond strength of pit and fissure sealant to enamel(P<0.01), 2) type of material, acid etching and moisture condition of enamel influenced the bond strength significantly(P<0.01), 3) type and application methods of dentin bonding agents influenced on the bond strength significantly(P<0.05). 3. There were statistically significant correlation between the work up to fracture and the bond strength, and between the work up to fracture and the displacement at bond failure(P<0.01), and in these cases the correlation coefficient was 0.87.

        폐성 고혈압환자에서 Amrinone 과 Dobutamine 이 전신 및 폐순환에 미치는 영향

        Background : Dobutamine and amrinone, phosphodiesterase-III inhibitor, are known to have both inotropic and vasodilatory properties. We evaluated the effects of both drugs on systemic and pulmonary hemodynamics in patients with pulmonary hypertension(PH). Methods : With Institutional Review Board approval, 45 patients whose mean pulmonary arterial pressure was greater than 30 mmHg were studied. After sternotomy under the steady state of anesthesia and controlled ventilation(30 mmHg<PaCO2<40 mmHg), patients recieved one of following drugs for 30minutes(min); dobutamine 5.0ug/kg/min(Group I), low dose amrinone(loading dose 1.0 mg/kg, followed by infusion 7.5 g/kg/min, Group II) or high dose amrinone (loading dose 2.0 mg/kg, followed by infusion 10 g/kg/min, Group III). Hemodynamic variables were measured at 10 min and 30 min after start of infusion. Results : Dobutamine didn't decrease pulmonary arterial pressure(PAP) and cause no hemodynamic change while low and high dose amrinone reduced PAP and especcial decrease of PAP in low dose amrinone group was statistically significnat. High dose amrinone increased cardiac index(CI) and decreased both systemic vascular resistance index(SVRI) and central venous pressure(CVP) more significantly than control value. Conclusions : In patients with chronic right ventricular failure associated with PH, amrinone may decrease the PAP and improve cardiac performance more effectively than dobutamin does. Increment of dosage of amrinone may not result in significant reduction of PAP. (Korean J Anesthesiol 1997; 33: 928∼936)

        Cutaneous, subcutaneous and systemic infection can be developed by genus Exophiala which is characterized by annellidic conidiogenesis. To identify genus Exophiala, morphological examination has proved unreliable in differentiation. The advent of gene sequencing technique including the analysis of sequence of the rDNA internal transcribed spacer (ITS) region and ITS-RFLP analysis permitted much more differentiation of genus Exophiala. Although a few cases of Exophiala (E.) spinifera infection identified by molecular techniques as well as morphological features have also been reported in the world, there was no one in Korea. An 89-year-old female presented a tender, dark gray, scaly and verrucous plaque measuring 3 cm in diameter on the right wrist for several months. She has been infected with HIV. The lesion occurred after traumatic injury. There were pseudohyphae on KOH preparation and dark black, velvety colony on fungal culture using potato dextrose agar. On lactophenol cotton blue stain, there were long annellophores and annelloconidia. Skin biopsy showed dark brown yeast-like fungal elements and granulomatous infiltration in the dermis. Also, the sequencing analysis of the ITS region of rDNA identified E. spinifera depending on GenBank BLAST. The lesions have been resolving by oral fluconazole 50 mg qd for 3 months. We report a case with phaeohyphomycosis caused by E. spinifera newly identified in Korea.

        Ursolic acid supplementation decreases markers of skeletal muscle damage during resistance training in resistance-trained men: a pilot study

        Ursolic acid (UA) supplementation was previously shown to improve skeletal muscle function in resistance-trained men. This study aimed to determine, using the same experimental paradigm, whether UA also has beneficial effects on exercise-induced skeletal muscle damage markers including the levels of cortisol, B-type natriuretic peptide (BNP), myoglobin, creatine kinase (CK), creatine kinase-myocardial band (CK-MB), and lactate dehydrogenase (LDH) in resistance-trained men. Sixteen healthy participants were randomly assigned to resistance training (RT) or RT+UA groups (n=8 per group). Participants were trained according to the RT program (60~80% of 1 repetition, 6 times/week), and the UA group was additionally given UA supplementation (450 mg/day) for 8 weeks. Blood samples were obtained before and after intervention, and cortisol, BNP, myoglobin, CK, CK-MB, and LDH levels were analyzed. Subjects who underwent RT alone showed no significant change in body composition and markers of skeletal muscle damage, whereas RT+UA group showed slightly decreased body weight and body fat percentage and slightly increased lean body mass, but without statistical significance. In addition, UA supplementation significantly decreased the BNP, CK, CK-MB, and LDH levels (p<0.05). In conclusion, UA supplementation alleviates increased skeletal muscle damage markers after RT. This finding provides evidence for a potential new therapy for resistance-trained men.

        Augmented Quantum Yield of a 2D Monolayer Photodetector by Surface Plasmon Coupling

        <P>Monolayer (1L) transition metal dichalcogenides (TMDCs) are promising materials for nanoscale optoelectronic devices because of their direct band gap and wide absorption range (ultraviolet to infrared). However, 1L-TMDCs cannot be easily utilized for practical optoelectronic device applications (e.g., photodetectors, solar cells, and light-emitting diodes) because of their extremely low optical quantum yields (QYs). In this investigation, a high-gain 1L-MoS<SUB>2</SUB> photodetector was successfully realized, based on the surface plasmon (SP) of the Ag nanowire (NW) network. Through systematic optical characterization of the hybrid structure consisting of a 1L-MoS<SUB>2</SUB> and the Ag NW network, it was determined that a strong SP and strain relaxation effect influenced a greatly enhanced optical QY. The photoluminescence (PL) emission was drastically increased by a factor of 560, and the main peak was shifted to the neutral exciton of 1L-MoS<SUB>2</SUB>. Consequently, the overall photocurrent of the hybrid 1L-MoS<SUB>2</SUB> photodetector was observed to be 250 times better than that of the pristine 1L-MoS<SUB>2</SUB> photodetector. In addition, the photoresponsivity and photodetectivity of the hybrid photodetector were effectively improved by a factor of ∼1000. This study provides a new approach for realizing highly efficient optoelectronic devices based on TMDCs.</P> [FIG OMISSION]</BR>
