        Evaluation of salt and root-zone temperature stresses in leafy vegetables using hydroponics as a clean production cultivation technique in northeastern Brazil

        Hydroponics has represented an emerging solution for regions with freshwater scarcity. Besides requiring a lower volume of water for production, this cultivation technique allows the use of brackish waters with high salinity levels. Under normal cultivation conditions (i.e., without any environmental control), plants are exposed to other abiotic stresses, such as tem perature, which can further enhance the salinity eff ect. Given the above, three experiments were conducted using a nutrient f i lm technique (NFT) hydroponic system: from April to May (autumn) with endive ( Cichorium endivia L., cv. Gigante Barbarella), from May to July (autumn/winter) with lettuce ( Lactuca sativa L., cv. Rubinela and cv. Veneranda), and from August to October (winter/spring) with chicory ( Cichorium intybus L., cv. Pão de Açúcar and cv. Radiche Todo Ano), in 2021. The trials aimed to evaluate the isolated and/or combined eff ect of salt and root-zone temperature stresses on yield, water use effi ciency, and visual quality of these vegetables. In all experiments, plants were subjected to stresses for 25 days in a randomized block design with a 2 × 4 factorial arrangement: two levels of electrical conductivity of water –ECw (0.30 and 5.50 dS m –1 for endive and chicory; 0.25 and 6.50 dS m −1 for lettuce) combined with four root-zone temperatures – RZTs (ambient-ARZT, ARZT + 2 °C, ARZT + 4 °C, and ARZT + 6 °C, corresponding to the mean temperatures of 24.72, 26.90, 28.77, and 30.73 °C for endive; 24.20, 26.00, 28.00, and 29.99 °C for lettuce; 27.12, 28.93, 31.01, and 32.86 °C for chicory). The isolated eff ects of RZT stresses or ECw levels on endive traits were observed, with leaf fresh matter losses of approxi mately 17 and 44% at higher ARZT + 6 °C (mean of 30.73 °C) and salinity (ECw 5.50 dS m –1 ), respectively. Shoot fresh matter losses of around 32 and 52% (Veneranda and Rubinela lettuces, respectively) and 65% (Radiche Todo Ano chicory) occurred only due to salt stress. The interaction aff ected the shoot fresh matter of Pão de Açúcar chicory, with approximate losses of 80% at higher ARZT + 6 °C (mean of 32.86 °C) and salt stress (ECw 5.50 dS m –1 ) than ARZT and ARZT + 2 °C and without salt stress (ECw 0.30 dS m –1 ). Despite these reductions, visual quality of plants was not depreciated. Therefore, the present study suggested cultivating leafy vegetables such as lettuce, chicory, and endive using brackish waters, even under high temperatures of nutrient solutions.

        Effect of feed restriction on the maintenance energy requirement of broiler breeders

        Objective: This study aimed to evaluate the effect of the ad libitum and restricted feeding regimen on fasting heat production (FHP) and body composition. Methods: Twelve Hubbard broilers breeders were selected with the same body weight and submitted in two feeding regimes: Restricted (T1) with feed intake of 150 g/bird/d and ad libitum (T2). The birds were randomly distributed on the treatments in two runs with three replications per treatment (per run). The birds were adapted to the feed regimens for ten days. After that, they were allocated in the open-circuit chambers and kept for three days for adaptation. On the last day, oxygen consumption (VO<sub>2</sub>) and carbon dioxide production (VCO<sub>2</sub>) were measured by 30 h under fasting. The respiratory quotient (RQ) was calculated as the VCO<sub>2</sub>/VO<sub>2</sub> ratio, and the heat production (HP) was obtained using the Brower equation (1985). The FHP was estimated throughout the plateau of HP 12 hours after the feed deprivation. The body composition was analyzed by dual-energy X-ray absorptiometry scanning at the end of each period. Data were analyzed for one-way analysis of variance using the Minitab software. Results: The daily feed intake was 30 g higher to T2 (p<0.01) than the T1. Also, the birds of the T2 had significatively (p<0.05) more oxygen consumption (+3.1 L/kg<sup>0.75</sup>/d) and CO<sub>2</sub> production (+2.2 L/kg<sup>0.75</sup>/d). That resulted in a higher FHP 359±14 kJ/kg<sup>0.75</sup>/d for T2 than T1 296±17.23 kJ/kg<sup>0.75</sup>/d. In contrast, the RQ was not different between treatments, with an average of 0.77 for the fasting condition. In addition, protein and fat composition were not affected by the treatment, while a tendency (p<0.1) was shown to higher bone mineral content on the T1. Conclusion: The birds under ad libitum feeding had a higher maintenance energy requirement but their body composition was not affected compared to restricted feeding.

        Influence of the Adhesive, the Adherend and the Overlap on the Single Lap Shear Strength

        The single lap joint is the most studied joint in the literature in terms of both theory and practice. It is easy to manufacture and the lap shear strength is a useful value for strength assessment and quality control. Simple design rules exist such as the one present in standard ASTM 1002 or in a recent paper by Adams and Davies. The main factors that have an influence on the lap shear strength are the type of adhesive, i.e. ductile or brittle, the adherend yield strength and the overlap length. The overlap increases the shear strength almost linearly if the adhesive is sufficiently ductile and the adherend does not yield. For substrates that yield, a plateau is reached for a certain value of overlap corresponding to the yielding of the adherend. For intermediate or brittle adhesives, the analysis is more complex and needs further investigation. In order to quantify the influence of the adhesive, the adherend and the overlap on the lap shear strength, the experimental design technique of Taguchi was used. An experimental matrix of 27 tests was designed and each test was repeated three times. The influence of each variable could be assessed as well as the interactions between them using the statistical software Statview. The results show that the most important variable on the lap shear strength is the overlap length followed by the type of adherend.

        Fractal dimension analysis as an easy computational approach to improve breast cancer histopathological diagnosis

        Histopathology is a well-established standard diagnosis employed for the majority of malignancies, including breast cancer. Nevertheless, despite training and standardization, it is considered operator-dependent and errors are still a concern. Fractal dimension analysis is a computational image processing technique that allows assessing the degree of complexity in patterns. We aimed here at providing a robust and easily attainable method for introducing computer-assisted techniques to histopathology laboratories. Slides from two databases were used: A) Breast Cancer Histopathological; and B) Grand Challenge on Breast Cancer Histology. Set A contained 2480 images from 24 patients with benign alterations, and 5429 images from 58 patients with breast cancer. Set B comprised 100 images of each type: normal tissue, benign alterations, in situ carcinoma, and invasive carcinoma. All images were analyzed with the FracLac algorithm in the ImageJ computational environment to yield the box count fractal dimension (Db) results. Images on set A on 40x magnification were statistically different ( p = 0.0003), whereas images on 400x did not present differences in their means. On set B, the mean Db values presented promissing statistical differences when comparing. Normal and/or benign images to in situ and/or invasive carcinoma (all p < 0.0001). Interestingly, there was no difference when comparing normal tissue to benign alterations. These data corroborate with previous work in which fractal analysis allowed differentiating malignancies. Computer-aided diagnosis algorithms may beneficiate from using Db data; specific Db cut-off values may yield ~ 99% specificity in diagnosing breast cancer. Furthermore, the fact that it allows assessing tissue complexity, this tool may be used to understand the progression of the histological alterations in cancer.

        Graviola Fruit Bar Added Acerola By-Product Extract Protects Against Inflammation and Nociception in Adult Zebrafish (Danio rerio)

        Studies involving foods associated with pain reversal and anti-inflammatory effects using zebrafish are rarely reported in the literature. This study aimed to evaluate the effect of graviola (Annona muricata L.) fruit bar (GFB) and GFB added with acerola (Malpighia glabra L) seed extract (ASE) on acute nociception and abdominal inflammation in adult zebrafish (Danio rerio). Acute nociception was induced by formalin, capsaicin, cinnamaldehyde, acidic saline, glutamate (cutaneous models), and hypertonic saline (corneal model), and inflammation was induced by carrageenan. Both GFB and ASE exhibited antinociceptive effect modulated by the nitrergic system, guanylate cyclase, and transient receptor potential ankyrin 1 and acid-sensing ion channels. The antinociceptive effect of GFB also appears to be modulated by the opioid system and glutamatergic receptors (N-methyl-D-aspartate receptor). Only ASE presented corneal antinociceptive effect. Both samples showed anti-inflammatory effect, being more significant the effect of GFB. The addition of acerola by-product extract in GFB results in a product with greater biological potential.

        Influence of polishing systems on roughness and color change of two dental ceramics

        PURPOSE. To evaluate the polishing effect on roughness and color change of pressed and layering ceramics after immersion in coffee solution. MATERIALS AND METHODS. 88 ceramic discs (1.0 mm × 10.0 mm) were manufactured - 44 nano-fluorapatite layering ceramics (IPS e.max Ceram. Group C) and 44 pressed lithium disilicate ceramic discs (IPS e. max Press - Group P). Each group was divided into 4 subgroups according to surface treatments: (G) Glaze, (S) Shofu polishing system (Shofu Inc.), (E) Edenta AG polishing System, (KG) 30-μm diamond granulation tip. Surface roughness (Ra) and color change (ΔE) measurings after the surface treatments were performed, before and 12 days after the immersion in coffee solution. A samples’ qualitative analysis was conducted with a scanning electron microscopy (SEM). Data were statistically-treated with oneway-ANOVA and Duncan’s tests, apart from paired t-test and Pearson’s correlation test (α=5%). RESULTS. The decrescent order, both for surface roughness (Ra) and ΔE for both ceramics were: KG > E > S > G (P<.05). With exception for PG and CG subgroups, which did not present statistical difference between them, all other pressed ceramics subgroups presented smaller Ra values and greater ΔE values than the layering ceramics subgroups (P<.05). CONCLUSION. Although mechanical polishing systems presented intermediate Ra values, their colors were considered clinically acceptable. There is a strong correlation between the surface roughness and the color change of tested ceramics.

        Application of the Fuzzy Logic for the Development of Automnomous Robot with Obstacles Deviation

        This paper proposed to elaborate a navigation system for an autonomous mobile robot, able to deviate from obstacles, from the study and application of Fuzzy Logic. With the algorithm in operation, it was verified that the Fuzzy logic offers a smoother transition in the movements. In order to validate the efficiency of the navigation system created, simulations were performed with the robot according to the rules inserted in the Fuzzy controller, where the input values of the sensors and the output values in the PWM of the board were analyzed. The results obtained were consistent with the responses given by the simulation in MatLab, following the same trend of behavior. With the realization of this project, it was concluded that the Fuzzy methodology presents a solution to the problems of navigation in real environments, allowing to implement a controller for an autonomous robot that can deflect obstacles avoiding their collision. One of the problems encountered is the angle of actuation of the ultrasonic sensors. This type of sensor works with an angle of actuation of 15◦, which leaves the robot with a low vision area in the use of three sensors. As a result, there may be no reading on objects entering zones without detection, leading to a collision with these obstacles. The responses were satisfactory, following the same trend behavior of the simulations of the Fuzzy controller.

        Susceptibility of Mice to Trypanosoma evansi Treated with Human Plasma Containing Different Concentrations of Apolipoprotein L-1

        The aim of this study was to test the susceptibility of mice to Trypanosoma evansi treated with human plasma containing different concentrations of apolipoprotein L-1 (APOL1). For this experiment, a strain of T. evansi and human plasma (plasmas 1, 2, and 3) from 3 adult males clinically healthy were used. In vivo test used 50 mice divided in 5 groups (A to E) with 10 animals in each group. Animals of groups B to E were infected, and then treated with 0.2 ml of human plasma in the following outline: negative control (A), positive control (B), treatment with plasma 1 (C), treatment with plasma 2 (D), and treatment with plasma 3 (E). Mice treated with human plasma showed an increase in longevity of 40.9±0.3 (C), 20±9.0 (D) and 35.6±9.3 (E) days compared to the control group (B) which was 4.3±0.5 days. The number of surviving mice and free of the parasite (blood smear and PCR negative) at the end of the experiment was 90%, 0%, and 60% for groups C, D, and E, respectively. The quantification of APOL1 was performed due to the large difference in the treatments that differed in the source plasma. In plasmas 1, 2, and 3 was detected the concentration of 194, 99, and 115 mg/dl of APOL1, respectively. However, we believe that this difference in the treatment efficiency is related to the level of APOL1 in plasmas.

        Macroscopic, Histological, and Microbiological Characterization of Contact Lesions at the Tibiotarsal Region of Broilers

        Brazil is considered as a great broiler feet exporter, especially for the Chinese trade. Contact lesions at the tibiotarsal region are responsible for economic losses and there is no model for its classification, thereby this study presents a fast and practical grade system to be used in the poultry industry and proposes these lesion characterizations into three different grades. For this, correlation was made between macroscopic, histological findings and microbiological quantification (Escherichia coli, Staphylococcus spp., Streptococcus spp. and sulphite-reducing clostridia) from contact lesions in the tibiotarsal region of 112 broiler carcasses, divided in four groups (n=28), accordingly to the lesion’s intensity. There were no significant differences in microbiological quantification among the groups (p>0.05) except for the grade 3 group, as grade 1 and 2 lesions were in the early stages and histopathological changes such as ulceration were not observed. In grade 3 lesion group, it was observed bacterial cocci grume and ulceration at the articular region and significantly higher microbiological count (p<0.05) for E. coli and Staphylococcus spp. In conclusion, the visual standard proposed in this work, correlated and confirmed by the histopathologic, and microbiologic characterization, allows to precise and fast ascertainment of the contact lesion grade in the tibiotarsal regions of broiler carcasses. Moreover, it should be highlighted that grades 1 and 2 alterations are not caused by an inflammatory process caused by pathogenic agents and should not be considered a public health risk.
