The unreduced (2n) gametes have long been used in triploid breeding of citrus. In lemon, the previously reported mechanisms for 2n megagametophyte formation were controversial, whereas that for 2n pollen production is still unknown. Herein, the frequency of and mechanism underlying 2n megagametophyte and 2n pollen formation in ‘Eureka’ lemon were investigated based on cytological observation and genotyping of the triploid hybrids between ‘Eureka’ lemon and ‘Early gold’ sweet orange. As a result, 4.79% of the viable pollens of ‘Eureka’ lemon were identified as the 2n pollen with a larger diameter (70.16 ± 3.92 μm). The 2n pollen might be resulted from the formation of parallel spindles at meiosis stage II. Among the 204 plantlets regenerated from embryo rescue following the sexual cross, 12 were triploids as identified by flow cytometry. According to the analysis of heterozygosity transmission using 13 pericentromeric single nucleotide polymorphism (SNP) markers and 20 randomly distributed simple sequence repeat (SSR) markers, 11 triploids were identified to be originated from the fertilization of 2n megagametophytes of ‘Eureka’ lemon, with a frequency of 5.39%. Among them, nine 2n megagametophytes were supposed to be arisen from the second division restitution (SDR), whereas the other two were from postmeiotic genome doubling (PMD). These results to understand the mechanism underlying 2n gamete formation in lemon are valuable for its efficient polyploid breeding.

Radiofrequency ablation (RFA) is a minimally invasive management strategy that has been widely applied for benign and recurrent malignant thyroid lesions as an alternative to surgery in Taiwan. Members of academic societies for specialists in interventional radiology, endocrinology, and endocrine surgery collaborated to develop the first consensus regarding thyroid RFA in Taiwan. The modified Delphi method was used to reach a consensus. Based on a comprehensive review of recent and valuable literature and expert opinions, the recommendations included indications, pre-procedural evaluations, procedural techniques, post-procedural monitoring, efficacy, and safety, providing a comprehensive review of the application of RFA. The consensus effectively consolidates advice regarding thyroid RFA in clinical practice for local experts.

Creep Failure and Damage Mechanism of Inconel 718 Alloy at 800–900 °C

The creep behavior and damage mechanisms of 718 alloys were investigated at 800–900 °C in air. The fracturemorphology and microstructure evolution were observed by optical, scanning and transmission electron microscope. Besides,the creep damage tolerance (λ) and creep strain evolution curve were also calculated. The results showed that the creep curvesof 718 alloys at 800 or 850 °C consisted of primary and tertiary stages, while the steady-state region became apparent at900 °C. The apparent creep activation energy of 718 alloy was in the range from 446.3 to 491.8 kJ/mol. The alloy presentedductile fracture at 800 °C due to the nucleation, growth and linkage of creep voids. However, the failure of alloys at 850 or900 °C presented necking to a point due to the microstructure degradation. Further investigations showed the softening ofmaterials and the loss of mechanical performance could be mainly attributed to the coarsening or decrease of strengtheningprecipitates. Above 850 °C, it was found that γ′ phases would dissolve into matrix and stress promoted the re-dissolution ofγ′ phases or led to the break of δ phases. Moreover, the creep strain evolution curves indicated that 718 alloys kept a relativestable state at 800–900 °C when the strain fraction was below 1.
TOSS: Telecom Operations Support Systems for Broadband Services

Due to the convergence of voice, data, and video, today's telecom operators are facing the complexity of service and network management to offer differentiated value-added services that meet customer expectations. Without the operations support of well-developed Business Support System/Operations Support System (BSS/OSS), it is difficult to timely and effectively provide competitive services upon customer request. In this paper, a suite of NGOSS-based Telecom OSS (TOSS) is developed for the support of fulfillment and assurance operations of telecom services and IT services. Four OSS groups, TOSS-P (intelligent service provisioning), TOSS-N (integrated large-scale network management), TOSS-T (trouble handling and resolution), and TOSS-Q (end-to-end service quality management), are organized and integrated following the standard telecom operation processes (i.e., eTOM). We use IPTV and IP-VPN operation scenarios to show how these OSS groups co-work to support daily business operations with the benefits of cost reduction and revenue acceleration.

<P>CAPE1, a conserved peptide elicitor derived from tomato PR-1, was induced by wounding and found to regulate immune responses against biological threats. As PR-1 is highly conserved across many organisms and the putative peptide from AtPR1 was also found to be bioactive in <I>Arabidopsis</I>, the results suggest that this peptide may be useful for enhancing resistance to stress in other plant species.</P><P>Many important cell-to-cell communication events in multicellular organisms are mediated by peptides, but only a few peptides have been identified in plants. In an attempt to address the difficulties in identifying plant signaling peptides, we developed a novel peptidomics approach and used this approach to discover defense signaling peptides in plants. In addition to the canonical peptide systemin, several novel peptides were confidently identified in tomato (<I>Solanum lycopersicum</I>) and quantified to be induced by both wounding and methyl jasmonate (MeJA). A wounding or wounding plus MeJA-induced peptide derived from the pathogenesis-related protein 1 (PR-1) family was found to induce significant antipathogen and minor antiherbivore responses in tomato. This study highlights a role for PR-1 in immune signaling and suggests the potential application of plant endogenous peptides in efforts to defeat biological threats in crop production. As PR-1 is highly conserved across many organisms and the putative peptide from At-PR1 was also found to be bioactive in <I>Arabidopsis thaliana</I>, our results suggest that this peptide may be useful for enhancing resistance to stress in other plant species.</P>

Purpose: Interleukin (IL)-17A plays a critical role in the pathogenesis of allergic airway inflammation. Yet, the exact roles of IL-17A in asthma are still controversial. Thus, the aim of this study was to dissect the roles of IL-17A in toluene diisocyanate (TDI)-induced mixed granulocytic asthma and to assess the effects of neutralizing antibody in different effector phases on TDI-induced asthma. Methods: IL-17A functions in allergic airway inflammation were evaluated using mice deficient in IL-17A (Il17a−/−) or IL-17A monoclonal antibody (IL-17A mab, intraperitoneally, 50 μg per mouse, 100 μg per mouse). Moreover, the effects of exogenous recombinant IL (rIL)-17A in vivo (murine rIL-17A, intranasally, 1 μg per mouse) and in vitro (human rIL-17A, 100 ng/mL) were investigated. Results: TDI-induced mixed granulocytic airway inflammation was IL-17A-dependent because airway hyperreactivity, neutrophil and eosinophil infiltration, airway smooth muscle thickness, epithelium injury, dysfunctional T helper (Th) 2 and Th17 responses, granulocytic chemokine production and mucus overproduction were more markedly reduced in the Il17a−/− mice or by IL-17A neutralization during the sensitization phase of wild-type (WT) mice. By contrast, IL-17A neutralization during the antigen-challenge phase aggravated TDI-induced eosinophils recruitment, with markedly elevated Th2 response. In line with this, instillation of rIL-17 during antigen sensitization exacerbated airway inflammation by promoting neutrophils aggregation, while rIL-17A during the antigen-challenge phase protected the mice from TDI-induced airway eosinophilia. Moreover, rIL-17A exerted distinct effects on eosinophil- or neutrophil-related signatures in vitro. Conclusions: Our data demonstrated that IL-17A was required for the initiation of TDI-induced asthma, but functioned as a negative regulator of established allergic inflammation, suggesting that early abrogation of IL-17A signaling, but not late IL-17A neutralization, may prevent the progression of TDI-induced asthma and could be used as a therapeutic strategy for severe asthmatics in clinical settings.
Detecting LDoS Attacks based on Abnormal Network Traffic

By sending periodically short bursts of traffic to reduce legit transmission control protocol (TCP) traffic, the low-rate denial of service (LDoS) attacks are hard to be detected and may endanger covertly a network for a long period. Traditionally, LDoS detecting methods mainly concentrate on the attack stream with feature matching, and only a limited number of attack patterns can be detected off-line with high cost. Recent researches divert focus from the attack stream to the traffic anomalies induced by LDoS attacks, which can detect more kinds of attacks with higher efficiency. However, the limited number of abnormal characteristics and the inadequacy of judgment rules may cause wrong decision in some particular situations. In this paper, we address the problem of detecting LDoS attacks and present a scheme based on the fluctuant features of legit TCP and acknowledgment (ACK) traffic. In the scheme, we define judgment criteria which used to identify LDoS attacks in real time at an optimal detection cost. We evaluate the performance of our strategy in real-world network topologies. Simulations results clearly demonstrate the superiority of the method proposed in detecting LDoS attacks.
