Crosstalk between G-protein signaling and glutamatergic transmission within the brain reward circuits is critical for long-term emotional effects (depression and anxiety), cravings, and negative withdrawal symptoms associated with opioid addiction. A previous study showed that Regulator of G-protein signaling 4 (RGS4) may be implicated in opiate action in the nucleus accumbens (NAc). However, the mechanism of the NAc-specific RGS4 actions that induce the behavioral responses to opiates remains largely unknown. The present study used a short hairpin RNA (shRNA)-mediated knock-down of RGS4 in the NAc of the mouse brain to investigate the relationship between the activation of ionotropic glutamate receptors and RGS4 in the NAc during morphine reward. Additionally, the shRNA-mediated RGS4 knock-down was implemented in NAc/striatal primary-cultured neurons to investigate the role that striatal neurons have in the morphine-induced activation of ionotropic glutamate receptors. The results of this study show that the NAc-specific knock-down of RGS4 significantly increased the behaviors associated with morphine and did so by phosphorylation of the GluR1 (Ser831) and NR2A (Tyr1325) glutamate receptors in the NAc. Furthermore, the knock-down of RGS4 enhanced the phosphorylation of the GluR1 and NR2A glutamate receptors in the primary NAc/striatal neurons during spontaneous morphine withdrawal. These findings show a novel molecular mechanism of RGS4 in glutamatergic transmission that underlies the negative symptoms associated with morphine administration.
Opposite Localization of Luteinizing Hormone Receptors and Galectin-3 in Mature Mouse Ovaries

본 연구는 성숙마우스의 난소에서 황체형성호르몬수용체(luteinizing hormone receptor, LHR)와 beta-galctoside-binding animal lectin인 galetin-3 (Gal-3)의 발현분포를 면역조직화학방법으로 평가하였다. LHR은 활동(성숙) 황체에서 강하게 발현되는 반면에, Gal-3의 발현은 퇴행중인 황체와 폐쇄난포에서 높게 나타났다. 또한, 임신중인 마우스의 황체에서는 LHR의 면역반응이 강하게 나타났으나, Gal-3의 면역반응은 현저히 낮았다. 위의 결과를 종합해 볼 때, 성숙마우스의 난소에서 LHR과 Gal-3는 난소의 특정한 시기에 따라 다른 분포양상을 나타냄을 알 수 있었으며, 이들 각각의 단백질의 발현으로 보아 황체의 형성과 퇴행에서 상반된 역할을 할 것으로 여겨진다. Received January 5, 2012 /Revised April 12, 2012 /Accepted May 8, 2012 The present study evaluated the localization of luteinizing hormone receptors (LHR) and galectin-3 (Gal-3), a beta-galactoside-binding animal lectin, in the mature mouse ovaries by immunohistochemical analysis. Intense LHR immunoreactivity was detected in the active corpus luteum (CL), whereas expression of Gal-3 was high in the regressing CL and atretic follicle. In the CL of pregnant mice, LHR immunoreactivity was intense, but Gal-3 expression was low. Thus, LHR and Gal-3 had opposite patterns of expression in mature mouse ovaries, suggesting that both proteins have stage-specific expression patterns and are possibly involved in CL formation and regression.
Fast neutron irradiation deteriorates hippocampus-related memory ability in adult mice

<P>Object recognition memory and contextual fear conditioning task performance in adult C57BL/6 mice exposed to cranial fast neutron irradiation (0.8 Gy) were examined to evaluate hippocampus-related behavioral dysfunction following acute exposure to relatively low doses of fast neutrons. In addition, hippocampal neurogenesis changes in adult murine brain after cranial irradiation were analyzed using the neurogenesis immunohistochemical markers Ki-67 and doublecortin (DCX). In the object recognition memory test and contextual fear conditioning, mice trained 1 and 7 days after irradiation displayed significant memory deficits compared to the sham-irradiated controls. The number of Ki-67- and DCX-positive cells decreased significantly 24 h post-irradiation. These results indicate that acute exposure of the adult mouse brain to a relatively low dose of fast neutrons interrupts hippocampal functions, including learning and memory, possibly by inhibiting neurogenesis.</P>
약용작물 잔대와 천궁 중 Pyraclostrobin의 잔류특성

본 연구는 pyraclostrobin의 잔대 및 천궁에 대한 잔류성을 구명하여 잔류농약의 잔류특성 및 안정성평가를 위한 기초자료로 활용코자 하였다. Pyraclostrobin은 strobilurin계로 식물병원균의 포자발아억제, 균사생육저지의 예방효과가 우수하며, 예방 및 치료용의 침투이행성 살균제로 사용되고 있다. 잔대의 시험 약제는 디메토모르프.피라클로스트로빈 25.5(16+9.5)% 액상수화제를 2,000배 희석하여 3회 살포 후 7일, 14일, 21일, 30일 경과후 수확하였으며, 천궁의 시험약제는 피라클로스트로빈 20% 입상수화제를 3,000배 희석하여 3회 살포 후 7일, 14일, 21일, 30일 경과후 수확하였다. Pyraclostrobin은 acetonitrile로 추출하고 응용된 QuEChERS 분석법을 이용하여 LC/MS/MSD로 기기분석하였으며, 분석법상 정량한계(method limit of quantitation; MLOQ)는 모두 0.01 mg/kg이었다. 분석의 정확성 및 재현성 검증을 위하여 약제를 처리하지 않은 공시료에 정량한계, 정량한계 10배 수준이 되도록 표준용액을 첨가하여 3반복 회수율 실험을 실시하였다. 회수율 시험 결과는 농촌진흥청에서 고시한 회수율의 유효 범위 및 변이계수(coefficient of variation, C.V.)를 만족하였다. Pyraclostrobin의 잔류량은 건잔대에서 정량한계(0.01 mg/kg) 미만∼0.07 mg/kg, 생잔대에서 정량한계(0.01 mg/kg) 미만∼0.01 mg/kg이었고, 건천궁의 잔류량은 0.21 mg/kg∼2.18 mg/kg, 생천궁 잔류량은 0.08 mg/kg∼0.14 mg/kg이었다. 생시료를 건시료로 건조하였을 때 잔대의 건조수율은 평균 22.8%, 천궁은 40.6%로 였다. 시험 작물 중 건조 농산물 잔류량이 생 농산물에 비해 높게 잔류되었으며, 건조 과정 중 수분제거로 인한 농약잔류량의 증가로 판단되었다.

WNT11 is a member of the non-canonical Wnt family and plays a crucial role in tumor progression. However, the regulatory mechanisms underlying WNT11 expression are unclear. Tumor necrosis factor-alpha (TNFα) is a major inflammatory cytokine produced in the tumor microenvironment and contributes to processes associated with tumor progression, such as tumor invasion and metastasis. By using site-directed mutagenesis and introducing a serial deletion in the 5’-regulatory region of WNT11, we observed that TNFα activates the early growth response 1 (EGR1)-binding sequence (EBS) in the proximal region of WNT11 and that the transcription factor EGR1 is necessary for the TNFα-induced transcription of WNT11. EGR1 bound directly to the EBSs within the proximal 5'-regulatory region of WNT11 and ectopic expression of EGR1 stimulated WNT11 promoter activity, whereas the knockdown of EGR1 expression by RNA interference reduced TNFα-induced WNT11 expression in T47D breast cancer cells. We also observed that mitogen-activated protein kinases (MAPK), extracellular signalregulated kinase (ERK), c-Jun N-terminal kinase (JNK), and p38 kinase mediated TNFα-induced transcription of WNT11 via EGR1. Our results suggest that EGR1 directly targets WNT11 in response to TNFα stimulation in breast cancer cells. [BMB Reports 2020; 53(12): 628-633]
