        논문(論文) : 論漢語詞彙與構式的綜合敎學 -作爲二語的漢語詞彙敎學的新思路

        The thesis fully investigates the errors made by foreign students who study Chinese from the view point of Construction Grammar. It finds out some common errors such as “same words with different construction”, “different words with same construction”, “different words with similar construction” and “different words with same meaning but different construction”. These errors are thought to be the result of inter language and the authors proposed a new synthetical language teaching method that is to combine lexicon and construction grammar. The thesis analyzes the differences between this new teaching method and the traditional method centering on sentence structure. It discusses the principles of this synthetical teaching method based on lexicon and Construction Grammar and the role and position of this new teaching method in the systematic teaching of Chinese lexicon.

        ORiginal Article : PNPLA3 rs738409 Polymorphism Associated with Hepatic Steatosis and Advanced Fibrosis in Patients with Chronic Hepatitis C Virus: A Meta-Analysis

        Background/Aims: The recognition of a correlation between patatin-like phospholipase domain containing-protein 3 (PNPLA3) rs738409 (C>G) and the severity of liver steatosis or fibrosis in chronic hepatitis C (CHC) has not reached a consensus. This meta-analysis sought to investigate with accuracy the association between the PNPLA3 rs738409 (C>G) polymorphism and liver steatosis and advanced fibrosis in CHC patients. Methods: We performed a comprehensive literature search from the PubMed, Embase, Web of Science, and Google Scholar databases up to December 31, 2014. Crude odds ratios (ORs) with 95% confidence intervals (CIs) were calculated. Statistical analyses were performed using Stata 12.0 software. Results: The meta-analysis revealed the severity of liver fibrosis was significantly higher in CHC patients with PNPLA3 rs738409 GG in Caucasians (versus CC+CG: OR, 2.29; 95% CI, 1.57 to 3.35; p<0.05) but not Asian populations. In Caucasians, liver steatosis was also more severe in CHC patients with rs738409 GG (versus CC+CG; OR, 4.33; 95% CI, 2.59 to 7.22; p<0.05). The sensitivity analysis indicated the results of this meta-analysis were stable and no publication bias was detected. Conclusions: PNPLA3 rs738409 (C>G) was associated with the risk of both advanced liver fibrosis and steatosis in patients with CHC, especially among Caucasian populations.         Antimycobacterial Activity of Fusaric Acid from a Mangrove Endophyte and its Metal Complexes

        Due to the increasing prevalence of multidrug-resistant Mycobacterium tuberculosis, there is an urgent need for new antituberculosis drugs that have novel mechanisms of action. As part of our ongoing search for antimycobacterial metabolites from mangrove endophytes, chemical analysis of the active extract of a strain of Fusarium sp. was performed, which led to the isolation of fusaric acid as the predominant constituent. A variety of metal complexes of fusaric acid were prepared. Antimycobacterial assays showed that Cadmium (II) and Copper (II) complexes exhibited potent inhibitory activity against the M. bovis BCG strain [minimum inhibitory concentration (MIC) = 4 μg/mL] and the M. tuberculosis H37Rv strain (MIC = 10 μg/mL), respectively. This is the first report of the antimycobacterial activity of the mangrove Fusarium metabolite and its coordinating metal complexes.

        New hydroperoxylated and 20,24-epoxylated dammarane triterpenes from the rot roots of Panax notoginseng

        Background: Root rot is a serious destructive disease of Panax notoginseng, a famous cultivated araliaceousherb called Sanqi or Tianqi in Southwest China. Methods: The chemical substances of Sanqi rot roots were explored by chromatographic techniques. MS,1D/2D-NMR, and single crystal X-ray diffraction were applied to determine the structures. Murinemacrophage RAW264.7 and five human cancer cell lines were used separately for evaluating the antiinflammatoryand cytotoxic activities. Results and Conclusion: Thirty dammarane-type triterpenes and saponins were isolated from the rot rootsof P. notoginseng. Among them, seven triterpenes, namely, 20(S)-dammar-25-ene-24(S)-hydroperoxyl-3b,6a,12b,20-tetrol (1), 20(S)-dammar-3-oxo-23-ene-25-hydroperoxyl-6a,12b,20-triol (2), 20(S)-dammar-12-oxo-23-ene-25-hydroperoxyl-3b,6a,20-triol (3), 20(S)-dammar-3-oxo-23-ene-25-hydroperoxyl-12b,20-diol (4), 20(S),24(R)-epoxy-3,4-seco-dammar-25-hydroxy-12-one-3-oic acid (5), 20(S),24(R)-epoxy-3,4-seco-dammar-25-hydroxy-12-one-3-oic acid methyl ester (6), and 6a-hydroxy-22,23,24,25,26,27-hexanordammar-3,12,20-trione (7), are new compounds. In addition, 12 known ones (12e16 and 19e25)were reported in Sanqi for the first time. The new Compound 1 showed comparable antiinflammatoryactivity on inhibition of NO production to the positive control, whereas the known compounds 9, 12, 13,and 16 displayed moderate cytotoxicities against five human cancer cell lines. The results will providescientific basis for understanding the chemical constituents of Sanqi rot roots and new candidates forsearching antiinflammatory and antitumor agents.

        Two Luminescent Cd(II) Coordination Polymers: Enhancement Activity on Pancreatic Cancer Combined with Biliary Stent Placement and 125I Particles

        With the reaction between bib ligands, 5-substituted isophthalic acid and Cd(II) ions, we produced two coordination polymers in success, that is, [Cd(5-meo-ip) (bib)0.5]n (2) and [Cd(5-me-ip)(bib)(H2O)]n·n(H2O) (1) (bib is 1,4-bis(imidazoly)butane, 5-meo-H2ip is 5-methoxyisophthalic acid, and 5-me-H2ip is 5-methylisophthalic acid). The luminescent performances and thermal stability of the two compounds were also explored. Their anti-pancreatic cancer activity combined with biliary stent placement and 125I particles was evaluated and the specific mechanism was explored as well. Firstly, after the model construction and compound treatment, the weight and volume of the tumor tissue was measured. Next, the apoptosis level of the cancer cells were evaluated with Annexin V-FITC/PI apoptosis assay.

      • Pathway and Network Analysis in Glioma with the Partial Least Squares Method

        Gene expression profiling facilitates the understanding of biological characteristics of gliomas. Previous studies mainly used regression/variance analysis without considering various background biological and environmental factors. The aim of this study was to investigate gene expression differences between grade III and IV gliomas through partial least squares (PLS) based analysis. The expression data set was from the Gene Expression Omnibus database. PLS based analysis was performed with the R statistical software. A total of 1,378 differentially expressed genes were identified. Survival analysis identified four pathways, including Prion diseases, colorectal cancer, CAMs, and PI3K-Akt signaling, which may be related with the prognosis of the patients. Network analysis identified two hub genes, ELAVL1 and FN1, which have been reported to be related with glioma previously. Our results provide new understanding of glioma pathogenesis and prognosis with the hope to offer theoretical support for future therapeutic studies.

        Establishment of Tripterygium wilfordii Hook. f. Hairy Root Culture and Optimization of Its Culture Conditions for the Production of Triptolide and Wilforine

        In order to solve the shortage of natural Tripterygium wilfordii Hook. f. plant resource for the production of the important secondary metabolites triptolide and wilforine, hairy roots were induced from its root calli by Agrobacterium rhizogenes. Induced hairy roots not only could be maintained and grown well in hormone-free half-strength Murashige and Skoog medium but also could produce sufficient amounts of both triptolide and wilforine. Although hairy roots produced approximately 15% less triptolide than adventitious roots and 10% less wilforine than naturally grown roots, they could grow fast and could be a suitable system for producing both secondary metabolites compared with other tissues. Addition of 50 μM methyl jasmonate (MeJA) could slightly affect hairy root growth, but dramatically stimulated the production of both triptolide and wilforine, whereas 50 μM salicylic acid had no apparent effect on hairy root growth with slightly stimulatory effects on the production of both secondary metabolites. Addition of precursor nicotinic acid, isoleucine, or aspartic acid at the concentration of 500 μM had varying effects on hairy root growth, but none of them had stimulatory effects on triptolide production, and only the former two had slightly beneficial effects on wilforine production. The majority of triptolide produced was secreted into the medium, whereas most of the produced wilforine was retained inside of hairy roots. Our studies provide a promising way to produce triptolide and wilforine in T. wilfordii hairy root cultures combined with MeJA treatment.

        A20 ameliorates disc degeneration by suppressing mTOR/BNIP3 axis-mediated mitophagy

        Background The pathological mechanism of intervertebral disc degeneration (IDD) is an unanswered question that we are committed to exploring. A20 is an anti-inflammatory protein of nucleus pulposus (NP) cells and plays a protective role in intervertebral disc degeneration. Objective This study aims to investigate the molecular mechanism by which A20 attenuates disc degeneration. Methods The proteins of interest were measured by immunoblotting, immunofluorescence, ELISA assay, and immunohistochemical technique to conduct related experiments. Immunofluorescence assays and mitochondrial membrane potential (JC-1) were used to assess mitophagy and mitochondrial fitness, respectively. Results Here, we demonstrated that A20 promoted mitophagy, attenuated pyroptosis, and inhibited the degradation of the extracellular matrix, consequently significantly ameliorating disc degeneration. Mechanistically, A20 reduces pyroptosis and further suppresses cellular mTOR activity. On the one hand, A20-induced mTOR inhibition triggers BNIP3-mediated mitophagy to ensure mitochondrial fitness under LPS stimulation, as a result of mitigating mitochondrial dysfunction induced by LPS. On the other hand, A20-induced mTOR inhibition reduces the loss of mitochondrial membrane potential and the generation of Mitochondrial ROS. Conclusion The study revealed that A20 promotes BNIP3-mediated mitophagy by suppressing mTOR pathway activation against LPS-induced pyroptosis.
