Currently, preliminary results have confirmed the existence of receptor-interacting protein kinase 3 (RIPK3) and mixed lineage kinase domain-like protein (MLKL)-dependent necroptosis of pancreatic acinar cells during early acute pancreatitis (AP), which might be a potential target for the effective regulation of necroinflammatory injury. However, the exact effect of receptor-interacting protein kinase 1 (RIPK1)-dependent regulated acinar cell necrosis on AP is still uncertain. In our study, we first explored the changes in the degree of local and systemic inflammation in AP rats when the activation of acinar cell RIPK1 was inhibited. The RIPK1 inhibitor Nec-1 was used to treat rats, and the levels of related inflammatory markers, necrosis indicators and apoptotic indicators were measured. Changes in pancreatic nuclear factor κB (NF-κB) and aquaporin 8 (AQP8) expression were noted. Next, the expression of AQP8 in AR42J cells was inhibited, and the degree of cell necrosis and inflammatory damage was found to be significantly reduced. Most importantly, we demonstrated that the RIPK1/NF-ĸB/AQP8 axis might be a potential regulatory pathway mediating RIPK1-dependent regulated acinar cell necrosis in early AP. Finally, we used the NF-κB inhibitor PDTC and Nec-1 to treat rats in different groups and measured the degree of pathological pancreatic injury, the activation of RIPK1, and the expression of NF-κB and AQP8. In summary, we hypothesized that there might be a RIPK1/NF-ĸB/AQP8 pathway controlling RIPK1-dependent regulated necrosis of acinar cells in AP, which might be a promising therapeutic target against AP-related injury.
Effect of processing parameters on the quality of fresh-cut button mushroom(Agaricus bisporus)

This study was conducted to investigate optimum processing procedure of fresh-cut button mushrooms. The experiments were done with different processing parameters including washing method, washing time, sanitation, and cutting time. Fresh mushrooms were washed by swirling water (SW), lift type-immersion washing (LIW), or combined LIW and water spray (LI+WS). Mushroom samples were washed by LIW for 0, 1, 3, and 5 minutes separately. Mushrooms were sanitized with 0, 50, or 100 ㎕L-1 chlorine solution (pH 7) for 60 seconds. Mushrooms were sliced at different times (before washing, after washing, or after drying). The combined washing treatment, LIW+WS was effective in maintaining better appearance and higher Lightness color value among treatments. Optimum washing time to remove foreign materials and maintain color was 3 minutes when mushrooms were washed by LIW. Samples sanitized with 50 ㎕L-1 chlorine reduced initial aerobic bacterial population and had only slight residual chlorine odor. Fresh-cut mushrooms sliced after washing had the lowest loss among samples. The optimized washing, sanitation, and cutting time parameters can be used for sequential processing of fresh-cut button mushrooms.

Pseudomonas syringae pv. actinidiae causes bacterial canker disease in kiwifruit. Owing to the prohibition of agricultural antibiotic use in major kiwifruit-cultivating countries, alternative methods need to be developed to manage this disease. Bacteriophages are viruses that specifically infect target bacteria and have recently been reconsidered as potential biological control agents for bacterial pathogens owing to their specificity in terms of host range. In this study, we isolated bacteriophages against P. syringae pv. actinidiae from soils collected from kiwifruit orchards in Korea and selected seven bacteriophages for further characterization based on restriction enzyme digestion patterns of genomic DNA. Among the studied bacteriophages, two belong to the Myoviridae family and three belong to the Podoviridae family, based on morphology observed by transmission electron microscopy. The host range of the selected bacteriophages was confirmed using 18 strains of P. syringae pv. actinidiae, including the Psa2 and Psa3 groups, and some were also effective against other P. syringae pathovars. Lytic activity of the selected bacteriophages was sustained in vitro until 80 h, and their activity remained stable up to 50oC, at pH 11, and under UV-B light. These results indicate that the isolated bacteriophages are specific to P. syringae species and are resistant to various environmental factors, implying their potential use in control of bacterial canker disease in kiwifruits.

Background: Medical staff may have difficulties in using conventional medicine to manage symptoms among terminally ill patients, including adverse effects of the treatment. Traditional Chinese medicine (TCM) is regarded as a complementary or alternative medicine, and has been increasingly used in the field of palliative medicine in recent years. This study aimed to investigate the experiences of and attitudes toward using TCM among palliative care professionals, and to provide preliminary information about its use in palliative care. Methods: This was a cross-sectional survey study conducted in eight inpatient hospice wards in Taiwan between December 2014 and February 2016. The questionnaire was self-administered, and was analyzed with descriptive statistics including Pearson’s Chi-square test and Fisher’s exact test. Results: A total of 251 palliative care professionals responded to the questionnaire, of whom 89.7% and 88.9% believed that the use of TCM could improve the physical symptoms and quality of life in terminally ill patients, respectively. Overall, 59.8%, of respondents suggested that TCM had rare side effects, and 58.2% were worried that TCM could affect the liver and kidney function of patients. In total, 89.7% and 88.0% of professionals agreed there were no suitable clinical practice guidelines and educational programs, respectively, for TCM use in palliative care. Conclusions: Most of the respondents agreed there was insufficient knowledge, skills-training, and continuing education on the use of TCM in terminally ill patients in Taiwan. These results show that to address patient safety considerations, guidelines about use of TCM in palliative care should be established.
Molecular cloning, chromosomal localization and expression profiling of porcine selenoprotein M gene

Selenoprotein M may regulate a myriad of biological processes through its redox function. In pigs, neither the nucleotide sequence nor the amino acid sequence is known. Furthermore,patterns of tissue expression and regulation by dietary selenium (Se) have not been examined. We determined the full coding sequence (CDS) and the chromosomal location of the porcine gene, SELM, and described its expression profile in vivo under different dietary Se concentrations. The cDNA sequence of porcine SELM from the start codon to the poly(A) tail was cloned by reverse transcription PCR. The CDS contained 429bases with a typical mammalian selenocysteine insertion sequence of form 2 (F2) located in the 3′-untranslated region. The gene was mapped to chromosome 14q21, where porcine SELM and its neighboring genes exhibited a similar organization to human homologues on chromosome 22q12.2. The expression pattern of SELM mRNA in muscle, thyroid, cerebral cortex, pituitary, testis, liver, and kidney was analyzed with real-time quantitative PCR in young male pigs fed a Se-deficient corn-soybean meal basal diet supplemented with 0.0, 0.3,or 3.0 mg Se/kg in the form of Se-rich yeast. Though the SELM mRNA abundance in each of the 7 tissues was not affected by the dietary Se concentrations, it was significantly higher in thyroid (P < 0.01) than in cerebral cortex, pituitary,testis, liver, and kidney at all of the 3 dietary Se concentrations.
