The first ORF of the ARV S1133 S1 segment encodes the nonstructural protein p10, which is responsible for the induction of cell syncytium formation. However, p10-dependent signaling during syncytium formation is fully unknown. Here, we show that dominant negative RhoA, Rho inhibitor C3 exoenzyme, ROCK/Rho-kinase inhibitor Y-27632 and Rac1 inhibitor NSC23766 inhibit p10-mediated cell fusion. p10 over-expression is concomitant with activation and membrane translocation of RhoA and Rac1, but not cdc42. RhoA and Rac1 downstream events, including JNK phosphorylation and transcription factor AP-1 and NF-B activation, as well as MLC expression and phosphorylation are simultaneously activated by p10. p10 point mutant T13M possessed 20% fusion-inducing ability and four p10 fusion-deficient mutants V15M, V19M, C21S and L32A reduced or lost their ability to activate RhoA and Rac1 signaling. We conclude that p10-mediated syncytium formation proceeds by utilizing RhoA and Rac1-dependent signaling.
Characterization of Protein Arginine Methyltransferases in Porcine Brain

Protein arginine methylation is a posttranslational modification involved in various cellular functions including cell signaling, protein subcellular localization and transcriptional regulation. We analyze the protein arginine methyltransferases (PRMTs) that catalyze the formation of methylarginines in porcine brain. We fractionated the brain extracts and determined the PRMT activities as well as the distribution of different PRMT proteins in subcellular fractions of porcine brain. The majority of the type I methyltransferase activities that catalyze the formation of asymmetric dimethylarginines was in the cytosolic S3 fraction. High specific activity of the methyltransferase was detected in the S4 fraction (high-salt stripping of the ultracentrifugation precipitant P3 fraction), indicating that part of the PRMT was peripherally associated with membrane and ribosomal fractions. The amount and distribution of PRMT1 are consistent with the catalytic activity. The elution patterns from gel filtration and anion exchange chromatography also indicate that the type I activity in S3 and S4 are mostly from PRMT1. Our results suggest that part of the type I arginine methyltransferases in brains, mainly PRMT1, are sequestered in an inactive form as they associated with membranes or large subcellular complexes. Our biochemical analyses confirmed the complex distribution of different PRMTs and implicate their regulation and catalytic activities in brain.

Direct conversion receiver is widely utilized in today’s communication system. However, it usually faces a common problem of front-end distortions such as IQ imbalance and frequency offset. When treated separately, effective algorithms exist for estimating and compensating for IQ imbalance as well as frequency offset. With both effects present, such algorithms do not lead to useful estimates of the related parameters. OFDM is sensitive to nonidealities in the receiver front-end. This leads either to stringent front-end specifications and, thus, an expensive device, or large performance degradations. A good summary, Guanbin [1] discusses the transmitter calibration briefly and proposes an estimation technique for calculating the imbalance parameters. For the IEEE 802.11a OFDM standard, Tubbax observe that additional effects from OFDM channel estimation and correction can affect the IQ correction circuits and proposes a smoothing procedure to compensate. The above procedure is not effective when the CFO is too small. This paper extends the previous results mentioned above by developing a simple and adaptive least-square (LS) algorithm to estimate and compensate for IQ imbalance assuming the IQ imbalance compensation is not affected by the CFO, then the CFO is estimated using any of several conventional algorithms. With the additional requirement of a known periodic training sequence (see Su and Chiu ) this process extends the results of to practical cases when CFO is small. Our compensation scheme eliminates the IQ imbalance based on one OFDM symbol and performs well in the presence of CFO. The compensation scheme has fast convergence and small residual degradations. The solution is implemented with an adaptive filter to adjust the parameters of estimation and is computationally relatively inexpensive. With the proposed method as shown in Fig. 1, we can estimate the effect of IQ imbalance without knowing CFO exactly by means of an repetitive training sequence as depicted in Fig.2. Two set of I/Q mismatch parameters, (εr=1.5㏈, ?φr=1.5˚) and (εr=3㏈, ?φr=3˚), are used in the simulations. Fig. 3 shows the performance comparison between the systems with and without I/Q imbalance compensation. It shows that the proposed algorithm can give a satisfactory performance for CFO > 5㎑. To reduce the drawback of high sensitivity under low CFO channel, we modify the adaptive scheme and the fine IQ estimation method to solve the problem of performance degradation when CFO is small. Therefore, our IQ imbalance estimation/compensation scheme potentially leads to low-cost and low-complexity receivers. The simulation results show that the modified scheme can achieve much better performance as shown in Fig. 4.

In the paper, the performance of a 5 kW DC/DC Wye-Delta-based series resonant converter designed for 400V input voltage and 48V output voltage server power supply is evaluated. The paper also performs operation modes analysis, circuit characteristics analysis, and use time domain analysis on Wye-Delta-based series resonant converter for circuit design to replace FHA analysis due to waveform distortion. Finally, a full-load efficiency up to 96.07% and power-density up to 2.84W/cm3 server power supply is implemented.

Background/Aims: With the recent progress in medical treatment, surgery still plays a necessary and important role in treating ulcerative colitis (UC) patients. In this study, we analyzed the surgical results and outcomes of UC in Taiwan in the recent 20 years, via a multi-center study through the collaboration of Taiwan Society of IBD. Methods: A retrospective analysis of surgery data of UC patients from January 1, 1995, through December 31, 2014, in 6 Taiwan major medical centers was conducted. The patients’ demographic data, indications for surgery, and outcome details were recorded and analyzed. Results: The data of 87 UC patients who received surgical treatment were recorded. The median post-operative follow-up duration was 51.1 months and ranged from 0.4 to 300 months. The mean age at UC diagnosis was 45.3±16.0 years and that at operation was 48.5±15.2 years. The 3 leading indications for surgical intervention were uncontrolled bleeding (16.1%), perforation (13.8%), and intractability (12.6%). In total, 27.6% of surgeries were performed in an emergency setting. Total or subtotal colectomy with rectal preservation (41.4%) was the most common operation. There were 6 mortalities, all due to sepsis. Emergency operation and low pre-operative albumin level were significantly associated with poor survival (P =0.013 and 0.034, respectively). Conclusions: In the past 20 years, there was no significant change in the indications for surgery in UC patients. Emergency surgeries and low pre-operative albumin level were associated with poor survival. Therefore, an optimal timing of elective surgery for people with poorly controlled UC is paramount.

Background/Aims: In Taiwan, due to budget limitations, the National Health Insurance only allows for a limited period of biologics use in treating moderate to severe Crohn’s disease (CD). We aimed to access the outcomes of CD patients following a limited period use of biologics, specifically focusing on the relapse rate and remission duration; also the response rate to second use when applicable. Methods: This was a multicenter, retrospective, observational study and we enrolled CD patients who had been treated with adalimumab (ADA) according to the insurance guidelines from 2009 to 2015. Results: A total of 54 CD patients, with follow-up of more than 6 months after the withdrawal of ADA, were enrolled. The average period of treatment with ADA was 16.7±9.7 months. After discontinuing ADA, 59.3% patients suffered a clinical relapse. In the univariate analysis, the reason for withdrawal was a risk factor for relapse (P=0.042). In the multivariate analysis, current smoker became an important risk factor for relapse (OR, 3.9; 95% CI, 1.2-14.8; P=0.044) and male sex was another risk factor (OR, 2.9; 95% CI, 1.1-8.6; P=0.049). For those 48 patients who received a second round of biologics, the clinical response was seen in 60.4%, and 1 anaphylaxis occurred. Conclusions: Fifty-nine percent of patients experienced a relapse after discontinuing the limited period of ADA treatment, and most of them occurred within 1 year following cessation. Male sex and current smoker were risk factors for relapse. Though 60.4% of the relapse patients responded to ADA again. (Intest Res 2017;15:487-494)
Max-throughput interference avoidance mechanism for indoor self-organizing small cell networks

Since mobile traffic has been growing recently, the deployment of indoor small cells has become an attractive solution to enhance coverage. However, the increasing density of cells makes inter-cell interference more considerable. In this paper, we propose a max-throughput Interference Avoidance (MTIA) centralized algorithm to improve the system’s throughput. Based on signaling and reports, a central controller connected to each base station can properly turn off base stations that may induce a relatively strong interference, and thus increase SINR. We implemented the MTIA algorithm in an LTE TDD network simulation and showed that MTIA effectively reduces inter-cell interference and improves the system’s throughput.
