[Purpose] Here, we aimed to determine the effect of Polygonum cuspidatum stem extract (PSE) on exorbital lacrimal gland-excised rat models and hyperosmotic stress-stimulated human conjunctival cells (HCCs). [Methods] Seven week old male Wistar rats were divided into six groups. Only the rats in the control group (NOR, n=5) did not undergo surgery. Three days after the surgery, the exorbital lacrimal gland-excised rats were randomly allocated to five groups: (1) vehicle- treated dry-eyed rats (DED, n=5); (2) PSE (10 mg/ kg) treated DED rats (PSE-10, n=5); (3) PSE (100 mg/ kg) treated DED rats (PSE-100, n=5); and (4) PSE (250 mg/kg) treated DED rats (PSE-250, n=5). In addition, the HCC line was co-treated with hyperosmolar media (528 mOsm) and PSE (1-100 μg/ml). [Results] PSE treatment restored the tear volume and goblet cell density by inhibiting severe corneal irregularities and damage. The treatment with PSE significantly attenuated the hyperosmolar stress-induced inflammation and cell death through the suppression of mRNA expression levels of Tumor necrosis factor-α (TNF-α), Interleukin-6 (IL-6), Interleukin-1β (IL-1β), and Interferon- γ (IFN-γ), and the expression of Bcl-2-associated X protein (Bax) as well as the activation of caspase-3 in vitro. [Conclusion] The inhibitory effects of PSE treatment on dry eye disease indicate the potential of nutritional intervention by PES against inflammatory diseases without adverse effects.

<P><B>Abstract</B></P> <P><B>Purpose</B></P> <P>Bisphenol A (BPA) has been regarded as a possible risk factor for reproductive health. We examined potential reproductive health benefits of <I>Lespedeza cuneata</I> ethanol extract (LCE). Previously, <I>Lespedeza cuneata</I> showed many therapeutic effects. However, the protective effect of LCE on BPA-induced testicular dysfunction and its mechanisms have not been precisely studied.</P> <P><B>Methods</B></P> <P>Mice were randomly divided into six groups (n = 7). Sperm counts and motility were measured by light microscope. Testosterone, total cholesterol, triglycerides, HDL, LDL-cholesterol, glucose, free fatty acids, hs-CRP, Angiotensinogen, Angiotensin II, GOT, GPT, TBARS, GSH, CAT, and SOD1 were measured in mouse serum. The potential protective effects of the LCE on mouse sertoli cells were evaluated.</P> <P><B>Results</B></P> <P>Oral administration of LCE in BPA-exposed male mice restored testis weight, sperm count, motility, and testosterone levels by inhibiting markers in serum. In addition, treatment with LCE in BPA-treated TM4 sertoli cells recovered cell viability by attenuating Bax expression and activating caspase 3 and PARP.</P> <P><B>Conclusions</B></P> <P>These results indicate that LCE prevented BPA-induced testicular dysfunction and cell viability in BPA-treated TM4 sertoli cells. The mechanical and electrical property of small-sized materials has been an important subject due to various applications such as semiconductors, MEMS devices, micro-nano sensors and flexible electronic devices. However, it has many difficulties because of not only fabrication and alignment of specimens, but also the measurement of strain and infinitesimal resistance of thin film. In this work, we used the real-time DIC strain measurement system in micro-tensile testing system. For measurement of electrical resistance, voltage/current sourcemeter is installed in the system. This system has advantages of real time strain monitoring up to 50 ㎚ resolution during micro-tensile test with the ability to measure a small amount of electrical resistance. For the measurement of electrical and mechanical properties, we fabricated micro-tensile specimens which are 200 ㎚ and 1.0 ㎛ thick and 70 and 200 ㎛ wide of gold thin film.

In recent years, the management of water pollution has been focused mainly on the post-treatment of point pollutions such as urban sewage and industrial waste water. Considerable improvement has been made to the water quality and the reduction of pollution. However, this post-treatment oriented technology has no countermeasures for non-point pollution that occupies considerable portion of water pollution The objective of this study was to investigate on field applicability through linked treatment of element technology for water purification From the field assessment, it was observed that the linked treatment improved the quality of the water and the removal efficiency is SS 41.6%, T-N 37.4%, T-P 40.8%, BOD 55.7.
Renal-protective effects of <i>n</i>-hexane layer from morning glory seeds ethanol extract

<P><B>Abstract</B></P> <P>Nephrotoxicity is a main problem in cancer patients using cisplatin. Oxidative stress, inflammation and apoptosis are the important mechanisms of cisplatin induced nephrotoxicity. In the present study, we investigated the effect of the extracts of morning glory on nephrotoxicity by cisplatin in human embryonic kidney cells 293 (HEK-293) and mice. Previous studies have reported that morning glory extracts showed potent activity on anti-inflammatory and anti-oxidant. However, the protective effects of the <I>n</I>-hexane layer of morning glory seed (MGs-Hx) on nephrotoxicity and its mechanisms have not been clearly understood. Oral administration with MGs-Hx showed protective effects <I>in vivo</I> experiments test and the treatment of MGs-Hx in a concentration of 100mg/kg/day had significant effect both of decreasing serum creatinine, BUN, serum uric acid level and reduced iNOS, COX-2 mRNA expressions with low side-effect. Moreover, cell viability was restored by MGs-Hx treatment compared to cisplatin-induced nephrotoxic HEK-293 cells. Co-treatment with MGs-Hx and cisplatin showed the significant effect to reduce inflammatory enzyme, iNOS expression and continuous production of NO. In addition, it exhibited a tendency to decreasing expression of apoptosis-related proteins, caspase-3, 8 and 9, and NF-κB translocation to nucleus as well as phosphorylation of p38, JNK, ERK in cisplatin-induced nephrotoxic HEK-293 cells. Our study provides insight into the underlying mechanisms of MGs-Hx and suggests that MGs-Hx might be a potential therapeutic agent to modulate inflammation and apoptosis in nephrotoxicity.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The present study aimed to evaluate protective effect of MGs-Hx on cisplatin-induced nephrotoxicity. </LI> <LI> MGs-Hx inhibited BUN, creatinine and uric acid levels in cispaltin-induced renal injury mice. </LI> <LI> MGs-Hx attenuated the cisplatin-induced inflammation and apoptosis in HEK-293 cells. </LI> <LI> Renoprotective effects of MGs-Hx were accompanied by suppression of MAPKs. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>
Quinic acid inhibits vascular inflammation in TNF-α-stimulated vascular smooth muscle cells

<P> Atherosclerosis is a chronic inflammatory disease, and the increased expression of adhesion molecules on vascular smooth muscle cells contributes to the progression of vascular disease. Quinic acid (QA) has been shown to possess radioprotection, anti-neuroinflammatory, and anti-oxidant activities; however, an anti-vascular inflammatory effect has not been reported. This study investigated the effect of QA on the expression of vascular cell adhesion molecule-1 (VCAM-1) stimulated by TNF-α in MOVAS cells. Pre-incubation of MOVAS cells, the mouse vascular smooth muscle cell line for 2 h with QA (0.1, 1 and 10 μg/mL) dose-dependently inhibits TNF-α-induced mRNA and protein expression of VCAM-1 and monocyte adhesion. QA inhibits TNF-α-stimulated phosphorylation of MAP kinase and NK-κB activation. Our results indicate that QA inhibits the TNF-α-stimulated induction of VCAM-1 in VSMC by inhibiting the MAP kinase and NF-κB signaling pathways and the adhesion capacity of VSMC, which may explain the ability of QA to inhibit vascular inflammation such as atherosclerosis. </P>

African swine fever (ASF) is one of the most complex infectious swine diseases and the greatest concern to the pig industry owing to its high mortality and no effective vaccines available to prevent the disease. Since the first outbreak of ASF in pig farms, ASF has been identified in 14 pig farms in four cities/counties in South Korea. The outbreak was resolved in a short period because of the immediate control measures and cooperative efforts. This paper reviews the ASF outbreak and the experience of successfully stopping ASF in pig farms in South Korea through active responses to prevent the spread of ASF. In addition, suitable changes to build a sustainable pig production system and collaborative efforts to overcome the dangerous animal disease, such as ASF, are discussed.
