        Development of a Label-Free LC-MS/MS-Based Glucosylceramide Synthase Assay and Its Application to Inhibitors Screening for Ceramide-Related Diseases

        Ceramide metabolism is known to be an essential etiology for various diseases, such as atopic dermatitis and Gaucher disease. Glucosylceramide synthase (GCS) is a key enzyme for the synthesis of glucosylceramide (GlcCer), which is a main ceramide metabolism pathway in mammalian cells. In this article, we developed a liquid chromatography-tandem mass spectrometry (LCMS/ MS) method to determine GCS activity using synthetic non-natural sphingolipid C8-ceramide as a substrate. The reaction products, C8-GlcCer for GCS, could be separated on a C18 column by reverse-phase high-performance liquid chromatography (HPLC). Quantification was conducted using the multiple reaction monitoring (MRM) mode to monitor the precursor-to-product ion transitions of m/z 588.6 → 264.4 for C8-GlcCer at positive ionization mode. The calibration curve was established over the range of 0.625-160 ng/mL, and the correlation coefficient was larger than 0.999. This method was successfully applied to detect GCS in the human hepatocellular carcinoma cell line (HepG2 cells) and mouse peripheral blood mononuclear cells. We also evaluated the inhibition degree of a known GCS inhibitor 1-phenyl-2-decanoylamino-3-morpholino-1-propanol (PDMP) on GCS enzymatic activity and proved that this method could be successfully applied to GCS inhibitor screening of preventive and therapeutic drugs for ceramide metabolism diseases, such as atopic dermatitis and Gaucher disease.

        Financial Development and Income Distribution : A System GMM Panel Analysis with Application to Urban China

        Financial development has strongly influenced the pattern of income distribution in post-reform China. In this paper, using Chinese provincial data over the period of 1986-2000 and applying the Generalized Method of Moment (GMM) techniques, we investigate the finance-inequality nexus in urban China. Empirical results show that China’s financial development significantly helps to reduce urban income inequality. However, these positive distributional gains from financial sector development have been severely offset by the increased urban unemployment and massive layoffs brought about by the implementation of radical urban reforms and the restructuring of state-owned enterprises. Financial development has strongly influenced the pattern of income distribution in post-reform China.       • An active learning method with difficulty learning mechanism for crack detection

        Crack detection is essential for inspection of existing structures and crack segmentation based on deep learning is asignificant solution. However, datasets are usually one of the key issues. When building a new dataset for deep learning, laborious and time-consuming annotation of a large number of crack images is an obstacle. The aim of this study is to develop an approach that can automatically select a small portion of the most informative crack images from a large pool in order to annotate them, not to label all crack images. An active learning method with difficulty learning mechanism for crack segmentation tasks is proposed. Experiments are carried out on a crack image dataset of a steel box girder, which contains 500 images of 320 × 320 size for training, 100 for validation, and 190 for testing. In active learning experiments, the 500 images for training are acted as unlabeled image. The acquisition function in our method is compared with traditional acquisition functions, i.e., Query-By-Committee (QBC), Entropy, and Core-set. Further, comparisons are made on four common segmentation networks: U-Net, DeepLabV3, Feature Pyramid Network (FPN), and PSPNet. The results show that when training occurs with 200 (40%) of the most informative crack images that are selected by our method, the four segmentation networks can achieve 92%-95% of the obtained performance when training takes place with 500 (100%) crack images. The acquisition function in our method shows more accurate measurements of informativeness for unlabeled crack images compared to the four traditional acquisition functions at most active learning stages. Our method can select the most informative images for annotation from many unlabeled crack images automatically and accurately. Additionally, the dataset built after selecting 40% of all crack images can support crack segmentation networks that perform more than 92% when all the images are used.

        HOMOLOGY AND SERRE CLASS IN D(R)

        Let 𝓢 be a Serre class in the category of modules and 𝖆 an ideal of a commutative Noetherian ring R. We study the containment of Tor modules, Koszul homology and local homology in 𝓢 from below. With these results at our disposal, by specializing the Serre class to be Noetherian or zero, a handful of conclusions on Noetherianness and vanishing of the foregoing homology theories are obtained. We also determine when Tor<sup>R</sup><sub>𝓼+t</sub>(R/𝖆, X) ≅ Tor<sup>R</sup><sub>𝓼</sub>(R/𝖆, H<sup>𝖆</sup><sub>t</sub>(X)).

      • A Visual Video Synthesis Tool Based on Video Materials

        Video matting is the process of extracting the foreground of a video and compositing it with another background to generate a new video. Currently, there are many ways to edit foreground image, yet we lack of an effective way to edit foreground video and merge it with a new background, forming a new, meaningful, realistic video. In this paper, we present a new video editing tool which fulfills this function effectively. Using this tool, we can easily edit the appearance of extracted object by resizing, rotation and recoloration. We provide a path specification tool to locate the object in an appropriate position at specified frame. Finally, by an illumination blending procedure, we generate a desired video with the objects composed realistically.

        Interaction Matrix Based Analysis and Asymptotic Cooperative Control of Multi-agent Systems

        In this paper, we investigate a decentralized asymptotic cooperative control problem of multi-agent systems with leader-follower configuration. We firstly develop a new method using a proposed “interaction matrix” for the analysis of cooperation convergence of multi-agent systems, i.e. both consensus of the agents states and trajectory tracking of the whole group can be instantaneously concluded only by observing the minimum eigenvalue of the interaction matrix. For a multi-agent system, the external given desired trajectory can be partially obtained (through sensing or detecting) by the leaders, but higher-order derivatives such as acceleration and jerk of the desired trajectory cannot be obtained. In this case, by using some conventional control methods, the trajectory tracking performance is always not satisfactory when a trajectory varies aggressively w.r.t. time. For the sake of asymptotic tracking of an arbitrary given external trajectory of a multi-agent system, we develop a nonlinear cooperative controller based on the robust integral of signum of cooperative error (RISCE) technique, where the interaction matrix is used. The simulation results show asymptotic convergence of cooperation by using the proposed control, and better performance compared to composited nonlinear feedback based PD (CNF-PD) control.

      • Theories of Religious Relationship and Christian-Buddhist Dialogue : A Christian Perspective

        This paper aims to examine the relationship between Buddhism and Christianity, especially the issue of how to see “the Other” through relationship theories of modern religions, e.g. Exclusivism, Inclusivism, Particularism, Pluralism and Comparativism. The exclusivists believe that conversion to Christianity is the only way for other religious adherents to salvation or liberation, thereby they deny the existence of identity among religious others, rendering the Christian-Buddhist Dialogue (CBD) impossible; the variant of Exclusivism-Particularism seeks to avoid the above impasse by making some room for dialogue through the distinction between language systems, the Otherness was translated into incommensurability arising from different languages; the Inclusivists subsume the Other as “Anonymous Christians” so that CBD turns into certain form of internal talk, the religious Others lose their own identities in this way; the pluralists respect the Other and their identities, then CBD enhances mutual understanding and enrichment for dialogue partners; the comparativists accept the Other and tries to go across the boundaries of religions, to be immered in the Other tradition and learn from the Other in the Other’s terminology and perspective. In this way, the understanding of one’s own tradition and faith are reinforced from the new insights.
