Proceedings of international Symposium on East-West Medicine, Seoul. 44-64, 1999.-Cordyceps sinensis is a major parasitic fungus on larva of Lepidoptera. It is one of the well know fungi used in traditional Chinese medicine for treatment asthma, lung and kideney diseases. Many fungi belonging to the genus Cordyceps have been demonstrated to produce natural products with various biological activities. Reported biological activities of genus cordyceps include:(a)inhibition of DNA and RNA synthesis;(b)enhancement of cell differentiation;(c)restruction of cytoskeleton;(d)inhibition of protein kinase activity;(e)antitumor activity on bladder, colon, lung carcinoma as well as fibroblastoma;(f)inhibition of the infection and revers transciptase activity of human immunodeficiency virus type 1; and(g) inhibition of methylation of nucleic acid. Thus, it is suggested that the extracts of Cordyceps sp. Contain antitumor, antiviral, and many other biological activities. In our previous studies, we proved that C. sinensis contains the antitumor and immunomodulatory agents. In the present studies, effects of C. sinensis on bronchoalveolar lavage fluids(BAL)cells were demonstrated. The crude methanolic extracts of C. sinensis were fractionated into 15 fractions by silica gel column chromatography. Effects of various fraction on BAL cells proliferation and lipopolysaccharide(LPS) stimulated interleukin-1β(IL-8),platelet activating factor(PAF), interleukin-6(IL-6), tumor necrosis factor-α(TNF-α),and interleukin-8(IL-8) production on BAL cells were determined. The result indicated that the CS-19-22 fraction suppressed BAL cells proliferation activated by LPS. The median inhibitory concentration(IC50) was 6㎍/ml. The CS-19-22 fraction also decreased IL-1β,PAF,IL-6, TNF-αand IL-8 production.The results of revers transcription-polymerase chain reaction(RT-PCR) demonstrated that the CS-19-22 fraction did not affect IL-1β,PAF,IL-6, TNF-α,and IL-8 mRNAs expression in BAL cells activated by LPS. By contrast, it inhibited the IL-10 mRNA expression but enhanced IFN-γand IL-12 mRNAs expression in activated BAL cells. Moreover, the CS-19-22 fraction blocked PAF-induced platelet aggregation. It is unlikely that cytotoxicity was involved. Because no cell deths were observable. We hypothesize that inhibitory mechanisms of CS-19-22 on BAL cells proliferation may be related to the impairments of gene expression and production of cytokines in BAL cells. The explanation of C. sinensis antiasthma function may involved the following(1)C.sinenesis contained immunomodulatory agents that stimulated IFN-γand IL-12 expression in TH1cells.(2)Enhanced secretion of IFN-γand IL-12 will inhibit the Tγcells immune responses Ex. Decreasing of IL-6 and IL-10 production.(3) The impairments of cytokines production in TH2 cells suppressed th BAL cells proliferation then more cytokines production Ex.IL-1β,PAF,IL-6,TNF-αand IL-8 in activated BAL cells were decreased. (4)The suppression of TH2cells immune responses will inhibit the growth and differentiation of B cells then IgE production will be decreased. (5)Reduced production of IgE will decreased asthma attack occurring in individuals. However, the results of the present study suggested that C. sinensis may also have acted to treat asthma in part by inhibiting BAL cells proliferation and cytokine gene expression and production. Future, experiments with treatment of animals with asthma with C. sinensis will be necessary to defined whether C. sinensis can reduce asthma injury. Plans are underway for the isolation of pure principal growth inhibitors and their mechanisms of action are subjected for further study.

      • Conductivity stability of carbon nanofiber/unsaturated polyester nanocomposites

        Carbon nanofiber (CNF)/unsaturated polyester resin (UPR) was prepared by a solvent evaporation method, and the temperature dependency of electrical conductivity was investigated. The CNF/UPR composites had quite a low percolation threshold due to CNF having a larger aspect ratio and being well dispersed in the UPR matrix. The positive temperature coefficient (PTC) was found in the CNF/UPR composites and it showed stronger effect around the percolation threshold. The electrical resistance of the CNF/UPR composites decreased and had lower temperature dependency with increasing numbers of thermal cycles.

        Separating machining errors of S-shaped samples based on the comprehensive error field of five-axis machine tools

        In the present study, a new method is proposed to separate machining errors of a 5-axis machine tool with a double turntable. Moreover, a high-precision compensation model is established to calculate machining errors of thin-walled parts in a 5-axis machine tool. In this regard, a comprehensive error model of the S-shaped sample part considering geometric, thermal, and machining errors is established. Then based on the homogeneous transformation matrix, the geometric error models of translational axes and the A and C axes of the rotation axis are established. Accordingly, geometric errors of each motion axes are identified and measured. Meanwhile, the thermal error of the spindle is measured, and the thermal error model of the machine tool along X/Y/Z directions is established using the multiple linear regression method. Finally, the S-shaped sample is processed and the on-machine measurement of the sample surface is carried out. Based on the obtained machining error from the separation method, the distribution of the total surface error at different curvatures of the S-shaped sample is analyzed, and the distributions of geometric and thermal errors of the machine tool are obtained. It is found that when the geometric and thermal errors of the machine tool are compensated, the measured machining error of each point of the S-shaped sample can be reduced by 10-15 μm compared with that before compensation.

        Processing Errors in an On-Machine Measurement Method Based on the Adaptive Triangular Mesh

        A novel, on-machine method applying the adaptive triangular mesh sampling measuring process on automotive parts mold was elaborated in order to obtain the processing errors of free-form surfaces. This process has firstly generated a triangular mesh through the advancing front method. This has allowed us to extract the coordinate data corresponding to the mesh nodes. Subsequently, the processed surfaces were fitted by the mean of NURBS surface reconstruction. The final stage has employed the determination of the normal distance between the reconstructed and theoretical surfaces using the generalized Newtonian method, which has eventually allowed us to obtain the free-form surface machining errors as well as the model surface profile errors corresponding to the experimental process. Therefore, we have effectively determined the free-form surface processing errors by using the processing surface reconstruction methodology based on the adaptive triangular mesh sampling.

        miR-140-3p Knockdown Suppresses Cell Proliferation and Fibrogenesis in Hepatic Stellate Cells via PTEN-Mediated AKT/mTOR Signaling

        Purpose: Liver fibrosis is a major cause of morbidity and mortality and the outcome of various chronic liver diseases. Activationof hepatic stellate cells (HSCs) is the key event in liver fibrosis. Studies have confirmed that miR-140-3p plays a potential regulatoryeffect on HSC activation. However, whether miR-140-3p mediates the liver fibrosis remains unknown. Materials and Methods: Expression of miR-140-3p was detected by real-time quantitative PCR (qPCR). Cell proliferation wasmeasured by MTT, while cell apoptosis rate was determined via flow cytometry. Western blot assay was used to detect the expressionof cleaved PARP. The fibrogenic effect was evaluated by expression of α-smooth muscle actin and desmin. Functional experimentswere performed in transforming growth factor β1 (TGF-β1)-induced HSC-T6 cells with transfection of anti-miR-140-3pand/or siPTEN. Target binding between miR-140-3p and PTEN was predicted by the TargetScan database and identified usingluciferase reporter assay and RNA immunoprecipitation. Results: TGF-β1 induced the activation of HSC-T6 cells, and miR-140-3p expression varied according to HSC-T6 cell activationstatus. Knockdown of miR-140-3p reduced cell proliferation and the expressions of α-SMA and desmin, as well as increasedapoptosis, in TGF-β1-induced HSC-T6 cells, which could be blocked by PTEN silencing. Additionally, inactivation of the AKT/mTOR signaling pathway stimulated by miR-140-3p knockdown was abolished when silencing PTEN expression. PTEN was negativelyregulated by miR-140-3p via direct binding in HSC-T6 cells. Conclusion: miR-140-3p is an important mediator in HSC-T6 cell activation, and miR-140-3p knockdown suppresses cell proliferationand fibrogenesis in TGF-β1-induced HSC-T6 cells, indicating that miR-140-3p may be a potential novel molecular targetfor liver fibrosis.

        A facile two-step preparation of compact and crystalline Sb2S3 thin film for efficient solar cells

        The two-step preparation of compact and crystalline Sb2S3 thin films was firstly reported using the pyrolysis of the Sb-butyldithiocarbamate complex solution in DMF. The porous and amorphous Sb2S3 thin films were successfully prepared at 170 ◦C for 30 min, and then can be converted to compact and crystalline Sb2S3 thin films at 200 ◦C for 30 min or 300 ◦C for 2 min. The corresponding solar cells with the architecture of FTO/TiO2 compact layer/Sb2S3/spiro-OMeTAD/Au achieved the photoelectric conversion efficiency of 4.16% at 200 ◦C and 5.05% at 300 ◦C. The two-step preparation of the compact and crystalline Sb2S3 thin films can provide the feasible approach for the fabrication of various microstructure thin film solar cells and the low preparation temperature of 200 ◦C was also attractive to assemble the flexible Sb2S3 thin film solar cells.

        Marker-Assisted Mating Applied in In-Situ Conservation of Indigenous Animals in Small Populations : (1) Choosing Mating Schemes for Maximum Heterozygosity

        Maintaining maximum genetic variability is of critical importance with in-situ conservation of animal species in small populations. Marker-assisted mating (MAM) was suggested to achieve maximum heterozygosity in offspring populations. The aims of this research was to investigate and decide the effectiveness and promising types of MAM to achieve this goal. Analysis of variance with simulation data revealed that the heterozygosity in offspring populations was significantly determined by sire heterozygosity from mating of non-inbred parent animals, and significantly by sire heterozygosity and percent parental difference in offspring reproduced by inbred parents. Seven types of marker-assisted mating schemes were examined, in which offspring exhibited heterozygosity that was -0.01 to 7.37% below or above that from random mating of non-inbred parent animals, and 0.00 to 16.39% above that from random mating of inbred parent animals. The great increase in offspring heterozygosity was observed with mating by tandem maximizing sire heterozygosity, percent parental difference, and dam heterozygosity. Random mating resulted in fluctuation of offspring heterozygosity. These results suggested that MAM was a promising method for maintaining maximum offspring variability in in-situ conservation of animal species in small populations.

        Resistant starch (RS), a novel endogenous inert marker for detecting glucose absorption of small intestine with sweeteners administration in mice

        Resistant starch could be degraded by the fermentation of colonic microorganisms in the large intestine of mammals, but not in the small intestine. In this study, we established a novel strategy by using resistant starch as an endogenous marker to determine the glucose absorption of the small intestine of laboratory animals. By optimization of the classical enzymatic method of starch measurement, the demand for the sample weight was reduced by 90%. Moreover, the amount of resistant starch in normal feed was detectable without any extra addition. The value of small intestine glucose absorption of mice was similar when using resistant starch and titanium dioxide as inert markers. The fermentation of resistant starch by intestinal microorganisms in the small intestine was demonstrated not disturbing the detection of glucose absorption significantly. Artificial sweeteners exposed ICR mice showed different glucose absorption which indicated, first, resistant starch can be used as a novel endogenous marker in the small intestine of small animals; second, although glucose tolerance did not change in mice after short-term exposure to artificial sweeteners, there were significant changes in glucose absorption associated with it; third, the short-term exposure resulted in no significant change in glucose tolerance.

        Distribution Method of Automotive Torque for Hub Motor Considering Energy Consumption Optimization

        To optimize the torque distribution of each drive wheel of a distributed In-wheel motor car, this paper proposes an in-wheel motor torque distribution method to consider the energy consumption and slip loss of the motor. First, a torque distribution model is established based on the improved quantum genetic algorithm of the disaster operation. Under NEDC operating conditions, the energy consumption of the in-wheel motor based on the torque distribution method of the improved quantum genetic algorithm is reduced by 11.62 % compared with that of the torque equivalent distribution method and 3.94 % compared with that optimized by the genetic algorithm. Finally, based on the in-wheel motor test bench, the motor torque and the battery SOC curve of the wheel motor under the NEDC condition and UDDS operating conditions are obtained. Experimental results show that the torque distribution method based on the improved quantum genetic algorithm can effectively reduce energy consumption, and it performs better than the ordinary genetic algorithm. Also, the energy consumption optimization effect is the most significant under NEDC conditions, with an energy consumption 11.4 % lower than that of the torque equivalent distribution method and 3.8 % lower than that of the genetic algorithm optimization distribution method.

        호텔브랜드 체험이 브랜드신뢰, 브랜드애착과 브랜드몰입에 미치는 영향

        본 연구는 감성 마케팅의 중요성이 부각되는 오늘날, 호텔브랜드 체험을 기반으로 브랜드에 대한 체험적 요소가 브랜드자산인 브랜드 신뢰, 브랜드 애착 및 브랜드 몰입에 미치는 영향을 살펴보고 그에 따른 실무적 제안을 하고자 하였다. 이에 따라 설문조사를 통해 얻는 자료로 분석한 결과 몇 가지 의미 있는 결과를 확인할 수 있었다. 첫째, 경로분석을 통해 호텔브랜드 체험의 하위구성요소들이 브랜드자산인 브랜드 신뢰와 브랜드 몰입에 미치는 영향을 확인한 결과 감각체험, 감성체험, 행동체험, 지적체험은 모두 긍정적 영향을 미치고 있음을 확인하였다. 둘째, 호텔브랜드 체험에 따른 브랜드자산인 브랜드 신뢰, 브랜드 애착 및 브랜드 몰입간의 영향관계를 검증한 결과 브랜드신뢰와 브랜드 애착이 브랜드 몰입에 긍정적인 영향을 미치고, 브랜드 신뢰가 브랜드 애착에 긍정적인 영향을 미치는 것이다. 이러한 연구를 통해 호텔 경영자에게 전략적인 시사점을 제공하고자 한다. The purposes of this study were to investigate the effect of experience in hotel on brand trust, brand attachment and brand commitment. It aims to contribute hotel brand marketing from analyzing and showing how brand experience factors affect brand equity factors. The findings and implications can be summarized as follows: the factor Sense Experience, Feel Experience, Act Experience and Think Experience of Brand Experience caused the positive impact on brand trust and brand attachment. Second, brand trust has a significant impact on brand attachment. Third, brand trust and brand attachment both caused a positive impact on brand commitment. Thus, the hotel managers need to establish a ongoing brand experiential marketing strategy which can improve brand commitment of hotel and ways to be recognized as motivational factors for offered services and products which customer can perceive and experience.
