      • Interleukin-2가 Mouse 자궁내막의 Acid Phosphatase 활성에 미치는 영향

        Interleukin-2, a kind of lymphokine produced by the T-lymphocyte stimulated by antigen or mitogen, has been studied intensively with the development of immuno-oncology and chemoimmunotherapy of the malignancy. Interleukin-2 is known to exert on antitumor effect by stimulating the proliferation of helper and cytotoxic T lymphocytes, augmenting the cytolytic activities of the cytotoxic T lymphocyte and differentiating a precursor cell into a lymphokine-activated killer cell in vivo. Though interleukin-2 suppresses the metastasis of the malignant tumor into the adjacent organs, it develops many side effects including fever, chills, malaise, mental confusion, diarrhea, vomiting and oliguria. Interleukin-2 also causes pulmonary, hepatic and renal toxicities and, especially, increase of capillary permeability with weight gain, pulmonary edema and ascites. In this experiment, the author studied the effect of interleukin-2 on the mouse endometrium, observing histochemically the changes in the activity of acid phosphatase following the estrous cycle. Female, DDY strain, weighing 20gm and exhibiting normal estrous cycle, were used as experimental animals. The estrous cycle was recognized by the vaginal smear. The experimental animals were treated with 2 million units of interleukin-2/0.2ml per Kg of body weight and with 0.2ml of normal saline per Kg of body weight in the control groups for 5 days. And animals were sacrificed 5th day after administration of interleukin-2. The specimens obtained from the corpus uteri were fixed in 10% neutral buffered formalin solution for 24 hours at 4℃ and sliced at 14μm thickness in a frozen cryostat. The activity of acid phosphatase was observed by the Gomori's method for histochemical study. The result obtained was as follows: The acid phosphatase activity was increased at estrus and decreased at diestrus in interleukin-2 treated group. There was, however, no marked differences in acid phosphatase activity of the proestrus and metestrus between control and interleukin-2 treated groups. Consequently, it was suggested that interleukin-2 develop the changes of the acid phosphatase of the endometrium of the mouse.

        It is well known that the endometrium of uterus changes histologically during the estrous cycle in rodents. Interleukin-2, a kind of lymphokine produced by the T lymphocyte activated by antigen or mitogen, induces antitumor effect by stimulating the proliferation of helper and cytotoxic T lymphocytes, augmenting the cytolytic activities of the cytotoxic T lymphocyte and differentiating a precursor cell into a lymphokine-activated killer cel in vivo. Although interleukin-2 suppresses the metastasis of the malignant tumor into the adjacent organs and the tumor itself, it develops many side effects including fever, chills and decreased hepatic functions. Interleukin-2 also causes increase of capillary permeability with weight gain, pulmonary edema and ascites. In this experiment, the author studied the effect of interleukin-2 on the mouse uterine glandular cells, observing electron microscopically the changes in cytoplasmic organelles of uterine glandular epithelial cells in mice. Totals of 20 healthy female mice, DDY strain, weighing about 20 gm and exhibiting normal estrous cycle, were used as experimental animals were injected with 2 million units of interleukin-2 diluted in 0.2ml distilled water per kg of body weight and with 0.2ml of normal saline per kg of body weight in the control group for 5 days. And animals were sacrificed at 5th day after administration of interleukin-2. The specimens obtained from te corpus uteri were pre-fixed in 2% glutaradehyde-2.5% paraformaldehyde buffered in Millonig's phosphate solution (pH 7.2) and post-fixed in the 1% osmium tetroxide. After dehydration the specimens were embedded in Epon 812. Ultrathin sections, 600-800A in thickness, were made and double stained with uranyl acetate and lead citrate. And these preparations were observed with the electron microscope. The results obtained were as follows. 1. Microvilli were reduced and microfilaments in microvilli were degenerated. 2. The pronounced dilatation, sacculation and fragmentation of the cisternae of rough endoplasmic reticulum was associated with detachment of the membrane-bound ribosomes. 3. The formation of secondary lysosomes such as autophagic vacuoies and multivesicular bodies were observed. 4. Cristae of the some mitochondria were arranged irregularly and dissociated. Consequently, the results obtained in the present study suggest that interleukin-2 appears to be cytotoxic to the uterine glandular cells of the mouse and induces the degenerative changes of the uterine glandular cells.

      • 水稻의 穗發芽性에 關한 硏究 (Ⅱ) 栽培時期 移動에 따른 穗發芽性

        This study was conducted in 1987 to investigate the viviparous germination in rice. Six varieties, i. e., Daeseongbyeo, Sangpungbyeo Hwaseongbyeo, Taebagbyeo, Chilseongbyeo and Samgangbyeo were used for this experiment. Seedlings were transplanted on May 25 and dune 5 and panicles were harvested 5 times with 5 days interval beginning 25 days after heading. Panicles were incubated at normal temperature right after harvest to observe the germination. Rate of viviparous germination of Japonica type was higher than that of Indica type. The early varieties such as Daeseongbyeo, showed high viviparous germination. Rate of viviparous germination of May 25 transplanting was higher than that of June 5 transplanting. Viviparous germination rate of Japonica type appeared to be increased as the harvesting was postponed. On the other hand, Indica type didn't show viviparous germinate. Based on this study the optimum harvesting time of tested varieties appeared to be between 40 to 45 days after heading.

      • 5-Fluorouracil이 흰쥐 肝臟의 燐酸鹽 分解酵素의 活性에 미치는 影響

        The 5-Fluorouracil (5-FU) has been used as a synthetic antineoplastic agent with the synthesis of nucleic acid in the cell. Unfortunately this drug is known to cause toxic reaction in normal tissue also. In this experiment, the authors studied the effect of 5-FU on the rat liver histochemically observing the change in the activities of acid phosphatase and adenosine triphosphatase. Healthy albino male rats (Wistar strain) were used as experimental animals which were divided into the control and 5-FU treated groups. All experimental animals were given food and water unlimited. The animals of experimental group were administered 5-FU, in a dose of 50mg per kg of body weight by intraperitoneal injection. The animals of control groups were administered only water for injectio. The experimental animals were sacrificed at 6, 12, 24 and 36 hours after administration of 5-FU, respectively. The liver specimens, sectioned by cryostat were stained by Gomori method and by Wachestein and Meisel method for studying changes in the activities of acid phosphatase and that of ATPase, respectively. The results obtained were as follows: 1. The acid phosphatase activity was increased in the hepatic parenchymal cells of the 5-FU treated rat liver. 2. The adenosine triphophatase activity was decreased in the intralobular bile canaliculi of the 5-FU treated rat liver. Consequently, it is suggested that 5-FU would induce the change in the activities of possibly from production of toxic metabolites and subsequent damage in rat liver.

        Toeye,s "Therapeutic Method without Medicine" can be regararded as fundamedal method of oriental medicine. It has three categories : natural therapy, spiritual therapy, and living therapy. First, natural therapy means the philosophy of man and natural as one unity by which human beings adjust themselves to nature, including nutritional diets. Second, spritural therapy implies one's own self realization and some techniques leading to today's "abdomen (commonly called Tan-Jeon) breathing." Third, living therapy is based on the principle that our own natural environment and food (commonly called Sin-To-Bul-Yi) are most suited to our body, practically concering all our lives in food, clothing and shelter. Therefore, Toegye Yi hwang's"Therapeutic Method without Medicine"(especilly his "17 Principles and 5 Secrets of Nutrition and Exercise Therapy") will serve as a good modern people to keep fit.

      • 흰쥐 肝臟의 組織發生에 관한 組織化學的 및 電子顯微鏡的 硏究

        Numerous workers have intensively studied for physological and histochemical aspects on the histogenesis and differentiation of mammalian livers focusing attention on its hematopoietic function. In early fetal life hepatic diverticulum formed caudal part of the foregut in separated into two parts by the mesodermal septum each being developed into primordia later. The author has undertaken histological and histochemical studies to persue the morphological and functional changes to occur in hepatic lobules during development and differential period. The pregnant albino rats, Wistar strain, were sacrificed every day starting from 15th to 20th day of gestation. The specimens obtained from the fetal liver were fixed with 10% formalin and stained with hematoxylin-eosin to observe the morphological development and differentiation of the liver. The activities of acid and alkaline phosphatase were observed by the Gomori's methods(1931, 1941) and ATPase activity by the Wachstein-Meisel's method(1957). And also another liver specimens fixed with 4% glutaraldehyde in 0.1M cacodylate buffer(pH 7.4) at 4℃ by the Eranko's method(1952) were examined with electron microscope to investigate phosphatase activities in the hepatic parenchymal cells. The results were as follows. 1. In the liver of the 15th day of the fetus, undifferentiated hepatic parenchymal cells were constituted and a few nucleated erythocytes were spread at the intercellular region, and at the 16th and 17th day of the fetus a number of hematocytoblast were distributed and primitive hepatic cord and hepatic sinusoid were formed. 2. In the Liver of the 18th, 19th and 20th day of the fetus, hematopoietic cells were reduced but the differentiation of he hepatic parenchymal cells, hepatic cord and hepatic sinusoid were observed. 3. The activity of the acid phosphatase increased from the 17th to 20th day of the fetus. High activity of the enzyme at the peripheral and central zone and trace activity at the lysosome and rough endoplasmic reticulum were observed. 4. Moderate activity of the alkaline phosphatase was observed in the liver of the 19th and 20th day of the fetus at the nuclear membrane of the parenchymal cell and at the villi of the bile canaliculi. 5. The activity of the adenosine triphosphatase was observed in the liver of the 18th day of the fetus, and positive activity at the plasma membrane and weak activity at the rough endoplasmic reticulum were observed at the 19th and 20th day of the fetus, respectively.

      • 水稻의 穗發芽에 關한 硏究 (Ⅰ) 收穫 및 置床時期의 影響

        This study was conducted in 1986 to investigate the vivaparous germination in rice. Two varieties, Daeseongbyeo and Chilseongbyeo, were transplanted on May 25 and June 5 and panicles were harvested 5 times with the interval of 5 days beginning 30 days after heading. Panicles were seeded in incubator at 20℃ right after harvest and after drying and storage to observe the germination. Rate of viviparous germination of Daeseongbyeo was higher than that of Chilseongbyeo regardless of the seeding time. Rate of vivaparous germination of May 25 transplanting was higher than that of June 5 transplanting. Germination of Daeseongbyeo was not afftected by seeding time. Viviparous germination rate of Daeseongbyeo appeared to be increased as the harvesting was posponded. On the other hand, Chilseongbyeo did'nt germinate when seeded right after harvesting and rate of vivaparous germination was decreased as the harvesting was postponded. Vivaparous germination was higher in the order of top > middle > bottom part on a panicle, while grains on the primary branches showed higher vivaparous germination than those on the secondary branches.

      • Malathion이 Mouse 肝細胞의 微細構造에 미치는 影響

        Malathin (0,0-dimethyl 5-[1,2 dicarboethoxyethyl] dithiophosphate) is one of the organic phosphorus compounds and has been widely used as an insecticide. It is well known that the organophosphorus compounds inactivate an enzyme, cholinesterase, and resultant accumulation of acetylcholine effects on a pattern of cholinergic stimulation. Present study has demonstrated the hepatotoxicity caused by malathion, an organophosphorus compound, observing the ultrastructural changes in the cytoplasmic organelles of hepatocytes in mice. The experimental animals were given 0.2ml of malathion diluted in olive oil(100mg/kg) by intraperitoneal injection. The animals of control group were killed at 6 hours after olive oil treatment, and the animals of the experimental groups were killed at 12, 24 and 36 hours after administration of malathion, respectively. The specimens obtained from the left anterior lobes of the liver were stained with uranyl acetate and lead citrate double contrast methods and observed with JEM 100B model electron microscope. The cytoplasmic organelles of hepatocyte in malathion trated mice showed the fallowing significant changes. 1) A slight morphological changes such as dilatation, sacculation and fragementation of the cisternae of rough endoplasmic reticulum associated with detachment of membrane-bound ribosomes and disaggregation of the free ribosomes are recognized. 2) Primary lysosomes were increased in number and formation of secondary lysosomes (autophagic vaculoes and residual bodies) were observed. 3) The smooth endoplasmic reticulum associated with depletion of the glycogen particles were noted. 4) Large amount of lipid droplets were originated. The results obtained in the present study suggested that malathion appeared to by cytotoxic to the liver cells of mice and that the organophosphorus compound would induce retrograde changes characterized by destruction of small organelles of the hepatocytes.

      • Mitomycin이 Mouse의 肝細胞 내 Pyronin 好性顆粒에 미치는 影響

        Mitomycin, one of the mitosane antibiotics, is isolated from Streptomyces caespitosus which has anticancer effect. It is well known that mitomycin inhibites DNA synthesis in mammalian cells. The major machanism accounting for the inhibitory effect of mitomycin is interference of the template activities of the DNA by cross linking to DNA double helix. The auther has investigated the effect of mitomycin on the liver of mice, observing the distribution and disaggregation of the pyroninophilic granules in the hepatic parenchymal cells. A total of 52 healthy mice of ICR strain (the weight 20g) were divided into two groups; The control and the mitomycin treated groups. All experimental animals were given food and water ad libitum. The animals of the mitomycin treated groups were administered 4mg per kg of body weight of mitomycin diluted in the water for injection by intraperitoneal injection. The animals of the MC treated groups were sacrified at intervals of 6, 12, 24 and 36 hours after administration of mitomycin Histochemical preparations were stained with the methyl green-pyronin method for the pyroninophilic granules in the hepatic parenchymal cells. A marked reduction or disappearance of the pyroninophilic granules were observed in the entire hepatic lobule at 24 hours after administration of mitomycin. Consequently, it is suggested that mitomycin causes the loss of pyroninophilic granules in the hepatic parenchymal cells of the liver in the mitomycin treated mice.

Growth of service trade of developed countries including America are on trend to exceed that of goods trade currently among advanced countries, specially attached importance to America, recognition to attempt to establish integrated rule on service trade have been arise and a group for service trade have been set up in process of Uruguay Round talks held on Sep. 1986. On the other hand, considering our country's economy being, reached level of advanced economy and looking into the admission toward OECD in realily. It is the time for our country to accelerate open our service market to foreigners in turn of America's persistent demand to open our service market. Accordingly, at present, as problems faced us we will need to catch up real circumstance of present domestic service industries in view of national economy and prepare countermeasure on service market opening. This paper comprises four chapters. First chapter deal with introduction, second chapter a deseription analysis of service in the world economy is developed, third chapter the background of America's demand toward our service market opening were treated, in last chapter real state of our country's service trade are countermeasures against America's persistent opperssion for our service market opening were delieved.
