        Sibship reconstruction and effective population size estimation in mass spawning ark shell, Scapharca broughtonii based on microsatellite analysis

        Five highly polymorphic microsatellite loci were utilized to reconstruct sibship during a mass spawning event of ark shell, Scapharca broughtonii and to estimate the genetic variability between the D-shaped larvae and metamorphosed juvenile stage. The allelic diversity, and observed and expected heterozygosities exhibited similarity between the two sampling stages. The sibship reconstruction results of 180 offspring in each stage demonstrated that the effective population size was 20 in the D-shaped larvae stage and decreased to 16 in the metamorphosed juvenile stage. The inferred number of parents at the metamorphosed juvenile stage decreased 16.13 % and the inbreeding rate increased 0.6 % comparing to the D-shaped larvae stage. Multiple matings were detected both in males and females,and the reproductive success of both sexes in the two stages was positively correlated to the number of individuals with which it had mated. The results lend insight to S. broughtonii cultivation in China and provide a genetic guide for the development of more effective spawning techniques in the future.

        CircBRMS1L Participates in Lipopolysaccharide-induced Chondrocyte Injury via the TLR4/NF-κB Pathway through Serving as a miR-142-5p Decoy

        Osteoarthritis (OA) is the main cause of pain and disability in the elderly. It is reported that circular RNA BRMS1L (BRMS1 like transcriptional repressor) (circBRMS1L) is upregulated in the synovial fluid of OA patients. Nevertheless, the biological function of circBRMS1L in OA has not been validated. Lipopolysaccharide (LPS)- induced chondrocytes (C28/I2) were used as OA cell models in vitro. Expression patterns of circBRMS1L, microRNA (miR)-142-5p, and Toll-like Receptor 4 (TLR4) mRNA were verified by quantitative real-time polymerase chain reaction. Loss-of-function experiments were performed to explore the influence of circBRMS1L on LPS-induced extracellular matrix (ECM) degradation, apoptosis, and inflammation in C28/I2 cells. Protein levels were detected by western blotting. The regulatory mechanism of circBRMS1L in OA was explored by bioinformatics analysis, dualluciferase reporter, RNA immunoprecipitation, and/or RNA pull-down assays. CircBRMS1L was prominently upregulated in OA cartilage tissues and LPS-stimulated C28/I2 cells. Inhibition of circBRMS1L reduced LPS-induced ECM degradation, apoptosis, inflammation, and oxidative stress in C28/I2 cells. MiR-142-5p was downregulated while TLR4 was upregulated in OA cartilage tissues and LPS-stimulated C28/I2 cells. CircBRMS1L adsorbed miR- 142-5p to regulate TLR4 expression. MiR-142-5p inhibitor reversed circBRMS1L silencing-mediated effects on LPSinduced ECM degradation, apoptosis, inflammation, and oxidative stress in C28/I2 cells. TLR4 elevation overturned the inhibitory influence of miR-142-5p mimic on LPSinduced ECM degradation, apoptosis, inflammation, and oxidative stress in C28/I2 cells. Notably, circBRMS1L activated the nuclear factor-κB (NF-κB) pathway by the miR-142-5p/TLR4 axis. LPS-induced ECM degradation, apoptosis, inflammation, and oxidative stress in chondrocytes were at least partially regulated by the circBRMS1L/miR- 142-5p/TLR4/NF-κB pathway, highlighting a new mechanism responsible for OA advancement.

        Study on transmission error and torsional stiffness of RV reducer under wear

        In order to discuss the influence of wear dynamic evolution on the degradation of core transmission performance, transmission error and meshing torsional stiffness of RV reducer, taking BX-40E reducer as the research object, based on Langkali-Nikraves contact force model and Archard wear theory, the wear coefficients with different position conditions were obtained through equivalent wear experiments. The RV transmission dynamic model considering wear, meshing stiffness, meshing force, meshing tooth pairs and transmission error linkage was established by using centralized parameter method and dynamic subsystem method. Compared with the traditional tooth profile wear simulation, which distribution and size of the wear depth curves were more and more different with wear dynamic evolution, theoretically concluded that the dynamic update of wear coefficient is necessary and correct for the accurate simulation of tooth surface wear. The transmission error shows a "double hump" distribution mapped to the wear depth of the tooth profile, and deteriorates with the wear accumulation. The equivalent torsional stiffness of cycloid pin teeth decreases piecewise approximately linearly due to wear. In the previous stage, the reduction rate is small because the tooth detachment first occurs at the tooth top and root, and the single tooth torsional stiffness near the tooth top and root is smaller. The research results provide a new idea and theoretical basis for improving the transmission accuracy, stability holding ability, wear reduction and life extension of RV reducer.

        After implementation of the merit pay, the quantity and quality of teaching staff's scientific research projects, papers and monographs, teaching reform projects, teaching achievements would definitely affects their post promotion and appointment and salary treatment, etc. The current office automation system, education administration management system, scientific research management system mainly focused on daily office use, teaching and scientific research business process and daily affairs processing. It is unable to provide scientific quantification, accurate statistics and decision support for the teaching research projects and scientific research projects. As for these cooperation projects involve multiple majors and departments, repeated recording, repetitive statistics, missing statistics or misinformation are easy to happen and that will affect the statistical results. With all kinds of information of teaching research and scientific research projects (data) doubly increases and constantly changes year by year, there is a tough problem that the university administrator and decision-making departments are facing. How to standardize the entry (or batch import) of research data, and realize multi condition query and dynamic update? And how to realize the implementation of teaching, scientific research merit automatic quantification, distribution according to proportion, and do automatic summation by name. “University teaching and research and scientific research performance quantitative statistics system based on Web “emerges as the times require. This project aims to develop a set of network management system with multiple users and graded rights, which embodies teaching research scientific research project, standardized management, merit points, automatic quantification statistics, data analysis and warning and report output. Its accurate and fast statistical results can provide the decision-making basis for their department annual summary, and parts of the system module can be applied to other universities after slightly modified.

        Effect of Workplace Ostracism on Knowledge Hiding: Mediating Role of Job Insecurity and Moderating Role of Psychological Ownership

        [연구목적] 본 연구의 목적은 직장 내 집단따돌림이 지식은폐 행동에 미치는 영향, 직무불안정성의 매개효과 및 심리적 주인의식의 조절효과를 검증하는 것이다. [연구방법] 본 연구는 이론적 연구를 바탕으로 세 가지 주요한 연구 가설을 제시하고 연구 모델을 수립하였다. 데이터는 설문조사 방법에 의해 중국 내 직원으로부터 수집되었고, SPSS 24.0은 연구 모델의 가설을 검증하기 위해 사용되었다. [연구결과] 직장 내 집단따돌림은 지식은폐 행동에 긍정적인 영향을 미치고, 직무불안정성는 두 변수의 관계에서 매개효과를 갖고 있으며, 심리적 주인의식은 두 변수의 관계에 조절효과를 갖게 된다는 연구 결과가 나왔다. [연구의 시사점] 본 연구는 이론과 데이터를 기반으로 직장 내 따돌림과 지식 은폐 사이의 역할 관계를 심층적으로 연구한다. 직원들의 지적 은폐 행위를 억제하기 위한 새로운 발상을 제안한다. 기업들은 직장 따돌림이 주는 악영향을 지양해야하여 직원들의 업무불안을 줄인다. 또한 직장 내 따돌림을 유발하는 전치 요소를 찾아, 조직 환경 개선, 직원들의 지식 공유와 작업 안정감을 증진시켜 기업 효율성을 높일 수 있도록 엄격한 관리와 인문 배려를 결합한다. [Purpose] The aim of this study is to verify the impact of Workplace Ostracism on Knowledge Hiding, the mediating effect of Job Insecurity and the moderating effect of Psychological Ownership. [Methodology] This study proposes three research hypotheses and established a research model as well. Data were collected by the internet survey method. SPSS 24.0 were used to verify the hypotheses of the research model. [Findings] Workplace Ostracism is positively related to Knowledge Hiding, Job Insecurity as the mediating variable and Psychological Ownership as the moderating variable effect Workplace Ostracism and Knowledge Hiding. [Implications] This research refers to the in-depth analysis of the relationship between workplace exclusion and knowledge hiding under the joint support of theory and data. Provides new ideas for inhibiting employees’ knowledge hiding behavior. Companies should avoid the negative effects of workplace exclusion and reduce employee insecurity. Identify pre-factors that induce workplace rejection and improve the organizational environment. Combining strict management with humanistic care to enhance employees’ knowledge sharing and job security, and improve corporate efficiency.

        High-level Expression of an Acidic and Thermostable Chitosanase in Pichia pastoris Using Multi-copy Expression Strains and High-celldensity Cultivation

        Chitin is a linear homopolymer of acetylated β- (1,4)-linked glucosamine residues and among the most abundant polysaccharides in the world. Here, we identified and purified a novel chitosanase (CCHA) from Aspergillus oryzae NKY2017 obtained from Hu’bei province in China. Construction of a cDNA library from this strain revealed the gene sequence subsequently expressed in Pichia pastoris and subsequent construction of multi-copy expression plasmids (CCHA1/2/3/4). The results demonstrated elevated levels of CCHA expression in multi-copy strains, with strain CCHA4 chosen for high-density fermentation and enzyme-activity experiments. High-density fermentation achieved a CCHA yield of 22,500 U/mL, and temperature and pH optimization resulted in the highest CCHA activity at 40°C and 4.0, respectively. We used this enzyme for a large-scale preparation of oligosaccharides: 4 g enzyme could convert 150 kg chitosan into oligosaccharides in 24 h at 40°C. These results demonstrated abundant CCHA expression in P. pastoris and suggested the efficacy of CCHA for use in industrial applications.

        “남방은 본래 독이 많아, 북쪽의 客이 언제나 침입할까 두려워 한다”는 것은 唐代문인의 영남에 대한 이미지(Image)이다. 영남은 환경이 무덥고,식물자원이 풍부하며, 瘴氣, 射工, 毒草, 蠱毒, 毒蛇등 독을 가진 생물이 살기에 적합하고, 옛날 이곳으로 이주해온 중원 인사들이 그 환경에 적응을 하는데 영향을 주어, 그들이 이 지역의 환경이 매우 나쁘다고 보게 되었다. 중원 지역과 비교해 볼 때, 영남 경제 문화의 낙후는 중원사회로부터 편견과 경시를 받게 만들게 되었고, 그들의 관념 속에 “蠻夷”의 땅으로 자리 잡게 되었다. 영남 경제문화 및 생태 환경의 특성들 때문에 쫓겨 난 문 인들의 주요 유배지가 되게 되었고, 이들 문인들은 그들의 좌절과 죽음에 대한 공포 속에서 영남의 매우 열악한 생태 환경을 반복적으로 그려냈던 것이다. 때문에 문인들의 영남에 대한 이미지의 형성은 영남의 생태환경, 사회환경 그리고 그들 자신이 처한 환경이 함께 작용한 결과였다. 당대 중기로부터 문인의 桂林, 連州와 韶州등에 대한 인식에 변화가 발생하였다. 이 지방의 환경은 사람의 마음에 들었으며 사람의 생존에 적합하였다. 문인의 이들 지방에 대한 인식의 변화는 그것들의 특별한 생태환경, 경제문화 발전의 성과와 함께 밀접한 관계가 있었다. 한편으로는, 桂林등 영남 북부의 산지 구릉지대의 비교적 높은 지세에 위치하고 있고, 영남 남부의 지대가 낮고 습기가 많은 땅의 환경과는 일정한 차별이 있다. 또 한편으로는 이들 지방은 중원과 영남의 육로교통선상에 위치하는데 지리 위치상으로 매우 중요하여 그 경제 문화는 북방으로부터의 이민의 큰 추세 속에서 신속한 발전이 일어났다. 그 지역의 경제 문화적 발전 과정은 사람들이 그 사회 환경과 생태 환경을 새롭게 개조하고 다시 새롭게 인식하는 과정이었다. 비록 이 지방의 인식에 대한 문인들의 변화가 발생하였다고 하지만, 전체적으로 볼 때 이러한 변화는 문인의 영남에 대한 이미지를 근본적으로 변화시킨 것은 아니어서 영남은 여전히 공포의 땅이고 사람들로 하여금 매우 무서워하는 땅으로 남아있었다. There were all kinds of poison in the south, the people from the north had always feared them. This was the literati’s Lingnan image in the Tang dynasty. It was warmer than the north and there were rich plants in Lingnan. This ecological environment fit malaria, akamushi disease, poison ivy, parasitic tympanties, viper and so on. The people from the north did not adapted Lingnan so that they thought it was very horrible. Compared with the Center China, economic and culture lag in Lingnan made that the literati was full of prejudice and discrimination. So the literati thought that Lingnan was uncivilized. Lingnan became the most important place that the banished literati were exiled because of its economy, culture and ecological environment. To these literati, they expressed their frustration and fear of death by writing poetry on abominable ecological environment of Lingnan. Therefore, that ecological and social environment in Lingnan interacted with the literati’s encounter had resulted the literati’s Lingnan image. The literati’s concept to Guilin, Lianzhou and Shaozhou began to change from the middle Part of Tang Dynasty, these place were delightful and suitable for the people from the north. This concept closely connected with their special ecological environment, the development of their economic and culture. On the one hand, Guilin, Lianzhou and so on located in the north of Lingnan. There was mountain and upland. Their ecological environment was different with the south of Lingnan that was wetlands. On the other hand,these place were along land transport line between the Central China and Lingnan, geography position was very important, their economy and culture developed rapidly with the help of the people from the north. The process that their economy and culture developed was that people changed and knew ecological environment again. Although the literati’s concept about Guilin, Lianzhou and so on shifted, they did not fundamentally change the literati’s Lingnan image. It was still horrible and very daunting.
