        Nelumbinis Semen Reverses a Decrease in $5-HT_{1A}$Receptor Binding Induced by Chronic Mild Stress, a Depression-like Symptom

        Depression is associated with a dysfunctional serotonin (5-hydroxytryptamine; 5-HT) system. More recently, several lines of evidence suggest that an important factor in the development of depression may be a deficit in the function and expression of $5-HT_{1A}$ receptors. The present study assessed if Nelumbinis Semen (N. s.) had an anti-depression effect through reversing a decrease in $5-HT_{1A}$receptor binding in rats with depression-like symptoms induced by chronic mild stress. Using a $5-HT_{1A}$ receptor binding assay, with a specific $5-HT_{1A}$receptor agonist, 8- OH-DPAT (8-hydroxy-2-(di-n-propylamino) tetralin), the mechanism of the anti-depression effect of N. s. on rats was investigated, and the effects compared with two well-known antidepressants, Hyperium Perforatum (St. Johns Wort) and fluoxetine (Prozac). Animals were divided into five groups: the normal (N) group without chronic mild stress (CMS), the control (C) group under CMS for 8 weeks, the Nelumbinis Semen (N. s.) treatment group under CMS for 8 weeks, the Hyperium Perforatum (H. p.) treatment group under CMS for 8 weeks and finally, the fluoxetine (F) treatment group under CMS for 8 weeks. Each treatment was administered to rats during the last 4 weeks of the 8-week CMS. A sucrose intake test was performed to test the anti-depression effect of N. s. The N. s. treatment significantly reversed the decreased sucrose intake under CMS (P<0.05 compared to control group under CMS). In the CA2 and CA3 regions of the hippocampus, both N. s. and H. p. reversed the CMS-induced decrease in $5-HT_{1A}$receptor binding. In the I to II regions of the frontal cortex, N. s. and H. p. also reversed the CMS-induced decrease in$5-HT_{1A}$receptor binding, and even showed a significant increase in $5-HT_{1A}$receptor binding compared to the F treatment group (N. s. vs. P, p<0.05, H. p. vs. P, p<0.05). However, in the hypothalamus, all treatments reversed the CMSinduced decrease in $5-HT_{1A}$receptor binding. This reversal effect of N. s. on the decrease in $5-HT_{1A}$receptor binding in the frontal cortex, hippocampus and hypothalamus of rat brains was very similar to that of H. p, but different from that of F. It is concluded that N. s. presents an anti-depression effect through enhancing $5-HT_{1A}$receptor binding.

        급성 신우신염 치료시 아미노글라이코사이드(Micronimicin Sulfate)에 의한 유사 Bartter씨 증후군의 발생에 관한 평가

        The aminoglycoside antibiotics is widely used in the treatment of infectious caused by gram-negative bacteria and for synergistic effect with(β-lactam antibiotics. However, its therapeutic usefulness is limited by this potential nephrotoxicity and by disturbance of electrolyte homeostasis resulting in hypomagnesemia, hypokalemia, hypocalcemia such as Bartter-like syndrome. Many case repots have been reported on development of Bartter-like syndrome after aminoglycosides administration. But these reports had the many differences of such as types of aminoglycosides, age of patients, duration and total dose of treatment, combined antibiotics and baseline diseases. Therefore, the purpose of this study is to assess the effects of micronomocin sulfate on magnesium, calcium and potassium status of patients in acute pyelonephritis. Twenty one patients in acute pyelonephritis(18 female/3 male, ages 20-75) was treated with single or combined antibiotics. Eleven of twenty one patients as study group were treated with boh micronomicin sulfate(aminoglycoside, 4mg/kg/day, during 5-8days) and flomoxef sodium (3rd cephalosporine, 2g/day, during 5-8days), and ten of twenty one patients as control group were treated only with flomoxef sodium(3rd cephalosporine. 2g/day. during 5-8days). Renal values, plasma and urinary electrolytes were measured before and at the end of IV antibiotic therapy. After micronomicin sulfate administrated for 6.4±1.5days, serum Mg, Ca, K, FEMG (fractional excretion of Mg), TTKG(transtubular K concentration gradient) and FEca.(fractional excretion of Ca) did not significantly change(p$gt;0.05). Therefore, those results suggest that micromonicin sulfate therapy within dose of 24mg/day(4mg/kg/day) for 6.4±1.5days may not cause disturbance of electrolyte homeostasis such as Bartter-like syndrome in acute pyelonephritis. Howerever, electrolyte disturbance is an important complication when aminoglycosides is given in larges doses over extended periods. Therefore, monitoring of blood concentration and urinary loes of electrolyte should be carried out along with careful observation of Bartter-like syndrome.

      • Cytoprotective Roles of a Novel Compound, MHY-1684, against Hyperglycemia-Induced Oxidative Stress and Mitochondrial Dysfunction in Human Cardiac Progenitor Cells

        <P>Diabetic cardiomyopathy (DCM) is tightly linked to heart disorders and dysfunction or death of the cardiomyocytes including resident cardiac progenitor cells (CPCs) in diabetic patients. In order to restore loss of function of resident or transplanted CPCs, much research has focused on novel therapeutic strategies including the discovery of novel function-modulating factors such as reactive oxygen species (ROS) scavengers. Here, we developed and defined a novel antioxidant, MHY-1684, for enhancing the angiogenic potential of CPCs against ROS-related DCM. Short-term treatment with MHY-1684 restored ROS-induced CPC cell death. Importantly, MHY-1684 decreased hyperglycemia-induced mitochondrial ROS generation and attenuated hyperglycemia-induced mitochondrial fragmentation. We observed that the activation process of both Drp1 (phosphorylation at the site of Ser616) and Fis-1 is drastically attenuated when exposed to high concentrations of D-glucose with MHY-1684. Interestingly, phosphorylation of Drp1 at the site of Ser637, which is an inhibitory signal for mitochondrial fusion, is restored by MHY-1684 treatment, suggesting that this antioxidant may affect the activation and inhibition of mitochondrial dynamics-related signaling and mitochondrial function in response to ROS stress. In conclusion, our finding of the novel compound, MHY-1684, as an ROS scavenger, might provide an effective therapeutic strategy for CPC-based therapy against diabetic cardiomyopathy.</P>

      • Identification of subtypes of refractory asthma in Korean patients by cluster analysis.

        <P>Refractory asthma, a subtype of asthma with uncontrolled symptoms despite antiasthma medications, is a heterogeneous syndrome with variable clinical features, presumably different etiologies, and pathophysiological mechanisms. The heterogeneity of refractory asthma, however, is poorly understood. We aimed to characterize refractory asthma and to improve our understanding of the heterogeneity of refractory asthma patients. We identified refractory asthma patients (n = 96) as defined by the American Thoracic Society's criteria from a cohort of Korean asthma patients (n = 2,187). Then, cluster analysis was conducted to classify subtypes of refractory asthma. Among the study group, 4.4 % (n = 96) of all asthma patients had refractory asthma. Cluster analysis identified four distinct groups of refractory asthma. Age at onset was younger in clusters 1 and 2 than in clusters 3 and 4. Patients in cluster 1 had the most well-preserved pulmonary function; patients in cluster 2 had a female predominance and the most severe airway obstruction; patients in cluster 3 were mostly female and had the most enhanced bronchial hyperresponsiveness; and patients in cluster 4 were most male and tended to be cigarette smokers. The current results suggest that refractory asthma is a heterogeneous syndrome and could be classified into four subtypes. Underlying pathogenesis and therapeutic approaches may differ for the different subtypes and further research is needed.</P>

        경추와 요추에 동반한 척추관협착증의 1례

        Spinal stenosis denoting a critical reduction of the A-P diameter of spinal canal, foramina and lateral recesses can be existed in both cervical and lumbar regions. But it is relatively rare that cervical and lumbar spinal stenosis are coexisted simulataneously. The authors report a case of coexisting cervical and lumbar spinal stenosis with historical background and discussion. The patient was 49 years old male who complained intermittent neurogenic claudication, paraparesis and voiding disturbance. A successful result was obtained through one stage decompressive laminectomies in cervical and lumbar reaions.

        A Fuzzy System Representation of Functions of Two Variables and its Application to Gray Scale Images

        An approximate representation of discrete functions {f$_{i,j}\mid$|i, j=-1, 0, 1, …, N+1}in two variables by a fuzzy system is described. We use the cubic B-splines as fuzzy sets for the input fuzzification and spike functions as the output fuzzy sets. The ordinal number of f$_{i,j}$ in the sorted list is taken to be the out put fuzzy set number in the (i, j) th entry of the fuzzy rule table. We show that the fuzzy system is an exact representation of the cubic spline function s(x, y)=$\sum_{N+1}^{{i,j}=-1}f_{i,j}B_i(x)B_j(y)$ and that the approximation error S(x, y)-f(x, y) is surprisingly O($h^2$) when f(x, y) is three times continuously differentiable. We prove that when f(x, y) is a gray scale image, then the fuzzy system is a smoothed representation of the image and the original image can be recovered exactly from its fuzzy system representation when it is a digitized image.e.

        시상및 기저핵 후부에 발생한 배아세포종 1례

        Intracranial germinoma usually occurs in the pineal and suprasellar regions. It is relatively rare that germinoma occurs in the thalamus and basal ganglia. The authors report a case of germinoma involving the posterior part of thalamus and basal ganglia involving the posterior part of thalamus and basal ganglia. The patient was 20 years old male who complained left side hemiparesis for 2 months. A successful result was obtained through radiation therapy after partial removal of tumor mass.

        Preparation and Characterization of Natural Material Extracted from Germinated Brown Rice

        Purpose: The aim of this study was to prepare and evaluate a natural material extracted from germinated brown rice (GBR). Herein, we evaluated whether the natural material could positively activate the biological effects seen during bone formation, including enhancement of metabolic activity, osteogenesis, and the expression of vascular endothelial growth factor (VEGF), one of the growth factors in human osteoblast-like cells. Methods: The natural material was created by a hot water extraction process after being soaked for 2~3 days in tap water and dried at 50°C. The material was characterized using field emission scanning electron microscopy (FE-SEM), energy dispersive X-ray spectroscopy (EDX), X-ray diffraction (XRD), and Fourier transformed infrared (FTIR) spectroscopy. The biological behaviors of the material were also investigated; we performed tests to assess cell cytotoxicity, metabolic activity, osteogenic markers related to bone formation, and VEGF. Results: The EDX, XRD, and FTIR results for the natural material indicated the presence of organic compounds. The natural material caused positive increases in cell metabolic activity and mineralized bone formation without cytotoxicity. The protein levels in the extract for the 6.25 μg/mL, 12.25 μg/mL, 25 μg/mL, 50 μg/mL, and 100 μg/mL groups were significantly different from that for the control. Conclusions: The GBR-based natural material was easy to prepare and had characteristics of a potential biomaterial. The biocompatibility of this natural material was evaluated using in vitro techniques; our findings indicate that this novel material is promising for agricultural and biological applications.

        폐루프 자속추정기를 이용한 철도차량의 유도 전동기 센서리스 벡터제어

        본 논문에서는 유도전동기를 인버터와 같은 전력변환장치를 사용하여 정상상태의 동작특성은 물론 과도특성까지 해석하기 위해서는 유도전동기와 부하를 포함하는 전체 시스템의 정확한 동적 모델링을 이용하여 시스템 방정식으로 표현해야 한다. 또한 최대 견인력 제어를 위해서 속도센서리스벡터제어와 부하 토크 외란 관측기를 통하여 점착력 계수를 추정한다. 이와 같은 시스템의 제어 알고리즘을 구현하기 위하여 철도 모의 장치를 이용하여 제안된 알고리즘을 시뮬레이션과 실험을 통하여 확인하고, 철도 차량의 속도 가감에 따른 공전속도에 대한 점착력의 관계 등 제반 사항을모의 장치에 의해서 구현하였다. Displayed system equationally using accurate dynamic modeling of whole system including induction motor and load to analyze induction motor to normal condiction’s action characteristic as well as transient characteristic using power converter device such as inverter in this paper. Also, presume adhesive power calculation through speed sensorless vector control and load torque disturbance observer for maximum tractive force control. Confirmed proposed algorithm through simulation and an experiment using railroad experiment equipment to embody control algorithm of such system. With relation of adhesive power about the wage speed by speed addition and subtraction of railway vehicle, embodied all sorts item by experiment equipment.
