In the present study, the changes in the production of the superoxide radicals and the activities of superoxide dismutase (superoxide: superoxide oxidoreductase, EC 1.15.1.1, SOD) and catalase (hydrogen peroxide: hydrogen peroxide oxidoreductase, EC 1.11.1.6) in the rat brains that were exposed to 95% O_2 or air following the intratracheal administration of catalase (3500 units) were investigated. The results were summarized as follows: 1. In most of experimental groups, activities of Cu, ZN-SOD were higher than that in controls (p<0.01, p<0.05), and the activities the rats exposed to 95% O_2 were higher than those of rats exposed to air after catalase administration (p<0,01). 2. Activities of Mn-SOD in most of experimental groups were similar to or higher than that in controls (p<0.01, p<0.05), and the activities of Mn-SOD in air exposed groups were higher than those of 95% O_2 exposed groups after catalase administration (p<0.01). 3. In all experimental groups treated with catalase., the production of superoxide radicals appeared to be negligible or very low in comparison with those in the control groups, 4. The activities of catalase in air exposed groups decreased at 30 min.,1hr, 2hr and 6hr in comparison with those of the control groups (p<0.01, p<0.05). But in all 95% O_2 exposed groups, activities of catalase were higher than that in controls (p<0.01). In conclusion, it seems likely that the intratracheal administration of catalase, on one hand, results in the increase of the activity of SOD and catalase in the brains of rats which were exposed to 95% O_2 and it suppresses the production of the superoxide radicals. Accordingly the SOD and catalase could protect the brain effectively from the harmful effects following the exposure to hyperoxic state.
