        비정상 간기능 환자에서 Pringle 술식시 혈역학 변화

        Background : During the Pringle maneuver (PM), the increase of systemic vascular resistance (SVR) and the active constriction of the intrahepatic capacitance vessels could minimize arterial blood pressure change. Pressor reactivity to sympathetic agonists is impaired and blood volume buffering capability is less efficient in a cirrhotic liver. Accordingly, we evaluated the relations between hemodynamics during PM and preoperative liver function test (LFT) by serum aminotransferase and Indocyanine Green (ICG) clearance. Methods : Twenty-seven patients undergoing hepatectomy with PM were classified into two groups according to the liver function state assigned by serum aminotransferases and ICG clearance test. Sequential changes of hemodynamics were measured with Doppler flowmeter during PM. Hemodynamic data were analyzed by using ANOVA for repeated measurement. Correlation between LFTs were sought using Pearson correlation and logistic regression. Results : During the PM, cardiac output decreased significantly compared to the preclamping period in the abnormal LFT group. There were no significant changes in any other hemodynamic variables in the normal LFT group. When comparing the two groups during PM, mean arterial blood pressures and cardiac output were significantly lower in the abnormal LFT groups compared to the normal LFT groups (P< 0.05). Conclusions : These differences may suggest that cardiovascular responsiveness to reflex autonomic stimulation during the PM is significantly impaired in patients with abnormal LFT compared with normal LFT subjects.         Antilipidemic effects and gene expression profiling of the glycosaminoglycans from cricket in rats on a high fat diet

        Glycosaminoglycan (GAG) from cricket (Gryllusbimaculatus) was studied as a potential health supplement. Antiatherosclerotic and antilipidemic effects of theGAG of G. bimaculatus (GbG, 5 or 10 mg/kg) wereinvestigated in 15-week old Wistar rats treated with GbGfor over a month. GbG produced a meaningful anti-edemaeffect with inhibition of C-reactive protein (CRP). Also,the weights of abdominal and epididymidal fat were alsoreduced in conjunction with a mild increase in bodyweight. Furthermore, the sero-biochemical parametersshowed an antihyperlipidemic effect with decreased levelsof phospholipid, AST, ALT, total cholesterol and glucosein a dose-dependent manner. In addition anticoagulant andantithrombotic effects were seen: platelet, thrombin time,prothrombin time and Factor I were increased with GbGtreatment. Furthermore, the GbG treated rat group (at10 mg/kg) compared to control, showed that 588 genes(test/control ratio[2.0) including lipocalin 2 (Lcn2) andalpha 2-macroglobulin (A2 m) were up-regulated, and 569genes (test/control ratio[0.5) including stearoyl-coenzymeA desaturase 1 (Scd1) were down-regulated. Basedon these results, GbG could potentially prevent or treatfatty liver or hyperlipidemia in rats on a high-fat diet.

        Immune modulation of glycosaminoglycan derived from P. lewisi in TNF-a stimulated cells

        Poecilocoris lewisi (Korean name: ‘‘Kwangdaenolinjae’’)is a red-striped gold stink bug (insect) whichhas been used as a crude drug in traditional medicine ofEast Asia and Korea. In this study, ethanol extract andglycosaminoglycan from P. lewisi (Pl GAG), as an activesubstance among its components, were investigated fortheir potential anti-inflammatory actions. They were foundto be a potent inducer of nitric oxide (NO) production fromcalf pulmonary artery endothelial (CPAE) cells and astimulator of endothelial nitric oxide synthase in a dosedependentmanner. The anti-inflammatory activities werealso evaluated by determining the level of adhesionmolecules related to atherogenesis and pro-inflammatorycytokines, such as vascular cell adhesion molecule-1(VCAM-1), intercellular adhesion molecule-1 (ICAM-1),secretory phospholipase A2, and prostaglandin E2,stimulated by tumor necrosis factor (TNF)-a in humanumbilical vein endothelial cells (HUVEC). They alsoshowed inhibitory effects on vascular endothelial growthfactor (VEGF) production in HUVECs. Matrix metalloproteinases(MMP-2 and 9) were also inhibited by treatmentwith this extract or glycosaminoglycan. Furthermore,this GAG showed cytotoxicity against CT-26 colon cancercells whereas having no cytotoxicity in CHO normal cells. Monosaccharide (amino, acidic, neutral monosaccharides)composition of used GAG was characterized bytrimethylsilylated GC–MS analysis method.

        Anti-aging Effect and Gene Expression Profiling of Aged Rats Treated with G. bimaculatus Extract

        Extract from Gryllus bimaculatus crickets inhibits oxidation at the DNA level, with reduced production of 8-hydroxy-2'-deoxyguanosine (8-OHdG). Microarray analyses were performed with a rat 28K cDNA clone set array to identify the gene expression profiles of aged (10 months old) Wistar Kyoto rats treated for one month with 100 mg/kg G. bimaculatus ethanol extract to assess the effects. The extract produced a meaningful anti-edema effect, evident by the inhibition of creatinine phosphokinase activity. The weights of abdominal and ovarian adipose tissues were reduced and the proportion of unsaturated fatty acids in adipose tissues was increased in an extract dose-dependent manner. Compared with untreated control rats, rats treated with the extract displayed the upregulation of 1053 genes including Fas (tumor necrosis factor receptor superfamily, member 6), Amigo3 (adhesion molecule with an immunoglobulin-like domain), Reticulon 4, 3-hydroxy-3-methylglutaryl-coenzyme (Hmgcr; a reductase), related anti-fatigue (enzyme metabolism), and Rtn antioxidant, and the downregulation of 73 genes including Ugt2b (UDP glycosyltransferase 2 family), Early growth response 1, and Glycoprotein m6a. Data suggest that G. bimaculatus extract may have value in lessening the effects of aging, resulting in a differential gene expression pattern indicative of a marked stress response and lower expression of metabolic and biosynthetic genes.

        Immune Modulation of B. terrestris Worker (a Type of Bumblebee), Extract on CFA- induced Paw Edema in Rats

        To develop a composition for enhancing immunity, based on alcohol extracts of thebumblebee as an active ingredient, bumblebee ethanol extracts were evaluated fortheir protective effect in chronic models of inflammation, adjuvant induced rat arthritis. B. terrestris worker extract (SDIEX) and, B. hypocrita sapporoensis lava and pupaextract (SPDYBEX), significantly decreased paw edema in arthritic rats, at a dose100 mg/kg, respectively. The cytokine levels related inflammation of COX-2, sPLA2,VEGF, and TNF-α, were decreased, compared to positive control, indomethacin(5 mg/kg). Histopathological data demonstrated decreases inflammatory activity, hind paw edema, and repaired hyaline articular cartilage in DRG over a 2 wkadministration. HPLC and GC-MS analysis of SDIEX and SPDYBEX revealed thepresence of cantharidin.

        Anti-hypertensive Effect of the Dongchunghacho, Isaria sinclairii, in the Spontaneously Hypertensive Rats

        The present study examined the effect of the methanol extract of Isaria sinclairii, a kind of Donchunghacho (Tochukaso), on blood pressure in spontaneously hypertensive rats (SHR). Blood pressure and heart rate were measured after treatment with the methanol extract of I. sinclairii by the indirect tail-cuff method and the direct in vivo model. Starting at 12 weeks of age, male SHR were treated with the extracts for 2 or 4 weeks. We found that, when compared to untreated control SHR, oral treatment with I. sinclairii methanol extract (30 mg/kg/day) remarkably decreased systolic blood pressure from 200 to 112 mmHg and decreased diastolic blood pressure from 114 to 88 mmHg. Furthermore, efficacy of methanol extract of I. sinclairii was superior to captopril (30 mg/kg/mL, positive control), an angiotensin-converting enzyme inhibitor, with a lowering effect that dropped systolic blood pressure from 201 to 130 mmHg and diastolic blood pressure from 102 to 92 mmHg. However, in normal Wistar Kyoto rats, I. sinclairii methanol extract did not significantly change the normal blood pressure, suggesting that this type of Dongchunghacho has a selective effect against hypertension. Therefore, methanol extract of I. sinclairii may be used as an anti-hypertensive food/agent. Furthermore, this extract also has multiple actions such as No production in endothelial cells, inhibiting thrombininduced blood coagulation by thrombin and mildly decreasing in prostaglandin E2 levels in cultured macrophage cells, all of which might contribute to protection against atherogenesis and thrombus formation. HPLC and MS analysis of methanol extract of I. sinclairii revealed the presence of adenosine.
