Osteosarcoma is a malignant bone tumor that develops in the osteoblast cells that form the outer covering of bone. It is the most frequent malignant bone tumor found at 10-14 years and at >65 years of age. Atmospheric pressure plasma has advantages of high density and rich chemical agents without elevation of the substrate temperature. These non-equilibrium characteristics show promising applications in the biomedical field, opening a new research area called “Plasma Medicine”, which includes sterilization, coagulation, wound healing, and cancer treatment. However, the mechanism of apoptosis caused by plasma treatment in osteosarcoma is not well understood. In this study, the researchers show that microwave plasma causes selective cell cycle arrest and apoptosis in osteosarcoma cells in vitro. An inhabitation of cell growth of microwave plasma showed that this plasma has selective cytotoxic effects on MG-63 cells in comparison to hFOB 1.19 cells. Also, the resesearchers observed that microwave plasma treatmenst significantly showed disruption and aggregation of F-actin. The results demonstrated that treatment of MG-63 cells with microwave plasma induced cell cycle arrest in the G2/M phase, using flow cytometry and western blot assay. To detect PARP and DFF-45 cleavage or a decrease as a result of caspase-3 activation, MG-63 cells were treated either with various times of microwave plasma. The research studies demonstrated that microwave plasma induced G2/M cell phase arrest and triggered apoptosis.

Early growth response (Egr)-1 is a $Cys_2-His_2-type$ zincfinger transcription factor. It has been shown to induce survival and proliferation of immature and mature B cells, respectively, but its role in the differentiation of B cells into plasma cells remains unclear. To examine the effects of Egr-1 deficiency on the activation of B cells, naive B cells from $Egr1^{-/-}$mice and their wild-type (WT) littermates were activated to proliferate and differentiate, and then assayed by FACS. Proportions of cells undergoing proliferation and apoptosis did not differ between $Egr1^{-/-}$ and WT mice. However, $Egr1^{-/-}$ B cells gave rise to fewer plasma cells than WT B cells. Consistently, $Egr1^{-/-}$ mice produced significantly lower titer of antigen-specific IgG than their WT littermates upon immunization. Our results demonstrate that Egr-1 participates in the differentiation program of B cells into plasma cells, while it is dispensable for the proliferation and survival of mature B cells.

Plasma cells are usually considered short-lived plasma cells. This view is changing because plasma cells can survive for a long time in bone marrow. Long-lived plasma cells may be important in allergy because of the persistence of immunoglobulin E in allergic animal models after immune suppression. Allergic therapies efficiently control allergic symptoms but do not provide a cure, especially in adults. Elimination of long-lived plasma cells that secrete specific immunoglobulin E may provide a cure in allergic patients. The purpose of this review is to describe the creation of long-lived plasma cells and their importance in allergic diseases.

Many researchers studied cell culturing on surfaces with chemical functional groups. Previously, we reported surface properties of plasma polymerized ethylenediamine (PPEDA) films deposited by plasma enhanced chemical vapor deposition with various plasma conditions. Surface properties of PPEDA films can be controlled by plasma power during deposition. In this work, to analyze correlation of cell adherence/proliferation with surface property, we cultured rat intestinal epithelial cells-18 on the PPEDA films deposited with various plasma powers. It was shown that as plasma power was decreased, density of cells cultured on the PPEDA film surface was increased. Our findings indicate that plasma power changed the amine density of the PPEDA film surface, resulting in density change of cells cultured on the PPEDA film surface.

Purpose: Chronic inflammatory gingival lesions occur as pyogenic granulomas or non-specific chronic suppurative lesions. Methods: Of the 59 chronic inflammatory gingival lesions examined, plasma cell granuloma (n=14), which showed an intense antibody-mediated immune reaction with the increased infiltration of plasma cells, was observed as a pseudotumor-like gingival overgrowth and myofibroblastic or fibrohistiocytitc proliferation of stromal cells with a heavy collection of plasma cells. The levels of CD3, CD20, CD31, CD68, RANKL, cathepsin G, cathepsin K, lysozyme, TNF${\alpha}$, MMP-2, and MMP-9 in the 14 cases of gingival plasma cell granuloma with immunohistochemical detection were measured to determine the pathogenetic progresses of the plasma cell granuloma compared to the common pyogenic granuloma (n=45) in the gingiva. Results: The gingival lesions of the plasma cell granuloma could be divided into three histological types, plasma cell predominant type (PPT, n=8), mixed inflammatory cell type (MICT, n=2), and sclerosed fibrosis type (SFT, n=4). The PPT showed a condensed infiltration of plasma cells into the perivascular spaces of the granulomatous lesion with frequent formation of Russel's body in their cytoplasm. The MICT showed the concomitant infiltration of many macrophages together with plasma cells, resulting in the diffuse destruction of stromal fibrous tissue. The SFT showed granulomatous lesions replaced gradually by thick collagenous fibrous tissue, resembling an inflammatory pseudotumor. The SFT expressed strongly the lymphocytic markers, CD3 and CD20, and the macrophage/monocyte markers, CD31 and CD68, but showed reduced expression of common inflammatory markers, TNF${\alpha}$, cathepsin G, lysozyme, MMP-2, and MMP-9, as well as the reduced expression of osteoclastogenic markers, RANKL and cathepsin K. Conclusion: These results suggest that a gingival plasma cell granuloma shows variable gene expression for cell-mediated immunity and stromal tissue degeneration, undergoing sclerotic fibrosis with a persistent inflammatory reaction.

Plasma cell leukemia is a rarely occurring plasma cell disease which is classified to primary plasma cell leukemia and secondary plasma cell leukemia. The clinical manifestations and pathological features are similar to acute leukemia and the prognosis is poor with median survival rate of 4-6 months after diagnosis. When diagnosing plasma cell leukemia, it is often difficult to differentiate plasma cell leukemia from chronic lymphoid leukemia due to the similar cellular morphology of lymphocytes and plasma cells. We report a case of plasma cell leukemia with unusual morphological feature diagnosed by a cell surface marker study. A 81 year old man was presented with renal failure, anemia, thrombocytopenia and atypical lymphoid leukocytosis. Bone marrow examination and peripheral blood smear revealed a large number of pleomorphic cells with some grooving nuclei. Immunophenotype feature of these cells were suggestive of plasma cell origin with positivity for CD38. Furthermore an urine & serum protein electrophoresis showed monoclonal gammopathy of lambda light chain type. The disease initially responded to melphalan and prednisolone, but eventually relapsed.

Angioimmunoblastic T-cell lymphoma (AITL) is a lymphoproliferative disorder of mature T follicular helper cells. Atypical lymphoid cells were observed in the bone marrow of an 80-year-old woman, and the flow cytometric determined immunophenotypes of B-cells were unusual, that is, CD19+, CD20-, and CD22- with lambda light chain restriction. Initially, we suspected BM involvement of B-cell lymphoma based on the presence of abnormal B-cells. However, the patient was diagnosed with AITL involving BM. A re-analysis of flow cytometric immunophenotyping revealed a minor, aberrant T-cell population, and the lambda light chain restriction observed by surface staining was considered non-specific binding. This case demonstrates B-cells in patients with EBV-positive T-cell lymphoma may exhibit immunophenotypes resembling those of plasma cells, and that proliferation of abnormal B-cells or plasma cells could also potentially mask underlying T-cell lymphoma. A more integrated approach is required for accurate diagnosis.
