Association of Apolipoprotein E Polymorphisms with White Matter Lesions and Brain Atrophy

Objective Apolipoprotein E (ApoE) is mainly synthesized in the liver. So far, it is unknown the relationship among APOE gene polymorphisms and WML, brain atrophy. Therefore, the aim of the study was to assess the associations of APOE gene polymorphisms in patients with WML and brain atrophy. Methods A total of 58 patients with WML, 128 patients with brain atrophy, 112 patients with co-occurrence of WML and brain atrophy and 95 healthy elderly volunteers were recruited from Renmin Hospital of WuHan University. Results Allele E3 was the most common allele. The alleles E2 had significantly higher levels of ApoB and lower age in WML group. The alleles E2 was associated with the lower level of ApoB, LDL-Ch, TCh, and sdLDL in co-occurrence group. The E3/E3 genotype has higher level of sdLDL, but lower age and female frequency in WML. The E3/E4 genotype had higher level of TG, but lower age in WML. Gender, Age, E2, Hyperhomocysteinemia and UA were also significantly associated with disease progression. Conclusion This study found that clinical data, lipids and metabolic complications were closely related to ApoE genotypes and alleles, and also disease progression and type.

Ardipusilloside-I is a natural triterpenoid saponin, which was isolated from Ardisia pusilla A. DC. The aim of the study was to evaluate the stimulation of ardipusilloside-I on gastrointestinal motility in vitro and in vivo. The experiment of smooth muscle contraction directly monitored the contractions of the isolated jejunal segment (IJS) in different contractile states, and the effects of ardipusilloside-I on myosin were measured in the presence of Ca2+-calmodulin using the activities of 20 kDa myosin light chain (MLC20) phosphorylation and myosin Mg2+-ATPase. The effects of ardipusilloside-I on gastro emptying and intestinal transit in constipation-predominant rats were observed, and the MLCK expression in jejuna of constipated rats was determined by western blot. The results showed that, ardipusilloside-I increased the contractility of IJS in a dose-dependent manner and reversed the low contractile state (LCS) of IJS induced by low Ca2+, adrenaline, and atropine respectively. There were synergistic effects on contractivity of IJS between ardipusilloside-I and ACh, high Ca2+, and histamine, respectively. Ardipusilloside-I could stimulate the phosphorylation of MLC20 and Mg2+-ATPase activities of Ca2+- dependent phosphorylated myosin. Ardipusilloside-I also stimulated the gastric emptying and intestinal transit in normal and constipated rats in vivo, respectively, and increased the MLCK expression in the jejuna of constipation-predominant rats. Briefly, the findings demonstrated that ardipusilloside-I could effectively excite gastrointestinal motility in vitro and in vivo.

Acetylcholinesterase (AChE) is an importantenzyme in the pathogenesis of Alzheimer’s disease (AD). Comparative quantitative structure-activity relationship(QSAR) analyses on some huprines inhibitors againstAChE were carried out using comparative molecular fieldanalysis (CoMFA), comparative molecular similarityindices analysis (CoMSIA), and hologram QSAR(HQSAR) methods. Three highly predictive QSAR modelswere constructed successfully based on the training set. The CoMFA, CoMSIA, and HQSAR models have valuesof r2 = 0.988, q2 = 0.757, ONC = 6; r2 = 0.966, q2 =0.645, ONC = 5; and r2 = 0.957, q2 = 0.736, ONC = 6. The predictabilities were validated using an external testsets, and the predictive r2 values obtained by the threemodels were 0.984, 0.973, and 0.783, respectively. Theanalysis was performed by combining the CoMFA andCoMSIA field distributions with the active sites of theAChE to further understand the vital interactions betweenhuprines and the protease. On the basis of the QSAR study,14 new potent molecules have been designed and six ofthem are predicted to be more active than the best activecompound 24 described in the literature. The final QSARmodels could be helpful in design and development ofnovel active AChE inhibitors.

<P>Contemporary science is witnessing a rapid expansion of the two-dimensional (2D) materials family, each member possessing intriguing emergent properties of fundamental and practical importance. Using the particle-swarm optimization method in combination with first-principles density functional theory calculations, here we predict a new category of 2D monolayers named tellurene, composed of the metalloid element Te, with stable 1T-MoS2-like (alpha-Te), and metastable tetragonal (beta-Te) and 2H-MoS2-like (gamma-Te) structures. The underlying formation mechanism is inherently rooted in the multivalent nature of Te, with the central-layer Te behaving more metal-like (e.g., Mo), and the two outer layers more semiconductorlike (e.g., S). We also show that the alpha-Te phase can be spontaneously obtained from the magic thicknesses divisible by three layers truncated along the [ 001] direction of the trigonal structure of bulk Te, and both the alpha- and alpha-Te phases possess electron and hole mobilities much higher than MoS2. Furthermore, we present preliminary but convincing experimental evidence for the layering behavior of Te on HOPG substrates, and predict the importance of multivalency in the layering behavior of Se. These findings effectively extend the realm of 2D materials to group-VI elements.</P>

Reliability analysis techniques combining with various surrogate models have attracted increasing attention because of their accuracy and great efficiency. However, they primarily focus on the structures with continuous response, while very rare researches on the reliability analysis for structures with discontinuous response are carried out. Furthermore, existing adaptive reliability analysis methods based on importance sampling (IS) still have some intractable defects when dealing with small failure probability, and there is no related research on reliability analysis for structures involving discontinuous response and small failure probability. Therefore, this paper proposes a novel reliability analysis method called AGPC-IS for such structures, which combines adaptive Gaussian process classification (GPC) and adaptive-kernel-density-estimation-based IS. In AGPC-IS, an efficient adaptive strategy for design of experiments (DoE), taking into consideration the classification uncertainty, the sampling uniformity and the regional classification accuracy improvement, is developed with the purpose of improving the accuracy of Gaussian process classifier. The adaptive kernel density estimation is introduced for constructing the quasi-optimal density function of IS. In addition, a novel and more precise stopping criterion is also developed from the perspective of the stability of failure probability estimation. The efficiency, superiority and practicability of AGPC-IS are verified by three examples.
Architecture of LTE-WIFI Integration for SDN Towards IP Systems

Against the background of a surge in software-defined network (SDN)as well as urgent demands for LTE-WIFI internetworking and IPv6 network integration, the paper integrates LTE mobile telecommunication technology, wireless local area network and 5G (the fifth generation mobile communication system)into one, in an attempt to complete routing, distribution, and transmission of different radio frequency signals (i.e.LTE, WIFI) on the SDN basis. As a result, large-area coverage of LTE-WIFI network signals is realized in a way that meeting the demands of reliability, safety,flexibility and extendibility for mobile internet. What is more, different complementary wireless telecommunication systems are closed combined with each other. Through the use of SDN integration technology, the paper designs network integration architecture of IP-LTE mobile internet and WIFI, and also studies on the application of SDN-based IP optical integration network, which has broad prospects.

The spike gene of porcine epidemic diarrhea virus (PEDV) was sequenced from 55 South China field strains isolated from pigs with symptoms of diarrhea. The sequences were compared within the set of field strains as well as with reference strains available in GenBank. Within the 55 South China PEDV field strains, the deduced amino acid sequence identities ranged from 93.8% to 99.9 % and ranged from 90.7% to 99.5% when compared with the foreign reference strains in GenBank. Our phylogenetic analysis showed that 10 of the 55 South China PEDV strains belonged to G1b and 45 belonged to G2b.
