        HCV-Specific Interleukin-21+CD4+ T Cells Responses Associated with Viral Control through the Modulation of HCV-Specific CD8+ T Cells Function in Chronic Hepatitis C Patients

        Interleukin-21 (IL-21)+CD4+ T cells are involved in the immune response against hepatitis B virus (HBV) by secreting IL-21. However, the role of IL-21+CD4+ T cells in the immune response against chronic hepatitis C (CHC) virus infection is poorly understood. This study aimed to investigate the role of IL-21+CD4+ T cells in CHC patients and the potential mechanisms. The study subjects in-cluded nineteen CHC patients who were grouped by viral load (low, < 106 RNA copies/ml, n = 8; high, > 106 RNA copies/ml, n = 11). The peripheral frequency of HCV-specific IL-21+CD4+ T cells was higher in the low viral load group and was negatively correlated with the serum HCV RNA viral load in all CHC patients. Meanwhile, IL-21+ cells accumulated in the liver in the low viral load group. In vitro, IL-21 treatment increased the expression of proliferation markers and cytolytic molecules on HCV-specific CD8+ T cells. In summary, these findings suggest that HCV-specific IL-21+CD4+ T cells might contribute to HCV control by rescuing HCV-specific CD8+ T cells in CHC patients.

        Core prescription pattern of Chinese herbal medicine for depressive disorders in Taiwan: a nationwide population-based study

        Background: Depressive disorders (DD) affect not only mood and behavior but also various physical functions. Traditional Chinese medicine (TCM) has been shown to have some benefits in treating DD. However, one formula or one single herb might be not show high efficacy when used to treat depression. Thus, this study aimed to examine the core prescription pattern of Chinese herbal medicine (CHM) among patients with DD in Taiwan as a reference for related research and clinical applications. Methods: All patients, who had been diagnosed with major depressive disorder or minor depression or dysthymia without any other baseline diseases and had at least one CHM outpatient clinical visit from 2002 to 2011, were extracted from three randomly sampled cohorts, namely the 2000, 2005 and 2010 cohorts of the National Health Insurance Research Database (NHIRD) of Taiwan. The collected data was analyzed to explore the patterns of herbal products. Results: There were 197,146 patients with a diagnosis of DD and of these 1806 subjects had only a diagnosis of DD and utilized CHM. The most common formula was Gan-Mai-Da-Zao-Tang (12.19%), while Suan-Zao-Ren (3.99%) was the most commonly prescribed single herb. The core pattern of prescriptions consisted of a combination of Gan-Mai-Da-Zao-Tang, Jia-Wei-Xiao-Yao-San, Chai-Hu-Jia-Long-Gu-Mu-Li-Tang, He-Huan-Pi, Yuan-Zhi and Shi-Chang-Pu. Conclusions: This study describes the CHM core prescription pattern used to treat patients in Taiwan with DD and it is a potential candidate for study in future pharmacological or clinical trials targeting DD.

        Effect of Tea Brewing Containers of Different Materials on the Taste and Quality Components of Tieguanyin and Pu’er Tea

        Several previous works showed that the brewing temperature, time, ratio of tea and water had significant effects on the quality of different tea infusions. The appropriate tea brewing condition is vital for a cup of delicious tea. In China, the tea brewing container is also thought to be important for tea brewing. However, there are few works on the effect of tea brewing containers of different materials on tea infusions. In this work, Tieguanyin and Pu’er tea infusions were prepared with tea cups of glass, white porcelain, and purple clay, the effect of different container materials on the taste and quality components of Tieguanyin and Pu’er tea were studied. The tea cups made of glass, white porcelain, and purple clay in same style were used for brewing Tieguanyin and Pu’er tea to obtain different treated tea infusions. The main components such as tea polyphenols, amino acids, total sugars, water extracts, theabrownins, and caffeine in different treated tea infusions were analyzed. All tea infusions were carried out in sensory tests. Among three different cups, the cup made of purple clay had a higher ability in keeping temperature. The content of tea polyphenols and caffeine of Tieguanyin and Pu’er tea were significantly high infused with the purple clay cup. The amino acids and catechins in Tieguanyin, theabrownins in Pu’er tea, are positively associated with the heat property of tea cups. Tieguanyin tea brewing with glass cup and Pu’er tea brewing with purple clay cup are much more welcomed by panelists. In the present work, Tieguanyin tea prepared with glass cups and Pu’er tea prepared with purple clay cup are suggested for a better taste tea infusion.

        The miR-145-5p/CD36 pathway mediates PCB2-induced apoptosis in MCF-7 cells

        Background Procyanidin B2 (PCB2) can increase the levels of anti-infammatory and immune mediators. Objectives However, its molecular mechanism in human breast cancer remains unclear. This study aimed to investigate the antitumor efect of PCB2 on MCF-7 cells and to examine the underlying mechanism. Methods The fow cytometry and EdU incorporation assays were measured the PCB2-induced BMECs. The expression levels of infammatory factors and immune response genes were upregulated in MCF-7 cells, high-throughput sequencing was used to detect diferentially expressed genes in blank and PCB2-treated MCF-7 cells. Results The results showed that PCB2 induced the apoptosis of MCF-7 cells. CD36 profles were afected in MCF-7 cells. Additionally, prediction software identifed a miR-145-5p binding site in the CD36 sequence. Luciferase reporter assays and Western blot analysis were used to verify the regulatory relationships between the diferentially expressed miRNA miR145-5p and CD36. MiR-145-5p and its key target (CD36) constitute a potential miRNA-mRNA regulatory pair. Functional studies in MCF-7 cells revealed that CD36 promotes but miR-145-5p inhibits apoptosis. Conclusion Overall, these data suggest that miR-145-5p inhibits the enhancing efect of PCB2 on CD36 expression by binding CD36 and subsequently regulating apoptosis, the immune response and anti-infammatory pathways. These results provide theoretical and experimental support for the treatment of breast cancer.

        Stem Cell-Derived Exosomes: A New Method for Reversing Skin Aging

        Senescence is an inevitable natural life process that involves structural and functional degeneration of tissues and organs. Recently, the process of skin aging has attracted much attention. Determining a means to delay or even reverse skin aging has become a research hotspot in medical cosmetology and anti-aging. Dysfunction in the epidermis and fibroblasts and changes in the composition and content of the extracellular matrix are common pathophysiological manifestations of skin aging. Reactive oxygen species and matrix metalloproteinases play essential roles in this process. Stem cells are pluripotent cells that possess self-replication abilities and can differentiate into multiple functional cells under certain conditions. These cells also possess a strong ability to facilitate tissue repair and regeneration. Stem cell transplantation has the potential for application in anti-aging therapy. Increasing studies have demonstrated that stem cells perform functions through paracrine processes, particularly those involving exosomes. Exosomes are nano-vesicular substances secreted by stem cells that participate in cell-to-cell communication by transporting their contents into target cells. In this chapter, the biological characteristics of exosomes were reviewed, including their effects on extracellular matrix formation, epidermal cell function, fibroblast function and antioxidation. Exosomes derived from stem cells may provide a new means to reverse skin aging.

        On the Surface Tension and Coulomb Energy of Neutron-star Ma

        We briefly report the results from a study of the structure, the surface and the Coulomb energyof the core-crust transition layer of neutron stars with global charge neutrality. The results showthat the surface tension is a power-law function of the baryon number density in the core region. We also present analyses of the influence of the gravitational field on the transition layer andthe instability against Bohr-Wheeler surface deformations in the case of neutron stars obeyingglobal charge neutrality. If the core-crust transition is assumed to occur at the nuclear densitycore 2.7×1014 g cm−3, the instability sets the upper limit to the crust density, critcrust 1.2×1014g cm−3.

        Adipose-Derived Stem Cell Exosomes Promoted Hair Regeneration

        Background: Some scholars have found that dermal papilla spheroid–derived exosomes could promote the development of hair follicles. However, whether adipose-derived stem cell exosomes (ADSC-Exos) have a similar effect on hair growth has not been determined yet. Thus, the purpose of this article was to detect whether ADSC-Exos could promote hair regeneration. Methods: Adipose-derived stem cells (ADSCs) were isolated from 6-week-old C57BL/6 mice. Then, ADSC-Exos were isolated from the ADSCs. Western blotting was used to detect specific exosome markers. The particle size and distribution of the exosomes were analyzed by NanoSight dynamic light scattering. A total of 12 nude mice were randomly divided into two groups (n = 6 each): the ADSC-Exos group and the control group. For the control group, a mixture of freshly isolated dermal cells (DCs) and epidermal cells (ECs) was grafted. For the ADSC-Exos group, a mixture of DCs, ECs, and 50 μg/ml of ADSC-Exos was grafted. Gross evaluation of the hair regeneration was carried out 2–3 weeks after the transplantation, and the graft site was harvested for histology at the third week. Results: The existence of exosomes derived from ADSCs was evidenced by CD63, ALX1, and CD9 expression. Two or three weeks after the grafting, the number of regenerated hairs in the ADSC-Exos group was higher than that in the control group (p < 0.001). Histologically, the terminal hairs were remarkable in the ADSC-Exos group, whereas the hair follicles observed in the control group were comparatively immature. The ADSC-Exos group had a higher number of regenerated follicles than the control group (p < 0.001). In addition, we found that the skin tissues in the ADSC-Exos group had higher PDGF and vascular endothelial growth factor expressions and lower transforming growth factor beta 1 levels than those in the control group. Conclusion: Our results indicated that ADSC-Exos could promote in vivo hair follicle regeneration.       • Association of GSTP1 and RRM1 Polymorphisms with the Response and Toxicity of Gemcitabine-cisplatin Combination Chemotherapy in Chinese Patients with Non-small Cell Lung Cancer

        Background: Previous studies showed that genetic polymorphisms of glutathione S-transferase P1 (GSTP1) were involved in glutathione metabolism and genetic polymorphisms of ribonucleotide reductase (RRM1) were correlated with DNA synthesis. Here we explored the effects of these polymorphisms on the chemosensitivity and clinical outcome in Chinese non-small cell lung cancer (NSCLC) patients treated with gemcitabine-cisplatin regimens. Materials and Methods: DNA sequencing was used to evaluate genetic polymorphisms of GSTP1 Ile105Val and RRM1 C37A-T524C in 47 NSCLC patients treated with gemcitabine-cisplatin regimens. Clinical response was evaluated according to RECIST criteria after 2 cycles of chemotherapy and toxicity was assessed by 1979 WHO criteria (acute and subacute toxicity graduation criteria in chemotherapeutic agents). Results: There was no statistical significance between sensitive and non-sensitive groups regarding the genotype frequency distribution of GSTP1 Ile105Val polymorphism (p>0.05). But for RRM1 C37A-T524C genotype, sensitive group had higher proportion of high effective genotype than non-sensitive group (p=0.009). And according to the joint detection of GSTP1 Ile105Val and RRM1 C37A-T524C polymorphisms, the proportion of type A (A/A + high effective genotype) was significantly higher in sensitive group than in non-sensitive group (p=0.009). Toxicity showed no correlation with the genotypes between two groups (p>0.05). Conclusions: Compared with single detection of genetic polymorphisms of GSTP1 Ile105Val or RRM1 C37A-T524C, joint detection of both may be more helpful for patients with NSCLC to receive gemcitabine-cisplatin regimens as the first-line chemotherapy. Especially, genetic polymorphism of RRM1 is more likely to be used as an important biomarker to predict the response and toxicity of gemcitabine-cisplatin combination chemotherapy in NSCLC.

        Influence of Cu2S Precipitates Dissolution on Ferrite Grain Growth During Heat Treatment in the Non-Oriented Electrical Steel Sheet

        The factor to deduce grain growth of non-oriented electrical steel sheet during heat treatment was researched in thispaper. Scanning electron microscope equipped with electron backscatter diffraction (EBSD), X-ray diffractionand transmission electron microscope were used to characterize the microstructure, dislocation density and precipitate,respectively. The EBSD results indicated that the grain size increased from 18.7 μm to 56 μm after heattreatment. Meanwhile, the characterization of grain size distribution, dislocation density and precipitates revealedthat the dissolution of Cu2S precipitates, which act as inhibitor, may lead to the grain growth. In addition, the pinningforce of grain boundary and the driving force of grain growth were calculated. Based on experiments resultsand theoretical calculations, the Cu2S precipitates with diameter of less than 39 nm and volume fraction of 1.74 ×10-4would be sufficient to inhibit the ferrite grain growth. Heat treatment provides an efficient way to prompt thegrain growth.
