        M2-like macrophages transplantation protects against the doxorubicin-induced heart failure via mitochondrial transfer

        Aims: The alternatively activated macrophages have shown a cardioprotective effect in heart failure. However, the effect of M2 adoptive transfer in non-ischemic heart failure is unknown. In this study, we evaluated the efficacy of M-CSF plus IL-4 induced M2-like macrophages transplantation in doxorubicin-induced cardiotoxicity. Methods: Bone marrow mononuclear cells were polarized as CCR2+CD206+ M2-like macrophages by a combination of M-CSF plus IL-4 treatment. C57BL/6 mice received a single intraperitoneal njection of doxorubicin (15 mg/kg). The treatment group were treated with M2-like macrophages (1 × 10^6 cells per mouse; i.v.) once a week for 2 weeks. After 3 weeks, we examined the percentage of resident cells and cardiac function. Furthermore, we evaluated cardiac fibrosis, cardiomyocyte apoptosis and circulating inflammatory factors. Finally, we investigated the mitochondria transfer in vitro in a direct and indirect co-culture conditions. Results: Cardiac function was significantly improved in doxorubicin-induced heart failure by adoptive transfer of M2-like macrophages. Besides, M2-like macrophages treatment attenuated cardiac fibrosis and cardiomyocyte apoptosis, as well as increased the level of circulating IL-4 and Th2 response. In vitro, M2-like macrophages could transfer mitochondria to injured cardiomyocytes in a direct and indirect way. Conclusions: In our study, adoptive transfer of M2-like macrophages could protect against the doxorubicin-induced cardiotoxicity, which may be partly attributed to mitochondria transfer. And M2-like macrophages transplantation could become a treatment for non-ischemic heart failure in the clinical practice.

        A universal ZVS design for a family of quadratic boost converters

        In some application scenarios of new energy, high voltage gain DC–DC converters are widely employed especially for photovoltaic systems, fuel cell systems, and electric vehicles. The two-stage cascade boost integrated with a coupled-inductor and diode-capacitor voltage multiplier cell has been widely researched due to the advantages of a quadratic voltage gain and low voltage stress for the semiconductor devices. In this paper, the universal ZVS design for a family of quadratic converter is given. The proposed method maintains an extremely high voltage conversion ratio and low voltage stress on the power devices. In addition, the problem of the voltage gain being infl uenced by the load due to a small excitation inductance selection is solved. Moreover, ZVS operation for all of the switches and ZCS operation for most of the diodes are achieved to lower switching loss and suppress voltage spikes. The active clamp circuit is applied to lower the voltage stress and to control the ZVS operation. The design and operation principles, along with steady state and effi ciency analyses of a specifi c converter are discussed in detail. In addition, a detailed comparison is presented among some similar topologies. Finally, a 320 W prototype circuit is implemented in laboratory to verify the performance of the proposed converter.

        Nonlinear simulation of tunnel linings with a simplified numerical modelling

        A high-efficiency simplified modelling approach is proposed for investigating the nonlinear responses of reinforced concrete linings of shield tunnels. Material and geometric nonlinearities are considered in the analysis of the lining structures undergoing large deformation before ultimately losing the load-carrying capacity. A beam-spring element model is developed to capture the force-transfer mechanism between lining segments and radial joints. The developed model is validated by comparing analyzed results to experimental results of a single-ring lining structure under two loading conditions: the ground overloading and the lateral unloading respectively. The results show that the lining structure under the lateral unloading due to excavation on the both sides of the tunnel is more vulnerable compared to the case of ground overloading on the top of the tunnel. A parameter study is conducted and results indicate that the lateral pressure coefficient has the greatest influence on the behaviour of the lining structure.

        A study of synergetic carrier emulsion liquid membrane for the extraction of amoxicillin from aqueous phase using response surface methodology

        Amoxicillin (AMX) is the most commonly used antibiotic in life. AMX was extracted from aqueoussolution by synergetic carrier emulsion liquid membrane (ELM) technology for thefirst time. Themembrane phase of ELM was mainly composed of kerosene and butyl acetate. The carrier has consisted ofAliquat-336 and TOA, the surfactant was Span-80, and the membrane enhancer was liquid paraffin. AndNa2CO3 and NaCl were used as the internal aqueous phase for extraction. Firstly, the basic structure andmechanism of emulsion were characterized and analyzed. And then, the center composite design (CCD)of response surface methodology (RSM) was used to optimize the operating conditions. The optimalvalues were: Aliquat-336 concentration (1.1% v/v), trioctylamine (TOA) concentration (8.6% v/v), andextraction time (15 min), AMX concentration (75 mg/L). A polynomial model wasfitted to predict theextraction yield of AMX. Under the optimal operating conditions, the optimal extraction prediction valuewas 98.58%, the extraction actual value was 98.2%, and the recovery rate of AMX was 75.3%.
