漢方藥의 成分 및 藥效에 關한 硏究(第2報) : Berberine 骨格變換에 對한 硏究 Studies on the Variation of Skeletons of Berberine

Previously we reported the simple photochemical valance automerization of berberinephenolbetaine derived from berbine in to the 8-methyl-8,14-cycloberbine. Treatmetn of the 8-methyl-8,14-cycloberbine with 10% acetone and Borontrifluorideethylether at room temperature for 3hr. afforded 8-hydro-8-methylspirobenzylisoquinoline(Ⅰ) Reduction of the substance (Ⅰ) with sodiumborohydride was converted easily to the 8,13-dihydroxy-8-menthyl-spirobenzylisoquinoline (Ⅱ) in good yields. On acidic treatment the 8-methyl-8,14-cycloberberne with thiophenol was afforded also 8-methyl-8-thiophenylspirobenzylisoquinoline (Ⅲ). The substance (Ⅲ) was strirred in methanol with sodiumborohydride at room temperature for 4hr. to give the 13-hydroxy-8-methyl-8-thiophenylspirobenzylisoquineoline (Ⅳ).
Berberinephenolbetaine 誘導體合成 및 活性硏究[Ⅲ]

Nucleophilic additon of allyl magnesium bromide to berberine chloride resulted in the 8-allyl-9, 10-dimethoxy-2, 3-methylenedioxy-13, 13a-dehydroberbine[Ⅰ]. And oxidation of [Ⅰ] with m-chloroperbenzoic acid in a stream of Argon at-20∼-30℃ afforded the 8-allylberberinephenolbetaine [Ⅱ], and cycloaddition of [Ⅱ] with dialkylacetylenedicarboxylate(R; a=CH_3, b=C_2H_5) in THF gave 8-allyl-9, 10-dimethoxy-2, 3-ethylendioxy-13-oxo-8, 14-ethenodimethyldicarboxyberbine[Ⅲ a] and 8-allyl-9, 10-dimethoxy-2, 3-methylenedioxy-13-oxo-8, 14-ethenodiethyl-dicarboxy-berbine[Ⅲb]. Heating of 8, 14-ethenoberbines[Ⅲa and Ⅲb] in ethanol effected smooth isomerization leading to the 3, 4-[benzo(3",4"-dimethoxy)]-1, 8-[dihydre(2',3'-methylenedioxy) naphthalene]-5-allyl-6, 7-dimethylcarboxy-2-oxo-azocine[Ⅳa] and 3, 4-[benzo(3", 4"-dimethoxy)]-1, 8-dihydro(2', 3'-methylenedioxy)naphthalene]-5-allyl-6, 7-diethylcarboxy-2-oxo-azocine[Ⅳb]. Irradiation of berberine chloride in methanol containing rose bengal and sodium methoxide with O_2 stream afforded the 8, 9, 10, 14-tetramethoxy-2, 3-methylenedioxy-13-oxoberberine[Ⅳ], and recrystallization of [Ⅳ] from methanol gave 8-methoxyberberinephenolbetaine[Ⅶ]. And cycloadditon of [Ⅶ] with dimethyl-acetylenedicarboxylate gave 8, 9, 10-trimethoxy-2, 3-methylene-dioxy-13-oxo-8, 14-ethenodimethyldicarboxyberbine[Ⅷ], and heating of [Ⅷ] in ethanol effected smooth isomerization leading to 3, 4-[benzo(3" 4"-dimethoxy)]-1, 8-[dihydro(2',3'-methylene-dioxy) naphthalene]-5-methoxy-6, 7-dimethylcarboxy-2-oxo-azocine[Ⅸ]. The in vitro biological evaluation showed that compounds[Ⅷ] and [Ⅸ] had antibacterial effect against Pseudomonas aeruginosa, and compounds [Ⅰ], [Ⅱ], [Ⅲb] and [Ⅶ] completely had anticancer activity to P-388 mouse lymphocyle leukemia at concentration of 100㎍, and [Ⅰ] almost had anticancer activity at concentration of 20㎍.
수종의 새로운 N-치환 Aminocyclohexanone 유도체 합성 연구

As the starting compound for synthesis of morphane derivatives, several new com-pounds, P-acetaminocyclohexanone, N-acetyl-4-ethylaminocyclohexanone, N-benzoyl-4-methylaminocycloheanone and P-phthalimidocyclohexanone are synthesized in this paper. The method^∽ of synthesis for the above compounds are indicated at Figure I in the original paper. It will be studied in future on synthesis of new morphane derivatives which will be studied as analgic action of central nerve system respectively.

Methyl 3-[(phenylsulfonyl)methyl]-1,4,8-trimethoxy-2-naphthoate(7) preparad by 3 steps was condensed with 5-ethyl-2-cyclohexen-1-one(8) to afford the teracyclic aromatic ring system compound 10, which was stepwise converted into the precursor of 11-Deoxy-β-rhodomycinone by means of various functional groups transformation.

We have described the synthesis and biological activity of novel retinamide . The retinamide derivatives were synthesized by introducing functional side chains into the 4-hydroxy group of 4-HPR. The activities could be dependent on the side chain length, functional group, and hetero atom. The antiprolife-rative potential of the derivatives was assessed by MTT assay in HCT116 cancer cell lines.

Data of IR spectra on a component medicine have been reported, and many drugs were frequently identified by this simple method. However, for the determination of pharmaceutical preparations this method has not been fully employed. Thus, authors report in this paper the data from the determination on bulk powder of antipyretic analgestics among the Korean pharmacopoea II. The research on the qualitive methods by IR spectra are on progress.
열분해 온도가 음식폐기물 바이오차의 이화학적 특성에 미치는 영향

현재 우리나라 음식 폐기물은 15,000톤/일 가량이 발생하고 있으며 처리 비용만 연간 1조원 이상이 소요되는 것으로 알려져 있다. 기존의 처리방식인 직매립 및 해양투기는 법 개정과 런던협약에 의해 금지되었고, 이에 따라 음식 폐기물의 자원화 기술 개발의 필요성이 대두되었다. 현재 퇴비화, 사료화 등을 통한 자원화를 진행하고 있으나 식물의 염류 장해, 가축 질병 발생 및 긴 처리시간 소요 등의 문제점이 잇따라 발생하며 보다 실용적이고 효율적인 방안이 필요한 실정이다. 이에 본 연구는 열분해를 통해 음식 폐기물 바이오차를 제조한 후, 열분해 온도에 따른 이화학적 특성을 분석하여 자원으로써 활용 가능성을 평가하였다. 음식 폐기물(이하 FW) 일정량을 각각 350℃, 500℃ 및 700℃(이하 FWB350, FWB500, FWB700)에서 4시간 동안 열분해 하여 바이오차를 제조하였고, 각각의 이화학적 특성을 분석하였다. 분석 결과 유기물 함량은 40.85%(FWB700)<46.83%(FWB500)<72.26%(FWB350)<82.69%(FW)로 나타났으며, pH는 5.11(FW)<6.94(FWB350)<10.14 (FWB500)<11.74(FWB700)로 나타났다. 무기성분 함량은 FW에서 T-N(2.58%), P(0.9%), K(0.8%), Ca (3.66%), Mg(0.16%), Na(1.44%)로 나타났으며, FWB에서는 T-N(2.22-2.89%), P(1.78-3.28%), K(1.63- 2.75%), Ca(8.02-13.7%), Mg(0.31-0.56%), Na(2.27-4.26%)로 열분해 온도가 높아질수록 함량이 더 높아지는 경향을 나타냈다. 유해 중금속인 As, Cd, Pb, Hg는 미검출 되었다. FT-IR 분석결과 FW에서는 보이지 않던 C-Cl 결합이 FWB에서 565cm<sup>-1</sup>과 605cm<sup>-1</sup>에서 나타났다. FW, FWB350에서는 관찰되던 2923cm<sup>-1</sup>의 NH<sub>4</sub><sup>+</sup> 작용기는 FWB500, FWB700에서는 사라졌다. 1040cm<sup>-1</sup>에서 C-OH결합, 1634cm<sup>-1</sup>에서 C=C 결합, 3433cm<sup>-1</sup>에서 OH작용기는 공통적으로 나타났으며 온도가 높아질수록 강도가 세지는 경향을 나타냈다. 위의 결과를 토대로 식물 생장에 필수적인 무기성분을 다량 함유하고 있는 양질의 FWB는 식물 생육 증진 및 토양 개량제로서의 잠재성을 가지고 있다고 평가될 수 있다.
