        ZNF424, a novel human KRAB/C2H2 zinc finger protein, suppresses NFAT and p21 pathway

        Zinc finger-containing transcription factors are the largest single family of transcriptional regulators in mammals, which play an essential role in cell differentiation, cell proliferation, apoptosis, and neoplastic transformation. Here we have cloned a novel KRAB-related zinc finger gene, ZNF424, encoding a protein of 555aa. ZNF424 gene consisted of 4 exons and 3 introns, and mapped to chromosome 19p13.3. ZNF424 gene was ubiquitously expressed in human embryo tissues by Northern blot analysis. ZNF424 is conserved across species in evolution. Using a GFP-labeled ZNF424 protein, we demonstrate that ZNF424 localizes mostly in the nucleus. Transcriptional activity assays shows ZNF424 suppresses transcriptional activity of L8G5-luciferase. Overexpression of ZNF424 in HEK- 293 cells inhibited the transcriptional activity of NFAT and p21, which may be silenced by siRNA. The results suggest that ZNF424 protein may act as a transcriptional repressor that suppresses NFAT and p21 pathway to mediate cellular functions. [BMB reports 2010; 43(3): 212-218]

      • MiR-675 Promotes the Growth of Hepatocellular Carcinoma Cells Through Cdc25A Pathway

        Background: MicroRNAs (miRNAs) have fundamental roles in tumorigenesis. MiR-675 is upregulated in hepatocellular carcinoma(HCC) cells. However, the roles of miR-675 in hepatocellular carcinogenesis are still not fully elucidated. In this study, we focus on investigating the effect and mechanism of miR-675 in proliferation of HCC cells. Materials and Methods: The cell proliferation was measured by MTT assays after transfection with miR-675 inhibitor and miR-675 mimics in HCC cells. The expression level of miR-675 was detected by real-time quantitative reverse transcription polymerase chain reaction. Protein expression of Cdc25A was measured by western blotting analysis. Results: In MTT assays, overexpression of miR-675 promoted the proliferation of HCC cells(P<0.05. at 48 hours, P<0.01. at 72 hours) compared with the miR-675mimics control group. Downexpression of miR-675 inhibited the proliferation of HCC cells(P<0.05. at 48 hours, P<0.01. at 72 hours) compared with the miR-675inhibitor control group. In western blotting analysis, the expression level of Cdc25A was significantly increased (p<0.05) after treatment with miR-675 mimics. The expression level of Cdc25A was significantly decreased (p<0.05) after treatment with miR-675 inhibitor. Conclusions: Our results indicate that miR-675 promotes the proliferation in human hepatocellular carcinoma cells by associating with Cdc25A signaling pathway.

        Maximum Likelihood and Signal-Selective TDOA Estimation for Noncircular Signals

        This paper addresses the issue of time-difference-ofarrival (TDOA) estimation for complex noncircular signals. First,under the wide-sense stationary assumption, we derive the maximum likelihood (ML) estimator and the Cramer-Rao lower bound for Gaussian noncircular signals in Gaussian circular noise. The ML estimator uses the second-order statistics information of a noncircular signal more comprehensively when compared with the cross-correlation (CC) and the conjugate CC estimators. Further,we present a scheme to modify the traditional signal-selective TDOA methods for noncircular signals on the basis of the cyclostationarity of man-made signals. This scheme simultaneously exploits the information contained in both the cyclic cross-correlation (CCC) and the conjugate CCC of a noncircular signal.

        Mechanical and Thermal Properties Enhancement and Swelling Behavior of Bacterial Cellulose/Collagen/Polyvinyl Alcohol Nanofiber Hydrogel Film

        Composite hydrogel films of bacterial cellulose/collagen/polyvinyl alcohol were prepared by soaking methodusing bacterial cellulose hydrogel (BC) was as the base material, collagen (COL) and polyvinyl alcohol (PVA) wereintroduced as the reinforcement material to further improve the tensile strength especially elongation at break. Morphology,mechanical property, and thermal stability of composite hydrogel films were also investigated. The swelling behavior wasstudied using Schott’s second-order swelling model in detail. Research results show that BC/PVA/COL composite showed181.9 % improvement in elongation at break to that of BC. It further improved the tensile strength of the obtained BC/PVA/COL composite film by 12.9 % after crosslinking treatment. Importantly, further investigation demonstrated that thestructural stability and thermal stability of the composite hydrogel films were remarkably enhanced. Swelling behavior of BCand the composite nanofiber hydrogel films in DI water followed the Schott’s second-order swelling model, the swelling ofcomposites significantly reduced after crosslinking treatment.

        중국 네트워크 마케팅 산업에서 제품 애용자가 독립사업자로 활동하려는 의도에 관한 연구: 확장된 계획 행동이론을 중심으로*

        Purpose – This study aims to investigate the motivations of product enthusiasts in China's network marketing industry to engage in independent business activities, using an extended Theory of Planned Behavior as the theoretical framework. Additionally, it explores the moderating role of prior knowledge of network marketing. Design/Methodology/Approach – By integrating perception as a variable, this research assesses its influence on the entrepreneurial intentions of product loyalists within China's network marketing sector. Furthermore, the study examines how prior knowledge of network marketing affects the relationships among attitude, subjective norms, perceived behavioral control, perception of network marketing, and intent. Findings – The results indicate that variables derived from the extended Theory of Planned Behavior, including attitude, subjective norms, perceived behavioral control, and awareness, positively impact individual intentions to become independent network marketers. Notably, perceived control and awareness also influence actual behavior. Moreover, prior knowledge serves as a moderator for all pathways, except in the relationship between perceived control, awareness, and intention. Research Implications – Network marketing companies should prioritize maintaining high product quality to establish consumer trust. Engaging in corporate social responsibility initiatives can enhance company image. Consistent education for consumers about network marketing can help foster entrepreneurial intentions among potential independent business owners.

        Human Leptin Protein Induces Proliferation of A549 Cells via Inhibition of PKR-Like ER Kinase and Activating Transcription Factor-6 Mediated Apoptosis

        Purpose: To investigate the anti-apoptotic mechanism of leptin in non-small cell lung cancer. Materials and Methods: The influences of leptin on apoptosis were investigated,analyzing the mechanism that triggers growth of A549 cells. The effects of leptin on cell proliferation were examined by XTT analysis. Leptin, C/EBP homologous protein (CHOP), phosphorylated-PKR-like ER kinase (p-Perk), inositol requiring proteins-1, spliced X-box transcription factor-1 (XBP1), cleaved activating transcription factor-6 (ATF6), eukaryotic translation initiation factor-2α, caspase-12 and CHOP protein were detected in four groups by western blot, and endoplasmic reticulum (ER) stress related mRNA were detected by reverse transcription PCR. Results: The expression of leptin in A549 and leptin transfected cells inhibited cisplatin activated ER stress-associated mRNA transcription and protein activation. Two ER stress unfolded protein response pathways, PERK and ATF6, were involved, and XBP1 and tumor necrosis factor receptor-associated factor 2 (TRAF2) were increased significantly when treated with cisplatin in A549-siRNA against leptin cells. Furthermore, CHOP expression was inhibited upon leptin expression in A549, LPT-PeP and LPT-EX cells. Conclusion: Leptin serves as an important factor that promotes the growth of A549 cells through blocking ER stress-mediated pathways. This blocking is triggered by p-Perk and ATF6 via inhibition of CHOP expression.

      • A numerical framework of the phenomenological plasticity and fracture model for structural steels under monotonic loading

        In this study, the classical J2 flow theory is explicitly proved to be inappropriate to describe the plastic behaviour of structural steels under different stress states according to the reported test results. A numerical framework of the characterization of the strain hardening and ductile fracture initiation involving the effect of stress states, i.e., stress triaxiality and Lode angle parameter, is proposed based on the mechanical response of structural steels under monotonic loading. Both effects on strain hardening are determined by correction functions, which are implemented as different modules in the numerical framework. Thus, other users can easily modify them according to their test results. Besides, the ductile fracture initiation is determined by a fracture locus in the space of stress triaxiality, Lode angle parameter, and fracture strain. The numerical implementation of the proposed model and the corresponding code are provided in this paper, which are also available on GitHub. The validity of the numerical procedure is examined through single element tests and the accuracy of the proposed model is verified by existing test results.

        Effects of traffic characteristics on pavement responses at the road intersection

        Compared with pavement structures of ordinary road sections, pavement structures in the intersection are exposed to more complex traffic characteristics which may exacerbates pavement distresses such as fatigue-cracking, shoving, shear deformation and rutting. Based on a field survey about traffic characteristics in the intersection conducted in Shanghai China, a three dimensional dynamic finite-element model was developed for evaluating the mechanistic responses in the pavement structures under different traffic characteristics, namely uniform speed, acceleration and deceleration. The results from this study indicated that:(1) traffic characteristics have significant effects on the distributions of the maximum principal strain (MPS) and the maximum shear stress (MSS) at the pavement surface; (2) vehicle acceleration or deceleration substantially impact the MPS and MSS at pavement surface and could increase the magnitude of them by 20 percent to 260 percent; (3) in the vertical direction, with the increase of vehicle deceleration rate, the location of the MPS peak value and the MSS peak value changes from the sub-surface layer to the pavement surface.
