        Pulmonary inflammation caused by silica dioxide nanoparticles in mice via TXNIP/NLRP3 signaling pathway

        Background Silica dioxide nanoparticles (SiONPs) have been used for various medical applications, including therapeutics and imaging, and the use of SiONPs has increased gradually over the years. However, despite an increase in the use of SiONPs, not much is known about mechanism of action of SiONPs and their pulmonary toxicity. Objective The present study investigated the pulmonary toxicity of SiONPs and explored the underlying mechanism of action, primarily focusing on thioredoxin-interacting protein (TXNIP)/NOD-like receptor pyrin domain-containing 3 (NLRP3) in SiONPs-treated mice. We investigated the toxic effects of SiONPs in the lung of BALB/c mice administered 5, 10, and 20 mg/kg SiONPs for 3 days. Results Exposure to SiONPs markedly increased inflammatory cell counts, including those of neutrophils and macrophages, and levels of inflammatory mediators, such as interleukin (IL)-1β, IL-6, and tumor necrosis factor-α in a dose-dependent manner in the bronchoalveolar lavage fluid. Moreover, the inflammation was verified upon histopathological analysis. In addition, exposure to SiONPs increased the expression of TXNIP in a dose-dependent manner and, in turn, upregulated NLRP3 inflammasome proteins, which subsequently induced IL-1β production. Conclusion Collectively, exposure to SiONPs induced inflammation in the lungs of mice, which resulted in the activation of IL-1β production via the TXNIP-NLRP3 axis. Our results provide useful information on the pulmonary toxicity induced by SiONPs and provide insights into the underlying mechanism of action.

      • 랫드에서 신우황청심원의 급성 및 아급성독성시험

        The acute and subacute toxicity of New Woohwangchungsimwon(NWCH) which was used l-muscone as substitutive material of musk were investigated in S.D. rats. In intraperitoneal acute toxicity test, rats(Sprague-Dawley, SPF) were injected intraperitoneally with dosages of 0, 540, 750, 1,070, 1,500 and 3,000 ㎎/㎏. Body weights were significantly decreased at 540 ㎎/㎏ dose group in both sexes and abnormal autopsy findings were founded in both sexes at all treated groups. Intraperitoneal LD_50 of NWCH was 812.3 ㎎/㎏ in male and 872.3 ㎎/㎏ in female rats. In the subacute toxicity study, NWCH was administrated orally to both sexes of rats for 4 weeks as several doses(0, 320, 800 and 2,000 ㎎/㎏). There were neither dead animals nor significant changes of body weights during the experimental period. In addition, no differences were found between control and treated groups in clinical signs, urinalysis, hematology, serum biochemical analysis, and other findings. Above data strongly suggest that no observed adverse effect level of NWCH might be over 2,000 ㎎/㎏/day in this study.

        Teaching materials available for the effective performance of laboratory exp-erimental work in middle school were developed: Physics part ; A measuring apparatus of uniformly accelerated motion and normal force of inclined plane was developed. Chemistry part ; An imploved Eudiometer applicable in measuring reaction mole ratio and usful for a gas generating apparatus was developed. Biology part ; convenient teaching materials observing law of dominance, law of segregation, law of independence, incomplete dominance and dihybrid cross were developed. Earth science part ; A laboratory teaching materials observing formation of wave, current and graded bedding, and identification of sedimentary rocks were developed. Putting the above results of study together, the integrated suggestions were made to the effective development of science education in middle school : 1. The contents of science textbook should be simplified to the direction of the understanding of essential principles and concept. 2. The development and dirtrihbution of various supplementary teaching materials (film, T.P., cassettes film, supplementary textbook, program teaching materials individual learning materials) are required. 3. The experimental laboratory should be equipped with common instruments and equipements. 4. Efforts and time are needed to science teachers, and so remission of sundry duties and arrangement of assistant are hopefully considered. 5. Teacher's research activities should be encouraged by administrative route or relevant institute.

      • 랫드에서 l-muscone의 급성독성 및 아급성독성시험 연구

        l-Muscone is synthesized for use as substitutive material of musk which is the active ingredient of woohwangchungsimwon. The objective of this investigation was to evaluate the acute and subacute toxicity of l-muscone in rats. In oral acute toxicity test, SPF Sprague-Dawley male and female rats were gavaged with l-muscone of two doses(0, 5.0g/㎏). No dead animal and abnormal autopsy findings were found in control and treated group. Body weights were slightly decreased in both sexes of rats treated with 5.0g/㎏. Therefore, oral LD_50 of l-muscone was consider to be higher than 5.0g/㎏ in male and female rats. In intraperitoneal acute toxicity test, rats were injected intraperitoneally with dosages of 0, 1,000, 1,316, 1,732, 2,279 and 3,000 ㎎/㎏. Decreased body weights and motor activities were observed at high dose group. Intraperitoneal LD_50 of l-muscone were 1,920 ㎎/㎏ in male and female rats. In the subacute study. l-muscone was administrated orally to both sexes of rats for 4 weeks as several doses(0, 10, 100 and 1,000 ㎎/㎏). There were neither dead animals nor significant changes of body weights during the experimental period. In addition, no differences were found between control and treated groups in clinical signs, urinalysis, hematology, serum biochemical analysis, and other findings. Above data suggest that no observed adverse effect level of l-muscone in rats might be over 1,000 ㎎/㎏/day in this study.

      • 운동전략이 기립자세의 기능적 전방 팔뻗기에 미치는 영향

        The purpose of this study is to determine whether movement strategies affect functional forward reach distance in a standing position. Forty-seven healthy subjects were selected for this study: 23 men and 24 women, with an average age of 22.3. Functional forward reach distances were measured as hip strategy and squat strategy(included knee and ankle movement strategy) in a standing position, respectively. The mean values of functional forward reach distance in hip strategy, squat strategy were 33.57㎝, 29.48㎝, respectively. There was significantly difference in functional forward reach distance between hip strategy and squat strategy(p<.001). There was no difference of functional forward reach distance between male and female in hip strategy, but there was significant difference in other strategy(p<.05). These results suggest that movement strategies should be considered during functional forward reach test in standing. Further study is required to determine whether movement strategies affect functional reach distance in elderly and disabled groups.

We present the overview of PON structure and its current technology status which is one of the essentials for optical access network. For the commercial development, the technology standardization and industrial activity on Ethernet PON are investigated.

      • 조혈모세포이식 후 골성장인자의 변화 및 골대사에 미치는 영향 : Impact on Bone Mineral Metabolism

        연구배경: 각종 장기이식의 시행이 많아지고 이식 후 생존율이 증가함에 따라 이식 후 합병증에 대한 관심 또한 높아지고 있다. 조혈모세포이식 후에도 다양한 내분비적 합병증이 발생할 수 있으며 골격에 대한 합병증도 문제점으로 대두되고 있다. 조혈모세포 이식 후 발생하는 골소실에는 이식 후 초기의 골형성 저하와 골흡수 증가가 중요한 역할을 담당하리라고 추측되는데 이러한 골재형성불일치(biochemical uncoupling)에 골 성장인자들이 미치는 영향에 대해서는 알려진 바가 없다. 본 연구에서는 조혈모세포이식 전, 후로 말초 혈액에서 IGF-I, FGF-2, M-CSF같은 성장인자의 변화를 알아보고, 이들 성장인자의 변화가 조혈모세포이식 후의 골형과 골흡수에 미치는 영향 및 이식 후 발생되는 골량 소실과의 연관성을 확인해 보고자 하였다. 방법: 여러 가지 혈액질환으로 인해 동종 골수이식을 시행 받은 환자들을 전향적으로 관찰하였으며 이식 전 및 이식 후 1주, 2주, 3주, 4주 및 3개월, 6개월 1년에 말초 혈액에서 골교체표지자를 측정하였다. 이식 전 및 이식1년 후에 요추골 및 대퇴골 골밀도를 측정할 수 있었던 36명의 환자들을 대상으로 냉동 보관되어 있던 혈청을 이용하여 IGF-I, FGF-2 및 M-CSF를 시기별로 측정하였으며 이들 성장인자와 골교체표지자의 변화 및 골밀도 변화 사이의 상관관계를 확인하였다. 결과: 골흡수 표지자인 혈청 ICTP는 이식 전에 비해 이식 후 4주까지 점차 의의 있게 증가하다가 이후 6개월까지 더욱 증가한 후 감소하였다. 골형성 표지자인 osteocalcin은 이식 후 3주까지는 점차 감소하다가 이후 증가하여 이식 후 3개월 및 6개월에 기저치보다 통계적으로 유의하게 증가한 후 감소하였다. 혈청 IGF-I과 FGF-2는 각각 이식 후 3주 및 1주까지 의미있게 감소하다가 이후 증가하였으며 혈청 M-CSF는 이식 후 1주째에 기저치에 비해 의미 있게 증가하였다가 이후 기저치로 회복되었다. 이식 1년 후 평균 요추부 골밀도는 5.2% 감소하였고 평균 근위대퇴골 골밀도는 11.6% 감소하였다. 이식 전 및 이식 후 3주에 측정한 IGF-I과 같은 시기에 측정한 오스테오칼신 사이에 유의한 상관관계가 관찰되었으며 이식 후 3주째의 M-CSF와 골흡수표지자인 M-CSF 사이에서 의미 있는 양의 상관관계를 관찰할 수 있었다. 이식 후 3주 및 3개월에 IGF-I이 낮은 환자일수록 이식 1년 후 근위대퇴골에서의 골소실이 많은 것으로 분석되었다. 결론: 조혈모세포이식 후 발생하는 골소실에는 기존에 알려진 기저질환의 영향, 성호르몬의 감소, 면역억제의 투여, 골수기질세포와 조골세포의 손상 및 이식초기 사이토카인의 변화이외에도 골성장인자가 관련이 있음을 확인하였고, 이는 이식 후 발생되는 골량소실에 중요한 역할을 할 것이라고 사료된다. Background: A loss of bone mass is usually detected after a bone marrow transplantation (BMT), especially during the early post-transplant period. We recently reported that enhanced bone resorption following a BMT was related to both the steroid dose and the increase in IL-6. We also suggested damage to the marrow stromal microenvironment, by myoablation, partly explains the impaired bone formation following a BMT. It is well known that some growth factor play important role in bone growth and osteogenesis. However, the pathogenetic role of bone growth factors in post-BMT bone loss is unknown and data on the changes in the growth factors, in accordance with bone turnover markers and bone mineral density (BMD) changes are scarce. We investigated changes in bone growth factors such as IGF-I (Insulin-like growth factor-I), fibroblast growth factor-2 (FGF-2) and Macrophage colony stimulating factor (M-CSF), during the post-BMT period, and assessed whether the growth factor changes influenced the bone turnover and post-BMT bone loss. The present study is the first prospective study to describe the changes in bone growth factors following a BMT. Methods: We prospectively investigated 110 patients undergoing a BMT, and analyzed 36 patients (32.4±1.3 years, 17 men and 19 women) whose BMDs were measured before, and 1 year after, the BMT. The serum biochemical markers of bone turnover were measured before, 1, 2, 3 and 4 weeks, 3 and 6 months, and 1 year, after the BMT. The serum, FGF-2, IGF-I and M-CSF levels were measured before and 1 and 3 weeks, and 3 months after the BMT. The correlation between the changes of growth factors and various bone parameters was analyzed. Results: The mean bone losses in the lumbar spine and total proximal femur, calculated as the percentage change from the baseline to the level at 1 year, were 5.2(p<0.05) and 11.6%(p<0.01), respectively. the serum type I carboxyterminal telopeptide(ICTP), a bone resorption marker, increased progressively until 6 months after the BMT, but thereafter decreased, to the base value after 1 year. Serum osteocalcin, a bone formation marker, decreased progressively, until 3 weeks after the BMT but then increased transiently, and finally returned to the base level at 1 year. The serum IGF-I and FGF-2 also decreased progressively until 3 weeks 1 week after the BMT, respectively, then increased to the base values at 3 months. The serum M-CSF increased briskly at 1 week post-BMT, then decreased to the base level. There were positive correlations between the percentage changes from the baseline proximal femur BMD and the IGF-I levels 3 weeks and 3 months (r=0.52, p<0.01, r=0.41, p<0.05) post BMT. A significant correlation was found between the IGF-I and osteocalcin levels pre-BMT, and 3 weeks after the BMT. Another positive correlation was found between the M-CSF and the ICTP levels at 3 weeks post BMT (r=0.54, p<0.05). Conclusion: In conclusion, there were significant changes in the serum IGF-I, FGF-2 and M-CSF levels in the immediate post-BMT period, which were related to a decrease in bone formation and loss in the proximal femoral BMD during the year following the BMT (J Kor Soc Endocrinol 17:664∼674, 2002).

        도시 거주 노인 수면 습관 및 불면증상의 요인 : 인구학적,질병 및 정신증상의 관련성

        연구목적 : 본 연구는 국내 노인의 습관을 조사하고 불면 증상과 연관 있는 인구학적, 질병학적 요인을 알고자 했다. 방 법 : 1997년 12월부터 1998년 1월까지 약 1개월에 걸쳐 2단계 층화집락표본추출의 역학조사법을 통해 선정된 3개 4개동 거주의 만 65세 이상 노인을 대상으로 수행되었다. 대상자 중 723명에서 등답을 받았으며 응답률은 73.8였다. 대학생인 설문조사요원들을 훈련시킨 뒤, 이들로 하여금 대상지역의 노인거주가정을 방문하여 개개 노인을 대상으로 반구조화된 면담으로 수면습관 및 불면증상 설문을 실시하였으며 그 외 한국어판 노인정신장애평가척도, 삶의 질 척도를 이용한 설문조사와 과거 질병력을 청취조사하게 하였다. 결 과 : 국내 노인들은 평균 오후 10시 16분에 자리에 누워 오전 5시22분에 일어나서 총 7시간 6분의 수면시간을 보였다. 노인의 11.7%가 거의 매일 밤 불면증을 14.8%가 간헐적 불면증을 호소하였다. 남자에서는 연령이 증가할수록 침상에 누워있는 시간이 증가하고, 또, 입면시까지 소요되는 시간과 수면 중의 각성회수가 증가되었으나 여성에서는 명확하지 않았다. 단변수 분석에서 요통, 중풍 병력의 환자에서 불면증상이 심했다. 다변량분석 결과 불면 증상과 관련 깊은 요인으로 남자에게서는 고령과 우울증상이 여자에게서는 남자의 위험요인에다 신체통증과 피부병력이 추가로 관련이 있었다. 결 론 : 지역 거주 노인에서 불면 증상은 매우 흔한 증상이며 우울증상과 명확한 상관관계를 보이고 있었다. 불면증을 극복하기 위해 불면증 위험요인의 인과관계에 대한 추후 연구가 필요하다. Objectives : The purpose of this study is to analyze sleep habit and investigate the association between insomnia and demographic, health and psycho-behavioral symptomatology in Korean urban elderly Method : A total of 723 community residents aged 65 years or older were surveyed by two stage, random cluster sampling method. The response rate was 76.2%. Interviewers performed door to door visits and administered a questionnaire about sleep, past medical history, and several scales as the Psychogeriatric Assessment Scale(PAS), the short form Quality of Life Scale (SF-36) and etc. Results : The mean scores are as follows : retiring time was ten-sixteen p.m., wake-up time was five twenty two a.m., duration of sleep was seven hours and six minutes. Among the elderly, 11.6% and 14.8% of the subjects reported chronic and intermittent difficulty in sleeping, respectively. The patients with back pain and history of stroke complained of more insomnia symptoms in univariate analysis. In ANCOVA analysis, insomnia symptoms were associated with an old age, depressive symptoms in male. Bodily pain and history of dermatologic disease were additional risk factors in female. Conclusions : The data suggest insomnia is one of most common health problems among elderly Koreans living in the community. Strong positive relationship between depressive symptoms and insomnia were found. Further research is needed to explore the causal relationship between insomnia and risk factors.

        벼 담수표면직파재배와 손이앙재배의 도복발생 발생 양상

        본 시험은 벼 담수표면직파재배 (이하는 담수직파)에서 가장 문제가 되는 도복발생의 원인을 손이양재배와 비교, 구명하기 위하여 작물시험장 답물포장에서 1991~1992년에 수행되었다. 공시품종은 자포니카인 화정벼를 이용하였고, 벼 재배방법은 침수작파와 손이양으로 하였으며, 질소시비량은 4수준(50,100, 150,200kg/ha)으로 처리하였다. 1. 벼의 지상부생중은 출수후 20일경에 가장 무거웠으며, 그 후 계속 감소되었고, 중심고비율은 성숙기까지 점진적으로 증가되었다. 또 좌절중은 출수후 40일 까지 계속 감소되었으며, 도복지수는 출하후 30~40일 경에 가장 컸다. 2. 손이앙에서는 간장, 지상부 생중, 도복지수는 침수직파에서 보다 현저히 크고, 좌절종은 서로 비슷하였으나 실제 포장도복은 침수직파에서 더 심하였는데 이는 두 재배양식 간에 도복원인이 서로 다르다는 것을 나타낸다. 3. 손이양은 질소 200kg/ha에서 반복형 도복이 있었으나, 침수직파는 질소 150kg/ha 이상에서 뿌리도복이 심하게 발생되었다. 4. 침수직파에서 뿌리도복이 심했던 주된 요인은 벼 뿌리의 천근화 현상 때문인데, 토층표면(0~5cm)의 뿌리분포는 침수직파(65.2%)가 손이앙(51.6%)보다 현저히 많았으나 상층 5~10cm에서는 침수직파(18.5%)가 손이앙(28.0%)보다 적었다. 또 간기부의 매몰깊이가 손이앙은 4.0cm로 깊었으나 침수직파는 1.2cm로 얕기 때문에 등숙의 진전과 더불어 지상부의 무게중심이 높아질 때 침수직파의 경우 뿌리의 식물체 지지력이 양화되어 도복이 쉽게 발생되었다. 5. 침수직파는 손이양에 비하여 단위면적당 수수는 많았으나 수당입수가 적고 도복으로 인하여 등숙비율이 낮았다. 벼 수량은 평균적으로 침수직파는 손이앙의 약 82% 정도였으며, 침수직파의 수량제한요인은 도복으로 인한 낮은 등숙비율이었다.과 수확지수는 부토심 7cm에서 1cm, 3cm, 5cm에서 보다 다소 높았다.e 및 free fatty acid) 는 13.3~17.4%로 나타났다.로 빠른 시일에 집중적인 연구가 필요하다고 본다.다. 4. 저온저장(4℃ , RH 50%)한 벼는 2년반 저장한 벼도 밥맛의 변화가 거의 없었다. 5. 1988년산 및 1989년산 일반계를 10분도와 12분도로 도정하였을 때 도정도에 따른 밥맛의 차이는 없었다.X>CoOx는 Co3O4 로 존재하고, 반응 전의 경우에는 이와는 다른 chemical state를 보여주었다. XRD 및 XPS 결과를 바탕으로, 촉매표면에 존재하는 Co3O4 의 외부표면이 Co2TiO4 와 CoTiO3 같은 CoTiOx 로 encapsulation되어 있는 모델구조를 제안할 수 있고, 이는 반응시간의 함수로 나타나는 촉매활성에 있어서 전이영역의 존재를 잘 설명할 수 있을 뿐만 아니라, XRD와 XPS에서 얻어진 촉매의 물리화학적인 특성을 잘 반영할 수 있다. 나타냈고, 골격근과 눈 조직에서 피루브산에 대한 LDH의 친화력이 상당히 크므로 LDH가 혐기적 조건에서 효율적으로 기능을 하는 것으로 사료된다.5) and "Cleanliness of clothes & features" (p <0.05) of VIP ward were significantly higher than those of a general ward.tive to apply.아울러 고려(考慮)해야 한다. 이것은 고무기술자(技術者)가 당면(當面)해야할 과제(課題)에 속(屬)하며 바람직 한것은 본장(本章)의 내용(內容)이 여러 상황하(狀況下)에서 당면(當面)한 문제(問題 Broadcast-seeded rice in submerged paddy frequently lodge in the field. In general, the causes of lodging in rice cultivation differ with different cultural methods. This study was conducted to investigate the causes of lodging in broadcast-seeded rice (BSR) and hand-transplanted rice (HTR) under four nitrogen (N) levels. Lodging in BSR was mainly a root lodging due to shallow root distribution, while that in HTR showed a bending type owing to deep rooting system. At the upper soil layer (0-5cm from the surface of ground) the root distribution of BSR (65.2%) was much larger than that of HTR (51.6%), whereas at the 5-10cm soil layer the root distribution of BSR (18.5%) was much smaller than that of HTR (28.0%). The depth of buried culm base was much shallower in BSR (1.2cm) than in HTR (4.0cm). The plant height, fresh weight, lodging index, culm diameter and thickness in HTR were much greater than those in BSR, and the breaking strength of lower internode was similar in the two cultivation methods indicating that HTR would have more lodging causes than BSR. In spite of . the more advantages to lodging resistance in BSR it severely lodged in the field. The main lodging-inducing factors of BSR were the shallow root distribution and shallow depth of buried culm base. Besides these, the higher ratio of gravity center of culm was an important factor. This result suggested that for the fundamental prevention of lodging in BSR, an ideotype of rice plant with ' a deep-rooted behavior ' should be developed.

        EXPERIMENTAL STUDY ON MEASUREMENT OFEMISSIVITY FOR ANALYSIS OF SNU-RCCS

        SNU-RCCS is a water pool type RCCS (Reactor Cavity Cooling System) developed for VHTR (Very High TemperatureReactor) application by SNU (Seoul National University). Since radiation heat transfer is the major process of passive heatremoval in a RCCS, it is important to determine the precise emissivity of the reactor vessel. Review studies have used a constantemissivity in the passive heat removal analysis, even though the emissivity depends on many factors such as temperature, surfaceroughness, oxidation level, wavelength, direction, atmosphere conditions, etc. Therefore, information on the emissivity of agiven material in a real RCCS is essential in order to properly analyze the radiation heat transfer in a VHTR. The objectives of this study are to develop a method for compensation of the factors affecting the emissivity measurementusing an infrared thermometer and to estimate the true emissivity from the measured emissivity via the developed method,especially in the SNU-RCCS environment. From this viewpoint, we investigated factors such as the attenuation effect of thewindow, filling gas, and the effect of background radiation on the emissivity measurements. The emissivity of the vessel surfaceof the SNU-RCCS facility was then measured using a sight tube. The background radiation was subsequently removed fromthe measured emissivity by solving a simultaneous equation. Finally, the calculated emissivity was compared with the measuredemissivity in a separate emissivity measurement device, yielding good agreement with the emissivity increase with vesseltemperature in a range of 0.82 to 0.88.
