      • Poster Session : PS 1454 ; Hemato-Oncology(Oncology) : The Methyltransferases EZH1 and EZH2 Control Hepatocyte Homeostasis and Regeneration

        Background: Polycomb group protein enhancer of zeste homolog 2 (EZH2) controls cell proliferation and differentiation and is frequently up-regulated in hepatocellular carcinoma. Genetic inactivation of Ezh2 did not produce obvious phenotypic differences in knockout livers suggesting a functional redundancy with Ezh1 homolog. Methods: To investigate the role of both Ezh1 and Ezh2 in liver homeostasis, we generated double knockout mice (E1/2KO) by crossing Ezh1 null and albumin promoter- driven Ezh2 conditional-null mice.The combined loss of EZH1 and EZH2 in mouse hepatocytes caused a global and gene-specific depletion of H3K27me3 marks including genes associated with hepatocyte homeostasis and regeneration. Mice mutant for both Ezh1 and Ezh2 remained viable and overtly normal by 3 months of age. Thereafter, E1/2KO mice exhibited progressive liver abnormalities manifested by development of regenerative nodules and concomitant periportal fibrosis, infiammatory infiltration and activation of A6-positive hepatic progenitor cells (HPCs). Results: In response to chronic treatment with CCl4, E1/2KO mice showed increased hepatic degeneration associated with liver dysfunction, cholestasis, and reduced ability to proliferate. CCl4-induced hepatotoxicity triggered an activation of HPCs, which did not prevent early lethality of E1/2KO mice. After 2/3ds partial hepatectomy, E1/2KO livers showed an increased liver injury and a blunted regenerative response. RNA-seq analysis revealed dysregulation of genes related to regulation of cell survival, fibrosis, and proliferation including Cdkn2a and Cdkn2b. Conclusions: This work demonstrates a critical role of EZH1 and EZH2in maintaining hepatic homeostasis and ability to regenerate.

      • Effects of Stimulus Size and Position on the Performance of a Gaze Control Device

        Background: Eye mouse is operated by tracking the eye movements in which it acts as an alternative input device that has high degree of freedom. It can perform most of the tasks that general mouse can do. However, the existing studies related to operation method and task area are few, and the results from research and study that can improve them are limited. Objective: In this study, the most commonly used click operators, which are dwell and blink, were compared to understand the performance difference between two click methods in terms of on-screen task position and size. When carrying out the task using eye mouse, the optimized operation method and task area were taken into investigation. Method: Twenty seven participants were to perform clicking tasks with given targets in the experiment where its independent variables were sizes and positions of targets, and clicking methods. The experiment was performed repetitively with given tasks and targets. Results: The experiment results showed that blink method compared to dwell method in terms of time, number of error, and number of failure, showed better performance than dwell method. And blink method showed its excellence in terms of efficiency and effectiveness in contrast to dwell method. The result also revealed that both methods for clicking target on upper part of the screen is more suitable than clicking target on lower part of the screen. For these findings, medium-size folder showed the strongest effect while part of the results of small-size folder and large-size folder did not show clear effect. Conclusion: Better performance by eye mouse can be achieved by the use of blink method rather than dwell, and position of targets in upper part of screen rather than lower part. Application: The results of this study contribute to the establishment of guidelines for development in interface design and operation method for eye mouse application.

      • The Effect of Moving Direction on the Dragging Task Performance of a Gaze Control Device

        Objective: This study aims to investigate the effects of moving direction on the dragging task performance of a gaze control device (eye mouse). Background: Eye mouse is developed and utilized for disabled people as an alternative input device to control computers instead of a conventional mouse. Nevertheless, its usability has been investigated insufficiently. Method: Total of twenty seven subjects participated in the experiment and performed dragging tasks by eye mouse under given stimulus as target position and moving direction varied. Results: The result in order of performance level by movement variation follows: diagonal movement, horizontal movement, and vertical movement in highest to lowest order. In detail, the horizontal movement in upper part showed higher performance compared to the horizontal movement in lower part. Conclusion: The result in terms of efficiency can be applied in establishment of guidelines for eye mouse development such as interface and control design. Future studies on the effectiveness respect to more complex variables such as approach angle or displacement may potentially derive the impact and correlations among effectiveness and other variables. Application: Also, it is expected to deliver higher level of performance by eye mouse as a new interface design based on the results of this study.

        Self-Forgiveness Moderates the Effects of Depression on Suicidality

        Objective: Not all depressive individuals are suicidal. An increasing body of studies has examined forgiveness, especially self-forgiveness, as a protective factor of suicide based on that suicide is often accompanied by negative self-perceptions. However, less has been studied on how different subtypes of forgiveness (i.e., forgiveness-of-self, forgiveness-of-others and forgiveness-of-situations) could alleviate the effects of depression on suicide. Hence, this study examined forgiveness as a moderator of depression and suicidality. Methods: 305 participants, consisted of 87 males and 218 females, were included in the study. The mean age was 41.05 (SD: 14.48; range: 19–80). Depression, anxiety, and forgiveness were measured through self-report questionnaires, and suicidal risk was measured through a structuralized interview. Moderations were examined through hierarchical regression analyses. Results: Depression positively correlated with suicidality. Results of the hierarchical regression analysis indicated forgiveness as a moderator of depression on suicidality. Further analysis indicated only forgiveness-of-self as a significant moderator; the effects of forgiveness-of-others and forgiveness-of-situation were not significant. Conclusion: These findings suggest that forgiveness-of-self is essential in reducing of the effects of depression on suicidality. It is suggested that self-acceptance and the promotion of self-forgiveness should be considered as an important factor when developing suicide prevention strategies.

      • Akt3 knockdown induces mitochondrial dysfunction in human cancer cells

        <P>Akt/PKB plays a pivotal role in cell proliferation and survival. However, the isotype-specific roles of Akt in mitochondrial function have not been fully addressed. In this study, we explored the role of Akt in mitochondrial function after stable knockdown of the Akt isoforms in EJ human bladder cancer cells. We found that the mitochondrial mass was significantly increased in the Akt1-and Akt3-knockdown cells, and this increase was accompanied by an increase in TFAM and NRF1. Akt2 knockdown did not cause a similar effect. Interestingly, Akt3 knockdown also led to severe structural defects in the mitochondria, an increase in doxorubicin-induced senescence, and impairment of cell proliferation in galactose medium. Consistent with these observations, the mitochondrial oxygen consumption rate was significantly reduced in the Akt3-knockdown cells. An Akt3 deficiency-induced decrease in mitochondrial respiration was also observed in A549 lung cancer cells. Collectively, these results suggest that the Akt isoforms play distinct roles in mitochondrial function and that Akt3 is critical for proper mitochondrial respiration in human cancer cells.</P>

        To develop the classification method of Agricultural by-productions for biogas production

        The objective of this study was to develop the classification method of various organic wastes. Specifically, the effects of proximate composition on the biogas production and degradation rates of agricultural by-production was investigated and a new standards for mixture of various organic wastes based on proximate composition combination was developed. Agricultural by-products (ABPs) with medium total carbohydrate, medium crude protein and low fat contents demonstrated the single step digestion process. ABPs with low total carbohydrate, high crude protein and high fat contents demonstrated the two step digestion process of Diauxic growth. The single ABP (Class No. 15) and the mixed ABPs (Class No. 12+18, 6+12+22, 9+12+18) after 10days showed the similar biogas yield pattern. We can use the classification method for the more ABPs and organic wastes from factory and municipal waste treatment plant for the high efficient biogas production.

        Enhanced ceramides production by Lactobacillus rhamnosus IDCC 3201 and its proposed mechanism

        The use of probiotics has been applied for a variety of fields (e.g., immune system, mental health, and heart). In this study, the feasibility of lysates from L. rhamnosus IDCC 3201 for cosmetic ingredients was evaluated. More specifically, enhanced ceramides production in human epidermal keratinocytes by the lysates and its proposed machanism were investigated through in vitro and genome analysis. In results, enhanced spingomyelinase activity and thereby increased ceramides production by the lysates from L. rhamnosus IDCC 3201 was observed. Furthermore, it was found that the existence of glucosylceramdase in L. rhamonsus IDCC 3201 was attributed to enhanced ceramides production. Finally, it was verified that the lysates from L. rhamonsus IDCC 3201 was regarded as safe for its use as cosmetic materials. Thus, these findings have significant implications that might lead to the development of functional and safe cosmetic products from probiotics.

      • Effects of ICT Ability of the Elderly People for the Acceptability of Conversational Agent

        Objective: This research is an analysis of how ICT ability are affected in each acceptance of factors in conversational agent, to invigorate the use of conversational agent for elderly people. Background: With the rapid increase in the population of elderly people, the ability of having independence and participating in social activities are one of the main issues to provide solutions for the problems of the aged. The control method for conversational agent is intuitive as it does not require an input device. This allows easier adaptation of interaction method even for the elderly people with less experience with ICT. Method: We recorded eight different conversation scenarios formulated for this research in advance, using the text-to-speech program. The laboratory was set to allow conversational interaction by transmitting the voice converted to the speaker through computer, upon hearing the articulations of the participants over a partition. The ICT ability of the participants, and acceptance of conversational agent was measured through a survey by seven-point Likert scale. Results: In order to enhance the acceptance of conversational agent among elderly people, it is required to develop a new design technique allowing elderly people to form trust towards conversational agent by perceiving it as a social presence. Moreover, it is necessary to consider the importance of elderly people to acknowledge the sociability and usefulness of conversational agent. Conclusion: In this research we compared various factors of acceptance among elderly people, utilizing conversational agent. Conversational agent would henceforth function as an opportunity not only for elderly people, but also for the context and users with difficulty controlling devices with certain body parts.
