물리적 처리에 의한 강력분 밀가루 Gliadin의 항원성 변화

본 연구에서는 가압가열 및 microwave 처리가 gliadin의 항원성에 미치는 영향을 살펴보기 위해 강력분에 가압가열과 microwave를 단독 또는 병행으로 처리하여 Ci-ELISA, SDS-PAGE 및 immunoblotting을 실시하였다. 가압가열 처리의 경우 시간이 길어질수록 IgG와의 결합력이 감소하였으며, 특히 50분 처리구에서 약 87%로 가장 낮은 결합력을 보였다. 또한 SDS-PAGE와 immunoblotting 결과에서도 무처리구에서 강하게 보였던 gliadin band가 가압가열처리에 의해 거의 소실되고 항체와 반응하지 않았다. 가압가열 및 microwave를 병행 처리 시도 마찬가지로 gliadin의 결합력이 감소하였으며, 처리구 중 가압가열 50분, microwave 5분 처리구에서 약 93%로 가장 낮은 결합력을 보였다. 반면 microwave를 단독으로 처리한 경우에는 일부 단백질의 변화는 관찰되었으나, 항원성 감소에는 효과가 없음을 확인하여 단백질 변화가 항원성에는 큰 효과를 준 것 같지 않다. 이상의 결과를 통해 가압가열 단독 처리 및 가압가열 및 microwave의 병행처리 시 gliadin의 항원성이 감소함을 확인하였다. This study was conducted to evaluate the effects of physical treatments on the antigenicity of gliadin in strong wheat flour. Strong wheat flour was treated with an autoclave (5, 10, 30, 50 min), a microwave (1, 5, 10 min), or both (10, 30, 50 min/ 5, 10 min), followed by SDS-PAGE, immunoblotting, and Ci-ELISA using anti-gliadin IgG. The results indicated that the binding ability of IgG to gliadin in strong wheat flour slightly decreased after autoclaving or autoclaving/microwaving. In particular, the binding ability was reduced to about 87% after autoclaving for 50 min and to 89% after autoclaving/microwaving (50/5 min). In addition, gliadin bands in the 50 min autoclaved group disappeared in both SDS-PAGE and immunoblotting. On the other hand, the antigenicity of gliadin was unaffected by microwaving alone. In conclusion, the results of this study suggest that autoclaving may reduce the antigenicity of gliadin in strong wheat flour.

Electromagnetic wave absorption properties of multistacked hexaferrite-epoxy composites

We report on electromagnetic wave (EM) absorption properties of multistacked hexaferrite-epoxy composites. Co-Ti substituted M-type hexaferrite powders, SrFe_12-2xCo_xTi_xO₁₉ (x=1.1, 1.2, 1.3) were prepared by conventional solid-state reaction method. Each composition of powder was calcined at 1300°C for 2 hours in air. The calcined powders were ground into fine powders, mixed with 10 wt% of epoxy binder, pressed to form toroidal-shaped (inner and outer diameters of 3.04 and 7.00 mm, respectively) green compacts, and finally cured at 180°C in air for 20 min. The real and imaginary parts of permittivities (ε', ε") and permeabilities (μ', μ") were measured on the toroidal shaped composites using a vector network analyzer (VNA) with an airline kit in the frequency range from 0.1 to 18 GHz. RL spectra for the composites were calculated based on transmission line theory and measured ε', ε" μ', μ" data. Also, the RL spectra were measured from S-parameter method using the VNA with airline kit and one-port Cu threaded hole socket. The minimum RL value (RL_min) < -20 dB could be obtained for the series of composites samples at the optimal thickness. The frequencies of RL_min were 14 GHz, 10 GHz, and 6 GHz for x = 1.1, 1.2, and 1.3 samples, respectively. The RL spectra were also measured on the stacked three samples (A, B, C) with varying stacking orders as ABC, ACB, BAC, BCA, CAB, CBA. The RL spectra were slightly varied depending on the stacking order. However, it was very far from the sum of RL spectra in each sample. The measured RL spectra of the stacked samples were well fitted with calculated RL spectra at total thickness of three samples. Here, the RL calculation was based on volume-averaged ε', ε" μ', and μ" values for each sample.

지연성 운동장애와 양성, 음성 정신분열병 환자에서의 인지기능 장애

저자들은 정신분열병 환자에서 양성, 음성 증후군에 따라 각각 특징적으로 어떤 인지기능의 장애를 보이는지 알아보며, 또 지연성 운동장애의 존재 여부가 인지기능에 어떤 영향을 미치는지 조사해보고자 지연성 운동장애 환자 31명과 대조군 32명에게 PANSS 검사를 시행하였고, 인지기능 검사로는 KWIS 검사와 Grooved Pegboard test, Trail making test A/B, Wisconsin card sorting test, Wecshler memory scale을 시행하였다. 그 결과 다음과 같은 성적을 얻었다. 1) KWIS 지능검사상 이해 소검사에서 양성 증후군 환자들이 음성 증후군에 비해 우수한 수행도를 보였으며 빠진 곳 찾기 소검사에서는 지연성 운동장애가 없는 집단이 있는 집단에 비해 우수한 수행도를 보였다. 2) Trail making test A형에서 양성 정신분열병 환자 집단이 음성 정신분열병 환자 집단에 비해 전반적으로 빠르고 효율적인 수행을 보였다. 3) 그밖의 다른 인지기능 검사상에서는 양성, 음성 정신분열병과 지연성 운동장애 유무에 따른 검사 결과의 차이는 보이지 않았다. 결론적으로 음성 증후군 환자들의 경우 양성 증후군에 환자들에 비해 실행 기능과 같은 전두엽 기능의 장애와 기억 능력의 저하가 좀더 분명하게 나타나 음성 증상과 인지기능 장애 사이에 통계적으로 의미있는 상관관계가 있는 것으로 나타났다. 반면 지연성 운동장애의 경우는 음성 증후군과 함께 나타날 때만 현실 검증력, 주의력의 저하가 두드러져서 지연성 운동장애 자체만으로 인지기능의 장애가 유발된다고 보기는 어려웠다. Objective : The goal of this study was to examine cognitive dysfunctions in positive and negative schizophrenic patients and investigate the effect of tardive dyskinesia on cognitive dysfunctions in them. Methods : Thirty two schizophrenic patients with tardive dyskinesia and thirty two schizophrenic patients without tardive dyskinesia as a control group were selected from psychiatric inpatients at a lunatic asylum and the cognitive dysfunctions of these patients were assessed with some neuropsychological tests including KWIS test, Grooved Pegboard test, Trail making test A and B, Wisconsin card sorting test, and Wechsler memory scale test. From these patients, we classified eighteen schizophrenic patients as a positive syndrome group and twenty five patients as a negative syndrome group with positive and negative syndrome scale and compared the results of neuropsychological tests between these two groups. Results : Patients without tardive dyskinesia showed better performances than those with tardive dyskinesia on picture completion subtest of KWIS. Positive schizophrenic patients showed better performances than negative schizophrenic patients on comprehension subtest of KWIS and trail making test A. Conclusion : Negative schizophrenic patients showed frontal lobe dysfunction and more memory impairment than positive schizophrenic patients. In conclusion, negative syndrome of schizophrenia is significantly associated with cognitive dysfunctions although tardive dyskinesia is not.

Identification of the genes which related cold (low temperature) stress in Bombyx mori

Stress may be defined as any modification of environmental parameters that leads to a response by biological organisms. Stresses that affect biolpgical structures may be nonthermal, such as ultraviolet radiation, pH, or salinity, or thermal. Temperture is one of the major stresses that all living organism face. The major effects of cold (low emperature) are decrease of membrane fluidity and the stabilization of secondary structures of RNA and DNA in the cells, which may effect the efficiency of translation, transcription, and DNA replication. In this study, we focus on discovering the genes that are expressed by the cold (low temperature) stress in the silkworm. In cold(low temperature) stress test, we found 100% survive from cold stress at 0℃ up to 12h and -5℃ up to 2h, and then, survive rate was rapidly decrease in silkworm. Thereafter two whole genes have selected by SSH (Suppression subtractive hybridization). (GenBank accession : GQ149511, GQ338156)

A sequence analysis of cDNA revealed a single open reading frame (ORF) of 1233 nucleotides. The deduced 410 amino acid sequence of B. mandarina hemolin contains 4 imunoglobulin (Ig) C-2 type domains. B. mandarina hemolin cDNA showed the highest sequence homology to known those of B. mori. The developmental profile in terms of expression level of hemolin mRNA was determined in the absence of a bacterial challenge. Hemolin mRNA was detected only in mid-gut, but not in hemocytes, fat body, testis, and silkglands. Hemolin mRNA in mid-gut was not detected until the spinning stage of the last instar larva, however, lit dramatically increased at the beginning of spinning and gradually decreased until pupal stage.

S-536 The Effect of PTD-mediated BMP-7 on EMT in Peritoneal Mesothelial Cells

Objective:?We investigated the effect of PTD-mediated BMP-7 (tissue-regeneration polypeptide 2, TRP2) on TGF-β1-induced EMT in cultured human peritoneal mesothelial cells (HPMCs). In addition, we investigated how to deliver the drugs to peritoneum in vivo models. Design and Method:?In vitro, HPMCs were cultured in M199 media containing 5.6 mM glucose (normal glucose, NG), NG + TGF-β1 (2 ng/ml) with or without TRP2 (100 ng/ml). After 72 hours, cells were harvested. In vivo, PD rat-O-port inserted into 11 Sprague-Dawley rats, and saline (control group, n=3), 4.25 % PD solution (PD group, n=3) or 4.25 % PD solution + TRP2 (PD + TRP2 group, n=5) were infused for 4 weeks. After 4 weeks, rats were sacrificed and the peritoneal tissues were removed. E-cadherin, ZO-1, α-smooth muscle actin (α-SMA), snail, vimentin, type I collagen, and fibronectin protein expression in HPMCs and the peritoneum were estimated by western blot analysis, and fibronectin expression was evaluated by immunohistochemistry staining. PF was assessed by Masson’s trichrome (MT) staining. Results:?In vitro, protein expression of E-cadherin and ZO-1 (epithelial marker) were significantly decreased, while α-SMA, snail, vimentin (mesenchymal marker), type I collagen and fibronectin were significantly increased in TGF-β1-stimulated HPMCs compared to control group, and these changes were significantly improved by TRP2 treatment. In vivo, peritoneal EMT and PF were significantly increased in PD rats compared to control rats. The thickness of mesothelial layer and the intensity of MT staining in the peritoneum of PD rats were also significantly higher compared to control rats. These changes of the peritoneum in PD rats were significantly ameliorated by the administration of TRP2. Conclusions:?This study suggests that TRP2 directly inhibits the process of TGF-β1-induced PF via peritoneal EMT in HPMCs. In addition, TRP2 mitigates PF in PD rats. The effect of PTD-mediated recombinant protein delivery system may be a potential therapeutic strategy for prevention of PF in PD patients.

In this study in compliance with the cold temperature stress about selection of the useful gene is contents from the silkworm which is been revealed. The survival rate which is caused by with the cold temperature stress until 12 hours was 100% in 0℃, until 2 hours was 100% in -5℃. A total of 960 clones were randomly selected from the subtraction cDNA library, and then performed a differential display hybridization analysis with forward and reverse probes. In conclusion, selected 53 partial clones and novel 2 full-length clones were identified as differential expressed genes. We assumed that the novel gene related with transmembrane.
