      • 種繭의 保護溫度가 蠶蛾의 産卵性에 미치는 影響

        The study is conducted to clarify the difference of oviposition ability of the silkworm mother moths sampled from race, Bombyx mori. which had been preserved in various temperatures in pupal stages. The results obtained are summarized as fellows; The number of mature eggs and the ration of total eggs laid in low temperature preservation in the preceeding term of the pupal stage are higher than those in high temperature preservation. Among the treatments the high and low temperature in the late term of pupal stage revealed the worst results. Ratio of normally oviposited eggs is exceedingly low if kept at low temperature in previous term and it is higher in the Chinese race than in the Japanese. However preservation temperatures affected not only oviposition ability, but states of egg laying. Ratio of unfertilized eggs is higher at low and high temperature during the pupal period and low temperature is more effective than high temperature in previous term. Correlation between the number of mature eggs in oviduct and normally oviposited eggs is high significance in every races and every treatments.

      • Foreign gene induction into Bombyx mori egg by electroporation

        Electroporation is well known today as a powerful transfection technique and is useful for the study of gene expression. The advantage of the electroporation method is that large quantity of silkworm (Bombyx mori) eggs can be transformed in a very short time. However, how to use it for introducing foreign gene into silkworm eggs needs systematical investigation. In our silkworm transgenesis program, we needed an efficient technique to evaluate the functionality of transgenes before their injection into eggs. The goal of this experiment was to find an alternative efficient method of generating transgenic silkworm eggs using a commercially available electroporation device. The Gene Pulser Xcell (Bio-Rad Laboratories, USA) were used. In contrast to other electroporation devices, which are based on a single pulse with exponential decay or square wave technology. We investigated pigmentation-rate and hatching-rate of the silkworm eggs of electroporation. We used foreign gene LacZ, EGFP, Ds-red induced vector system with selection marker for transgenic silkworm. The LacZ gene in 3rd instar larva DNA can be detected by β-galactosidase stain. During these technical studies we found that optimizing parameters such as electrical voltage, number of pulses and their frequency, and conductivity of the buffer was important. These results confirmed that electroporation is available technique for transfecting B. mori egg.

        This studf was carried out to investigate the elucidation of gene expression in embryo formation of the Kidney mutant, especially yolk proteins in unfertilized eggs and template activity of m·RNA extracted from them. The results were summarized as follows: There was no recognized qualitative difference in yolk proteins of unferilized eggs between the Kidney mutant and normal. There was not any difference, between Kidney mutant and normal in the molecular species of m-RNA derived from maternal origin with the template activity in vitro.

      • 現存하는 韓國 在來種 蠶品種의 形質特性

        The objective of this research was to characterize of the silkworm that might be recognized to the Korea native strains. The eleven strains of Korean race used in this study. which is cultured in Korea and Japan seri-cultural research organs. Most of Korean varieties were three molting and univoltine, comparatively longer larval duration than the trimolter general. The egg characters of varieties showed shor-elliptic shpe and dark brown egg color except for a few varieties which shows greenish dark brown. Also, most of varieties were showed the plain(p) and moricaud(pM) in larval markings. Moricaud marking of the varieties also consist of innumerable dark grayish brown lines and dots, though somewhat darker and lighter than that of the wild silkworm, Bombyx mandarina. Cocoon characters variations of varieties were seen in the size, color and shape. 8 varieties were colored cocoon, t.e..yellow, greenish yellow and light green etc..the others were white cocoon. The shape of cocoos were consisted of constricted shallowly in the middle and attenuated at one or both ends f cocoon, t.e..spindle. The results of this study is sufficient evidence that Korea strains were shown as the one of regional origin strain of domesticated silkworm such as the Chinese and Japanese etc.

      • 누에의 후배자발생 초기에 있어서 돌연변이 불면잠계통간의 형질발현 특성

        Studies were carried out to investigate phenotypic expression, mortality and biochemical analysis of haemolymph proteins of nm-d, nm-f, nm-i, nwr-k and nmn non-molting mutants of the silkworm, Bombyx mori. The non-molting mutants characters were expressed in the homozygote of each mutant genes. All strains of non-molting mutants were similar with each other in physiological characteristics, but the expression varied with each strains. The larvae of nm-d, nm-i and nwfn died between day 5 and day 9 after hatching without the first molt. The nm-f and nm-k mutants died between day 5 and day 16 with a slight increase of body weight and, more than 90% of the mutants larvae died before the first molt and a few of them survived to the 2nd and the 3rd instar and died. The haemolymph protein components of nm-d,nm-i and nmn were rapidly reduced , and on the other hand those of nm-f and nm-k consistently until they died. And there were no distinguishable difference in haemolymph components of non-molting mutants, as compared to those of normals.

      • 腎藏形卵에 있어서 卵殼構造의 解析

        The structure ki mutant chorion is described by the ultrastructural level with the scanning electron microscope. The micropyle on the anterior, posterior, leteral and dorsal side region surface patterns do not so much differ from ki mutant and normal strains. The surface structure of ventral side of ki mutant, which is the presumptive embryonic region in oocytes, were netted part corresponding to the boundary of follicle cells that is similar to lateral side structure of egg·shell. The egg-shell thickness of ventral side on the ki mutant is mostly thicker. However, the structure of transverse section of normal and ki mutant egg-shell are composed of the outer, middle and inner layers. The ki not effects the formation of the lamellar layer in the egg-shell. It is observed that the characteristic structure in ki mutant egg chorion may be attributed to a areal specificity of epithelium in ventral side of egg-shell formation, and it affects the process of the accumulation of secreted chorionic material and the egg-shell architecture.

        Molecular conformation and crystallinity of white colored silkworm cocoons with different silkworm varieties

        Bombyx mori silkworm has many silkworm varieties and the structure and properties of silk can be varied depending on the silkworm variety. Therefore, the choice of proper silkworm variety can be an effective way to control and improve the performance of silk materials in biomedical and cosmetic applications. As a preliminary study, in the present study, thirty nine Bombyx mori silkworm cocoons were prepared and their molecular conformation and crystallinity were examined. Molecular conformation and the crystallinity of silkworm cocoons were different depending on the silkworm variety. Interestingly, the crystallinity index of outside of cocoon was different from that of inside of cocoon and the difference between the crystallinities of outside and inside of cocoon was varied depending on the silkworm variety. In this study, the silkworm variety cocoons were classified into three groups (Group 1, Group 2, and Group 3) based on the difference of crystallinities between outside and inside.

        Crystallinity of yellow colored silkworm variety cocoons

        The structure and properties of silk polymers (fibroin and sericin) can be satisfactorily controlled by choosing a suitable silkworm variety and, hence, this parameter (i.e., silkworm variety) has attracted increasing attention. A previous study reported that the crystallinity of white colored silkworm cocoons depends on the silkworm variety. In the present study, sixteen yellow colored silkworm variety cocoons were produced and their molecular conformation and crystallinity were investigated. The conformation of the silkworm cocoons varied with the silkworm variety. Most cocoons exhibited ${\beta}$-sheet conformation, although random coil and ${\beta}$-sheet conformations co-existed in some cocoons (e.g., 21 and D90). The crystallinity of the silkworm cocoons varied with the silkworm variety and the measurement position of the cocoon (i.e., outer surface or inner surface). However, the difference in the crystallinity indices of the outer and inner surfaces comprising the cocoons varied with the silkworm variety, but was <2% for all cocoons, except for MAL.

        Morphology and crystallinity of silkworm cocoons with different rearing seasons

        The silkworm cocoon has recently attracted the attention of researchers because of its usefulness as a biomedical material (e.g., usage in the development of the membrane for guided bone regeneration and usage as a starting material for the fabrication of natural silk nonwoven fabric). The silkworm variety and strain have been reported as one of the important factors affecting the structure and properties of silk materials. This study examines the morphology and the molecular conformation of the silkworm cocoon at different cocoon measurement points, as well as the effect of the silkworm strain and rearing season on the morphology and crystallinity of the silkworm cocoon. The results show that the morphology of the outside of the cocoon does not differ depending on the cocoon measurement points, but that of the inside does. The silkworm cocoon crystallinity is not affected by the cocoon division. Interestingly, the crystallinity of the outside of the Jam101 cocoon differs depending on the rearing season, whereas it does not in other silkworm strains.
