        Significant enhancement of bromate removal in drinking water: Implications for the mechanism of sonocatalytic reduction

        <P><B>Abstract</B></P> <P>Bromate (BrO<SUB>3</SUB> <SUP>−</SUP>) is a potential carcinogenic compound that can form during the disinfection of drinking water. For the first time, the sonocatalytic reduction of BrO<SUB>3</SUB> <SUP>−</SUP> were studied through examining the effect of the important operational parameters such as ultrasound (US) frequency, TiO<SUB>2</SUB> loading, pH, temperature and other anions. By observing sonoluminescence (SL) at various frequencies and measuring H<SUB>2</SUB> production rates, a new reduction mechanism for BrO<SUB>3</SUB> <SUP>−</SUP> is also proposed. The kinetic results presented that the BrO<SUB>3</SUB> <SUP>−</SUP> reduction rates enlarged as the measured H<SUB>2</SUB> production rate increased for all parameters, and the slopes between the BrO<SUB>3</SUB> <SUP>−</SUP> reduction and H<SUB>2</SUB> production rates for sonocatalysis were much higher than those for sonolysis. Interestingly, sonolysis could be limited by the amount of H<SUB>2</SUB> production, but sonocatalysis could increase the BrO<SUB>3</SUB> <SUP>−</SUP> reduction rate because of additional electrons (e<SUP>−</SUP>) from the surface of TiO<SUB>2</SUB> via SL. The highest SL intensities for H<SUB>2</SUB> production and BrO<SUB>3</SUB> <SUP>−</SUP> reduction rates occurred at an ultrasound frequency of 500kHz, implying that an increase in SL intensity caused by ultrasound cavitation enhanced H<SUB>2</SUB> production and increased the e<SUP>-</SUP> released to the conduction band of TiO<SUB>2</SUB>. At 500kHz, BrO<SUB>3</SUB> <SUP>−</SUP> reduction rate (6.84×10<SUP>−2</SUP> min<SUP>−1</SUP>) by sonophotocatalysis was 5.2 times higher than that (1.32×10<SUP>−2</SUP> min<SUP>−1</SUP>) of sonolysis. Investigation into the effect of anion species showed that BrO<SUB>3</SUB> <SUP>−</SUP> reduction was inhibited or enhanced depending on the type of anions present.         In this study, ibuprofen(IBP) degradation by the photochemical (UV/S2O82-) and sonochemical (US/S2O82-) processes was examined under various parameters, such as UV (10~40±5 W/L) and US (50~90±5 W/L) power density, optimum dosage of persulfate ion (S2O82-), temperature (20~60℃) and anions effect (Cl-, HCO3-, CO32-). The pseudo?first?order degradation rate constants were in the order of 10-1 to 10-5 min-1 depending on each processes. The synergistic effect of IBP degradation in UV/S2O82- and US/S2O82- processes could investigated, due to the generation of SO4- radical. This result can confirm from the produced H2O2 and SO42- concentration in each processes. IBP degradation rate affected by the S2O82- dosage, temperature, power and anion existence parameters. In particular, IBP degradation rate increased with the increase of the temperature (60℃) and applied power density (UV:40±5 W/L, US:90±5 W/L). On the other hand, anions effect on the IBP degradation was negative, due to the anion play as a the scavenger of radical.

        초음파 (US)와 다양한 파장범위의 자외선 (UV) 조사에 따른 DEP 분해특성에 관한 연구

        Diethyl phthalate (DEP) is widely spread in the natural environment as an endocrine disruption chemicals (EDs). Therefore, in this study, ultrasound (US) and ultraviolet (UVC), including various applied power density (10-40 W/L), UV wavelengths (365 nm, 254 nm and 185 nm) and frequencies (283 kHz, 935 kHz) were applied to a DEP contaminated solution. The pseudo-first order degradation rate constants were in the order of 10-1 to 10-4 min-1 depending on the processes. Photolytic and sonophotolytic DEP degradation rate also were high at shortest UV wavelength (VUV) due to the higher energy of photons, higher molar absorption coefficient of DEP and increased hydroxyl radical generation from homolysis of water. Sonolytic DEP degradation rate increased with increase of applied input power and the dominant reaction mechanism of DEP in sonolysis was estimated as hydroxyl radical reaction by the addition of t-BuOH, which is a common hydroxyl radical scavenger. Moreover, synergistic effect of were also observed for sonophotolytic degradation with various UV irradiation.

      • Bioconversion of ginsenoside Rc into Rd by a novel α-L-arabinofuranosidase, Abf22-3 from Leuconostoc sp. 22-3: cloning, expression, and enzyme characterization.

        <P>A novel α-L-arabinofuranosidase (Abf22-3) that could biotransform ginsenoside Rc into Rd was obtained from the ginsenoside converting Leuconostoc sp. strain 22-3, isolated from the Korean fermented food kimchi. The gene, termed abf22-3, consisting of 1,527 bp and encoding a protein with a predicted molecular mass of 58,486 Da was cloned into the pMAL-c2x (TEV) vector. A BLAST search using the Abf22-3's amino acid sequence revealed significant homology to that of family 51 glycoside hydrolases. The over-expressed recombinant Abf22-3 in Escherichia coli BL21 (DE3) catalyzed the hydrolysis of the arabinofuranoside moiety attached to the C-20 position of ginsenoside Rc under optimal conditions of pH 6.0 and 30 C. This result indicated that Abf22-3 selectively converts ginsenoside Rc into Rd, but did not catalyze the hydrolysis of glucopyranosyl groups from Rc or other ginsenosides such as Rb1 and Rb2. Over-expressed recombinant enzymes were purified by two steps with amylose-affinity and DEAE-cellulose chromatography and then characterized. The kinetic parameters for α-L-arabinofuranosidase showed apparent Km and Vmax values of 0.95 0.02 μM and 1.2 0.1 μmol min(-1) mg of protein(-1) against p-nitrophenyl-α-L-arabinofuranoside, respectively. Using a purified MBP-Abf22-3 (10 μg/ml), 0.1 % of ginsenoside Rc was completely converted to ginsenoside Rd within 20 min.</P>

        Evaluating the Degree of Macrodispersion of Carbon Nanotubes using UV-VIS-NIR Absorption Spectroscopy

        We measured the degree of macrodispersion of the various single-walled carbon nanotubes (SWCNTs) and multi-walled carbon nanotubes (MWCNTs) using UV-VIS-NIR absorption spectroscopy. CNTs were dispersed with SDS of 2 wt % in deionized water using the homogenizer and then were further centrifugated at 6000 g for 10 min. The degree of macrodispersion, expressed by Dm(λ)=Aa(λ)/Ab(λ)*100 (%), where λ is the wavelength and Aa(λ) and Ab(λ) are the absorbance of the sample after and before centrifugation, respectively. In the case of MWCNTs, we evaluated the degree of macrodispersion by the average degree of macrodispersion (Dm(λ)) between 1000 and 1200 nm. The degree of macrodispersion of SWCNTs was evaluated at the wavelength in which van Hove singularity-related transition regions were excluded, i.e., the range was chosen between E11S and E22S peaks. We have estimated six samples with the same method. The standard deviation of each sample was lower than 5. Therefore, we presented a reliable evaluation method for the macrodispersion of CNTs for standardization.

        Effects of Vitamin D Supplementation on Children with Autism Spectrum Disorder: A Systematic Review and Meta-analysis

        The effect of vitamin D supplementation on individuals with autism spectrum disorder (ASD) is inconclusive. We aimed to conduct a meta-analysis of the available randomized controlled trials (RCTs) to explore whether vitamin D supplementation can improve core symptoms and coexisting conditions in children with ASD. Data were obtained by searching the PubMed, Embase, Web of Science, CINAHL and Cochrane Library databases up to February 2022 following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines. Using a random-effects model, mean differences with 95% confidence intervals (CIs) were calculated through a meta-analysis. There were eight RCTs with 266 children with ASD in the present review, among which six RCTs were included in the meta-analysis. Children who received vitamin D supplementation showed a significant improvement in stereotypical behavior scores (pooled mean difference (MD): −1.39; 95% CI: −2.7, −0.07; p = 0.04) with low heterogeneity (I2 = 34%), and there was a trend toward decreased total scores on the Social Responsiveness Scale (SRS) and Childhood Autism Rating Scale (CARS, p = 0.05); however, there were no other significant differences in the core symptoms of ASD and coexisting conditions between groups as measured by the Aberrant Behavior Checklist (ABC). Vitamin D supplementation appears to improve stereotypical behaviors but does not improve other core symptoms and coexisting conditions. Further randomized controlled trials with large sample sizes and individualized doses are needed.

        Evaluation of Ultrasonic Oxidation of Diethyl Phthalate Using Box-Behnken Design with Response Surface Methodology

        The kinetic constant of diethyl phthalate (DEP) oxidized by ultrasound was measured as a function of ultrasound power density, $Cl^-$ concentration and solution temperature. The ultrasonic oxidation kinetic of DEP could be described by the pseudo-first-order reaction d[DEP]/dt = -kCDEP, where k ($min^{-1}$) was identified using Box-Behnken Design coupled with Response Surface Methodology. The final equation for describing the kinetic constant k is as follows: Analysis of variance (ANOVA) tests showed that the second-order polynomial regression model could properly interpret the experimental data with a high determination coefficient ($R^2$, 0.998). The P-value of the model was 0.0001 which is less than 0.05, implying that the model is significant. Besides, the response surface showed that ultrasound power density gave much more effect than another two factors (temperature and $Cl^-$ ion concentration), which provides a possible evidence for the future design.

      • Effects of Low-Intensity Ultrasound on Chondrogenic Differentiation of Mesenchymal Stem Cells Embedded in Polyglycolic Acid: An <i>in Vivo</i> Study

        <P>In this study we investigated the effects of LIUS on chondrogenic differentiation of bone marrow-derived mesenchymal stem cells (BM-MSC). Our hypothesis is that LIUS may be a noninvasively effective stimulant to a biological system in vivo by turning on differentiation of MSCs and promotion of chondrogenesis. MSCs were isolated from the bone marrow of New Zealand white rabbits and cultured in monolayer for 2 weeks. They were then harvested and seeded into polyglycolic acid (PGA) non-woven mesh at a number of 5 x 10(6) cells. Cultured with a chondrogenic-defined media for 1 week, the PGA/MSCs constructs (n = 4) were implanted subcutaneously in the back of nude mice (n = 9, each group). The ultrasound (US) group received US stimulation at a frequency of 0.8 MHz and intensity of 200 mW/cm(2) for 10 min every day up to 4 weeks, while the control group had no US stimulation. Analyses of histological, immunohistochemical, biochemical, and mechanical characteristics were made at 1, 2, and 4 weeks post-stimulation, respectively. Total DNA contents showed no significant difference between the two groups. Total collagen and glycosaminoglycan (GAG) increased more significantly in the US-stimulated group than in the control. Histology of Safranin O/Fast green confirmed more intense and spreading extracellular matrix (ECM) at 2 and 4 weeks in the US-stimulated specimens. Mechanical tests exhibited that compressive strengths were also significantly higher in the US-stimulated cells at later times. This study strongly suggests that it may be possible for ultrasound to have some stimulatory effects in vivo on the chondrogenesis of MSCs.</P>

        1-Oxoeudesm-11(13)-ene-12,8α-lactone-induced Apoptosis via ROS Generation and Mitochondria Activation in MCF-7 Cells

        A novel eudesmane-type sesquiterpene compound, 1-oxoeudesm-11(13)-eno-12,8a-lactone (OEL), was isolated from Aster himalaicus. Its effect on apoptosis in human breast adenocarcinoma (MCF-7) cells was investigated. MTT assay showed that OEL substantially reduced the viability of KB, MCF-7, U87, A172, and MG-63 cells. MCF-7 cells were used to further evaluate the antitumor effects and anticancer mechanisms of OEL. OEL-induced apoptosis was characterized by chromatin condensation, formation of apoptotic bodies, and phosphatidylserine on extracellular surface; these effects were confirmed by DAPI nuclear staining and flow cytometry. Increased expression of Bax and deceased expression of Bcl-2 were also observed in OELtreated MCF-7 cells. Moreover, OEL induced the loss of mitochondria membrane potential, release of cytochrome C, activation of caspase-9, and generation of reactive oxygen species. These findings indicate that reactive oxygen species generation and mitochondria activation were involved in apoptosis induced by OEL in MCF-7 cells. The results from our study demonstrated that OEL may be a promising pro-apoptotic compound that could be used to develop novel anticancer drugs.
