        RAPD Variation and Genetic Distances among Tibetan, Inner Mongolia and Liaoning Cashmere Goats

        Relationship among Tibetan cashmere goats, Inner Mongolia cashmere goats and Liaoning cashmere goats was studied using the technique of random amplified polymorphic DNA (RAPD). One primer and four primer combinations were screened. With the five primers and primer combinations, DNA fragments were amplified from the three breeds. Each breed has 28 samples. According to their RAPD fingerprint maps, the Nei's (1972) standard genetic distance was: 0.0876 between Tibetan cashmere goats and Inner Mongolia cashmere goats, 0.1601 between Tibetan cashmere goats and Liaoning cashmere goats, 0.0803 between the Inner Mongolia cashmere goats and Liaoning cashmere goats. It coincides with their geographic location. The genetic heterogeneity of Tibetan cashmere goats, Inner Mongolia cashmere goats and Liaoning cashmere goats is 0.3266, 0.2622 and 0.2475 respectively. It is also consistent with their development history.

      • Determinants of Switching Behavior of New Energy Vehicles in China: Evidence from Subjective Norms

        Purpose - This study investigates the determinants of the switching behavior of new energy vehicles in China. With the help of the Theory of Planned Behavior (TPB) and technology acceptance model (TAM), the moderating effect of subjective norms is discussed. Design/Methodology/Approach - We develop and test conceptual models using data collected from 240 respondents in China. Then, SPSS 25.0, AMOS 21.0, PROCESS v3.3, and SmartPLS 4.0.9.2 were used for empirical tests. Findings - The results show that perceived risk has a significant negative impact on switching intention, while perceived value and perceived ease of use have a significant positive impact on switching intention. Switching intention has a significant positive impact on user switching behavior. In addition, social identity has a positive correlation effect on attitude toward switching. Moreover, subjective norms have a moderating effect on the relationship between perceived risk, social identity, perceived value, perceived ease of use, and switching intention. Research Implications - The theoretical significance of this study lies in the technology acceptance model (TAM), theory of planned behavior (TPB), and the theory of perceived risk, perceived value, and social identity are introduced into the model to expand the model. Its practical significance is to provide feasible suggestions for the new energy automobile industry to meet user expectations, playing a guiding and reference role for the subsequent rise of renewable energy cars, and promoting the outlawing of fuel-powered autos and the sustainable development of renewable energy vehicles.

        Reservoir characteristics of and potential analysis to Lower Palaeozoic shale in Xiuwu Basin, Lower Yangtze Plate

        In order to evaluate reservoir characteristics and gas potential of organic-rich shale in Bottom-Lower Cambrian Hetang Formation and Upper Ordovician Xinkailing Formation, Xiuwu Basin, Jiangxi Province, Lower Yangtze Plate of China, researches regarding reservoir characteristics of organic-rich shale thickness, moderate burial depth, abundance and maturity of organic matter, fracturability, organic-matter pores and absorption performance have been conducted to Hetang Formation and Xinkailing Formation through field survey, shallow drilling, analysis to hydrocarbon-generation potential, scanning electron microscopy, measuring specific surface area and isothermal adsorption experiments. The results show that (1) Hetang Formation reservoir, although with extremely rich material basis, is of unfavorable hydrocarbon generation conditions due to late-stage thermal maturity, besides of undeveloped micropore structure, reservoir space development mainly by macroscopic crack, so further exploration should focus on locating favorable areas of low maturity and developed cracks, which is considered distributing in the core of Yanshan-Lingangshan-Shiziyan Mountains syncline; (2) Xinkailing Formation reservoir is of better organic matter abundance, pore development and absorption performance than Xinkailing Formation and hence further exploration should focus on surveying favorable areas of better sedimentary facies belt and stable structure for major breakthrough in shale-gas exploration, which is considered distributing in Wuning-Lixi and Jinkou-Tuolin areas.

        AEG-1 aggravates inflammation via promoting NALP3 inflammasome formation in murine endometriosis lesions

        Endometriosis (EMs) is one of the most common gynaecological diseases in women of childbearing age. Astrocyte elevated gene-1 (AEG-1) is associated with the invasion, migration, apoptosis and prognosis of various cancers. However, the roles of AEG-1 in EMs and its corresponding molecular mechanism are still unknown. In this study, animal models of EMs were established and mice were divided into two groups (n = 10): Sham group and EMs group. The EMs cells were isolated from EMs model. The AEG-1 gene was knocked down by shRNA, while the SOCS1 gene was knocked down by siRNA. Histological changes, AEG-1 expression in tissues and inflammatory factors level were detected by H&E staining, immunohistochemistry and ELISA, respectively. RTqPCR and western blotting were used to determine the expression level of related proteins. The present study found AEG-1 was up-regulated in the EMs model. Enhanced AEG-1 promoted inflammatory cell infiltration, and elevated the levels of IL-1β, IL-6, and TNF-α in EM group (p < 0.05). Besides, AEG-1 overexpression promoted the expression of NALP3, ASC and Cleavedcaspase- 1, while decreased SOCS1 level (p < 0.05). Decrease of SOCS1 further promoted the formation of NALP3 inflammasome. The inhibitory effect of AEG-1 on SOCS1 was weakened after the addition of MG-132 (p < 0.01). Furthermore, silencing AEG-1 alone increased SOCS1 level, decreased the levels of inflammatory cytokines, thereby inhibited the formation of NALP3 inflammasome. All these results demonstrated that AEG-1 aggravated inflammation via promoting NALP3 inflammasome formation in murine endometriosis lesions.

        Study on activity of the signaling pathways regulating hepatocyte differentiation during rat liver regeneration

        To understand the mechanism of hepatocyte differentiation during rat liver regeneration (LR) at the genetranscription level, Rat Genome 230 2.0 Array was used to determine the gene expression changes ofhepatocytes. Then the genes associated with cell differentiation and their signaling pathways were searched byNCBI, QIAGEN and KEGG webs, and their signaling activities were calculated by spectral function Ep(t)based on the expression profiles. The key genes associated with the signaling pathways and cell differentiationwere analyzed and verified by the same kind extraction method. The mechanism of hepatocyte differentiationduring rat LR was analyzed by Ingenuity Pathway Analysis 9.0 (IPA). The results showed that 42 signalingpathways regulated cell differentiation and the Ep(t) values of IL-9 signaling pathway and its JAK/STAT andPI3K/AKT branches, PEDF signaling pathway and its NF-κB and Ras/ERK branches were significantlyincreased in the progression phase (6–72 h after partial hepatectomy [PH]) of rat LR, and the Ep(t) values ofhepatocyte differentiation were also significantly increased. ERK/MAPK signaling pathway was significantlyincreased in the priming (0.5–6 h after PH) and progression phase, and hepatocyte differentiation were alsosignificantly increased. In conclusion, among the 42 signaling pathways that regulated cell differentiation, thesignaling activity of IL-9 signaling pathway and its JAK/STAT and PI3K/AKT branches, PEDF signalingactivity and its NF-κB and Ras/ERK branches, and ERK/MAPK signaling activity were significantly increasedand promoted hepatocyte differentiation in rat LR. Meanwhile, the expression of MYC involved in mentionedsignaling pathways and expression of AGT, BCL2, IL10, CCND1, PLAT, and NF1 related to celldifferentiation were increased significantly.
