        Multidisciplinary design optimization of external engagement cylindrical gear flowmeter

        Many factors affect the performance of an engagement cylindrical gear flowmeter (EGF). Each factor can simultaneously affect multiple performance indicators (e.g., flow pulsation, internal leakage and the total power loss) in different ways, and each indicator affects each other, which increases the difficulty of optimization design and precision compensation. In this paper, an optimization design method is proposed. First, the accurate displacement calculation formula was obtained by integrating the tooth profile equation. Second, the important factors that affect each performance index and the influence trend between each performance index were obtained by the design of experiments (DOE) and sensitivity analysis (SA). Third, based on the SA results, the optimization equations were established and the optimal structural parameters were obtained by using MIGA and NLPQL combined optimization algorithm. The results show that this optimization strategy can be applied to the optimization design of EGF.

        Cell Type-Specific and Inducible PTEN Gene Silencing by a Tetracycline Transcriptional Activator-Regulated Short Hairpin RNA

        Inducible and reversible gene silencing in desired types of cells is instrumental for deciphering gene functions using cultured cells or in vivo models. However, efficient conditional gene knockdown systems remain to be established. Here, we report the generation of an inducible expression system for short hairpin RNA (shRNA) targeted to PTEN, a well-documented dual-specificity phosphatase involved in tumor suppression and ontogenesis. Upon induction by doxycycline (DOX), the reverse tetracycline transcriptional activator (rtTA) switched on the concomitant expression of GFP and a miR-30 precursor, the subsequent processing of which released the embedded PTEN-targeted shRNA. The efficacy and reversibility of PTEN knockdown by this construct was validated in normal and neoplastic cells, in which PTEN deficiency resulted in accelerated cell proliferation, suppressed apoptosis, and increased invasiveness. Transgenic mice harboring the conditional shRNA-expression cassette were obtained; GFP expression and concurrent PTEN silencing were observed upon ectopic expression of rtTA and induction with Dox. Therefore, this study provides novel tools for the precise dissection of PTEN functions and the generation of PTEN loss of function models in specific subsets of cells during carcinogenesis and ontogenesis.

        An Improved Method for Rapid Preparation of Oligodendrocytespecific Rabbit Polyclonal Antibody

        Antibodies are important tools in the study of protein function and diagnostic tests. However, traditional antiserum preparation requires a time-consuming immunization protocol and subsequent purification of polyclonal antibodies. In this study, a rapid and efficient method for polyclonal antibody preparation has been developed. Juxtanodin (JN) and silent information regulator-2 (Sirt2),both of which are oligodendrocyte-specific proteins, were used for antibody preparation. The N-terminal 170 amino acids of JN (JN170) and amino acids 231-351 of Sirt2 (Sirt2-121) were expressed as GST-tagged proteins from a pET-41a(+) vector in E. coli strain BL21 (DE3) cells. The fusion proteins were purified and used to immunize rabbits following both a traditional protocol, in which antigen was presented biweekly, and a modified rapid protocol, in which the immunization on day 1 was boosted on days 5and 28. ELISA, Western blot analysis and immunofluorescent staining showed that antibodies produced via the rapid protocol could recognize these two oligodendrocytespecific proteins in vitro and in the rat central nervous system (CNS), respectively, similar to those produced with the traditional protocol. Thus, our study provides a novel rapid method to prepare high specificity antibodies via a modified immunization protocol and subsequent antibody purification.

        Bias load analysis and asymmetric modification of rollers in the main drive bearing of large roadheader

        An asymmetric modification method for the roller profile was proposed to address the bias load effect of rollers in the main drive bearing of a large roadheader. A contact mechanics model of the bearing was solved, and line contact theory was used to explore the effect of combined load and convexity on the bias load effect. The contact stress was found to be asymmetrically distributed along the contact line. This trend tended to be remarkable with the decrease in axial load or the increase in overturning moment. Symmetrically modified rollers can be used to improve the bias load effect by increasing convexity. However, this process led to the contact stress concentrating in the roller center. The influence of the design parameters for asymmetric modification on the roller profile and the contact stress was considered. The ideal design parameters were determined by using a genetic algorithm. The contact condition and fatigue life were remarkably increased.

        A Novel Identification Approach for Palm Bio-impedance Spectroscopy

        Aiming at the properties of the palm bio-impedance spectroscopy (PBIS) data and shortage of the existing algorithms, this paper proposed a kind of the PBIS identification method based on immune clone. This method mainly consisted of three parts, which are data normalization, feature selection and matching. The PBIS algorithm constructed a new classifier based on the immune clone selection algorithm (ICSA) and the hidden space support vector machine (HSSVM), called the ICSA-HSSVM. It was compared with other classifiers such as Euclidean distance, cosine distance and support vector machine (SVM). The experimental results show that the proposed classification method can achieve more than 95% accuracy in identity recognition. The PBIS biometric identification tech depends on the human body inherent characteristics to carry out the status identification so as to overcome the drawbacks of traditional information safety tech status identification. The main advantages of PBIS algorithm include four aspects. The first is the uniqueness, universality, stability, easy collection, and it is basically not being subject to the influence of pathological changes in the human body positions. The second is not sensitive to the environment and the excellent recognition performance can be obtained through the method easily. The three is high stability and uniqueness when it is served as the biometric potentiality, being in agreement with biometric identification requirements. (4) The four is able to resist against counterfeiting attacks effectively. The method proposed in this paper is of great theoretical and practical values in the fields of national public security, internet information security, military communication security, micro-blog micro-message public opinion supervision.

      • A Single Fluorescent Chemosensor for Simultaneous Discriminative Detection of Gaseous Phosgene and a Nerve Agent Mimic

        <P>A fluorescent chemosensor has been developed for discriminative detection of phosgene and a nerve agent mimic diethyl chlorophosphate (DCP), which was comprised of an anthracene-carboxyimide fluorophore and <I>o</I>-phenylenediamine (OPD) reaction site. Upon phosphorylation of OPD, the chemosensor displays an obvious fluorescence turn-on response toward DPC at 588 nm with instant response and a low detection limit (88 nM). By contrast, the chemosensor exhibits a colorimetric and fluorescence enhancement response at 500 nm toward phosgene with fast response (<2 min), high selectivity, and a low detection limit (72 nM). Furthermore, chemosensor-loaded test membrane was fabricated for real-time, portable and efficient discriminative detection of trace amounts of gaseous phosgene and DCP vapor with different optical responses.</P> [FIG OMISSION]</BR>

        A Quantitative Method for Seismic Robustness of RC Frame Considering Resistance Vulnerability of Column and Storey Drift Ratios

        The concept of seismic robustness is proposed by combining the concept of seismic performance and structural robustness. The existing qualitative, quantitative and evaluation methods of seismic robustness are all direct researched on the whole structure, and the influence mechanism of its internal components on the overall seismic robustness is still unclear. It is very important to establish a clear relationship between component design and structural seismic robustness for the structural design, reinforcement design and final evaluation of structural seismic robustness. Based on this, taking the column as the starting point, a quantitative method for the seismic robustness of RC frame by the seismic robustness index is proposed, which takes into account the resistance vulnerability of column and influence of column on the storey drift ratios (SDRs). In which, the resistance vulnerability is represented by the defined control vulnerability coefficient Pimax, and the influence on the SDRs are represented by the storey drift ratio importance coefficients (SDRCs) . The method not only reflects the essential mechanical properties of the column, but also reflects the effects brought about by different SALs. The feasibility of the method is demonstrated by numerical examples of two types of failures (assuming single column and two columns failure), and four optimization design proposals are proposed for it. The analysis shows that  of the target columns to the floor where the target columns located are obviously greater than those on the remaining floors of the target frame. The seismic robustness index R decreases sharply with the increase of the seismic action level (SAL). R is different compare single column failure with two columns failure under 4 SALs. The most effective way to improve the R of the frame under a certain SAL is to retrofit its control column.

        Systemic Reactions to Dust Mite Subcutaneous Immunotherapy: A 3-Year Follow-up Study

        Purpose: The incidence of allergen specific immunotherapy-related systemic reactions (SRs) varies among different studies, and many factors are likely to contribute to SRs. This study aims to investigate the incidence, characteristics, and risk factors of SRs to standardize dust mite-specific subcutaneous immunotherapy (SCIT) in Central China. Methods: All patients receiving standardized dust mites (100-100,000 SQ-U/mL; Alutard SQ, Hørsholn, Denmark) immunotherapy were followed up. Recorded data included demographics, diagnosis, patient status, pulmonary function testing results before and after each injection, allergen dosage, and details of SRs. Results: From June 2011 to August 2014, a total of 208 patients received 4,369 injections; 27 (13.0%) patients experienced 48 (1.1%) systemic reactions. Most of the SRs were grade 2 reactions (n=30, 62.5%), followed by grade 1 (n=11, 22.9%), grade 3 (n=7, 14.6%), and no fatal reactions occurred. Forty-six SRs (95.8%) occurred within 30 minutes. Higher SR rates were associated with high concentration extracts (100,000 SQ-U/mL), injections with concomitant local reactions (LRs), children, asthma and high sensitivity (skin prick test 3+/4+ and/or sIgE≥17.5 kUA/L) (P<0.05). The estimated odds of SRs increased in children (OR=6.57; 95% CI: 1.88-22.97, P=0.003), asthmatic patients (OR=4.10; 95% CI: 1.72-9.80, P=0.002), and injections with LRs (OR=2.41; 95% CI: 1.33-4.36, P=0.004).Conclusions: The incidence of SRs to dust mite SCIT was low, and multiple factors were associated with the increased incidence of SRs. Children, asthmatics and patients with concomitant LR may be prone to develop SRs.

        Novel cross‑switch seven‑level inverter with triple boost capability and self‑balancing

        A novel single-phase seven-level inverter using a cross-switch cell (CSC) is presented in this paper. Compared with conventional inverters, the proposed topology can generate seven-level output, triple boost gain with one dc source and a reduced number of switches. The CSC provides different charging paths for the capacitors to realize self-balance. Hence, the proposed inverter can realize self-balancing without additional control methods or sensors. Moreover, the proposed structure inherently generates negative output voltage levels without a back-end H-bridge circuit, which reduces the voltage stress on switches. This paper has fully illustrated the operation principles, and presents capacitance calculation and loss analysis. Afterward, a comparative study against recently proposed seven-level inverters is introduced, illustrating the merits of the proposed topology in terms of reduced switches, high voltage-boosting capacity, and self-balancing. Ultimately, a laboratory prototype is built for the validation of the proposed inverter.
