        RPTOR methylation in the peripheral blood and breast cancer in the Chinese population

        Background: Altered regulatory-associated protein of mTOR, complex 1 (RPTOR) methylation levels in peripheral blood was originally discovered as breast cancer (BC)-associated risk factor in Caucasians. Objective: To explore the relationship between RPTOR methylation and BC in the Chinese population, we conducted two independent case-control studies. Methods: Peripheral blood samples were collected from a total of 333 sporadic BC cases and 378 healthy female controls for the DNA extraction and bisulfite-specific PCR amplification. Mass spectrometry was applied to quantitatively measure the levels of methylation. The logistic regression, Spearman's rank correlation, and Non-parametric tests were used for the statistical analyses. Results: In our study, we found an association between BC and RPTOR_CpG_4 hypomethylation in the general population (per-10% of methylation, OR 1.29, P = 0.012), and a weak association between BC and RPTOR_CpG_8 hypomethylation in the women with older age (per-10% of methylation, OR 2.34, P = 0.006). We also identified age as a confounder for the change of RPTOR methylation patterns, especially at RPTOR_CpG_4, which represented differential methylation comparing age groups especially in the BC cases (age < 50 years vs age ≥ 50 years by Mann-Whitney U test, P < 0.0001 for BC cases and P = 0.079 for controls). Conclusion: Our study validated the association between hypomethylation of RPTOR and BC risk in the Chinese population also with weak effect and mostly for postmenopausal women. In addition, our findings provided novel insight for the regulation of DNA methylation upon aging or the change of hormone levels.

        Theoretical Analysis of Laser-Locked Spectroscopy Employing a Confocal Fabry-Perot Cavity

        A theoretical analysis of laser-locked enhanced absorption spectroscopy employing a confocal Fary-Perot cavity (CFPC) is presented. The signal-to-noise ratio and the minimum detectable absorbance, which are limited by either the shot noise or the amplitude noise due to the loose laser lock loop, are also discussed in detail. The results show that the effective absorption path length of a CFPC configuration is the same as that of the conventional nonconfocal Fary-Perot cavity configuration, with the CFPC configuration being more convenient to align without deliberate mode matching. Thus, the CFPC configuration should greatly simplify the complications of conventional laser-locked spectroscopy.

      • Nonlinear thermal post-buckling analysis of graphene platelets reinforced metal foams plates with initial geometrical imperfection

        Although some scholars have studied the thermal post-buckling of graphene platelets strengthened metal foams (GPLRMFs) plates, they have not considered the influence of initial geometrical imperfection. Inspired by this fact, the present paper studies the thermal post-buckling characteristics of GPLRMFs plates with initial geometrical imperfection. Three kinds of graphene platelets (GPLs) distribution patterns including three patterns have been considered. The governing equations are derived according to the first-order plate theory and solved with the help of the Galerkin method. According to the comparison with published paper, the accuracy and correctness of the present research are verified. In the end, the effects of material properties and initial geometrical imperfection on the thermal post-buckling response of the GPLRMFs plates are examined. It can be found that the presence of initial geometrical imperfection reduces the thermal post-buckling strength. In addition, the present study indicates that GPL-A pattern is best way to improve thermal post-buckling strength for GPLRMFs plates, and the presence of foams can improve the thermal post-buckling strength of GPLRMFs plates, the Foam- II and Foam- I patterns have the lowest and highest thermal post-buckling strength. Our research can provide guidance for the thermal stability analysis of GPLRMFs plates.

        Theoretical Analysis and Optimization of Extrinsic Fabry-Perot Interferometer Optical-fiber Humidity-sensor Structures

        The theoretical analysis and optimization of extrinsic Fabry-Perot interferometer (EFPI) opticalfiber humidity sensors are deeply investigated. For a typical dual-cavity structure composed of an optical fiber and a humidity-sensitive membrane (HSM), the changes in refractive index (RI) and initial length are discussed for polymer materials and porous oxide materials when relative humidity (RH) increases. The typical interference spectrum is simulated at different RH using MATLAB. The spectral change caused by changing HSM RI and initial length are simulated simutineously, showing different influences on humidity response. To deeply investigate the influence on RH sensitivity, the typical response sensitivity curves for different HSM lengths and air-cavity lengths are simulated. The results show that the HSM is the vital factor. Short HSM length can improve the sensitivity, but for HSM RI and length the influences on sensitivity are opposite, because of the opposite spectral-shift trend. Deep discussion and an optimization method are provided to solve this problem. According to analysis, an opaque HSM is helpful to improve sensitivity. Furthermore, if using an opaque HSM, a short air cavity and long HSM length can improve the sensor’s sensitivity These results provide deep understanding and some ideas for designing and optimizing highly sensitive EFPI fiber humidity sensors.

        Complete Genome Sequence of Salmonella enterica Serovar Pullorum Multidrug Resistance Strain S06004 from China(s)

        As Salmonella enterica serovar Pullorum remains a major economic problem for the poultry industries of countries with no efficient control measures, we presented a multidrug resistance strain S06004 (isolated from a clinically sick chicken in China in 2006) for genome sequencing. The genome comparison showed that the strain contained two prophages, the ST104 and prophage-4 (Fels2) of E. coli LF82, which were not detected in the only published genomes of S. Pullorum RKS5078 and CDC1983-67. In addition, the GyrA Ser83 point mutation, drugresistant genes, and many antibiotic pump systems that are present in S06004 may be contributing to the multidrug resistance of this strain.

        Active noise cancellation frequency‑locked loop with a notch filter

        To improve the performance of frequency-locked loops (FLLs) under distorted grid conditions, a series of pre-filtering techniques have been added to remove harmonics. One of them is the FLL-based comb filter (COMB-FLL), which offers a high disturbance rejection capability. However, it has some disadvantages, such as approximating a fractional delay and compensating an accumulated round-off error in the digital implementation of COMB-FLL. To alleviate these problems, this study proposes an active noise cancellation (ANC) FLL with a notch filter (NF), which incorporates an NF and an ANC to improve the frequency-locking ability. In this research, the structure of a comb filter is simplified into an NF, and an ANC is creatively introduced to eliminate harmonics from the frequency signal obtained by FLL. Furthermore, ANC has been improved to make it suitable for electricity-related applications. The proposed FLL features a unique cascade structure, which has excellent frequency-locking ability and dynamic characteristics under normal, abnormal, and harmonically distorted grid conditions. The simulations and experimental results verify the validity and reliability of the proposed FLL.

        Overexpression of the AtSTK Gene Increases Salt, PEG and ABA Tolerance in Arabidopsis

        AtSTK (At5g02800), which is a serine-threonineprotein kinase gene of Arabidopsis thaliana, was cloned, andits function was studied. The study found that the overexpressionof AtSTK could significantly improve the ability of A. thaliana to tolerate salt, PEG, and ABA stresses. RT-PCRanalysis revealed that the expression of the AtSTK genecould be obviously induced by salt, PEG, and ABA. Theexamination of the physiological characteristics showed thatthe overexpression of AtSTK in Arabidopsis significantlyreduced the plasma membrane permeability, significantlyincreased the proline content, and decreased the MDA content. These changes may reflect the physiological mechanismsthrough which AtSTK overexpression improves stress resistancein Arabidopsis. In addition, the overexpression of the AtSTKgene significantly antagonised the inhibitory effect of highconcentrations of exogenous ABA on Arabidopsis seedgermination. The subcellular localisation results showed thatAtSTK is located in both the cytosol and the nucleus. Theexamination of its tissue-specific expression showed thatAtSTK is expressed in various Arabidopsis tissues and isparticularly strongly expressed in the vessels. The signallingpathway analysis indicated that AtSTK might transfer thesalt stress signal in Arabidopsis through the MAPK pathway.

        Sliding Mode Control for Servo Motors Based on the Differential Evolution Algorithm

        A sliding mode control (SMC) for servo motors based on the differential evolution (DE) algorithm, called DE-SMC, is proposed in this study. The parameters of SMC should be designed exactly to improve the robustness, realize the precision positioning, and reduce the steady-state speed error of the servo drive. The main parameters of SMC are optimized using the DE algorithm according to the speed feedback information of the servo motor. The most significant influence factor of the DE algorithm is optimization iteration. A suitable iteration can be achieved by the tested optimization process profile of the main parameters of SMC. Once the parameters of SMC are optimized under a convergent iteration, the system realizes the given performance indices within the shortest time. The experiment indicates that the robustness of the system is improved, and the dynamic and steady performance achieves the given performance indices under a convergent iteration when motor parameters mismatch and load disturbance is added. Moreover, the suitable iteration effectively mitigates the low-speed crawling phenomenon in the system. The correctness and effectiveness of DE-SMC are verified through the experiment.

        A Novel Approach in Spinal Cord Stimulation for Enhancing Gastric Motility: A Preliminary Study on Canines

        Background/Aims Gastroparesis is commonly seen in patients with diabetes and functional dyspepsia with no satisfactory therapies. Dysautonomia is one of the main reasons for the imbalanced motility. We hypothesized that spinal cord stimulation (SCS) is a viable therapy for gastroparesis via the autonomic modulation to improve gastric motility. The aim is to find an optimal method of SCS for treating gastroparesis. Methods Eight healthy-female dogs were implanted with a gastric cannula, a duodenal cannula, 2 multi-electrode spinal leads, and an implantable pulse generator. Gastric motility index (MI) was used to determine the best stimulation location/parameters of SCS. Optimized SCS was used to improve glucagon-induced gastroparesis. Results With fixed parameters, SCS at Thoracic 10 (T10) was found most effective for increasing gastric MI (37.8%, P = 0.013). SCS was optimized with different parameters (pulse width: 0.05-0.6 msec, frequency: 5-500 Hz, motor threshold: 30-90%) on T10. Our findings revealed that 0.5 msec, 20 Hz with 90% motor threshold at T10 were the best parameters in increasing MI. Glucagon significantly delayed gastric emptying, and this inhibitory effect was partially blocked by SCS. Gastric emptying at 120 minutes was 25.6% in the control session and 15.7% in glucagon session (P = 0.007 vs control), while it was 22.9% with SCS session (P = 0.041 vs glucagon). SCS with the optimal parameters was found to maximally enhance vagal activity and inhibit sympathetic activity assessed by the spectral analysis of heart rate variability. Conclusions SCS with optimized stimulation location and parameters improves gastric motility in healthy-dogs and accelerates gastric emptying impaired by glucagon via enhancing vagal activity.
