To construct transfurmed Bm5 cells, Autographa californica nuclear polyhedrosis virus(AcNPV) IE1 gene, an immediate early viral gene was firstly used in this study. AcNPV IE1 gene, which shares on 95.3% nucleotide sequence homology with Bombyx mori nuclear polyhedrosis virus (BmNPV) IE1 gene, was isolated and cloned into pBluescript. Neomycin gene from pKO-neo was inserted under the control of the IE1 promoter to yield pAcIE1-neo. The plasmid pAcIE1-neo was transfected into Bm5 or Sf9 cells, and neomycin-resistant cells were selected in TC100 medium containing 10% fetal bovine serum (FBS) and 1 mg/ml G4l8 for two weeks. Individual clones were picked and each was amplified for further characterization. The genomic DNA from neomycin-resistant cells was isolated and characterized by PCR using AcNPV IEI gene-specific primers and by Southern blot analysis using neomycin gene probe. We concluded that AcNPV IE1 gene was functional in B. mori-derived Bm5 cells as well as Spodoptera fugjprrda-derived Sf9 cells to produce stably-transformed insect cells
A Fault Location Algorithm Based on Circuit Analysis for Untransposed Parallel Transmission Lines

<P>This paper proposes a fault-location algorithm for ultra-high-voltage untransposed parallel transmission lines that only use the voltages and currents at the local end. The proposed algorithm uses the voltage equation for the faulted phase of the faulted line. The equation contains the fault distance, fault resistance, and fault current. To obtain the fault current, Kirchhoff's voltage law is applied on the loops of three phases consisting of the faulted line and the adjacent parallel line. The fault current can be represented in terms of the fault distance. Inserting the fault current into the voltage equation results in an equation that contains only two parameters (i.e., the fault distance and fault resistance). The fault distance is estimated by solving the equation. Test results indicated that the algorithm accurately estimates the fault distance regardless of the fault resistance and mutual coupling effects.</P>

<P>To our best knowledge, monodispersed beta-Ga2O3 nanospheres were successfully synthesized for first time via morphology-controlled gallium precursors using the forced hydrolysis method, followed by thermal calcination processes. The morphology and particle sizes of the gallium precursors were strongly dependent on the varying (R = SO42-/NO3-) concentration ratios. As R decreased, the size of the prepared gallium precursors decreased and morphology was altered from sphere to rod. The synthesized S2 (R = 0.33) consists of uniform and monodispersed amorphous nanospheres with diameters of about 200 nm. The monodispersed beta-(GaO3)-O-2 nanospheres were synthesized using thermal calcination processes at various temperatures ranging from 500 to 1000 degrees C. Monodispersed beta-(GaO3)-O-2 nanospheres (200 nm) consist of small particles of approximately 10-20 nm with rough surface at 1000 degrees C for 1 h. The UV (375 nm) and broad blue (400-450 nm) emission indicate recombination via a self-trapped exciton and the defect band emission. Our approach described here is to show the exploration of beta-Ga2O3 nanospheres as an automatic dispersion, three-dimensional support for fabrication of hierarchical materials, which is potentially important for a broad range of optoelectronic applications.</P>

Cyclophilins are originally identified as cytosolic binding protein of the immunosuppressive drug cyclosporine A. They have an activity of peptidyl prolyl cis/trans-isomerases (PPIase), which may play important roles in protein folding, trafficking, assembly and cell signaling. In this study, we report the cloning and characterization of a Bombyx mori cyclophilin A (bCypA) cDNA. The full-length cDNA of bCypA consist of 947 nucleotides with a polyadenylation signal sequence AATAAA and contain an open reading frame of 498 nucleotides encoding a polypeptide of 166 amino acids. The deduced amino acid sequence of bCypA shares a central peptidyl prolyl cis/trans-isomerase and a cyclosporin-A-binding domain with other cyclophilin sequences. Relative quantification real-time (RT) PCR analysis shows that mRNA transcripts of bCypA are detected in all the investigated tissues and highest expression level in the skin of 3-day-old 5 instar larva. Also, bCypA had PPIase activity on the proline-containing peptides. Accordingly, we suggest that bCypA is a new member of the cyclophilin A (CyPA) family and will be useful for quality control of bioactivity recombinant proteins with proline-containing peptides.

목적 : 속 산전 염색체 이수성 진단을 위한 미배양 양수세포를 이용한 FISH 검사는 최근 많은 세포유전검사실의 중요한 업무 중의 하나가 되고 있다. 이에 본 저자들은 의뢰된 양수검체 943례에 대하여 산전 염색체 이수성 진단에 있어서 미배양 양수 FISH의 임상적 유용성을 알아보고자 한다. 방법 : 2004년에서 2006년까지 의뢰된 943례의 양수검체에 대하여 염색체 13번, 18번, 21번에 대한 간기 FISH검사를 시행하였고, 산모의 나이, 임신주수와 적응증을 분석하였다. FISH 결과는 고전적 염색체 핵형분석과 비교분석하였다. 결과 : 양수 검체 943례에 대해 염색체 핵형분석을 시행한 결과 45례(4.8%)에서 염색체 이상이 발견되었고 이를 간 기 FISH결과와 비교하였다. 가족성 염색체 상호전좌와 역위 20례를 제외한 염색체 이상이 25례에서 발견되어 2.7%를 차지하였는데 그 중 6례(0.7%)는 FISH로 검출되지 않았고 19례(2.0%)는 FISH로 검출되었다. 핵형분석결과 다운증후군이 14례(Classic형 13례, 전좌형 1례)로 관찰되었다. 에드워드 증후군은 5례로 모두 FISH로 검출되었고 위양성, 위음성은 없었다(특이도와 민감도, 100%). 결론 : 본 연구결과는 FISH검사가 염색체 이수성을 진단하는데 있어 고전적 핵형분석법을 보완할 수 있는 신속하고 예민한 방법이지만 FISH검사로 모든 염색체이상을 검출할 수 없으며 위음성결과를 보일수 있는 FISH 검사의 한계에 대하여 신중한 유전상담이 중요하다고 사료된다. Purpose : Fluorescence in situ hybridization (FISH) on uncultured amniotic fluid cells offers the opportunity for rapid screening of aneuploidies and has become an integral part of the current practice in many clinical cytogenetics laboratories. Here, we retrospectively analyzed the results of interphase FISH in 943 amniotic fluid samples and assessed the efficiency of FISH for rapid detection of aneuploidies. Methods : Interphase FISH for chromosome 13, 18, and 21 was performed in 943 consecutive amniotic fluid samples for rapid diagnosis of aneuploidies referred from 2004 to 2006. Karyotypes from standard cytogenetic analysis were compared to the FISH results. Results : A total of 45 chromosomal rearrangements (4.8%) were found after conventional cytogenetic analysis of the 943 amniotic fluid. After exclusion of known familiar chromosomal rearrangements and inversions (2.1%, 20/943), 2.7% (25/943) were found to have chromosomal abnormalities. Of this group, 0.7% (6/943) were chromosomal abnormalities not detectable by FISH and 2.0% (19/943) were numerical abnormalities detectable by FISH. All 14 cases of Down syndrome (Classic type, 13 cases; Robertsonian type, 1 case) and 5 cases of trisomy 18 were diagnosed and detected by FISH and there were no false-positive or -negative results (specificity and sensitivity=100%). Conclusion : The present study demonstrates that FISH can provide a rapid and sensitive clinical method for prenatal identification of chromosome aneuploidies. However, careful genetic counseling is essential to explain the limitations of FISH, including the inability to detect all chromosomal abnormalities and the possibilities of uninformative or false-negative results in some cases.

Zinc has been considered as a vital constituent of proteins, including enzymes. Mobile reactive zinc (Zn<sup>2+</sup>) is the key form of zinc involved in signal transductions, which are mainly driven by its binding to proteins or the release of zinc from proteins, possibly via a redox switch. There has been growing evidence of zinc’s critical role in cell signaling, due to its flexible coordination geometry and rapid shifts in protein conformation to perform biological reactions. The importance and complexity of Zn<sup>2+</sup> activity has been presumed to parallel the degree of calcium’s participation in cellular processes. Whole body and cellular Zn<sup>2+</sup> levels are largely regulated by metallothioneins (MTs), Zn<sup>2+</sup> importers (ZIPs), and Zn<sup>2+</sup> transporters (ZnTs). Numerous proteins involved in signaling pathways, mitochondrial metabolism, and ion channels that play a pivotal role in controlling cardiac contractility are common targets of Zn<sup>2+</sup>. However, these regulatory actions of Zn<sup>2+</sup> are not limited to the function of the heart, but also extend to numerous other organ systems, such as the central nervous system, immune system, cardiovascular tissue, and secretory glands, such as the pancreas, prostate, and mammary glands. In this review, the regulation of cellular Zn<sup>2+</sup> levels, Zn<sup>2+</sup>-mediated signal transduction, impacts of Zn<sup>2+</sup> on ion channels and mitochondrial metabolism, and finally, the implications of Zn<sup>2+</sup> in health and disease development were outlined to help widen the current understanding of the versatile and complex roles of Zn<sup>2+</sup>.
