        ATP6V0d2 Suppresses Alveoli Macrophage Alternative Polarization and Allergic Asthma via Degradation of PU.1

        Purpose Macrophages are important regulators of environmental allergen-induced airway inflammation and asthma. ATP6V0d2 is a subunit of vacuolar ATPase highly expressed in macrophages. However, the functions of ATP6V0d2 in the regulation of pathogenesis of allergic asthma remain unclear. The aim of this study is to determine the function and related molecular mechanisms of macrophage protein ATP6V0d2 in allergic asthma. Methods We compared the disease severity between female C57BL/6 wild-type and ATP6V0d2−/− mice in an ovalbumin (OVA)-induced asthma model. We also investigated the association of expression of ATP6V0d2, PU.1 and CCL17 with disease severity among asthmatic patients. Results The expression of ATP6V0d2 in sputum cells of asthmatic patients and in the lungs of OVA-challenged mice was enhanced compared to healthy subjects and their counterparts, respectively. However, ATP6V0d2-deficient mice exaggerated inflammatory cell infiltration as well as enhanced alternative activated macrophage (AAM) polarization and mucus production in an OVA-induced asthma model. Furthermore, we found that Atp6v0d2 promoted lysosomal degradation of Pu.1, which induced AAM polarization and Ccl17 production. Among asthma patients, ATP6V0d2 expression was inversely associated with disease severity, whereas PU.1 and CCL17 expression was positively associated with disease severity. Conclusions Our results identify macrophage Atp6v0d2, as an induced feedback inhibitor of asthma disease severity by promoting Pu.1 lysosomal degradation, which may in turn leads to reduced AAM polarization and Ccl17 production.

        Heat transfer characteristics in regenerator cell for gaseous organic compound treatment

        Regenerative combustion technology can efficiently decompose organic gases with high thermal efficiency. This capability is attributed to the regenerator and the periodic gas switching technology. However, published findings regarding the regenerator were inconsistent with some important parameters, and investigations into the regenerative chamber did not provide a comprehensive explanation of the heat transfer characteristics. Therefore, a regenerator cell was investigated in this study. The temperature distribution pattern inside the cell was simulated after model verification. The effects of the superficial velocity, switching time, side width, and wall thickness of the regenerator cell on the outlet temperature, energy recovery ratio, and heat-transfer coefficient were investigated. The outlet temperature, heat transfer, and energy recovery ratio of the regenerator cells varied monotonically during each period. The average energy recovery ratio and heat transfer coefficient indicated that the side width of the regenerator cell was the most significant factor. Meanwhile, the switching time and wall thickness did not significantly affect the energy recovery ratio. The superficial velocity and wall thickness did not significantly affect the heat transfer coefficient.

        Metabolic Syndrome with Hyperglycemia and the Risk of Ischemic Stroke

        Purpose: The association of ischemic stroke and metabolic syndrome (MetSyn) with or without diabetes mellitus (DM) is not clear. The present study aimed to identify the impact of diabetes or hyperglycemia on the risk of MetSyn-associated ischemic stroke. Materials and Methods: This study comprised an Asian population of 576 patients with acute nonembolic cerebral infarction and 500 controls. MetSyn was defined according to the criteria of the International Diabetes Federation. MetSyn patients were further subgrouped according to their glucose levels: MetSyn with DM, MetSyn with impaired fasting glucose (IFG) and MetSyn with normal glucose tolerance (NGT). The impact of MetSyn on cerebral infarction was then evaluated. Results:At baseline, the prevalence of MetSyn in patients with cerebral infarction was higher than that of the controls (57.29% vs. 10.00%, p<0.01). In the stroke group, the prevalences of MetSyn with DM, IFG, and NGT were 25.69%, 8.85% and 22.74%, respectively, all of which were higher than that of the controls (all p-values <0.05). By multiple logistic regression analysis, we discovered that MetSyn was associated with an increased risk of cerebral infarction (odds ratio: 5.73, p<0.01). After adjustment for all the components of MetSyn, the odds ratios of MetSyn with DM, IFG, and NGT were 5.70, 2.24 and 2.19 (all p-values <0.05), respectively. Conclusion: In Asian population, patients with MetSyn accompanied by T2DM are at the greatest risk for acute non-embolic stroke. Additionally, IFG was not observed to be associated with an increased risk for MetSyn-related ischemic stroke.

        Ginsenoside Rg1 ameliorates Alzheimer's disease pathology via restoring mitophagy

        Background: Alzheimer's disease (AD) is a common form of dementia, and impaired mitophagy is a hallmark of AD. Mitophagy is mitochondrial-specific autophagy. Ginsenosides from Ginseng involve in autophagy in cancer. Ginsenoside Rg1 (Rg1 hereafter), a single compound of Ginseng, has neuroprotective effects on AD. However, few studies have reported whether Rg1 can ameliorate AD pathology by regulating mitophagy. Methods: Human SH-SY5Y cell and a 5XFAD mouse model were used to investigate the effects of Rg1. Rg1 (1µM) was added to β-amyloid oligomer (AβO)-induced or APPswe-overexpressed cell models for 24 hours. 5XFAD mouse models were intraperitoneally injected with Rg1 (10 mg/kg/d) for 30 days. Expression levels of mitophagy-related markers were analyzed by western blot and immunofluorescent staining. Cognitive function was assessed by Morris water maze. Mitophagic events were observed using transmission electron microscopy, western blot, and immunofluorescent staining from mouse hippocampus. The activation of the PINK1/Parkin pathway was examined using an immunoprecipitation assay. Results: Rg1 could restore mitophagy and ameliorate memory deficits in the AD cellular and/or mouse model through the PINK1-Parkin pathway. Moreover, Rg1 might induce microglial phagocytosis to reduce β-amyloid (Aβ) deposits in the hippocampus of AD mice. Conclusion: Our studies demonstrate the neuroprotective mechanism of ginsenoside Rg1 in AD models. Rg1 induces PINK-Parkin mediated mitophagy and ameliorates memory deficits in 5XFAD mouse models.

        미성년자 범죄는 국가와 사회가 특히 관심하고 있는 화제 중 하나로서 그 중에서도 미성년자 재산형 범죄가 많은 편으로 여기에 대한 관심과 연구는 미성년자로 하여금 건전한 금전관을 가지게 할 것이며 미성년자 범죄를 효과적으로 억제할 수 있으리라 기대된다. 가정의 영향, 대중매체의 홍보, 법적 규범 등의 미흡한 점은 대부분 미성년자 재산형 범죄의 주요 원인이라는 분석이 있다. 본고에서는 범죄의 예방교육의 시각에서 이러한 원인에 대한 연구를 진행하고자 한다. 교육은 미성년자 범죄에 있어서 가장 중요한 예방으로 가정교육과 더불어 대중매체가 전달하는 가치관은 중요한 역할을 하고 있다. 현대의 미성년자들은 다원적 문화환경에서 생활하고 있으며 물질적으로 생활이 풍부하여 그에 못지않게 여러 유혹들도 다양하다. 그래서 재산형 범죄는 미성년자 범죄 중 자주 발생하는 죄가 되여 있으며 여기에 대한 방도로서는 우선적으로 종합적 범죄 발생의 원인을 찾아내야 할 것이다. 구체적으로는 미성년자의 가정경제상황, 가족관계, 대중매체의 악의적 유도, 플랫폼의 결함, 미흡한 법규 등은 모두가 미성년자의 재산형 범죄를 초래하는 주요 원인임을 본고에서는 지적할 것이다. 그리고 이러한 모든 요인들을 중요시하여 미성년들에게 양호한 환경의 조성과 합리한 법적 규범을 보완하여 미성년자의 재산형 범죄의 예방교육에 조금이나마 도움되기 기원하는 바이다. Juvenile crime has always been a topic of great concern to the country and society, among which juvenile crime against property is the mainstream. Property crime is a frequent crime among juvenile crimes. Paying attention to property crime is conducive to cultivating their correct money concept and effectively restraining juvenile crimes. The influence of family conditions, the noise of mass media and the lack of legal norms are the main reasons for juvenile property crimes. This paper, from the perspective of education, aims at these reasons to carry out preventive research. Education is the fundamental for a minor crime prevention, it not only need to family education, the spread of the mass media constantly values for the youth of this age but also has a very important education meaning, the minor crime prevention law should continue to refine and improve, strengthen the community and school's daily education and education after the sin and punitive measures. Only in this way can we reduce juvenile delinquency effectively.

      • A New Expectation Trust Benefit Driven Algorithm for Grid Environments

        Since the resource of nodes can access and leave the Grid system freely, it is possible some malicious nodes exist in the Grid environment. In addition, the numbers and qualities of nodes’ resource can be changed dramatically and optionally, so these malicious nodes must be affecting the utilization of Grid resource. Hence, the trust management mechanisms are developed and widely used in the resource scheduling for Grid environments. However, most of the trust management models are mainly focus on the nodes historic behaviors and few of them consider the future trust of the node based on its previous activities. This paper proposes a new method to detect the nodes behaviors of resource providers in Grid environments based on D-S theory. Through simulating experiments, the new mechanism could record the behaviors exactly and prevent the malicious ones accessing the Grid system effectively. It would give a strong support for the success of resource scheduling in Grid environments.

        A triage strategy in advanced ovarian cancer management based on multiple predictive models for R0 resection: a prospective cohort study

        Objective: To present the surgical outcomes of advanced epithelial ovarian cancer (AEOC) since the implementation of a personalized approach and to validate multiple predictive models for R0 resection. Methods: Personalized strategies included: 1) Non-invasive model: preoperative clinico-radiological assessment according to Suidan criteria with a predictive score for all individuals. Patients with a score 0–2 were recommended for primary debulking surgery (PDS, group A), or otherwise were counseled on the choices of PDS, neoadjuvant chemotherapy (NAC, group B) or staging laparoscopy (S-LPS). 2) Minimally invasive model: S-LPS with a predictive index value (PIV) according to Fagotti. Individuals with a PIV <8 underwent PDS (group C) or otherwise received NAC (group D). Intraoperative assessment (with Eisenkop, peritoneal cancer index [PCI], and Aletti scores) and surgical results were prospectively collected. Results: Between September 2015 and August 2017, 161 pathologically confirmed epithelial ovarian cancer patients were included. A total of 52 (32.3%) patients had a predictive score of 0–2, and 109 (67.7%) patients had a score ≥3. Among these individuals, 41 (25.5%) patients received S-LPS. Finally, 110 (68.3%) patients underwent PDS (A+C), and 51 (31.7%) patients received NAC (B+D). The R0 resection rates in PDS and NAC patients were 56.4% and 60.8%, respectively. The area under the curve (AUC) of Suidan criteria was 0.548 for group (A+C). The AUC of Fagotti score was 0.702 for group C. The AUC of Eisenkop, PCI, and Aletti scores were 0.808, 0.797, and 0.524, respectively. Conclusion: The Suidan criteria were not effective in these AEOC patients. S-LPS was helpful in decision-making for PDS and should be endorsed in the future.

        Spoilage Lactic Acid Bacteria in the Brewing Industry

        Lactic acid bacteria (LAB) have caused many microbiological incidents in the brewing industry, resulting in severe economic loss. Meanwhile, traditional culturing method for detecting LAB are time-consuming for brewers. The present review introduces LAB as spoilage microbes in daily life, with focus on LAB in the brewing industry, targeting at the spoilage mechanism of LAB in brewing industry including the special metabolisms, the exist of the viable but nonculturable (VBNC) state and the hop resistance. At the same time, this review compares the traditional and novel rapid detection methods for these microorganisms which may provide innovative control and detection strategies for preventing alcoholic beverage spoilage, such as improvement of microbiological quality control using advanced culture media or different isothermal amplification methods.
