CTCF is required for maintenance of auditory hair cells and hearing function in the mouse cochlea

<P><B>Abstract</B></P> <P>Auditory hair cells play an essential role in hearing. These cells convert sound waves, mechanical stimuli, into electrical signals that are conveyed to the brain via spiral ganglion neurons. The hair cells are located in the organ of Corti within the cochlea. They assemble in a special arrangement with three rows of outer hair cells and one row of inner hair cells. The proper differentiation and preservation of auditory hair cells are essential for acquiring and maintaining hearing function, respectively. Many genetic regulatory mechanisms underlying hair-cell differentiation and maintenance have been elucidated to date. However, the role of epigenetic regulation in hair-cell differentiation and maintenance has not been definitively demonstrated. CTCF is an essential epigenetic component that plays a primary role in the organization of global chromatin architecture. To determine the role of CTCF in mammalian hair cells, we specifically deleted <I>Ctcf</I> in developing hair cells by crossing <I>Ctcf</I> <SUP> <I>fl/fl</I> </SUP> mice with <I>Gfi1</I> <SUP> <I>Cre/+</I> </SUP> mice. <I>Gfi1</I> <SUP> <I>Cre</I> </SUP> <I>; Ctcf</I> <SUP> <I>fl/fl</I> </SUP> mice did not exhibit obvious developmental defects in hair cells until postnatal day 8. However, at 3 weeks, the <I>Ctcf</I> deficiency caused intermittent degeneration of the stereociliary bundles of outer hair cells, resulting in profound hearing impairment. At 5 weeks, most hair cells were degenerated in <I>Gfi1</I> <SUP> <I>Cre</I> </SUP> <I>; Ctcf</I> <SUP> <I>fl/fl</I> </SUP> mice, and defects in other structures of the organ of Corti, such as the tunnel of Corti and Nuel's space, became apparent. These results suggest that CTCF plays an essential role in maintaining hair cells and hearing function in mammalian cochlea.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

<P>Sound perception via mechano-sensation is a remarkably sensitive and fast transmission process, converting sound as a,mechanical input to neural signals in a living organism. Although knowledge of auditory hair cell-functions has advanced over the past decades, challenges remain in understanding their biornechatics, partly, because of their :biophysical complexity and the lack of appropriate probing tools. Most current studies of hair cells have been conducted in a relatively low-frequency range (<1000 Hz); therefore, fast kinetic study of hair cells has been difficult, even though mammalians have sound perception of 20 kHz or higher.. Here, Here, we demonstrate that the magnetic force nanoprobe (MFN) has superb spatiotemporal capabilities to mechanically stimulate spatially targeted individual hair cells with a temporal resolution of up to 9 its, which is equivalent to approximately SO :kHz; therefore, it possible. to investigate avian hair cell biomechanics at different tonotopic regions of the cochlea covering a full hearing frequency range of 50 to 5000 Hz. We, found that the variation of the stimulation frequency and amplitude of hair bundles creates distinct mechanical responsive features along the tonotopic axis, where the kinetics of the hair bundle recovery motion exhibits unique frequency-dependent characteristics: basal, middle, and apical hair bundles can effectively respond at their respective ranges of frequency. We revealed that such recovery kinetics possesses two different time constants that are closely related to the passive and active motilities of hair cells. The use of MFN is critical, for the kinetics study of free-standing hair cells in a spatiotemporally distinct tonotopic organization.</P>

This study is about a pattern design on LGP(Light Guide Panel). Using an LED, there is 2 kinds of system to make FFL. One of the systems is direct type which arranges an LED in a PCB substrate. Direct type has higher Light efficiency and more uniformity of illumination distribution then other system, but direct type has dazzling and a problem in a hot spot. To reduce such problems, This paper studied method to put pattern in a LGP upper part.
Construction of Hyper Enhanced Baculovirus Expression Vector System

The baculovirus expression vector system (BEVS) is an effective and widely used method for the production of recombinant proteins in insect cells or larvae. However, the expression efficiency of foreign proteins using the polyhedrin promoter could not obtain the protein yields observed for native polyhedrin. To enhance the production efficiency of foreign protein in baculovirus expression system, the effects of various enhancer factor were investigated by fusion expressing them with the enhanced green fluorescent protein (EGFP). As a hyper expression factor, the optimal hyper BEVS was constructed by combining Hr5 sequence, VP39 promoter and Burst sequences. Additionally, the proteins expressed by the hyper expression system was markedly increased. This study suggests a new option for higher expression of useful foreign recombinant protein using the BEVS.

We report a patient with life-threatening left main coronary artery complication during diagnostic coronary angiography were successfully treated with left main stenting. One year after angioplasty the patient was asymptomatic, the exercise ECG stress test and phamacological stress thallium-201 SPECT was negative for ischemia and follow-up coronary angiography showed no restenosis.
Prevention of DDoS Attacks for Enterprise Network Based on Traceback and Network Traffic Analysis

With the wide usage of internet in many fields, networks are being exposed to many security threats, such as DDoS attack and worm/virus. For enterprise network, prevention failure of network security causes the revealing of commercial information or interruption of network services. In this paper, we propose a method of prevention of DDoS attacks for enterprise network based on traceback and network traffic analysis. The model of traceback implements the detection of IP spoofing attacks by the cooperation of trusted adjacent host, and the method of network traffic analysis implements the detection of DDoS attacks by analyzing the traffic characteristic. Moreover, we present the result of the experiments, and compare the method with other methods. The result demonstrates that the method can effectively detect and block DDoS attacks and IP spoofing attacks.

PURPOSE: To investigate the surgical results of Ahmed valve implantation with intraoperative bevacizumab injection in patients with neovascular glaucoma (NVG). METHODS: A retrospective comparative case series review was conducted on 52 eyes with NVG who underwent Ahmed glaucoma valve implantation with or without intraoperative bevacizumab intravitreal injection. In the intraoperative intravitreous bevacizumab injection group (IVB group, 20 eyes), 1.25 mg of bevacizumab was injected into the vitreous cavity during Ahmed valve implantation. In the control group (32 eyes), only Ahmed valve implantation was performed. Surgical failure was defined when (1) the postoperative intraocular pressure was over 21 mm Hg at consecutive clinic visits, (2) the visual acuity became light perception negative, (3) additional antiglaucomatic surgery was required, or (4) devastating operative or postoperative complications were noted. RESULTS: Although the success rate in the IVB group (70.0%) was higher than that in the control group (62.5%) 1 year after operation, the differences were not statistically significant (P=0.828 by log-rank test). Mean intraocular pressures in the IVB group were significantly lower than those of the control group at 12 and 15 months (P<0.05 by the Mann-Whitney U test). Postoperative complications were similar between the 2 groups. Preoperative history of trabeculectomy was a significant risk factor for surgical failure of Ahmed valve implantation in NVG (relative risk=4.618; P=0.018 by Cox regression model). CONCLUSIONS: Intraoperative IVB injection does not seem to be helpful for better surgical outcomes of Ahmed valve implantation in NVG. A history of trabeculectomy is a risk factor for failure after Ahmed valve implantation in patients with NVG.
Relationship between Growth Factors and Spectral Characteristics of Satellite Imagery in Korea

This study attempts to analyze the relationship between forest volume and age based on 5th NFI data and spectral characteristics of satellite imagery using ASTER sensor in Korea. Forest stand volume and age had the negative correlation with the spectral reflectance in all of the band (Blue, Green, Red, SWIR). With increasing of stand volume and age, spectral reflectance decrease. The spectral reflectance of band1 showed the highest correlation between stand volume and spectral reflectance among the VNIR wavelength. The spectral reflectance band 1, 2 (visible wavelength) and stand age have high correlation compared to other bands. The correlation coefficients between forest volume and vegetation indices have low relationship. This result indicates that the reflectance of blue band may be important factor to improve the potential of optical remote sensing data to estimate forest volume and age.
