        청가시덩굴 추출물의 기능성 원료 표준화를 위한 지표성분 Resveratrol, trans-Scirpusin A의 분석법 개발 및 검증

        본 연구는 HPLC를 이용하여 청가시덩굴 추출물을 개별인정형 건강기능식품의 기능성 원료로 개발하기 위한 원료 표준화의 일환으로, 청가시덩굴 추출물의 지표성분을 resveratrol과 trans-scirpusin A로 설정하고 이에 대한 HPLC 분석법을 확립하여 유효성의 검증을 실시하였다. 분석법 유효성 검증은 특이성, 직선성, 정확도, 정밀도, 검출한계 및 정량한계 등을 통해 분석법의 신뢰성을 검증하였으며, 그 결과 표준용액과 청가시덩굴 추출물 간의 HPLC 크로마토그램 및 UV spectrum의 일치 여부 등의 비교를 통해 다른 물질과 간섭 없이 피크가 분리된 것으로 특이성을 확인하였다. 또한 표준용액 검량선의 상관계수(R²)는 0.9999로 매우 우수한 직선성으로 관찰되어 분석에 적합한 것으로 확인되었으며, 검량선의 기울기 및 표준편차를 이용한 검출한계는 resveratrol이 0.98 μg/mL, trans-scirpusin A는 0.49 μg/mL였고 정량한계는 resveratrol이 2.98 μg/mL, trans-scirpusin A는 1.48 μg/mL로 각각 확인되었다. 청가시덩굴 추출물에 표준물질을 3개 농도 첨가하고 분석한 회수율은 resveratrol이 98.77~99.24%, trans-scirpusin A는 98.45~99.45%로 나타나 정확성이 있는 것을 확인할 수 있었다. 청가시덩굴 추출물의 조제 농도 2.2, 4.4 및 6.6 mg/mL에서 반복성은 resveratrol이 0.99~1.22%, trans-scirpusin A는 1.12~1.32%를, 실험실 내 정밀성에서는 일내 정밀성은 resveratrol이 0.67~0.87%, trans-scirpusin A는 1.18~1.33%로 나타났고 일간 정밀성은 resveratrol이 0.93~1.22%, trans-scirpusin A는 1.33~2.27%로 확인되어 본 분석법은 정밀성이 있음을 확인할 수 있었다. 이상의 분석결과를 통해 확립된 청가시덩굴 추출물의 지표성분인 resveratrol과 trans-scirpusin A의 HPLC 분석법은 적합한 시험법으로 검증되었으며, 본 시험법은 향후 청가시덩굴 추출물의 건강기능식품 기능성 원료 개발과 표준화를 위한 기초자료로 활용될 것으로 사료된다. This study was undertaken to establish an analytical method using high-performance liquid chromatography (HPLC). HPLC for the standard determination of resveratrol and trans-scirpusin A as functional ingredients in Smilax sieboldii extract. We evaluated the specificity, linearity, accuracy, precision, limit of detection (LOD), and limit of quantitation (LOQ) of various analytical methods for detecting resveratrol and trans-scirpusin A using HPLC. The specificity was confirmed by the chromatogram obtained using the HPLC analytical method. Also, the results of UV and the coefficient of correlation (R²) obtained was 0.999, which confirmed that this was a suitable analysis with high linearity. The LOD was 0.98, 0.49 μg/mL, and LOQ was 2.98, 1.48 μg/mL, which was confirmed as a suitable limit level for the analysis of resveratrol and trans-scirpusin A content in the S. sieboldii extract. The recovery of resveratrol and trans-scirpusin A content was determined to be 98.77±0.73∼99.24±1.47% and 98.45±1.18∼99.45±1.66%, respectively, indicating high accuracy. The intra-day repeatability and the intra-laboratory precision of the daily repetition were confirmed to be 0.67∼0.87%, 1.18∼1.33% and 0.93∼1.22%, 1.33∼2.27%, respectively, for trans-scirpusin A, for the relative standard deviation. These results indicate that the reported HPLC method is simple, reliable, and reproducible for the detection of resveratrol and trans-scirpusin A in S. sieboldii extract.

      • A Driving Method of Pixel Circuit Using a-IGZO TFT for Suppression of Threshold Voltage Shift in AMLED Displays

        <P>A driving method of pixel circuit using amorphous indium gallium zinc oxide (a-IGZO) thin-film transistor (TFT) is proposed to improve the image quality of active matrix light-emitting diode displays. The proposed pixel circuit employs a diode-connected structure to compensate for variation in threshold voltage (V-th) of the a-IGZO TFT. In addition, the proposed driving method adopts negative bias annealing to suppress the V-th shift. The annealing time is optimized based on the experimental observation of the minimum V-th shift. After a stress time of 30 000 s, the measurement results showthat the (Vth) shift is reducedby 29.6%, using an optimized annealing time of 5% of one frame time. In addition, the maximum deviation in the emission current using the proposed driving method wasmeasured to be less than 4.32% after a stress time of 30 000 s.</P>

        11β-수산화효소 결핍에 의한 선천성 부신증식증 1예

        저자들은 국내에서 처음으로 음핵 비대와 지속적인 질출혈을 주소로 내원한 19세 여성에서 고혈압이 동반되지 않으면서 11 -수산화효소 결핍으로 인한 선천성부신증식증을 경험하였기에 문헌 고찰과 함께 이를 보고하는 바이다. Congenital adrenal hyperplasia refers to a group of autosomal recessive disorders that is defective in the synthesis of cortisol. The enzymes most often affected are 21-hydroxylase and 11 - hydroxylase. The low levels of cortisol stimulate the pituitary gland to release ACTH. Chronic elevation of the ACTH level causes bilateral adrenal hyperplasia and a secondary increase in androgen formation. We examined a 19 year-old woman presented with clitoral hypertrophy and vaginal spotting. The subjects basal level of serum cortisol was low, but the serum levels of ACTH, 17a-hydroxyprogesterone, deoxy-corticosterone were elevated. The urinary excretions of 17-ketosteroids and 17-hydroxycorticosteroids were also increased. The karyotyping study and transrectal ultrasonography showed normal findings. The patient underwent clitoris reduction surgery and received hydrocortisone. To the best of our knowledge, this is the first case of 11 -hydroxylase deficiency in Korea (J Kor Soc Endocrinol 19:58∼63, 2004).

        Targeted presurgical decompensation in patients with yaw-dependent facial asymmetry

        Facial asymmetry can be classified into the rolling-dominant type (R-type), translation-dominant type (T-type), yawing-dominant type (Y-type), and atypical type (A-type) based on the distorted skeletal components that cause canting, translation, and yawing of the maxilla and/or mandible. Each facial asymmetry type represents dentoalveolar compensations in three dimensions that correspond to the main skeletal discrepancies. To obtain sufficient surgical correction, it is necessary to analyze the main skeletal discrepancies contributing to the facial asymmetry and then the skeletal-dental relationships in the maxilla and mandible separately. Particularly in cases of facial asymmetry accompanied by mandibular yawing, it is not simple to establish pre-surgical goals of tooth movement since chin deviation and posterior gonial prominence can be either aggravated or compromised according to the direction of mandibular yawing. Thus, strategic dentoalveolar decompensations targeting the real basal skeletal discrepancies should be performed during presurgical orthodontic treatment to allow for sufficient skeletal correction with stability. In this report, we document targeted decompensation of two asymmetry patients focusing on more complicated yaw-dependent types than others: Y-type and A-type. This may suggest a clinical guideline on the targeted decompensation in patient with different types of facial asymmetries.

      • Effects of Nicotine on Aβ or CT_(105)-induced Toxicity

        알츠하이머 치매의 병인기전에서 아밀로이드 베타 펩티드는 중요한 역할을 할 것이라는 연구결과가 많이 보고되어있다. 그러나 알츠하이머 치매의 진행과 아밀로이드 베타 펩티드의 생성에는 여러 가지 모순도 보고되었다. 그러므로 아밀로이드 전구단백질에서 생성된 아밀로이드 베타 펩티드 외에 여러 가지 대사물들이 알츠하이머 치매의 병인기전과 관련이 있으리라 여겨진다. 이미 본 연구팀은 니코틴이 신경보호효과가 있다고 알려진 분비형 아밀로이드 전구단백질의 발현을 시간, 농도 의존적으로 증가시킨다는 연구결과를 보고하였다. 이번 논문에서는 니코틴의 전처리로 아밀로이드 베타 펩티드 및 아밀로이드 C단 단백질에 의한 1차 신경세포에서의 세포독성 효과가 억제되며, 이러한 효과는 니코틴의 길항제인 알파붕가로톡신에 의해 상쇄된다는 연구결과를 얻었다. 뿐만 아니라 C단 단백질을 PC12세포에 유전자를 이입시켰을 때, 니코틴에 의하며 세포 독성이 억제되는 결과를 얻었다. 이러한 연구결과로 볼 때, 니코틴이나 니코틴 수용체는 콜린성 신경전달 물질 보상 뿐만 아니라 아밀로이드 베타 펩티드, 아밀로이드 C단 단백질의 독성을 저해함으로써 인지기능 향상에 영향을 줄 것으로 예상된다. Several lines of evidence indicate that Aβ may play an important role in the pathogenesis of AD. However, there are several discrepancies between the production of Aβ and the development of the disease. Thus, Aβ may not be the sole active fragment of β-amyloid precursor protein (βAPP) in the neurotoxicity associated with AD. Previously, findings from our experiments have shown that nicotine enhances the release of APPs, which has neurotrophic and neuroprotective activities in concentration and time-dependent manners. In this study, our results showed that Pretreatment of nicotine (>10μM, for 24hr) partially prevented Aβ or CT_(105)-induced cytotoxicity in primary cultured neuronal cells, and the effects of nicotine-induced protection was inhibited by the pretreatment with a nicotine receptor antagonist α-bungarotoxin. Nicotine (>10μM, for 24hr) partially inhibited CT_(105)-induced cytotoxicity when PC12 cells was transfected with CT_(105). From these results, we proposed that treatment of nicotine or nicotinic receptor agonist might improve the cognitive functions not only by supplementation of cholinergic neurotransmission but also by protecting Aβ-or CT_(105)-induced neurotoxicity probably through the increased release of APPs and the activation of nicotinic receptor.

        한국인 탈색모발에 블루계열 염색 시 지렁이 자가분해물의 처리 효과

        In Korean hair, blue-dyeing causes severe hair damage due to a pre-bleaching step. Due to such damage, color retention period of the hair becomes very short. The prsent study aimed to address this problem, and to this end, samples were prepared as follows: blue-dyed hair (control group), dyed hair after pre-treatment with earthworms autolysate on bleached hair (pre-group), dyed hair with a mixture of dye and earthworms autolysate (mixed group). After blue-dyeing bleached hair once and 3 times, L<sup>*</sup>a<sup>*</sup>b<sup>*</sup> values measured to examine the color retention after shampoo application, at frequency once, 10 times, 20 times. For blue-dyeing, the hair was dyed well, where a<sup>*</sup> was close to ‘0’ in proportion to b<sup>*</sup> as brightness (L<sup>*</sup>) and yellow (b<sup>*</sup>) were relatively low. L<sup>*</sup>a<sup>*</sup>b<sup>*</sup> values were found to be highest in a pre-group when bleached hair (once, 3 times) was blue-dyed and shampooed up to 20 times while the color retention was the best in the pre- and mixed-groups. In regard to the dependence of color retention on shampoo frequency, the pre-group was more than 3 times greater than the control group and mixed-group. In terms of ‘dyeing uniformity’, ‘vividness’, ‘glossiness’ and ‘smoothness’, the pre-treated group was the best for both one-time and 3-times bleached hair. With respect to ‘texture roughness’, the control group was the highest. Regarding ‘hair dye deodorization effects’, the mixed group was the greatest. Just like the spectrophotometer measurement, the pre-treated group was the best at an expert group questionnaire. Since the earthworms autolysate treatment was found to enhance hair coloration and color durability during blue hair dyeing, it is reasonable to say that such treatment is commercially valuable.

        금화규(Abelmoschus manihot) 꽃 추출물의 In Vitro 피부 미백 및 주름 개선 효능 연구

        본 연구는 화장품 소재로서 금화규(Abelmoschus manihot) 꽃의 미백 활성율 및 항주름 활성을 평가하기 위해 수행하였다. 금화규 꽃은 물과 70% 에탄올을 이용하여 추출하였다. Tyrosinase 억제 활성은 물과 70% 에탄올 추출물 모두 농도 의존적으로 억제 활성이 증가 된다고 평가되었으며, B16F10 세포주에서 생존율에 영향을 미치지 않은 농도인 10, 25, 50, 75, 100 μg/mL에서 멜라닌 생합성을 저해하는 것으로 확인되었다. 금화규 꽃의 항주름 효능으로는 collagenase와 elastase의 활성 저해 능력을 측정한 결과 70% 에탄올 추출물이 물 추출물보다 높은 항주름 활성을 보였다. 본 연구 결과를 통하여 금화규 꽃 추출물은 미백 활성과 항주름 활성을 가지는 기능성 화장품 소재로 사용 가능할 것으로 사료된다. In this study, the skin whitening and anti-wrinkle activity of Abelmoschus manihot (A. manihot) flowers were evaluated as a cosmetic material. A. manihot flowers were extracted using water and 70% ethanol. Tyrosinase inhibitory activity was evaluated to increase concentration-dependent inhibitory activity in both water (AMW) and 70% ethanol extracts (AME), and was found to inhibit melanin biosynthesis at concentrations that did not affect survival in B16F10 cell lines at 10, 25, 50, 75, and 100 μg/mL. As a result of measuring the anti-wrinkle effectiveness of A. manihot flowers, 70% ethanol extract (AME) showed higher anti-wrinkle activity than water extract (AMW). Through the results of this study, it is considered that the A. manihot flower extract can be used as a functional cosmetic material with whitening and anti-wrinkle activity.

        Molecular and Clinical Features of Fluconazole Non-susceptible Candida albicans Bloodstream Isolates Recovered in Korean Multicenter Surveillance Studies

        Acquired fluconazole resistance (FR) in bloodstream infection (BSI) isolates of Candida albicans is rare. We investigated the FR mechanisms and clinical features of 14 fluconazole non-susceptible (FNS; FR and fluconazole-susceptible dose-dependent) BSI isolates of C. albicans recovered from Korean multicenter surveillance studies during 2006–2021. Mutations causing amino acid substitutions (AASs) in the drug-target gene ERG11 and the FR-associated transcription factor genes TAC1, MRR1, and UPC2 of the 14 FNS isolates were compared with those of 12 fluconazole-susceptible isolates. Of the 14 FNS isolates, eight and seven had Erg11p (K143R, F145L, or G464S) and Tac1p (T225A, R673L, A736T, or A736V) AASs, respectively, which were previously described in FR isolates. Novel Erg11p, Tac1p, and Mrr1p AASs were observed in two, four, and one FNS isolates, respectively. Combined Erg11p and Tac1p AASs were observed in seven FNS isolates. None of the FR-associated Upc2p AASs were detected. Of the 14 patients, only one had previous azole exposure, and the 30-day mortality rate was 57.1% (8/14). Our data show that Erg11p and Tac1p AASs are likely to contribute to FR in C. albicans BSI isolates in Korea and that most FNS C. albicans BSIs develop without azole exposure.

      • Matrix Metalloproteinase-8 Plays a Pivotal Role in Neuroinflammation by Modulating TNF-α Activation

        <P>Matrix metalloproteinases (MMPs) play important roles in normal brain development and synaptic plasticity, although aberrant expression of MMPs leads to brain damage, including blood–brain barrier disruption, inflammation, demyelination, and neuronal cell death. In this article, we report that MMP-8 is upregulated in LPS-stimulated BV2 microglial cells and primary cultured microglia, and treatment of MMP-8 inhibitor (M8I) or MMP-8 short hairpin RNA suppresses proinflammatory molecules, particularly TNF-α secretion. Subsequent experiments showed that MMP-8 exhibits TNF-α–converting enzyme (TACE) activity by cleaving the prodomain of TNF-α (A<SUP>74</SUP>/Q<SUP>75</SUP>, A<SUP>76</SUP>/V<SUP>77</SUP> residues) and, furthermore, that M8I inhibits TACE activity more efficiently than TAPI-0, a general TACE inhibitor. Biochemical analysis of the underlying anti-inflammatory mechanisms of M8I revealed that it inhibits MAPK phosphorylation, NF-κB/AP-1 activity, and reactive oxygen species production. Further support for the proinflammatory role of microglial MMP-8 was obtained from an in vivo animal model of neuroinflammatory disorder. MMP-8 is upregulated in septic conditions, particularly in microglia. Administration of M8I or MMP-8 short hairpin RNA significantly inhibits microglial activation and expression/secretion of TNF-α in brain tissue, serum, and cerebrospinal fluid of LPS-induced septic mice. These results demonstrate that MMP-8 critically mediates microglial activation by modulating TNF-α activity, which may explain neuroinflammation in septic mouse brain.</P>

        Bioassay-guided isolation and identification of anti-obesity phytochemicals from fruits of Amomum tsao-ko

        Amomum tsao-ko (Zingiberaceae), an important traditional medicinal herb, possesses many biological activities, including anti-inflammatory effects. Though the anti-obesity properties of the crude ethanol extract of A. tsao-ko fruits have been reported, the anti-adipogenic properties of its phytochemical constituents have not been reported. Therefore, in the present study, we isolated the active constituents of A. tsao-ko and investigated their anti-adipogenic effects. The bioassay-guided isolation of the phytochemicals from the ethanol extract of A. tsao-ko fruits identified four bioactive compounds, comprising one fatty acid (1), one sesquiterpene alcohol (2), and two phenolic compounds (3 and 4). Their structures were elucidated by a combination of 1D and/or 2D nuclear magnetic resonance and mass spectrometry. The anti-adipogenic activities of the four compounds evaluated by Oil Red O staining in 3T3-L1 cells revealed that the treatment with the isolated compounds 1 and 3 reduced the lipid accumulation in 3T3-L1 adipocytes more strongly than the compounds 2 and 4, in a dose-dependent manner.
