      • YMCA 체육의 역사적 함의

        이 연구는 역대 YMCA 체육을 토대로 하여 구체적으로 살펴보고자 한다. 이에관해 생활체육 활성화 방안 모색을 제시하는데 논의 및 결론을 도출하였다. 이는 다음과 같이 확인할 수 있다. 첫째, 한국에서의 YMCA운동은 1899년 경성에서 선교활동을 시작되었다. 둘째, YMCA의 전인교육정신인 지(智)·덕(德)·체(體)의 정신과 통하는 정신으로서 비폭력, 민주주의의 실천교육과 상통하는 정신이었다. 셋째, 이상재의 교육사상은 인간의 본질적인 면에 치중하여 올바른 인격체로 양성하는 것으로 지·덕·체의 전인적 교육이었음 알 수 있었다. 넷째, 서울 YMCA의 창립과 역사적 배경을 보면 서울YMCA는 1903년 10월 28일 창립하였다. 다섯째, 서울YMCA를 도입, 회원들에게 지도한 스포츠로는 야구, 농구, 배구, 축구, 유도, 검도, 체조 등이 있었다. 이상과 같이 종합해 볼 때, 역대 YMCA 체육의 역사에서 보이듯이 그 정책을 통해 한국생활체육의 활성화를 위한 방안 모색에 따른 정체성을 제시할 수 있는 것이다. This research is looked at concretely is based on the generation of YMCA physical training. It derive a discussion and conclusion to grope a plan to activate living physical training. This can be confirmed by the following essay. First, the YMCA exercise in Korea started in Gyeong-sun, 1899 as a missionary activity. Second, the mind of former education of YMCA’s just·morality·formality was a mind connected to the communicating with anti-violence, democratic practice education. Third, Lee Sang-Jea’s educational method is to bring up the personality to the right side valued on true nature of a human being, which indicates that his educational method is for the whole man. Fourth, looking at the foundation and historical backround, Seoul YMCA was established on 28th October 1903. Fifth, sports that Seoul YMCA guided to members were baseball, basketball, volleyball, football, judo, kendo, gymnastics etc. To round up all the above statements, the identity following a plan to activate Korea living P.T can be derived through the policy as seen in YMCA physical training’s history in generations.

        플라즈마 정보인자를 활용한 SiO<sub>2</sub> 식각 깊이 가상 계측 모델의 특성 인자 역할 분석

        We analyzed how the features in plasma information based virtual metrology (PI-VM) for SiO2 etching depth with variation of 5% contribute to the prediction accuracy, which is previously developed by Jang. As a single feature, the explanatory power to the process results is in the order of plasma information about electron energy distribution function (PIEEDF), equipment, and optical emission spectroscopy (OES) features. In the procedure of stepwise variable selection (SVS), OES features are selected after PIEEDF. Informative vector for developed PI-VM also shows relatively high correlation between OES features and etching depth. This is because the reaction rate of each chemical species that governs the etching depth can be sensitively monitored when OES features are used with PIEEDF. Securing PIEEDF is important for the development of virtual metrology (VM) for prediction of process results. The role of PIEEDF as an independent feature and the ability to monitor variation of plasma thermal state can make other features in the procedure of SVS more sensitive to the process results. It is expected that fault detection and classification (FDC) can be effectively developed by using the PI-VM.

        Comparison of SARS-CoV-2 Antibody Responses and Seroconversion in COVID-19 Patients Using Twelve Commercial Immunoassays

        Background: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) antibody assays have high clinical utility in managing the pandemic. We compared antibody responses and seroconversion of coronavirus disease 2019 (COVID-19) patients using different immunoassays. Methods: We evaluated 12 commercial immunoassays, including three automated chemiluminescent immunoassays (Abbott, Roche, and Siemens), three enzyme immunoassays (Bio-Rad, Euroimmun, and Vircell), five lateral flow immunoassays (Boditech Med, SD biosensor, PCL, Sugentech, and Rapigen), and one surrogate neutralizing antibody assay (GenScript) in sequential samples from 49 COVID-19 patients and 10 seroconversion panels. Results: The positive percent agreement (PPA) of assays for a COVID-19 diagnosis ranged from 84.0% to 98.5% for all samples (>14 days after symptom onset), with IgM or IgA assays showing higher PPAs. Seroconversion responses varied across the assay type and disease severity. Assays targeting the spike or receptor-binding domain protein showed a tendency for early seroconversion detection and higher index values in patients with severe disease. Index values from SARS-CoV-2 binding antibody assays (three automated assays, one LFIA, and three EIAs) showed moderate to strong correlations with the neutralizing antibody percentage (r=0.517–0.874), and stronger correlations in patients with severe disease and in assays targeting spike protein. Agreement among the 12 assays was good (74.3%–96.4%) for detecting IgG or total antibodies. Conclusions: Positivity rates and seroconversion of SARS-CoV-2 antibodies vary depending on the assay kits, disease severity, and antigen target. This study contributes to a better understanding of antibody response in symptomatic COVID-19 patients using currently available assays.

        Expression and prognostic significance of SIRT1 in ovarian epithelial tumours

        <P>AIMS: Sirtuin1 (SIRT1) is a nicotinamide adenine dinucleotide-dependent deacetylase. Recently, some studies have suggested that SIRT1 could be over-expressed in breast, prostate and colon cancers and up-regulated SIRT1 inactivates p53 by deacetylation. Therefore, we investigated the prevalence and the prognostic impact of SIRT1 and p53 expression in ovarian epithelial tumours. METHODS: Immunohistochemical expression of SIRT1 and p53 were evaluated using tissue microarray in 40 cases of benign epithelial tumours, 36 cases of borderline tumours, and 90 cases of malignant tumours. RESULTS: Expression of SIRT1 was significantly increased in malignant epithelial tumours compared to benign and borderline epithelial tumours (p < 0.001). In particular, a high proportion of serous carcinoma expressed SIRT1 (86%, 55/64 cases). Despite the frequent expression of SIRT1 in malignant ovarian epithelial tumours, serous carcinomas of high FIGO stage showed less frequent SIRT1 expression compared to that of low stage serous carcinomas (p = 0.029). Moreover, increased expression of SIRT1 in serous carcinoma correlated with increased overall survival by univariate (p = 0.014) and multivariate analyses. CONCLUSIONS: Over-expression of SIRT1 may play an important role in the early stage of ovarian carcinogenesis.</P>

      • Comparison of the efficacy of allogenic dermal fibroblasts and platelet-rich plasma for repaired tendon-to-bone healing in a rabbit model of chronic rotator cuff tear

        Introduction and Background The purpose of this study was to compare the efficacy between allogenic dermal fibroblasts (ADFs) and platelet-rich plasma (PRP) for the tendon-to-bone healing in a rabbit model of a chronic rotator cuff tear. Material and Method A total of 32 rabbits were randomly allocated into four groups (n=8 each). In two groups, torn supraspinatus tendons which were detached and left for 6 weeks were repaired in a transosseous manner with the injection of 5x10<sup>6</sup> ADFs with fibrin on left shoulder and PRP with fibrin on right shoulder (Figure 1 and 2). The expression (relative ratio to control) of genes including type 1 collagen (COL1), type 3 collagen (COL3), bone morphogenic protein-2 (BMP-2), scleraxis (SCX), SOX9, and aggrecan (ACAN) was assessed at 4 weeks (Group A), and at 12 weeks (Group B) after repair. Same surgical procedures were performed with ADFs injection on both shoulder in Group C, and PRP injection in Group D for the histological and biomechanical evaluations of tendon-to-bone healing at 12 weeks after repair, Results Seven rabbits died before the evaluation (Group A,B,C,D;n=2,1,2,2, respectively). At four weeks after repair, mRNA expression of COL1 and BMP-2 was higher in ADFs injected shoulders (1.6±0.8 and 1.0±0.3, respectively) than PRP injected shoulders (1.0±0.3 and 0.6±0.3; p=0.019 and 0.013, respectively). At 12 weeks after repair, all genes did not show differences between ADFs and PRP injected shoulders (all p>0.05). In the histologic evaluation, the collagen continuity and maturation of tendon-to-bone interface were better in Group C than Group D (p=0.008, and 0.048, respectively) at 12 weeks after repair. The mean load-to-failure resulted 32.3±6.2 N/kg in Group C, and 24.4±5.2 N/kg in Group D at 12 weeks after repair (p=0.015). Conclusions ADFs seems to have better enhancement of healing than PRP in repaired tendon of chronic rotator cuff tear model of rabbits.

      • Inhibitory activity of isolated components from Peucedanum japonicum Thunb on SARS-CoV-2 papain-like protease

        Papain-like protease(PLpro) is one of the target enzymes to develop SARS-CoV-2 antiviral drug. The aim of the our study is to find PLpro inhibitor from Peucedanum japonicum Thunb. Ethanol extract of the roots of P. japonicum was chromatographied by silica gel and C-18 column chromatographies to obtain two compounds 1 and 2. These compounds were elucidated as khellactone (1) and (+)-3"-acetoxy-4"-(2-methylbutyroyl)khellactone (2) by analyzing 1H- and 13C-NMR spectra. Their inhibitory activity on PLpro was evaluated in the difference in the amount of the corresponding product derived in the substrate Z-RLRGG-AMC acetate. Two showed, in the doese-dependent manner, the inhibitory activity with IC50 values of 35.5±1.2 and 42.9±0.9 μM, respectively. Finally, two inhibitors (1 and 2) were eligible for the lead compounds to develop PLpro protease inhibitor.

        Our study aims to find papain like protease(PLpro) inhibitors from Sophora flavescens. The methanol extracts of the S. flavescens roots was subjected to column chromatography with silica gel, C-18, and sephadex LH-20 resins to achieve six compounds 1-6. Their structures were elucidated by analyzing the spectroscopic signals, and the six compounds were identified as kurarinone (1), sophoraflavanone G (2), kushenol B (3), maackiain (4), kushenol A (5), and isoxanthohumol (6). They were found to be 83.2, 106.0, 101, 97.1, -5, and 28.9% inhibitory activities on PLpro at 100 μM concentration, respectively. Inhibitors 1-4 were calculated, in dose dependent manner, to have IC50 values with 17.9. 13.6, 8.0 and 21.2 μM, respectively. Therefore, the potential inhibitor (3) could be possibly used as new PLpro inhibitor.

        Insulin Resistance as a Risk Factor for Gallbladder Stone Formation in Korean Postmenopausal Women

        Background/Aims: The objective of this study was to determine whether insulin resistance is associated with gallbladder stone formation in Korean women based on menopausal status. Methods: The study included 4,125 consecutive Korean subjects (30-79 years of age). Subjects who had a medical history of diabetes, hypertension, dyslipidemia, other cardiovascular disorders, or hormone replacement therapy were excluded. The women were subdivided into two groups according to their menopausal status. Results: Analysis of premenopausal women showed no significant differences in the homeostasis model of assessment-insulin resistance (HOMA-IR) index between the two groups in terms of gallstone disease. The associations between the occurrence of gallbladder stones and age, obesity, abdominal obesity, hyperinsulinemia, and high HOMA-IR index were statistically significant in the analysis with postmenopausal women. In a multiple logistic regression analysis, low high density lipoprotein-cholesterol was an independent predictor of gallbladder stone formation in premenopausal women. However, the multiple logistic regression analysis also showed that age and HOMA-IR were significantly associated with gallbladder stone formation in postmenopausal women. In an additional analysis stratified by obesity, insulin resistance was a significant risk factor for gallbladder stone formation only in the abdominally obese premenopausal group. Conclusions: Insulin resistance may be associated with gallbladder stone formation in Korean postmenopausal women with abdominal obesity.

        Protective effect of RIPK1-inhibitory compound in <i>in vivo</i> models for retinal degenerative disease

        <P><B>Abstract</B></P> <P>Receptor interacting protein kinase 1 (RIPK1) plays a key role in necroptosis, which is a type of programmed necrosis that is involved in ocular diseases, including glaucoma and dry age-related macular degeneration (AMD). We previously introduced RIPK1-inhibitory compound (RIC), which has biochemical characteristics and a mode of action that are distinct from those of the prototype RIPK1 inhibitor necrostatin-1. The intraperitoneal administration of RIC exerts a protective effect on retinal ganglion cells against a glaucomatous insult. In this study, we examined the protective effect of RIC on retinal pigment epithelium (RPE) against sodium iodate (SI) insult, which is associated with dry AMD pathogenesis. The eye drop administration of RIC that reached on the retina prevented RPE loss in SI-induced retinal degeneration. RIC consistently demonstrated retinal protection in the funduscopy and electroretinogram analyses in SI-injected rabbits and iodoacetic acid-treated mini-pigs. Moreover, the <I>in vivo</I> protective effects of RIC were superior to those of ACU-4429 and doxycycline, which are other medications investigated in clinical trials for the treatment of dry AMD, and RIC did not induce retinal toxicity following topical administration in rats. Collectively, RIC displayed excellent retinal penetration and prevented retinal degeneration in the pathogenesis of dry AMD with a high <I>in vivo</I> efficacy.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The eye drop administration of RIPK1-inhibitory compound (RIC) prevents RPE loss in <I>in vivo</I> models of dry AMD. </LI> <LI> RIPK1-dependent necrosis, necroptosis, might be a plausible target for treatment of dry AMD. </LI> </UL> </P>
