        Antiviral Efficacy of a Short PNA Targeting microRNA-122 Using Galactosylated Cationic Liposome as a Carrier for the Delivery of the PNA-DNA Hybrid to Hepatocytes

        Peptide nucleic acids (PNAs) that bind to complementary nucleic acid sequences with extraordinarily high affinity and sequence specificity can be used as antisense oligonucleotides against microRNAs, namely antagomir PNAs. However, methods for efficient cellular delivery must be developed for effective use of PNAs as therapeutic agents. Here, we demonstrate that antagomir PNAs can be delivered to hepatic cells by complementary DNA oligonucleotide and cationic liposomes containing galactosylated ceramide and a novel cationic lipid, DMKE (O,O'-dimyristyl-N-lysyl glutamate), through glycoprotein-mediated endocytosis. An antagomir PNA was designed to target miR-122, which is required for translation of the hepatitis C virus (HCV) genome in hepatocytes, and was hybridized to a DNA oligonucleotide for complexation with cationic liposome. The PNA-DNA hybrid molecules were efficiently internalized into hepatic cells by complexing with the galactosylated cationic liposome in vitro. Galactosylation of liposome significantly enhanced both lipoplex cell binding and PNA delivery to the hepatic cells. After 4-h incubation with galactosylated lipoplexes, PNAs were efficiently delivered into hepatic cells and HCV genome translation was suppressed more than 70% through sequestration of miR-122 in cytoplasm. PNAs were readily released from the PNA-DNA hybrid in the low pH environment of the endosome. The present study indicates that transfection of PNA-DNA hybrid molecules using galactosylated cationic liposomes can be used as an efficient non-viral carrier for antagomir PNAs targeted to hepatocytes.

      • MESSAGE TYPE STRATEGY IN CHARITABLE FUNDRAISING

        In appeals for donations, some charities highlight why their programs are important or abstract meanings that individuals' contributions can have. On the other hand, some other charities highlight how their programs are implemented or specific actions that are to be made. Drawing on construal level theory, we posit that abstract, “why” laden appeals will be more persuasive than concrete, “how” laden appeals when psychological distance (via physical distance) is high (vs. low). One experiment was conducted to test this premise. Specifically, we examined whether the location where the programs of a nonprofit organization were run was far (i.e., a foreign country) or near (i.e., domestically) would moderate the persuasiveness of abstract versus concrete messages. In terms of the procedure, participants were first asked to read a message from a nonprofit organization helping children suffering from rare diseases in their own country or in foreign countries. Then, they indicated the extent to which they were willing to help the nonprofit. To manipulate the message type, we varied the headline as well as the content of the message so as to make on version construed at a low level and the other at a high level. Further, to investigate a moderating role of disease types, we measured the changes in response to when disease names are suggested more specifically due to the variation in the degree of familiarity to the charity subject. The results of this experiment supported our prediction on the fit effect between physical distance and message type. To be specific, when the appeal was for a foreign cause, abstract messages generated a higher willingness to help the nonprofit. Conversely, when the appeal was for a local cause, concrete messages generated a higher willingness to help the nonprofit. These results indicate construal fit effect in a charitable giving context. Furthermore, we investigated a moderating role of disease types given that such fit effects can be stronger among low- (vs. highly) familiarity of disease. We observed a significant three-way interaction involving the degree of familiarity.

      • Filamin is essential for shedding of the transmembrane serine protease, epithin

        <P>Epithin is a type II transmembrane serine protease that exists in a soluble and membrane-bound form. Shedding is thought to be important in regulating its action, but little is known regarding the intracellular events that trigger such shedding. Here, we show that phorbol myristate acetate (PMA) causes the release of epithin. It also causes accumulation of the protein at the site of cell-cell contacts, and this accumulation is dependent on the formation of cortical actin. In addition, we have identified the actin-binding protein, filamin, as the linker between epithin and the actin cytoskeleton. The interaction of epithin and filamin was enhanced by PMA, and epithin was not released from filamin-deficient M2 cells. We also show that the release of epithin does not require its own activity and is blocked by a metalloprotease inhibitor, GM6001. These results show that filamin has an essential role in shedding by linking epithin to the as yet unidentified metalloprotease-shedding enzyme(s).</P>

      • 클러스터링 방법을 이용한 칼라 영상 분할

        영상분할(image segmentation)은 주어진 영상을 서로 비슷한 성질을 지니는 지역들로 나누는 과정을 말한다. 본 연구에서는 칼라영상에서 얻을 수 있는 빨강(red), 초록(green), 파랑(blue) 성분을 이용하여 얻어진 2차원 누적분포도(histogram)를 분석하여 칼라영상 내에 존재하는 적절한 클러스터의 갯수와 각 클러스터들의 초기 중심값을 구한 후, K-means 방법을 이용하여 영상을 분할하였다. 클러스터링(clustering) 방법을 이용하여 영상을 분할할 경우, 영상 내에 존재하는 클러스터(cluster)의 갯수를 적절히 결정하는 것이 중요한 문제가 된다. 또한, K-means 방법이나 Isodata 방법인 경우에는 그 결과가 초기에 선택된 클러스터의 중심값에 커다란 영향을 받는다. 본 연구에서는 2차원 누적분포도에 스케일-스페이스 필터링(scale-space filtering)을 적용하여 이러한 문제를 해결하였다.

        촉감을 활용한 체험 공간의 구조에 관한 연구

        (연구배경 및 목적) 학문적 영역에서 시각 중심적 사고에 비해 비 시각 감각은 그동안 큰 주목을 받아오지 못했다. 그러나 비 시각 감각은 과거 인간의 삶과 소통에 있어 중심적인 역할을 해왔던 만큼 그 중요성은 강조될 필요가 있다. 그 중에서 피부를 통해 인지하는 촉각은 인간이 태어나면서 최초로 느끼는 감각으로, 시각을 포함한 모든 감각들을 확장 시키는 중요한 감각이다. 최 근 들어 촉각에 대한 인식이 나아지면서 촉각적 경험을 바탕으로 형성된‘촉감’에 대한 연구가 활발하다. 그에 따라 다양해진 인간의 기호와 욕구를 충족시키기 위해 대상과의 직접적인 접촉을 유도하는‘체험’을 중심으로 하는 공간 전략이 부각되고 있다. 따라서 본 연구는 촉감을 활용한 체험 공간의 특성을 파악하고 그것을 유형화 하는데 목적이 있다. (연구방법) 먼저 대부분의 촉감의 인지는 피막을 통해 이루어지는 만큼 피부가 가지는 성질과 역할과 대해 알아보고 촉감의 개념을 정리했다. 다음은 이론적 고찰과 담론을 바탕으로 촉감이 가지는 주요한 3가지 특성을 파악한 후 촉지각과 감각온도, 시각적 촉감으로 분류했다. 또한 촉감이 활용된 체험 공간에서 나타나는 특징을 3개의 유형으로 정리하고 각 유형에 따른 속성 3가지씩 총 9개를 도출해 낸 결과 감촉체험(기억·지각·반응), 시간체험(과거·순간·가상), 소유체험(신뢰·애착·쾌락)으로 제시했다. 앞서 분석한 촉감의 특성과 체험 유형을 토대로 분석 매트릭스를 설정하고 다양한 매체의 복합적인 체험이 가능한 2000년 이후의 전시 공간을 범위로 선정하여 사례 연구를 진행했다. (결과) 종합적 고찰을 통해 앞서 분류한 촉감의 특성을 기준으로 각각의 모든 사례에서 두 개 이상의 특성을 복합적으로 체험한다는 결과를 도출했다. 여기서 주목할 점은 직접적인 피부의 접촉을 통한 촉지각보다 덜 직 접적인 촉감의 특성이라고 할 수 있는 감각온도와 시각적 촉감을 통한 체험의 빈도수가 더 높았다는 것이다. 이들을 통한 체험은 직접적인 피부의 접촉을 통해 물체의 질감을 느끼고 판단하는 것에서 멈추지 않고 실제공간이 아닌 과거의 어떤 순간이나 현실에서는 경험하지 못할 가상공간에서의 체험을 가능하게 했다. (결론) 오늘날 촉감의 역할이 단순히 판단을 위한 것에 그치지 않고 감각적 사고와 상상력을 촉발하는 수단으로 작용함에 따라 그에 부응할 수 있는 체험 공간들이 많이 생겨나는 추세이다. 다양한 매체들의 출현은 이에 중요한 수단이 되었고 인간이 공간과 상호작용하며 소통할 수 있는 체험의 장을 마련하는데 큰 역할을 하고 있다. 앞으로의 촉감을 활용한 체험 공간의 연구에 있어 고려해야 할 요소와 방향성을 검토하는 데 근거를 제공함에 본 연구의 의의를 가진다. (Background and purpose) Non-visual senses have not been noticed more than visual thinking in disciplines. However, non-visual senses should be emphasized as central to human life and communication. Among them, the tactile sense is the primal sense of humans at birth; it is the crucial sense that extends all senses, including vision. Recently, active research regarding the “haptic sense” has been conducted based on tactile experiences. Accordingly, spatial strategies based on experiences that induce direct contact with objects to fulfill various human tastes and needs are prominent. Thus, this study intends to identify and typify haptic experiential spaces. (Method) The first focus of this research was a review of the skin’s characteristics and roles to summarize the concept of the haptic sense, given that haptic perception is typically based on membranes. The next focus was on identifying and classifying three main qualities of the haptic sense based on theoretical investigations and discourses: tactile perception, effective temperature, and visual hapticity. The characters of haptic experiential space were typified into three, and three attributes of each of these were derived; thus, there were nine attributes ultimately identified: touching experience (memory, perception, reaction); temporal experience (past, moment, imagery); and possessive experience (trust, naffection, pleasure). With the analysis model founded on the haptic qualities and types of experiences described above, case studies of exhibition spaces have been conducted since 2000 to examine the complex experiences associated with various media. (Results) By comprehensive investigation, all cases showed complex experiences of two or more of the haptic qualities classified above. It should be noted that higher frequencies of such experiences occurred with effective temperatures and visual hapticity, which are probably less direct qualities of the haptic sense than the tactile experience. The experiences through the former two qualities went beyond simply feeling and judging the textures of objects through direct skin contact toward enabling experiences in virtual space, rather than in the past or present reality. (Conclusions) As the roles of the haptic sense go beyond simple judgments to sensuous thinking and imagination, many experiential spaces that reflect such changes tend to emerge. The advent of various media became an important means for them, playing the crucial role of preparing the experiential field in which humans can interact with space. The significance of this study is in providing the grounds to review the elements and direction of future research on haptic experiential space.

        포크너와 타자의 냄새

        This essay investigates how William Faulkner presents the olfactory experiences through which his characters recognize others as a process of othering. The stereotyped smell in his novels is revealed not as a reason for hatred but rather as a product. The “negro smell,” Faulkner frequently uses in his novels, is a part of “racial formation,” which occurs through the interactions between social structure and the significations of it in everyday lives. Southern white people in the Reconstruction and Jim Crow era attempted to construct the identity of whiteness by demarcating the color bar. The “negro smell” in Light in August, Go, Down Moses, and Intruder in the Dust is a sign of the parasitical nature of “pure” whiteness, which is only possible through fabricating the identity of African Americans as a “pollutant.” The forced segregation causes shame and anxiety in Faulkner’s characters because racial formation is a fundamentally ideological process that tries to delineate a factitious demarcation. In Intruder in the Dust, Faulkner unfolds the olfactory experience around “black smells’’ into a complicated process within which we should find the “idea” about the smells of others, the “conditions” of the residential inequalities, and social practices termed “acceptance” that solidify ideologies. In some other novels and films, the stereotyped smell of others and people’s hatred of such smell provoke resistance against the discrimination within the social hierarchy. Unlike these efforts, Faulkner’s contemplations on the stereotyped smell of others help one penetrate the constructiveness of the discrimination to challenge the social hierarchy itself.

        <P>We propose a facile fluorometric system for detection of gene mutations using graphene oxide (GO).
