      • Emodin Sensitizes Hepatocellular Carcinoma Cells to the Anti-Cancer Effect of Sorafenib through Suppression of Cholesterol Metabolism

        <P>Reduced therapeutic efficacy of sorafenib, a first-generation multikinase inhibitor, is often observed during the treatment of advanced hepatocellular carcinoma (HCC). Emodin is an active component of Chinese herbs, and is effective against leukemia, lung cancer, colon cancer, pancreatic cancer, and HCC; however, the sensitizing effect of emodin on sorafenib-based HCC therapy has not been evaluated. Here, we demonstrate that emodin significantly improved the anti-cancer effect of sorafenib in HCC cells, such as HepG2, Hep3B, Huh7, SK-HEP-1, and PLC/PRF5. Mechanistically, emodin inhibits sterol regulatory element-binding protein-2 (SREBP-2) transcriptional activity, which suppresses cholesterol biosynthesis and oncogenic protein kinase B (AKT) signaling. Additionally, attenuated cholesterol synthesis and oncogenic AKT signaling inactivated signal transducer and activator of transcription 3 (STAT3), an oncogenic transcription factor. Furthermore, emodin synergistically increased cell cycle arrest in the G1 phase and apoptotic cells in the presence of sorafenib. Animal models xenografted with HepG2 or SK-HEP-1 cells also showed that the combination of emodin and sorafenib was sufficient to inhibit tumor growth. Overall, these results suggested that the combination of emodin and sorafenib may offer a potential therapy for patients with advanced HCC.</P>

        Antimicrobial effect of phytochemicals to Listeria monocytogenes isolated from slaughterhouses

        본 연구에서는 도축장에서 분리된 Listeria monocytogenes를 저해할 수 있는 식물성 항균 물질을 탐색하고, 각 항균 물질의 최소살균농도(MBC)를 확인하였다. 잠재적인 항균 물질로 자몽종자 추출물, 감귤류 추출물, 생강 추출물, 배 추출물, 매실 농축액, 도라지 추출물, 대추 추출물, 오미자 추출물을 선정하였고, 대상 균주로는 국내 도축장에서 분리한 15 개의 L. monocytogenes 균주를 혼합하여 사용하였다. MBC 확인 결과, 최대 4.0 μg/mL 농도의 생강 추출물, 배 추출물, 매실 농축액, 도라지 추출물, 대추 추 출물, 오미자 추출물은 고농도의 L. monocytogenes (7 Log CFU/mL)에 대해 균 저해 효과가 전혀 나타나지 않았다. 반면, 7 Log CFU/mL의 L. monocytogenes에 대한 자몽종자 추출물과 감귤류 추출물의 MBC는 0.002 μg/mL인 것으로 확인되었다. 3 Log CFU/mL의 L. monocytogenes에 대한 MBC를 시험해 본 결과, 자몽종자 추출물과 감귤류 추출물의 MBC는 0.001 μg/mL로 나타났다. 이상의 결과로 볼 때 자몽종자 추출물과 감귤류 추출물은 L. monocytogenes를 제어하기 위해 적용될 수 있을 것으로 판단된다. This objective of this study was to investigate inhibitory effect of natural antimicrobials on Listeria monocytogenes isolated from Korean slaughterhouses. A mixture of 15 strains of L. monocytogenes at low (3 Log CFU/mL)and high (7 Log CFU/mL) concentration was exposed to various extracts (grapefruit seed extract, citrus fruits extract, ginger extract, pear extract, Japanese apricot concentrate, balloon flower extract, jujube extract, and omija extract) at 0.001-4.0μg/mL . Ginger extract, pear extract, Japanese apricot concentrate, balloon flower extract, jujube extract, and omija extract showed no antimicrobial effects to high-concentration of L. monocytogenes (7 Log CFU/mL). However, grapefruit seed extract and citrus fruit extract showed antibacterial effects against L. monocytogenes at 3 and 7 Log CFU/mL with MBCs of 0.001 and 0.002 μg/mL, respectively. These results indicate that grapefruit seed extract and citrus fruits extract can be used to control L. monocytogenes as natural antimicrobials.

        Prevalence of Pediatric Autism Spectrum Disorder in Korea Using National Statistics Including the COVID-19 Pandemic, 2011 to 2021

        Purpose: Regularly analyzing the prevalence of autism spectrum disorder (ASD) is important to inform policies and strategies for proper management. The present study aimed to esti-mate trends in ASD prevalence according to age and time in Korean children. Methods: We monitored the annual prevalence of ASD in children aged 2 to 18 years based on information from Statistics Korea and the Korean National Health Insurance Service. We estimated changes in prevalence between 2011 and 2021 after stratifying partici-pants into three age groups. Furthermore, we analyzed the prevalence of ASD by birth year. Results: The highest level of prevalence appeared in children aged 6 years, and thereafter, it de-clined with age. The prevalence of ASD in children aged 2 to 18 years increased from 0.05% in 2011 to 0.12% in 2021. During the same time, the prevalence in preschool children (2 to 5 years) remarkably increased from 0.04% in 2011 to 0.12% in 2021, while the prevalence in adolescents (13 to 18 years) increased from 0.05% in 2011 to 0.09% in 2021. The prevalence of ASD notably increased during the coronavirus disease 2019 (COVID-19) pandemic (2020 to 2021). Among children born between 2011 and 2015, the prevalence of ASD was higher for children with recent birth years, especially during the COVID-19 pandemic. Conclusion: The overall ASD prevalence in children and adolescents in Korea seems to be increasing, especially in preschool children during the COVID-19 pandemic. Early and more effec-tive interventions are necessary for Korean children.

        어성초 메탄올 추출물로부터 항산화 효능을 가진 활성물질의 확인

        본 연구는 어성초의 메탄올 추출물을 이용하여 항산화 효능이 가장 좋은 fraction을 찾고, 항산화 효능을 나타내는 성분을 분석하였다. 메탄올 추출에 의한 유기 용매 별 분획에서 항산화 효과가 가장 좋은 ethyl acetate 분획물을 칼럼 크로마토그래피하여 12가지의 fraction 중 가장 높은 항산화 효과를 보인 Fr. 10을 이용하여 DPPH 라디칼의 소거활성, 환원력, 지질과산화, 세포독성, DNA 산화 및 DCFH-DA를 이용한 세포내 과산화수소 제거효과를 조사하였다. DPPH radical scavenging activity, reducing power, TBARS, cell viability, DNA oxidation and DCF fluorescence 12가지의 fraction들 중 항산화 효능이 가장 좋은 Fr. 10의 DPPH radical 소거 활성 결과로 64 μg/ml 농도에서 양성대조군에 근접하는 60%의 억제능을 보였다. Reducing power 결과로 32 μg/ml의 농도에서 140%로 양성대조군과 비슷한 결과 값을 보였다. TBARS 결과로 2 μg/ml에서 양성대조군과 같은 값의 활성산소 억제능이 나타난다. 또한 cell viability 결과로 32 μg/ml 이상의 농도에서 세포 독성에 의해 생존율이 감소하였고, DCF fluorescence 결과로 농도의존적으로 H2O2에 의한 산화적 손상을 억제하는 것으로 나타났다. DNA oxidation 실험결과 1 μg/ml 이상의 농도에서 DNA의 손상을 감소시킴을 확인할 수 있었다. IR 및 LC-MS를 이용하여 Fr. 10의 유효성분을 조사한 결과 rutin (분자량, 610)으로 확인 되었다. 결론적으로 어성초의 메탄올 추출물로부터 분리한 Fr. 10은 농도 의존적으로 항산화 효능이 우수하게 나타났고, 어성초는 화장품 및 기능성 식품의 소재로 활용될 수 있음을 시사한다. This study was carried out to evaluate the antioxidant effect of methanolic extract of Houttuynia cordata (HCME) and to identify a compound having antioxidant effect. The ethyl acetate fraction of HCME showed the highest antioxidant effect in organic solvent fractions. The fraction was then separated into 12 fractions by open column chromatography. Among these fractions, the fraction 10 (Fr. 10) with the highest antioxidant activity was isolated, and its antioxidant effect was evaluated by DPPH radical scavenging activity, reducing power, TBARS, cell viability, DNA oxidation and DCF fluorescence. The Fr. 10 at a 64 μg/ml showed 60% of inhibitory effect similar to that of vitamin C at 10 μg/ml, compared with blank group. The Fr. 10 at 64 μg/ml showed 264% of reducing power, compared with blank group. TBARS assay showed that the Fr. 10 at 64 μg/ml had 35.5% of inhibitory effect similar to that of vitamin E at 1,000 μg/ml, compared with blank group. The Fr. 10 above 32 μg/ml displayed cytotoxicity. However, it was observed that the Fr. 10, above 1 μg/ml reduced DNA damage. DCF fluorescence assay showed that the Fr. 10 inhibited oxidative stress by H₂O₂ in a dose dependent manner. The compound of Fr. 10 was identified to be rutin whose molecular weight is 610 by the IR and LC-MS analyses. Therefore, these results suggest that the rutin of Fr. 10 could use as a natural antioxidant for development of cosmetics and functional foods.

        아버지와 어머니의 문해 신념 및 문해 상호작용이 유아의 어휘력에 미치는 영향

        This study focused on 120 children age four-five and their parents examining the parents’ literacy beliefs and literacy interaction and how the two variables affect their children’s vocabulary. The subjects were 120 children and 120 parents in Seoul, Gyeonggi Province and Sejong City. ‘Parental literacy beliefs’ and ‘literacy interaction of parents’ were measured using questionnaires by Lee and Kim (2011) and Son and Kim (2012). To estimate the vocabulary of the children, this study used the Receptive Expressive Vocabulary Test: REVT (Kim, Hong, Kim, Jang, & Lee, 2009). A researcher visited institutes and conducted the procedure individually. The data was analyzed using the t-test, Pearson’s correlation, and the multiple regression. The results of this study are as follow. First, there was a statistically significant difference between each parent’s literacy beliefs. There was also a statistically significant difference between each parent’s literacy interaction. Second, there were significant correlations between each parent’s literacy beliefs and literacy interactions. Interestingly, The higher the fathers’ beliefs in literacy, the more openly they interacted, and the more they read and interacted with pictures. Additionally, the higher the mothers’ beliefs, the more open interaction with picture-book reading interaction and the more directive interaction. Third, we examined the effects of each parent’s literacy beliefs and literacy interactions on children’s vocabulary skills. The father’s ‘parental role’ and the mother’s ‘parental reading influence’ showed significant influence over the vocabulary skills of the children. Also, the children’s vocabulary skills and father’s ‘picture-book reading interaction showed significant influence. In conclusion, the interaction between each parent’s literacy beliefs and father’s literacy interaction influences their children’s vocabulary skills.

        <P>Ternary logic circuit has been studied for several decades because it can provide simpler circuits and subsequently lower power consumption via succinct interconnects. We demonstrated a ternary full adder exhibiting a low power-delay-product of ~10<SUP>−16</SUP> J, which is comparable to the binary equivalent circuit. The ternary full adder was modeled using device parameters extracted from the experimentally demonstrated multi-V<SUB>th</SUB> ternary graphene barristors.</P>

        Study on the Improvement of Health and Nutrition Status After a 12-week Protein-Rich Supplementation Regimen in Children and Adolescents With Brain Lesions Disorder

        Through a survey on dietary intake of children and adolescents with brain lesions, the present study aimed to analyze the current status of nutrient intake and examine the effect of high-protein nutrient drink on their nutritional and muscle statuses. The study participants were 90 juvenile participants aged 8-19 years, with brain lesions. The participants were provided with a protein nutrient drink for 12 weeks and a questionnaire survey on dietary intake was performed to analyze the level of nutrient intake before and after ingestion. The physical measurements were taken to determine the improvements in nutrient and muscle statuses. The results showed that, before the intake of protein nutrient drink as a supplement, the participants exhibited lower height, weight, and body mass index than those of the standard levels of healthy individuals, and the level of nutrient intake through diet was lower than those of the required and recommended levels of nutrient intake for Koreans. Conversely, after the intake of protein nutrient drink for 12 weeks, the level of nutrient intake and physical statuses such as weight showed significant improvements. In addition, the muscle status had undergone approximately 10% of change during the intervention with no significant difference. Thus, to ensure an adequate level of nutrient supply to children and adolescents with brain lesions, there is an urgent need to develop a guideline of nutrient intake. The findings in this study are expected to serve as the basic data for such guidelines.
