This study aims to examine how current educational content should be transformed to enable readers to demonstrate their capabilities as practical knowledge. To this end, we examined our reading education in contrast to the Common Core State Standards (CCSS) in the United States. The results showed that reading curriculum is biased toward the reading function, and it is necessary to revise the educational content that was seemingly selected without evidence and focus on key functions. Furthermore, when selecting functions, it is necessary to enhance the system and affiliation between categories, as well as to increase the specificity, practicality and consistency of functional statements, by presenting them as principles of iterative deepening when arranging them. Accordingly, we proposed a content system table centered on “function” and “genre, text, attitude, and context” as categories of different layers. And decoding, reading fluency, changes of high school ‘reading’ subjects are required to enhance practicality of reading education, This is expected to increase the practicality of reading education as practical knowledge.
Isolation and Characterization of the Colletotrichum acutatum ABC Transporter CaABC1

Fungi tolerate exposure to various abiotic stresses, including cytotoxic compounds and fungicides, via their ATP-driven efflux pumps belonging to ATP-binding cassette (ABC) transporters. To clarify the molecular basis of interaction between the fungus and various abiotic stresses including fungicides, we constructed a cDNA library from germinated conidia of Colletotrichum acutatum, a major anthracnose pathogen of pepper (Capsicum annum L.). Over 1,000 cDNA clones were sequenced, of which single clone exhibited significant nucleotide sequence homology to ABC transporter genes. We isolated three fosmid clones containing the C. acutatum ABC1 (CaABC1) gene in full-length from genomic DNA library screening. The CaABC1 gene consists of 4,059 bp transcript, predicting a 1,353-aa protein. The gene contains the typical ABC signature and Walker A and B motifs. The 5'-flanking region contains a CAAT motif, a TATA box, and a Kozak region. Phylogenetic and structural analysis suggested that the CaABC1 is a typical ABC transporter gene highly conserved in various fungal species, as well as in Chromista, Metazoans, and Viridiplantae. We also found that CaABC1 was up-regulated during conidiation and a minimal medium condition. Moreover, CaABC1 was induced in iprobenfos, kresoxim-methyl, thiophanate-methyl, and hygromycin B. These results demonstrate that CaABC1 is necessary for conidiation, abiotic stress, and various fungicide resistances. These results will provide the basis for further study on the function of ABC transporter genes in C. acutatum.

Purpose The aim of this work was to evaluate a new polylactic acid microsphere depot formulation (Leuprolide Depot 3 M) prepared by a water-in-oil-in-water emulsion/solvent evaporation method. Methods The physical properties of the microspheres, including particle size and drug encapsulation efficiency, were characterized. The accelerated and long-term in vitro release profiles were examined. Pharmacokinetic and pharmacodynamic studies were conducted in male beagle dogs after a single subcutaneous injection of the microspheres at 11.25 mg/animal leuprolide acetate by measuring the plasma leuprolide and testosterone levels, respectively. Results Leuprolide Depot 3 M was spherical in shape with a smooth surface and showed an average diameter of 17.83 μm and a drug loading content of 7.58% (w/w). These properties were comparable to those of Lupron Depot 30 mg, a commercial product. In vitro release of leuprolide at 37 °C from both formulations was sustained for up to 126 days and showed a similarity factor (f2) of 51.0. In the pharmacokinetic study, the plasma leuprolide concentration after subcutaneous injection of Leuprolide Depot 3 M or Lupron Depot 30 mg remained at approximately 0.1 ng/mL for up to 98 days. There were no significant differences in the pharmacokinetic parameters between the two formulations. Moreover, the plasma testosterone level was effectively suppressed to less than 0.5 ng/mL until the end of the evaluation. Conclusion These results suggest that Leuprolide Depot 3 M and Lupron Depot 30 mg are expected to have comparable efficacy profiles in clinical settings, which should be confirmed in further clinical studies.
Genome wide analysis of rice blast pathogen, Magnaporthe oryzae, in Korea

Magnaporthe oryzae is a causal pathogen of the rice blast, the most destructive disease of rice worldwide. This ascomycete fungus has been intensively studied as a model organism in plant-pathogen interactions. Recently, the genome sequence of M. oryzae 70-15 was published by International Rice Blast Consortium. This led us to investigate comparative and functional analysis of this pathogen at genome-wide level. As a first step, sequencing project of M. oryzae Korean strain KJ201 was launched to face practical interests on diversity of plant pathogens. We constructed a fosmid library with a copy number controllable pCC1 vector. Over 13,000 end reads from 6637 fosmid clones were generated and anchored on genome sequence of the strain 70-15. Currently, 9,793 end reads, in which 2.684, clones with both ends and 1,757 clones with on end matched, are aligned to the sequence of strain 70-15. In the next step, shotgun sequencing of fosmid clones was conducted for fine scale comparative analysis of chromosome 7 that is well defined in genome sequencing of 70-15. Totally 119 fosmid clones were pooled in region and analyzed. For reasonable intra-species comparison in genome of M. oryzae, two strains, KJ201 and Guy11, were chosen for whole genome shotgun sequencing analysis with GS454 FLX system. About 8X genome coverage sequences were obtained for each strain and being merged for comparative analysis. Genome information of other strains is being also available from China and Japanese research groups and could be joined together for extended comparative analysis and the results give more detail in genome diversity in species level.

This study examined students' perceived importance of the benefits derived from an international multi-course collaborative (IMCC) project and how these perceptions impact students' extrinsic and intrinsic motivations for using the project Facebook (i.e., perceived usefulness and enjoyment), attitude toward the project, and intent to participate in future IMCC projects. The data were collected from 96 students who participated in the IMCC project and were enrolled in four different fashion merchandising courses in two different countries, the U.S. and South Korea. This study found that perceived enjoyment of the project Facebook influenced attitude toward the project and intent to participate in future IMCC projects. Perceived usefulness and enjoyment were predicted by the perceived importance of cultural understanding and feedback exchange among international group members. Attitude was influenced by the perceived importance of networking through the IMCC project, while intent to participate in future IMCC projects was predicted by perceived importance of cultural understanding and networking through the IMCC project. This study provides educators in the related disciplines insights about how to incorporate social network sites into course curricula to motivate students' project participation and learning.

<P>Specialized plant tissues, such as the epidermis of a leaf covered with stomata, consist of soft materials with deformability and electrochemical properties to achieve specific functions in response to various environmental stimuli. Stimulus-responsive hydrogels with electrochemical properties are good candidates for imitating such special functionalities in nature and thus have great potential in a wide range of academic and industrial applications. However, hydrogel-incorporated conductive materials are usually mechanically rigid, which limits their application in other fields. In addition, the fabrication technology of structured functional hydrogels has low reproducibility due to the required multistep processing. Here, inspired by nature, specifically the stimulus-responsive functionalities of plants, a new thermo-responsive multifunctional hybrid membrane (HM) is synthesized through the in situ hybridization of conductive poly(pyrrole) (PPy) on a photopolymerized poly(N-isopropylacrylamide) (PNIPAm) matrix. The morphological and electrical properties of the fabricated HM are investigated to characterize various aspects of its multiple functions. In terms of morphology, the HM can be easily fabricated into various structures by smartly utilizing photopolymerization patterning, and it exhibits thermo-responsive deformability. In terms of functionality, it exhibits various electrical and charge responses to thermal stimuli. This simple and efficient fabrication method can be used as a promising platform for fabricating a variety of functional devices.</P>
