Anchored protease-activatable polymersomes for molecular diagnostics of metastatic cancer cells

<P>Real-time quantitative and qualitative analyses of metastasis-associated proteases are critical for precise diagnosis and novel therapeutic treatment of advanced cancers. However, conventional methods based on DNA, peptides, and proteins require sophisticated chemistry and additional processes to expose detection moieties, and they lack elements of temporal control, which limit their applicability. We designed unique protease-activatable polymersomes (PeptiSomes) for high sensitivity,<I>in situ</I>quantitative analysis of activating membrane-type 1 matrix metalloproteinases (MT1-MMP, MMP14). To do this, we first synthesized an amphiphilic block polymer-peptide and a copolypeptide based on mPEG-<I>b</I>-pLeu and MT1-peptide-<I>b</I>-pLeu, respectively. Amphiphilic self-assembled PeptiSomes in water were capable of disassembling and releasing the encapsulated self-quenched fluorescence dye (calcein)<I>via</I>enzymatic activation by MT1-MMP. Our PeptiSome system may potentially prevent the initiation and progression of cancer metastasis. Furthermore, the PeptiSome approach described here is likely to facilitate the development of rapid protease assay techniques and further extend the role of proteases as metastasis indicators and therapeutic targets.</P>
High Involvement Work Practices and Temporary Employment Practices

본 연구는 미국 뉴저지 주의 소재 병원들에 종사하는 간호사들의 설문조사를 통해, 고성과(고참여)작업관행과 임시고용관행이 노동자들에게 어떤 영향을 미치는지 고찰했다. 이를 위해 임의적 노력이라는 개념이 개발되었는데, 이는 요소 분석을 통해 더 열심히 일하기, 더 현명하게 일하기와 더 친절하게 일하기라는 세 가지로 분류되었다. 고성과(고참여)작업관행은 이 중 더 현명하게 일하기로 부분적으로 매개되어 돌봄 서비스 질 인식과 긍정적 관계를 보여주었으며, 임시고용관행은 더 열심히 일하기로 부분적으로 매개되어 돌봄 서비스 질 인식과 부정적 관계를 보여주었다. 이 연구는 이렇게 상이한 상관관계를 가진 고성과(고참여)작업관행과 임시고용관행을 동시에 연구함으로써 현재 많이 사용되고 있는 이들 관행들의 사용을 재고해 볼 필요를 제기한다. This paper examines how two popular employment practices, high involvement work practices (HIWPs) and temporary employment practices, work on workers and firms, using a survey of nurses in New Jersey, U.S.A., hospitals. For this purpose, discretionary work effort construct is developed, resulting in three factors ? working harder, working smarter, and working kinder effort. Then, a positive relationship is found between HIWPs and perceived quality of work, partially mediated by working smarter effort, and a negative relationship is found between temporary employment practices and perceived quality of work through partial mediation by working harder effort. The simultaneous investigation of HIWPs and temporary employment practices, with these different relationships, gives a chance to reconsider on the use of these two employment trends.
A Study on the Home-Range and Habitat Use of Spot-Billed Duck (Anas poecilorhyncha) in Spring

The spring home range and habitat use of the spot-billed duck in Korea were studied using GPS-mobile phone-based telemetry (WT-300). The study areas were Anseong-si, Seosan-si, Nonsan-si, and Sejong-si. Analysis was performed using minimum convex polygon (MCP) and kernel density estimation (KDE) spot-billed ducks had an average home range of 70.28 km<sup>2</sup> (standard deviation [SD]=84.50, n=6), and a core habitat (50%) 2.66 km<sup>2</sup> (SD=2.60, n=6), according to MCP and KDE, respectively. Wetlands (41.5%) and rice fields (35.7%) were highly used as habitats. The rice field use rate was high during the day, and the wetland utilization rate was high at night. Rice fields and wetlands were the primary habitats in spring.
Another Look at Affirmative Action : Voices of U.S. Beneficiaries

적극적 고용조치는 평등의 개념을 ‘기회의 평등’에서 ‘결과의 평등’으로 확장하는 주요 정책의 하나로 설계되었다. 여성을 둘러싼 평등 대 차이 논쟁에 비추어 볼 때, 이 정책은 평등주의적 접근에 기반하고 있다. 적극적 고용조치가 비전통적 직업에 보다 많은 여성을 합류시켰다는 점에서는 긍정적으로 평가받고 있지만, 한편으로는 여성의 특성을 무시하고 남성 규범을 따르도록 했다는 비판도 받고 있다. 미국에서 적극적 고용조치를 통해 남성 중심 직업에서 일하게 된 여성들의 경험담을 통해, 이 논문은 다음과 같은 결론을 이끌어낸다. 비전통적 직업에의 여성의 존재 자체가 이들의 평등한 지위를 보장해 주지는 않는다. 그러나 이들 여성들이 모두 남성 규범을 따르는 것도 아니다. 이 여성들은 다양한 생존전략을 구사하는데, 그 중 일부는 남성성과 여성성이라는 기존 질서에 도전한다. 이들 여성들이 어떤 생존전략을 구사하는가에 관계없이 이들은 기존의 성 정체성을 해체하고 재구성하는 과정을 겪는다. 이들의 경험담은 한국에서도 적극적 고용조치가 평등하면서도 차이를 인정하는 노동 현장을 건설하는데 일조할 수 있다는 가능성을 제시한다. Affirmative Action was designed to expand the equality notion from ‘equality of opportunities’ to ‘equality of outcomes.’ Regarding the equality vs. difference debate on women, it is based on the equality side. While Affirmative Action is appraised to enable more women to get into non-traditional occupations, it is also criticized of ignoring women’s traits and making them imitate men. This paper, through voices of women in non-traditional occupations in the U.S., one group of beneficiaries of Affirmative Action, concludes as follows: The existence of women in nontraditional jobs in itself doesn’t guarantee the equal status for these women. However, it does not lead to these women accepting and following male norms, either. For survival, these women use different strategies, some challenging the given distinction between masculinity and femininity. In the process, they deconstruct the masculine and feminine identities and reconstruct them. The voices of Affirmative Action beneficiaries in the U.S. shed light on a potential of Affirmative Action for building “equal and different” workplaces in Korea.

<P>In the present study, a methanolic extract of Ecklonia cava and its solvent-partitioned fractions were evaluated for their antiadipogenic effect in 3T3-L1 adipocytes. One of them, the <I>n</I>-BuOH fraction, effectively reduced lipid accumulation and glucose consumption. In addition, the presence of the <I>n</I>-BuOH fraction in adipocytes suppressed the regulations of adipogenic transcription factors, PPARγ and SREBP1c, and adipogenic specific genes, FABP4, FABP1, FAS, LPL, HSL, and ACS1. Further purification of <I>n</I>-BuOH fractions led to the isolation of six phlorotannins (<B>1</B>–<B>6</B>). The six phlorotannins effectively suppressed triglyceride accumulation. <P>Plant homeodomain finger protein 2 (PHF2), which contains a plant homeodomain and a Jumonji-C domain, is an epigenetic regulator that demethylates lysine 9 in histone 3 (H3K9me2). On the other hand, runt-related transcription factor 2 (Runx2) plays essential roles in bone development and regeneration. Given previous reports that the PHF2 mutation can cause dwarfism in mice and that PHF2 expression is correlated with that of Runx2 in differentiating thymocytes, we investigated whether PHF2 regulates Runx2-mediated bone formation. Overexpression of PHF2 facilitated bone development in newborn mice, and viral shRNA-mediated knockdown of PHF2 delayed calvarial bone regeneration in adult rats. In primary osteoblasts and C2C12 precursor cells, PHF2 enhances osteoblast differentiation by demethylating Runx2, while suppressor of variegation 3-9 homolog 1 (SUV39H1) inhibits bone formation by methylating it. The PHF2-Runx2 interaction is mediated by the Jumonji-C and Runt domains of the two proteins, respectively. The interaction between Runx2 and osteocalcin promoter is regulated by the methylation status of Runx2, i.e., the interaction is augmented when Runx2 is demethylated. Our results suggest that SUV39H1 and PHF2 reciprocally regulate osteoblast differentiation by modulating Runx2-driven transcription at the post-translational level. This study may provide a theoretical basis for the development of new therapeutic modalities for patients with impaired bone development or delayed fracture healing.</P>
Cytotoxic Compounds from the Fruits of Vitex rotundifolia against Human Cancer Cell Lines

Activity-guided fractionation of an ethanol extract from the fruits of Vitex rotundifolia led to the isolation of three flavonoids (1–3), one lignan (4), and three phenolic compounds (5–7). The structures of compounds 1–7 were identified by NMR data. The efficacy of all compounds was evaluated by their cytotoxic activities against nine human cancer cell lines using an in vitro assay.
