The New Strategy of Object-Based Directional Query

To increase directional query efficiency, based on the study of existing algorithms of directional query, new pruning rules for directional query were given, combined with the new index structure MB-tree. The rules exclude the MBRs outside the query area and output all leaf nodes in the MBRs inside the query area. Based on the orders defined in MB-tree, a new algorithm of directional query is given combining with MB-tree by using recursive method, and the new algorithm can reduce I/O cost effectively. Experiment showed that the new directional query algorithm reduces the number of visited nodes, decreases I/O cost, and improves the efficiency of directional query.

Background: Evidence supporting an association between the 8q24 rs4242382-A polymorphism and prostate cancer (PCa) risk has been reported in North American and Europe populations, though data from Asian populations remain limited. We therefore investigated this association by clinical detection in China, and meta-analysis in Asian, Caucasian and African-American populations. Materials and Methods: Blood samples and clinical information were collected from ethnically Chinese men from Northern China with histologically-confirmed PCa (n=335) and from age-matched normal controls (n=347). The 8q24 (rs4242382) gene polymorphism was genotyped by polymerase chain reaction-high-resolution melting analysis. We initially analyzed the associations between the risk allele and PCa and clinical covariates. A meta-analysis was then performed using genotyping data from a total of 1,793 PCa cases and 1,864 controls from our study and previously published studies in American and European populations, to determine the association between PCa and risk genotype. Results: The incidence of the risk allele was higher in PCa cases than controls (0.222 vs 0.140, $P=7.3{\times}10^{-5}$), suggesting that the 8q24 rs4242382-A polymorphism was associated with PCa risk in Chinese men. The genotypes in subjects were in accordance with a dominant genetic model (ORadj=2.03, 95%CI: 1.42-2.91, $Padj=1.1{\times}10^{-4}$). Presence of the risk allele rs4242382-A at 8q24 was also associated with clinical covariates including age at diagnosis ${\geq}65$ years, prostate specific antigen >10 ng/ml, Gleason score <8, tumor stage and aggressive PCa, compared with the non-risk genotype ($P=4.6{\times}10^{-5}-3.0{\times}10^{-2}$). Meta-analysis confirmed the association between 8q24 rs4242382-A polymorphism and PCa risk (OR=1.62, 95%CI: 1.39-1.88, $P=1.0{\times}10^{-5}$) across Asian, Caucasian and African American populations. Conclusions: The replicated data suggest that the 8q24 rs4242382-A variation might be associated with increased PCa susceptibility in Asian, Caucasian and African American populations. These results imply that this polymorphism may be a useful risk biomarker for PCa in multi-ethnic populations.
Failure characteristics of combined coal-rock with different interfacial angles

In order to investigate the influence of the interfacial angel on failure characteristics and mechanism of combined coal-rock mass, 35 uniaxial/biaxial compressive simulation tests with 5 different interfacial angels of combined coal-rock samples were conducted by PFC2D software. The following conclusions are drawn: (1) The compressive strength and cohesion decrease with the increase of interfacial angle, which is defined as the angle between structure plane and the exterior normal of maximum principal plane, while the changes of elastic modulus and internal friction angle are not obvious; (2) The impact energy index $K_E$ decreases with the increase of interfacial angle, and the slip failure of the interface can be predicted based on whether the number of acoustic emission (AE) hits has multiple peaks or not; (3) There are four typical failure patterns for combined coal-rock samples including I (V-shaped shear failure of coal), II (single-fracture shear failure of coal), III (shear failure of rock and coal), and IV (slip rupture of interface); and (4) A positive correlation between interfacial angle and interface effect is shown obviously, and the interfacial angle can be divided into weak-influencing scope ($0-15^{\circ}$), moderate-influencing scope ($15-45^{\circ}$), and strong-influencing scope (> $45^{\circ}$), respectively. However, the confining pressure has a certain constraint effect on the interface effect.
Anti-proliferation Effects of Interferon-gamma on Gastric Cancer Cells

IFN-${\gamma}$ plays an indirect anti-cancer role through the immune system but may have direct negative effects on cancer cells. It regulates the viability of gastric cancer cells, so we examined whether it affects their proliferation and how that might be brought about. We exposed AGS, HGC-27 and GES-1 gastric cancer cell lines to IFN-${\gamma}$ and found significantly reduced colony formation ability. Flow cytometry revealed no effect of IFN-${\gamma}$ on apoptosis of cell lines and no effect on cell aging as assessed by ${\beta}$-gal staining. Microarray assay revealed that IFN-${\gamma}$ changed the mRNA expression of genes related to the cell cycle and cell proliferation and migration, as well as chemokines and chemokine receptors, and immunity-related genes. Finally, flow cytometry revealed that IFN-${\gamma}$ arrested the cells in the G1/S phase. IFN-${\gamma}$ may slow proliferation of some gastric cancer cells by affecting the cell cycle to play a negative role in the development of gastric cancer.
Experimental study of the association between sandstone size effect and strain rate effect

The association between sandstone size effect and strain rate effect were investigated experimentally with a split Hopkinson pressure bar (SHPB) system. The sandstone samples with Φ50 mm and different lengths were loaded under the constant ratio of punch velocity to sample length to study their size effects. Sandstone samples with constant length of 25 mm were taken as the reference to study their strain rate effects. Results indicate that, under the same velocity of the punch, strain rate of each sandstone sample is inversely proportional sample length; dynamic strength of sandstone increases with the strain rate and the length to diameter ratio (L/D), and presents a quadratic curvilinear relation with strain rate while presenting a cubic curvilinear relation with sample L/D; the reasonable L/D of Φ50 mm sandstone samples ranges from 0.5 to 0.8; that dissipated energy can present a fixed proportional relation with punch kinetic energy is unrelated to sample length.
Inhibition of SMP30 Gene Expression Influences the Biological Characteristics of Human Hep G2 Cells

Senescence marker protein 30 (SMP30), a hepatocellular carcinoma (HCe) associated antigen had been identified by our research group. To study its mechanisms of regulation and associations with the occurrence and development of HCe, we inhibited expression by RNAi technique, and observed effects on the biological characteristics of Hep G2 cells. In cell viability assays, cell growth in the experimental group (with siRNA transfection) was elevated. In Transwell invasion assays, compared with blank and control groups, numbers of invading cells in the experimental group were significantly increased, whereas in apoptosis assays, the percentage apoptosis demonstrated no differences, but after UV irradiation, that in the experimental group was higher than the other two groups. In a word, SMP30 can inhibit the proliferation and invasion of human hepatoma cells and thus can be regarded as a cancer suppressive factor.
