        광양만 유역 퇴적토 중의 다환방향족탄화수소류

        Polycyclic aromatic hydrocarbons(PAHs) in sediments of Kwangyang bay on Korea, have been investigated by gas chromatography-mass spectrometery (GC/MS). The average content of total PAHs in all samples was 2,211ppb, and the range was from 36 to 22,699pph. The higher concentrations were found in sediment sampled near Kwangyang iron processing plant, Taeindo. The resulting distributions of PAHs according to the number of aromatic ring and molecular ratios of specific aromatic compounds (phenanthrene, anthracene, fluoranthene and pyrene) have been discussed in terms of sample location, origin of the organic matter. Four, five and six rings of PAHs were mainly found near to Kwangyang iron processing plant, and three and four rings near to Yeosu chemical kombinat. This result typically indicated the local characteristics, depending on the origin of PAHs.

        광양만으로 유입하는 하천하구에서의 이화학적 특성 : 우수기 직후 유입하천에 따른 영향 effect of inputting rivers after rainy season in summer

        Physicochemical characteristics of Kwangyang Bay, have been investigated on the effects of inputting rivers just after rainy season in summer 2003. The results ranged from 4.96~8.01 mg/L for DO, 1.16~3.09 mg/L for COD, 7.21~67.71 μM for DIN and 0.12~121.84 μM for phosphate. The COD of Yeosu Area was 1.19~3.09 mg/L, Kwangyang Area was 1.23 ~ 2.76 mg/L. The COD of Wallae cheon in Yeosu Area was 3.09 mg/L and Donghoan in Kwangyang Area was 2.76 mg/L. The reason of high COD might be industrial sewage of organic chemical plants and iron foundry. DIN of Yeosu Area and Kwangyang Area were 7.21--63.43 μM and 8.43 67.71 μM respectively. The average phosphate of Yeosu Area was 23.65 μM. The source of phosphate might be industrial sewages of fertilizer plant and phosphate gypsum stocked with high amounts. The metal concentrations were measured in the range of 1.01~175.10 ㎍/L for Cu, 0.13~1.78 ㎍/L for Cd, 1.13~5.21 ㎍/L for Pb, 1.58~13.57 ㎍/L for Cr and 1.07~20.68 ㎍/L for As. The average Cu concentration was 78.99 ㎍/L in Kwangyang Area. The source of Cu might be industrial sewage of iron foundry. The average Cr concentration of Yeosu Area was 9.07 ㎍/L. That is 2 times higher than Kwangyang Bay 5.74 gg/L, because of sewage of chemical plants.

        대학생들의 생명의 기원과 진화에 대한 관점

        본 연구는 과학을 전공하고 있는 사범대 학생들의 생명에 대한 기원의 본성과 교수방법,기원 및 진화와 관련된 지식주장 에 대한 관점을 조사하고,전공,학년,종교와 관점과의 관련성에 대해 알아보고자 실시되었다. 본 연구는 광주광역시에 소재하는 한 대학의 사범계열 과학 전공 대학생 281명의 설문지 응답 결과를 대상으로 실시하였으며,설문지는 생명에 대한 기원의 본성과 교수방법에 관한 6개 문항과 기원과 진화에 관한 지식주장 11개 주제의 31개 문항으로 구성되어있다. 문항별 빈도와 평균값을 조사하고,배경별로 유의한 차이를 보인 경우 사후비교분석을 통해 관점의 차이 정도를 알아보았다. 분석 결과 대학생들은 생명에 대한 기원의 본성과 교수방법에 대하여 전반적으로 진화론과 창조론을 균등하게 바라보고 있으나,기원과 진화에 관한 지식주장에 대해 전반적으로 진화론적인 관점을 보이는 것으로 나타났다. 연구 대상이 예비 교사들이었다는 점을 감안할 때,학교에서 두 이론을 교수하는 방법이나 학생 스스로 선택하도록 하는 내용의 기원의 본성과 교수방법에 대한 경향은 진화론과 창조론을 균등하게 바라보는 관점이 우세한 것으로 나타난 것은 주목할 만한 점이며,따라서 앞으로 기원과 진화에 관한 두 이론과 그 교수방법에 대하여 예비교사들을 대상으로 하는 체계적인 연구의 수행 이 필요하다. The purpose of the study was to investigate college students' views of nature of ongms and teaching of origins and of knowledge claim of origins and evolution. A total of 281 college students who enrolled in an university located in a metropolitan city participated in the study. The instrument of this study was a questionnaire which consisted of 6 propositions on the nature of origins and teaching of origins, and 31 knowledge claims about evolution. Frequency analysis, one-way ANOVA, and post-hoc comparison analysis were conducted. Regarding the views of nature of origins and teaching of origins, college students tended to think two theories of origins equal. Regarding teaching of origins, Christian students tended to treat two theories, creationism and evolution, more equally than non-Christian students did. Biology major students showed more positive attitude toward teaching origins with two model approach than the other majors did. Regarding knowledge claims of origins and evolution, college students' views of origins were skewed toward evolution side. Views on origins were different according to major, school year and its combination.

      • 편마비 환자의 건측에 적용한 고유수용성 신경근 촉진법이 환측 근 긴장도에 미치는 영향

        The purpose of this study was to investigate the effect of proprioceptive neuromuscular facilitation (PNF) approach on the spasticity of affected side in patients with hemiplegia, Three subjects with hemiplegia participated in this study. In single-subject research design (a multiple baseline across individuals) was employed in this study. The intervention program including PNF to the unaffected side was introduced for 30 minutes each day during each intervention phase. Muscle tone of affected side was measured with Tone Assessment Scale, and active hip abduction distance of affected side was taken with the subject supine. And then the muscle tone and the hip abduction distance were measured again 30 minutes later following the intervention. The results showed that the PNF application had some beneficial effects on both muscle tone and active hip abduction. This results suggests that PNF application to non-paretic limb can be effective in reducing muscle tone and improving hip abduction range on paretic limb in persons with hemiplegia. However, further research is needed to prove the effect of PNF application on functional improvement.

      • 들민달팽이(Deroveras varians)의 배자발육과 난황단백질

        들민달팽이의 배자발육은 해부현미경으로 관찰하였다. 산란 후 4일이 경과면서 발육이 급격하게 진전되어, 10일이 지나면서 난 내에서 들민달팽이 유충의 형태를 갖추고, 12일째 부화하였다. 들민달팽이의 배자발육에 따른 난황단백질의 변화는 산란 1일째, 3일째 및 6일째 알의 난황단백질을 전기영동방법으로 분석하였다. 그 결과 배자발육과 함께 급격히 감소되는 난황단백질 관련 밴드를 관찰할 수 있었다. We characterized embryogenesis and egg proteins of Variable field Slug, Deroceras varians. The embryogenesis was observed by light microscopy. The eggs were hatched at the twelfth day after oviposition. The egg proteins of the first day, third day and sixth day after oviposition were analyzed by native-and SDS-PAGE analyses. The results showed that yolk proteins are gradually decreased during embryogenesis.

      • 누에 배자와 난소의 초대 배양에 있어서 세포의 분화형태

        곤충 세포의 초대 배양에 있어서 그 분화양상을 관찰하기 위하여 누에 배자와 난소를 초대 배양하였다. 그 결과 몇 가지 상이한 세포형이 관찰되었는데 배자 초대 배양의 분화형태는 상피형 세포와 망상조직을 형성하는 섬유상 세포가 관찰되었고, 난소에서는 주로 상피형 세포가 관찰되었다. 또한 두 조직 모두에서 거대 세포보다는 작지만 다소 큰 세포 형태가 관찰되었다. This study was carried out to investigation for differential charaterization of the cells in primary culture with insects. In the process of primary culture with embryos and larval ovaries of silkworm, Bombyx mori, three types of cells were observed. Fibroblast-like cells which formed network and epitherial-like cells migrated from embryos. But in case of ovaries, fibroblast-like cells were not found. Somewhat large cells but smaller than giant-cells were observed in the cultured embryos and ovaries.

      • Construction of Stably Transformed Bm 5 Cells by Using Autographa californica Nuclear Polyhedrosis Virus IE 0 Gene

        To construct transfurmed Bm5 cells, Autographa californica nuclear polyhedrosis virus(AcNPV) IE1 gene, an immediate early viral gene was firstly used in this study. AcNPV IE1 gene, which shares on 95.3% nucleotide sequence homology with Bombyx mori nuclear polyhedrosis virus (BmNPV) IE1 gene, was isolated and cloned into pBluescript. Neomycin gene from pKO-neo was inserted under the control of the IE1 promoter to yield pAcIE1-neo. The plasmid pAcIE1-neo was transfected into Bm5 or Sf9 cells, and neomycin-resistant cells were selected in TC100 medium containing 10% fetal bovine serum (FBS) and 1 mg/ml G4l8 for two weeks. Individual clones were picked and each was amplified for further characterization. The genomic DNA from neomycin-resistant cells was isolated and characterized by PCR using AcNPV IEI gene-specific primers and by Southern blot analysis using neomycin gene probe. We concluded that AcNPV IE1 gene was functional in B. mori-derived Bm5 cells as well as Spodoptera fugjprrda-derived Sf9 cells to produce stably-transformed insect cells

        Construction of Stably Transformed Bm5 and Sf9 Cells Displaying Green Fluorescence by Using Autographa californica Nuclear Polyhedrosis Virus IE1 Gene

        Transformed Bm5 or Sf9 cells displaying green fluorescence were constructed by using Autographa californica nuclear polyhedrosis virus (AcNPV) immediate early gene(iel). Green fluorecent protein(gfp) gane was introduced under the control of the AcNPV iel promotor to yield expression plasmid pAcIE1-GEP. It was transfected into Sf9 or Bm5 cells and cell clones expressing GFP were selected by fluorescence microscopy. Genomic DNA from transformed cells was isolated and integration of AcNPV iel gene harboring gfp gene was confirmed by PCR using AcNPV iel gene-specific primers. The GFP was successfully expressed in the cytoplasm of insect cells transformed with pAcIE1-GFP and the expressed GFP was maintained during cell division. Furthermore, GFP expression by AcNPV iel promoter in transformed cells was not interfered with viral replication. This suggests that transformed cells displaying foreign gene product by using AcNPV iel promoter will be useful for the diverse applications of the insect cells.

      • Analysis of Promoter Strength of Autographa californica Nuclear Polyhedrosis Virus IE1 Gene by Using Rreconmbinant Baculovirus

        To analysis a promoter strength of Atographa californica nucler polyhedrosis virus (AcNPV) IE1 gene, an immediate viral gene, ${\beta}$-glactosidase gene as a reporter gene was introduced under the control of the IE1 promoter. The restriction fragment containing IE1 promoter and ${\beta}$-galctosidase gene from pAcIE1-gal were inserter into pBacPAK9 to yield transfer vector pAcNPV-IE1-gal. The pAcNPV-IE1-gal was cotransfected with AcNPV genomic DNA BacPAK6 into Sf9 cells to produce recombinant baculovirus AcNPV-IE1-gal. In addition, recombinant bacvulovirus AcNPV-gal, which express ${\beta}$-galac-tosidase under the control of the polyhedrin promoter, was constrer, was constructed to compared with AcNPV-IE1-gal. The recombinant viruses were respectively infected into Sf9 cells and characterized by the virus titer and expression of ${\beta}$-galactoxidase in Sf9 cells. The promoter strength of IE1 and polyhedrin promoters was determined by the amount of ${\beta}$-galactosidase secreted into medium by viral infection. The titer of AcNPV-IE1-Gal determined by plaque assays in Sf9 cells was similar to that of AcNPV-gal. However, expression level of ${\beta}$-galactosidase by AcNPV-IE1-gal was significantly lower than that by AcNPV-gal. In conclusion, promoter strength of IE1 was approximately 25-fold lower than that of polyhedrin.
