      • MicroRNA Expression Profile Analysis Reveals Diagnostic Biomarker for Human Prostate Cancer

        Prostate cancer is a highly prevalent disease in older men of the western world. MicroRNAs (miRNAs) are small RNA molecules that regulate gene expression via posttranscriptional inhibition of protein synthesis. To identify the diagnostic potential of miRNAs in prostate cancer, we downloaded the miRNA expression profile of prostate cancer from the GEO database and analysed the differentially expressed miRNAs (DE-miRNAs) in prostate cancerous tissue compared to non-cancerous tissue. Then, the targets of these DE-miRNAs were extracted from the database and mapped to the STRING and KEGG databases for network construction and pathway enrichment analysis. We identified a total of 16 miRNAs that showed a significant differential expression in cancer samples. A total of 9 target genes corresponding to 3 DE-miRNAs were obtained. After network and pathway enrichment analysis, we finally demonstrated that miR-20 appears to play an important role in the regulation of prostate cancer onset. MiR-20 as single biomarker or in combination could be useful in the diagnosis of prostate cancer. We anticipate our study could provide the groundwork for further experiments.

        Modification of DC Flashover Voltage at High Altitude on the Basis of Molecular Gas Dynamics

        The effect of altitude on thermal conduction, surface temperature, and thermal radiation of partial arc was investigated on the basis of molecular gas dynamics to facilitate a deep understanding of the pollution surface discharge mechanism. The DC flashover model was consequently modified at high altitude. The validity of the modified DC flashover model proposed in this paper was proven through a comparison with the results of high-altitude simulation experiments and earlier models. Moreover, the modified model was found to be better than the earlier modified models in terms of forecasting the flashover voltage. Findings indicated that both the thermal conduction coefficient and the surface thermodynamics temperature of partial arc had a linear decrease tendency with the altitude increasing from 0 m to 3000 m, both of which dropped by approximately 30% and 3.6%, respectively. Meanwhile, the heat conduction and the heat radiation of partial arc both had a similar linear decrease of approximately 15%. The maximum error of DC pollution flashover voltage between the calculation value according to the modified model and the experimental value was within 6.6%, and the pollution flashover voltage exhibited a parabola downtrend with increasing of pollution.

        Background Nisoldipine can effectively suppress pulmonary arterial smooth muscle cell proliferation and c-Fos expression. Objective To identify the mechanism of the anti-inflammatory effects in monocrotaline-induced pulmonary arterial hypertension (PAH), focusing on the c-Fos/NLRP3/caspase-1 pathway. Results In a mice model of monocrotaline-induced PAH, miRNA-155 expression was increased. In an in vitro model, overexpression of miRNA-155 promoted inflammation and induced c-Fos, NLRP3, and caspase-1 protein expression. The inhibition of c-Fos reduced the effects of miRNA-155 on inflammation in an in vitro model of monocrotaline-induced PAH. The inhibition of NLRP3 reduced the effects of miRNA-155 on inflammation in an in vitro model of monocrotaline-induced PAH. Conclusions miRNA-155 increased inflammation in monocrotaline-induced PAH through c-Fos/NLRP3/caspase-1.

        Knockdown of the olfactory co-receptor Orco impairsmate recognition in Tenebrio molitor (Coleoptera: Tenebrionidae)

        The yellow mealworm beetle, Tenebrio molitor (Coleoptera: Tenebrionidae), is a common insect pest that damages stored grain. The insect olfactory co-receptor (Orco) is a transmembrane protein localized on the surface of the dendrites of olfactory sensory neurons and plays an essential role in the olfactory perception. In this study, an Orco ortholog (named as Tmol\Orco) inT.molitorwas characterized. The deduced amino acid sequence of Tmol\Orco shared significant high identities with Orcos in other insect species from distinct Orders. Reverse transcription quantitative PCR (RT-qPCR) analysis showed that Tmol\Orco was predominantly expressed in the antennae of both male and female adults, and the difference between male antennae and female antennae was not significant. Double-stranded RNA (dsRNA) directly against the Tmol\Orco mRNA was synthesized and injected into the T. molitor male adults, RT-qPCR results demonstrated that the transcription level of Tmol\Orco was significantly reduced at 6 d after the injection. The mate-seeking behavior of Tmol\Orco-silenced beetles was significantly inhibited. Compare to the control individuals, the percentage of Tmol\Orco-silenced beetles successfully find mateswas significantly decreased and the average time they spent for searching were significantly increased. These results provided first direct evidence that suppression of Tmol\Orco transcription contributes to the decline in mate recognition of T. molitor

        A Novel Ginsenosidase from an Aspergillus Strain Hydrolyzing 6-O-Multi- Glycosides of Protopanaxatriol-Type Ginsenosides, Named Ginsenosidase Type IV

        Herein, a novel ginsenosidase, named ginsenosidase type IV, hydrolyzing 6-O-multi-glycosides of protopanaxatrioltype ginsenosides (PPT), such as Re, R1, Rf, and Rg2, was isolated from the Aspergillus sp. 39g strain, purified, and characterized. Ginsenosidase type IV was able to hydrolyze the 6-O-α-L-(1→2)-rhamnoside of Re and the 6-O-β-D- (1→2)-xyloside of R1 into ginsenoside Rg1. Subsequently, it could hydrolyze the 6-O-β-D-glucoside of Rg1 into F1. Similarly, it was able to hydrolyze the 6-O-α-L-(1→2)- rhamnoside of Rg2 and the 6-O-β-D-(1→2)-glucoside of Rf into Rh1, and then further hydrolyze Rh1 into its aglycone. However, ginsenosidase type IV could not hydrolyze the 3-O- or 20-O-glycosides of protopanaxadioltype ginsenosides (PPD), such as Rb1, Rb2, Rb3, Rc, and Rd. These exhibited properties are significantly different from those of glycosidases described in Enzyme Nomenclature by the NC-IUBMB. The optimal temperature and pH for ginsenosidase type IV were 40℃ and 6.0, respectively. The activity of ginsenosidase type IV was slightly improved by the Mg2+ ion, and inhibited by Cu2+ and Fe2+ ions. The molecular mass of the enzyme, based on SDS-PAGE, was noted as being approximately 56 kDa.

        STING Negatively Regulates Double-Stranded DNA-Activated JAK1-STAT1 Signaling via SHP-1/2 in B Cells

        Recognition of cytosolic DNA initiates a series of innate immune responses by inducing IFN-I production and subsequent triggering JAK1-STAT1 signaling which plays critical roles in the pathogenesis of infection, inflammation and autoimmune diseases through promoting B cell activation and antibody responses. The stimulator of interferon genes protein (STING) has been demonstrated to be a critical hub of type I IFN induction in cytosolic DNA-sensing pathways. However, it still remains unknown whether cytosolic DNA can directly activate the JAK1-STAT1 signaling or not. And the role of STING is also unclear in this response. In the present study, we found that dsDNA directly triggered the JAK1-STAT1 signaling by inducing phosphorylation of the Lyn kinase. Moreover, this response is not dependent on type I IFN receptors. Interestingly, STING could inhibit dsDNA-triggered activation of JAK1-STAT1 signaling by inducing SHP-1 and SHP-2 phosphorylation. In addition, compared with normal B cells, the expression of STING was significantly lower and the phosphorylation level of JAK1 was significantly higher in B cells from MRL/lpr lupus-prone mice, highlighting the close association between STING low-expression and JAK1-STAT1 signaling activation in B cells in autoimmune diseases. Our data provide a molecular insight into the novel role of STING in dsDNA-mediated inflammatory disorders.

        Visual Attention Servo Control for Task-specific Robotic Applications

        This paper proposes a visual attention servo control (VASC) method which uses the Gaussian mixture model (GMM) for task-specific applications of mobile robots. In particular, low dimensional bias feature template is obtained using GMM to get an efficient attention process. An image-based visual servo (IBVS) controller is used to search for a desired object in a scene through an attention system which forms a task-specific state representation of the environment. First, task definition and object representation in semantic memory (SM) are proposed, and bias feature template is obtained using GMM deduction for features from high dimension to low dimension. Second, the features intensity, color, size and orientation are extracted to build the feature set. Mean shift method is used to segment the visual scene into discrete proto-objects. Given a task-specific object, top-down bias attention is evaluated to generate the saliency map by combining with the bottom-up saliency-based attention. Third, a visual attention servo controller is developed to integrate the IBVS controller and the attention system for robotic cognitive control. A rule-based arbitrator is proposed to switch between the episodic memory (EM)-based controller and the IBVS controller depending on whether the robot obtains the desired attention point on the image. Finally, the proposed method is evaluated on task-specific object detection under different conditions and visual attention servo tasks. The obtained results validate the applicability and usefulness of the developed method for robotics.

      • Expression of hPOT1 in HeLa Cells and the Probability of Gene Variation of hpot1 Exon14 in Endometrial Cancer are Much Higher than in Other Cancers

        To investigate the expression of hPOT1 in the HeLa cell line and screen point mutations of hpot1 in different tumor tissues a two step osmotic method was used to extract nuclear proteins. EMSA was performed to determine the expression of hPOT1 in the HeLa cell line. PCR was also employed to amplify the exon14 sequence of the hpot1 gene in various of cancer tissues. A SV gel and PCR clean-up system was performed to enrich PCR products. DNAStar was used to analyse the exon14 sequence of the hpot1 gene. hPOT1 was expressed in the HeLa cell line and the signal was gradually enhanced as the amount of extracted nuclear proteins increased. The DNA fragment of exon14 of hpot1 was successfully amplified in the HeLa cell line and all cancer tissues, point mutations being observed in 2 out of 3 cases of endometrial cancer (66.7%) despite the hpot1 sequence being highly conserved. However, the sequence of hpot1 exon14 do not demonstrate point mutations in most cancer tissues. Since hPOT1 was expressed in HeLa cell and the probability of gene point variants was obviously higher in endometrial cancer than other cancers, it may be involved in the pathogenesis of gynecological cancers, especially in cervix and endometrium.
