        김치유산균용 발현벡터 pSJE6c 개발과 이를 이용한 외래 유전자 발현

        본 연구실에서 개발한, 김치에서 분리한 Leu. mesenteroides SY2 유래 pFMBL1 을 바탕으로 구축한 셔틀벡터인, pSJE[7]를 외래유전자 발현에 적합하게 개량한 발현벡터를 구축하였다. Lactococcus lactis LM0230에서 분리한 프로모터 P6C를 pSJE에 도입하였다. P6C 염기서열을 지닌 oligonucleotide 쌍을 따로 제조한 후 annealing을 통해 짧은 DNA 단편을 얻어서 제한효소 처리후 pSJE에 도입하여 pSJE6c를 구축하였다. PSJE6c 효능 검증을 위해서 외래 유전자인 aga와 lacZ를 각각 pSJE6c에 도입하였다. P6C 프로모터와 비교를 위해 고유 프로모터를 지닌 유전자들도 각각 pSJE에 도입하였다. 재조합 plasmid들을 electroporation 방법으로 Lactobacillus brevis 2.14 균주에 도입하고 재조합균주들의 생육곡선과 효소역가 그리고 slot blot으로 전사체 농도를 측정하였다. 결과를 보면 PSJE6c에 클로닝 된 유전자들이 pSJE상의 유전자보다 효소역가들이 약 1.5배에서 2배 정도로 높았다. 전사체 농도 측정 결과도 pSJE6c 들에서 더 많은 전사체가 생성됨을 보여주었다. 이상 결과들은 효율적인 발현벡터들의 사용을 통해서 김치유산균에서 외래유전자 발현 효율을 높일 수 있음을 보여준다. Development of expression vectors is important for the basic and applied researches on kimchi LAB (lactic acid bacteria). An expression vector, pSJE6c was constructed by inserting P6C promoter sequence from Lactococcus lactis into pSJE, a shuttle vector for E. coli and Leuconostoc species. To test the efficiency of pSJE6c, aga ($\alpha$-galactosidase) and lacZ ($\beta$-galactosidase) genes were expressed in Lactobacillus brevis 2.14. Compared to the pSJE, expression levels of both genes were increased, indicating P6C promoter was better than indigenous promoters. Enzyme activities of L. brevis cells harboring pSJE6caga (pSJE6c with aga) or pSJE6Z (pSJE6c with lacZ) were 1.5-2 fold higher than those with pSJEaga (pSJE with aga) or pSJEZ (pSJE with lacZ). More RNA transcripts were detected in cells harboring pSJE6c based recombinant plasmid. The results indicated that heterologous gene expressions in kimchi LAB could be improved significantly by use of efficient expression vectors.

        유방암 치료에 대한 한의 임상 가이드라인 개발을 위한 기초연구

        Objectives : The purpose of this study was to present the clinical practice guidelines for breast cancer through the analysis of existing clinical practice guidelines in Western and Chinese medicine. Methods : The literature search was performed regarding conventional and integrative medical treatment of breast cancer using Google Scholar, KIOM OASIS, PUBMED and Jisan library of Daejeon university. Results : Breast cancer is the fifth most common cancer in Korea. Recent studies revealed that combining conventional and integrative medical treatment can reduce the sequela of surgical operation and improve survival rate. However there isn’t still a systemic clinical guideline for breast cancer in korean medicine. This study will be a preliminary study to establish clinical practice guidelines of Korean Medicine for breast cancer.Conclusions : Further objective and systemic clinical studies related to Korean Medicine are needed to develope more advanced clinical practice guidelines of breast cancer.

        한인진(韓茵蔯), 울금(鬱金), 산사(山査), 단삼(丹蔘) 발효복합추출물이 RAW264.7 및 HUVEC 세포에서 나타나는 이상지질혈증 관련 염증인자 발현 및 항산화에 미치는 영향

        Objectives: To investigate the effect of fermented extract of Artemisiae Iwayomogii Herba, Curcumae Longae, Crataegi Fructus and Salviae Miltiorrhizae Radix (FMH) on anti-inflammation associated with dyslipidemia and anti-oxidation in RAW264.7 and HUVEC cells. Methods: The total polyphenols, total flavonoids, DPPH radical scavenging activity, ABTS radical scavenging activity, and cytotoxicity of FMH were measured. RAW264.7 cells treated with FMH were tested for production of NO, and for cytokine and LTB4 levels and HUVEC cells treated with FMH were examined for production of cDNA of genes related to inflammation. Results: 1. FMH contained polyphenols and flavonoids. The DPPH and ABTS radical scavenging activity of FMH increased in a concentration-dependent manner. 2. FMH treatment inhibited the production of nitric oxide (NO), cytokines, and LTB4 in RAW264.7 cell when compared to the untreated control group. 3. FMH decreased the transcription of pro-inflammatory genes, whereas it increased transcription of anti-inflammatory genes, in HUVEC cells. Conclusion: FMH is effective as an antioxidant and for treatment and prevention of dyslipidemia, atherosclerosis, ischemic heart disease, stroke, and other cardiocerebrovascular diseases.

        복합단백발효사료와 어분의 산란계에 대한 사양가치 비교시험

        To compare the nutritive value of fish meal with that of fermented compound protein meal, the feeding trial with 288 laying hens of shaver strain was carried out for 8 weeks. The results obtained are as follows; 1. Experimental fermented compound protein meal contained less protein (39.5%) than imported fish meal (67%) and domestic fish meal (56%). However, moisture content of domestic fish meal (19.8%) was higher than that of fermented compound protein meal (10.6%) and imported fish meal (9.8%). It was found that the content of Ca, P and methionine in forign fish meal was superior to those in the fermented or domestic fish meal. 2. In growth rate, there was no statistical difference between the treatments. However, the amount of feed consumed was higher for the hens fed fermented compound protein meal than for those fed fish meal, although no significant difference was observed between the source of fish meal. 3. There was no statistical significance among the treatment in egg production and egg weight, but the hens feed fermented compound protein meal produced a little more eggs of heavier egg weight than those fed control diet. 4. These results indicate that fermented compound protein meal up to 10% level can be used in the laying hens diet satisfactorily.

        방사성 옥도 ( 131Ⅰ ) 가 Guinea Pig 의 난소 , 자궁 및 성주기에 미치는 영향

        The effects of radioiodine (^(131)I) administration on the histology of ovary and uterus and sexual cycle of the mature guinea pigs were observed. A single dose of 4.5 mC radioiodine per ㎏ body weight was injected subcutaneously. On the 1, 7, 14, 28, 42 and 56th day after radioiodine treatment, five treated and 5 control guinea pigs were sacrificed and their ovary and uterus were examined histologically. The variations in sexual cycle were examined by vaginal smear method through the experimental period. The results obtained were as follows: 1. No histological changes in the ovary were observed on the first day after treatment. On the 7th day after treatment, however, some atretic follicles were appeared and granulosa cells were changed with pyknosis and karyorrhexis. From the 14th day onwards, primordial follicles began to be damaged with a tendency to be severe gradually. After 42 days atretic follicles .and atrophy or hypoplasia of interstitial cells wore observed. On the 56th day after treatment, these ovaries began to be restored to the normal state. 2. The atrophy of myofibrils and multiplications of interstitial connective tissue of tunica muscularis were observed in the uterus from after treatment. 3. The change in sexual cycle of the treated guinea pig was apparent. The diestrus period extended in some guinea pigs on the 10th day after treatment. From the 21st day onwards, the distinct extension of diestrus period was observed.

        두부외상후 만성 기질성 정신장애 환자에서 Tc-99m HMPAO Brain SPECT 분석

        It is well known that Tc-99m HMPAO brain SPECT can reflect the functional lesions better than X-ray computerized tomography(CT) and magnetic resonance imaging(MRI) in the cerebral disorders. In order to evaluate the clinical utilities of Tc-99m HMPAO brain SPECT in patients with post-traumatic chronic organic mental disorder(OMD). We included 28 patients diagnosed as OMD in department of psychiatry after traumatic head injury. And we compared the results of Tc-99m HMPAO SPECT with those of MRI, EEG and MINI mental status examination(MMSE). The results were as follows 1) All patients diagnosed as OMD showed diffuse or focal decreased cerebral perfusion on Tc-99m HMPAO SPECT. 2) Most frequent lession on brain Tc-99m HMPAO SPECT was decreased perfusion on both frontal lobe. And most frequent lesion on brain Tc-99m HMPAO SPECT was decreased perfusion on both frontal lobe. And most frequent lesion on brain Tc-99m HMPAO SPECT showing normal brain MRI result was also decreased both frontal perfusion. 3) Eight of 28 patients showed focal brain MRI lesions(4 small frontal hygroma, 3small cerebral infarction and 1 cerebellar encephalomalacia) which were not detected in brain Tc-99m HMPAO SPECT. 4) The patients showing less than 20 points on MMSE disclosed abnormal results of EEG more frequently than those disclosing more than 20points. In conclusion, we think that Tc-99m HMPAO brain SPECT is sensitive method to detect functional lesions of the brains in patients with chronic post-traumatic organic mental disoder.

        연소도이득효과(BUC) 적용 사용후핵연료 운반용기의 임계 불확실도 평가

        In general, conventional criticality analyses for spent fuel transport/dry storage systems have been performed based on assumption of fresh fuel concerning the potential uncertainties from number density calculation of Transuranic and Fission Products in spent fuel. However, because of economic loss due to the excessive criticality margin, recently the design of transport/dry storage systems with Burnup Credit(BUC) application has been actively developed. The uncertainties in criticality analyses on transport/storage systems with BUC technique show strong dependance upon initial enrichment and burnup rate, whereas those in the conventional criticality evaluation based on fresh fuel assumption do not show such a dependance. In this study, regulatory-required uncertainties of the criticality analyses for BK 26 Cask, which is conceptually designed spent fuel transport cask with BUC corresponding to the limiting circumstances on nuclear power plants in Korea, are evaluated as a function of initial enrichment and burnup rate. Results of this study will be used as basic data for spent fuel loading curve of BK 26 Cask. 국내 외 수많은 수송 건식저장 시스템의 임계해석은 사용후핵연료내에 초우라늄물질(transuranic) 및 핵분열생성물(fission products) 계산의 불확실성을 이유로, 신연료로 가정된 가상연료를 적용하여 평가해왔다. 그러나 과도한 임계 여유도에 따른 경제적 손실이 크기 때문에 최근 들어 연소도이득(Burnup Credit, BUC)이 반영된 수송 건식저장 시스템의 설계 및 상용화가 추진되고 있다. 이러한 BUC 기술은 기존 임계해석 시요구되는 상수화된 불확실도와 달리 초기 농축도와 연소도 구간에 따라 상이한 불확실도를 갖게 된다. 이에 본 연구에서는 '국내 원전의 제한사항이 반영된 26다발 SNF 장전 BUC 적용 용기'(이하 BK 26 Cask)를 대상으로 관련 기술표준 및 설계요건에서 요구되는 불확실도를 평가하여 농축도 및 연소도의 함수로 계산하였다. 본 연구결과는 추후 BK 26 Cask 국내 사용후핵연료의 장전 수용률 분석의 기반자료로 활용된다.

        곤륜(崑崙).소부(少府) 침자(鍼刺)가 하장간막신경(下腸間膜神經) 활성(活性)과 혈압(血壓) 및 심박수(心搏數)에 미치는 영향(影響)

        Objectives : The aim of the present study is to observe effects of acupuncture related with the nerve system, Inferior mesenteric nerve activity(IMNA) was examined on BL60 acupuncture closely connected with the bladder and HT8 with little tied. Materials and methods : Acupunctures were performed during 100 seconds after stabilization. IMNA was measured by micromanipulator, preamplifier, mean arterial pressure(MAP) and heart rate were observed by blood pressure transducer, 4-channel transducer amplifier in anesthetized rat. Results : On BL60 acupuncture, the heart rate was not significantly decreased but IMNA, MAP were significantly decreased and Maximum peak of IMNA was increased during inserted acupuncture, decreased after ejected acupuncture respectively. On HT8 acupuncture, IMNA was decreased during acupuncture than before acupuncture but the others did not showed signigicant difference. Conclusions : Our results demonstrated a meridian interaction between BL60 acupoint in the bladder channel and Inferior mesenteric sympathetic nerve related to the bladder. This fact suggest that BL60 acupuncture have effect on the bladder through the nerve system. Meanwhile, IMNA was decreased during HT8 acupuncture. This result is supposed HT8 to have not meridian function but acupoint function by another mechanism. It needs to be closely examined other meridians and nerve by the optimal approaching method.
