Bioassay-guided isolation of an aqueous ethanolic extractfrom the unripe fruits of Malus pumila cv. Fuji using matrixmetalloproteinase-1 (MMP-1) inhibition and type-1 procollageninducing assays led to isolation of eight phenolic compounds,reynoutrin (1), quercitrin (2), isoquercitrin (3), phloridzin (4),phloretin-2'-O-xyloglucoside (5), 5-O-p-courmaroylquinic acid(6), chlorogenic acid methyl ester (7), and chlorogenic acid (8). The structures were established on the basis of nuclear magneticresonance and mass spectroscopic data interpretations. All isolateswere evaluated for their inhibitory effects on MMP-1, andcompounds 1, 2, and 6 exhibited potent inhibitory effects onMMP-1 with IC50 values of 1.3±0.1, 1.4±0.1, and 3.2±0.6 μM,respectively. When evaluated for their effects on type I procollagensynthesis on human fibroblast cells, compounds 1 and 3 wererespectively found to display potent activity for induction of type-1 procollagen by 78.5 and 78.6% at the tested concentration of12.5 M.
Approximate residual stress and plastic strain profiles for laser-peened alloy 600 surfaces

This paper presents approximate in-depth residual stress and plastic strain profiles for laser-peened alloy 600 surface via FE analysis. In approximations, effects of the initial welding residual stress and the number of shots are quantified. Based on FE analysis results, residual stress profiles are quantified by two variables; the maximum difference in stress before and after LSP, and the depth up to which the compressive residual stress exists. Plastic strain profiles are quantified by one variable, the maximum equivalent plastic strain at the surface. The proposed profiles are validated by comparing with published LSP experimental results for welded plates. Effects of the initial welding residual stress and the number of shots on these variables are discussed. The proposed profile can be directly applied to predict the mitigation effect of LSP on PWSCC and to efficiently perform structural integrity assessment of laser peened nuclear components.

This paper proposes strain-based failure model of A533B1 pressure vessel steel to simulate failure, followed by application to OECD lower head failure (OLHF) test simulation for experimental validation. The proposed strainbased failure model uses simple constant and linear functions based on physical failure modes with the critical strain value determined either using the lower bound of true fracture strain or using the average value of total elongation depending on the temperature. Application to OECD Lower Head Failure (OLHF) tests shows that progressive deformation, failure time and failure location can be well predicted.

This paper proposes a combined plastic and creep constitutive model of A533B1 pressure vessel steel to simulate progressive deformation of nuclear pressure vessels under severe accident conditions. To develop the model, recent tensile test data covering a wide range of temperatures (from RT to 1,100 ◦C) and strain rates (from 0.001%/s to 1.0%/s) was used. Comparison with experimental data confirms that the proposed combined plastic and creep model can well reflect effects of temperature and strain rate on tensile behaviour up to failure. In the companion paper (Part II), the proposed model will be used to simulate OECD lower head failure (OLHF) test data.

Tissue engineering using mesenchymal stem cells (MSCs) is a promising technology for treatment or regeneration of tissue defects. However, to put this technology into practical use, it is necessary to determine suitable biomaterials as well as to expand MSCs ex vivo. One major challenge of ex vivo expansion of MSCs is to avoid the use of fetal bovine serum (FBS), which is a fundamental culture supplement. In this study, we investigated effects of autologous serum on proliferation and osteoblastic differentiation of bone marrow stem cells (BMSCs) isolated from patients with skeletal diseases. We isolated BMSCs from 7 independent patients with avasular necrosis (AVN; 3 cases), osteoarthritis (OA; 2 cases), femoral head fracture (1 case) or hip dysplasia (1 case) and compared the effects of FBS, heat inactivated (HAS) and normal autologous serum (HA) on the proliferation of BMSCs. Proliferation analyses of BMSCs revealed that 5 out of 7 BMSCs cultured in 10 % HAS showed higher proliferation than cells cultured in 10 % FBS. Moreover, analysis of osteoblastic differentiation as assessed by alkaline phosphatase (ALP) staining, a marker for osteoblastic differentiation, showed that HAS, AS and FBS were capable of supporting osteoblastic differentiation of BMSCs at the similar level. Interestingly, mineralization was increased in BMSCs cultured in HAS (5 out of 6 BMSCs). These results demonstrate that HAS compared with conventional FBS has a better potential to stimulate proliferation and mineralization of BMSCs as well as to support their osteoblastic differentiation.
