열처리에 의한 Casein Micelle 의 성질변화에 관한 연구

This experiment was conducted to study the changes of physico-chemical properties and their contents of casein micelles by heat treatment at various temperatures and times. Fresh or heated casein micelles were fractionated into different sizes by various ultra-centrifugation times and the relative amount of protein of each fraction to the total protein was determined. Some properties of these fractions were also tested by rennet action. Heat-induced changes of casein micelles were observed by polyacrylamide gel electrophoresis. The results were summarized as follows; 1. When skimmilk was ultracentrifuged at 100,000xg for 60 minutes, the proportions of the casein micelles sedimented, serum casein and whey protein to the total protein were 69.97%, 9.86% and 20.47%, respectively. By the heat treatment at 95℃ for 10 minutes, the proportions of these fractions to the total protein were changed to 76.13%o, 20.31% and 3.56%, respectively. 2. When skimmilk was ultracentrifuged at 100,000xg for 10, 30 and 60 minutes, the proportions of pellet 1, 2, 3 and serum casein to the total casein were 27.17%, 45.52%, 14.91% and 12.40%, respectively. In case of heat treatment at 95℃ for 10 minutes, the proportions of these fractions to the total casein were changed to 17.42%, 28.64%, 32.88% and 21.06%, respectively. 3. Non-protein nitrogen soluble in 12% TCA released from ultra-centrifugal serum casein (100,000xg, 60 min) by rennet action was decreased with increase of heating time and temperature. 4. As the results of gel electrophoresis, a new band appeared between α_s-casein and β-casein and its density was increased gradually with the heat treatment at 75℃ or higher temperature.

The ferroelastic phase in hydrogen-bonded TlH2PO4 was investigated by means of impedance spectroscopy. The temperature-dependent dielectric susceptibility exhibited perfect agreement with that expected for strain-polarization coupling. The high-temperature dielectric susceptibility revealed the corresponding Curie-Weiss temperature to be far above the antiferroelectric phase transition temperature, indicative of a novel mixed phase between the antiferroelectric and the ferroelastic phase transitions.3Hb

This study was conducted to determine how analytic-holistic thinking style, one of the cultural characteristics, affects intentionality judgment and what role the crime typicality plays in the process. To this end, the psychological mechanism of intentionality judgment was confirmed in cases where the criminal typicality was manipulated by the moral characteristics of the crime perpetrator and victim. As a result, higher the criminal typicality lead to higher intentionality judgement. In addition, it was found that people with relatively holistic thinking style (with high causality and perception of change) judge less of the perpetrator intentionality. In particular, it was confirmed that the analytic-holistic thinking style interacts with criminal typicality and affects the intentionality judgment. Specifically, people with a holistic thinking style (high causality) judged the perpetrator to have high intentionality in typical cases, but judged in a reverse way in atypical cases. It means that, holistic thinkers, compared to analytic thinkers, strongly judge intentionality based on criminal typicality considering the moral characteristics of not only the perpetrators but also the victims. Along with the significance of this study, the relationships among analytic-holistic thinking style, criminal typicality and intentionality judgment were discussed.
향장기성 두경부 편평세포암종의 미세잔존암 모델에서 GM-CSF 유전자를 이입시킨 제한복제성 헤르페스바이러스 벡터를 이용한 종양백신의 유전자 치료

Background and Objectives: The Herpes Simplex type 2 Defective Infectious Single Cycle virus (DISC virus) is attenuated virus originally produced as viral vaccines but are also efficient gene transfer vehicle. The main goals of this study were to examine the efficiencies of the gene transfer using DISC vectors for various head and neck squamous cell carcinoma cell lines and to evaluate the efficacy of vaccination with DISC virus carrying a immunomodulatory genes (GM-CSF) as cancer therapy in a organotopic oral cavity squamous cell cancer model. Materials and Methods : We determinated the gene transfer efficiency of DISC virus by x-gal stain method and proved gene and protein expression of DISC-GMCSF transfected SCCVII cells by RT-PCR and ELISA method. Also we evaluated the ex vivo vaccination effects of SCCVII/GMCSF (DISC-GMCSF transfected SCCVII vaccine) vaccine on preventing the recurrence of micro-residual tumor. After the vaccination of SCCVII/GMCSF, specific cytotoxic T-cell responses was evaluated by CTL assay. Results: At an MOI of 10 DISC virus showed 64-88% of transfection rates in various head and neck squamous cancer cell lines. SCCVII cells transduced by DISC virus vector (MOI=10) carrying the GM-CSF gene, produced 4.5 nanogram quantities of GM-CSF per 10 6 cells. In vivo vaccination using tumor cells transduced ex vivo with DISC-GMCSF resulted in better protection rate against subsequent tumor recurrence in organotopic oral cavity cancer model. Although tumor free survival rate was not statistically significantly increased in vaccination group (p=0.078), tumor specific cytotocic T-cell responses were significantly increased in SCCVII/GMCSF vaccination group. Conclusion: These data demonstrate that; 1) The DISC virus vector is capable of efficient gene transfer to various head and neck squamous cancer cell lines, 2) GM-CSF secreting genetically modified tumor vaccine (SCCVII/GMCSF) efficiently protected against tumor recurrence in organotopic micro-residual oral cavity cancer model and produced tumor specific cytotoxic T-cell response. DISC virus-mediated, cytokine gene transfer may prove to be useful as a clinical therapy for head and neck cancers.
