Transient exposure to hydrogen peroxide inhibits the ubiquitination of phosphorylated IκBα in TNFα-stimulated HEK293 cells

        Yeji Lee,Jin Choi,Kyung-Ho Ha,주대명 생화학분자생물학회 2012 Experimental and molecular medicine Vol.44 No.8

During ischemia-reperfusion injury, brief pre-exposure to oxidative stress renders organs resistant to subsequent severe damage. NF-κB is a transcription factor that is involved in reperfusion-induced inflammatory and immune responses. The activity of NF-κB has been shown to be modulated by oxidative stress in various cell types through different pathways. We studied the effect of pre-exposure to oxidative stress on subsequent NF-κB activation in TNFα-stimulated HEK293 cells. The cells were transiently exposed to 0.5 mM H2O2 for 20 min, prior to stimulation with TNFα, and the subsequent expression of NF-κB-dependent genes and the levels of NF-κB signaling molecules were measured. Pre-exposure to H2O2 significantly delayed the TNFα-induced expression of an NF-κB reporter gene and inflammatory proteins (intercellular adhesion molecule-1 and IL-1β). The degradation of inhibitor of NF-κB α (IκBα) and the nuclear translocation of NF-κB were also delayed by H2O2 treatment, whereas IκBα phosphorylation and IκB kinase activity were not changed. When we examined the ubiquitin/proteosome pathway in H2O2-treated cells,we could not detect significant changes in proteosomal peptidase activities, but we were able to detect a delay of IκBα poly-ubiquitination. Our results suggest that transient exposure to oxidative stress temporally inhibits NF-κB-dependent gene expression by suppressing the poly-ubiquitination of phosphorylated IκBα in HEK293 cells.

Microbiota-derived lactate promotes hematopoiesis and erythropoiesis by inducing stem cell factor production from leptin receptor+ niche cells

        Lee Yong-Soo,Kim Tae-Young,Kim Yeji,Kim Seungil,Lee Su-Hyun,서상욱,Zhou Bo O.,Eunju O.,김광순,권미나 생화학분자생물학회 2021 Experimental and molecular medicine Vol.53 No.-

Although functional interplay between intestinal microbiota and distant sites beyond the gut has been identified, the influence of microbiota-derived metabolites on hematopoietic stem cells (HSCs) remains unclear. This study investigated the role of microbiota-derived lactate in hematopoiesis using mice deficient in G-protein-coupled receptor (Gpr) 81 (Gpr81 − /− ), an established lactate receptor. We detected significant depletion of total HSCs in the bone marrow (BM) of Gpr81 −/− mice compared with heterogenic (Gpr81 +/− ) mice in a steady state. Notably, the expression levels of stem cell factor (SCF), which is required for the proliferation of HSCs, decreased significantly in leptin receptor-expressing (LepR + ) mesenchymal stromal cells (MSCs) around the sinusoidal vessels of the BM from Gpr81 −/− mice compared with Gpr81 +/− mice. Hematopoietic recovery and activation of BM niche cells after irradiation or busulfan treatment also required Gpr81 signals. Oral administration of lactic acid-producing bacteria (LAB) activated SCF secretion from LepR + BM MSCs and subsequently accelerated hematopoiesis and erythropoiesis. Most importantly, LAB feeding accelerated the self-renewal of HSCs in germ-free mice. These results suggest that microbiota-derived lactate stimulates SCF secretion by LepR + BM MSCs and subsequently activates hematopoiesis and erythropoiesis in a Gpr81-dependent manner.

Factors Related to Smoking Status Among Young Adults: An Analysis of Younger and Older Young Adults in Korea

Lee, Yeji,Lee, Kang-Sook The Korean Society for Preventive Medicine 2019 Journal of Preventive Medicine and Public Health Vol.52 No.2

Objectives: Young adulthood represents a critical developmental period during which the use of tobacco may begin or cease. Furthermore, differences in smoking behaviors between younger (aged 18-24 years) and older (aged 25-34 years) young adults may exist. This study aimed to characterize patterns related to current smoking in younger and older young adults. Methods: This study used data acquired from the Sixth Korea National Health and Nutrition Examination Survey conducted from 2013 to 2014. A total of 2069 subjects were categorized as younger (712 subjects) and older (1357 subjects) young adults. The chi-square test was used to assess the relationships between smoking status and socio-demographic, health-related, and smoking-related factors. Multivariable logistic regression models were constructed to assess the factors affecting current smoking in these age groups. Results: The current smoking prevalence was 18.3% among the younger young adults and 26.0% among the older young adults. Sex, education level, occupation, perceived health status, alcohol consumption, and electronic cigarette use were related to current smoking in both age groups. Secondhand smoke exposure at home and stress levels showed significant relationships with smoking in younger and older young adults, respectively. Conclusions: Strong correlations were found between the observed variables and smoking behaviors among young adults. Determining the factors affecting smoking and designing interventions based on these factors are essential for smoking cessation in young adults.

Immunogenicity of lipid nanoparticles and its impact on the efficacy of mRNA vaccines and therapeutics

        Lee Yeji,Jeong Michaela,Park Jeongeun,Jung Hyein,Lee Hyukjin 생화학분자생물학회 2023 Experimental and molecular medicine Vol.55 No.-

Several studies have utilized a lipid nanoparticle delivery system to enhance the effectiveness of mRNA therapeutics and vaccines. However, these nanoparticles are recognized as foreign materials by the body and stimulate innate immunity, which in turn impacts adaptive immunity. Therefore, it is crucial to understand the specific type of innate immune response triggered by lipid nanoparticles. This article provides an overview of the immunological response in the body, explores how lipid nanoparticles activate the innate immune system, and examines the adverse effects and immunogenicity-related development pathways associated with these nanoparticles. Finally, we highlight and explore strategies for regulating the immunogenicity of lipid nanoparticles.

      • Hydrogen production from the photocatalytic hydrolysis of sodium borohydride in the presence of In–, Sn–, and Sb–TiO2s

Lee, Yeji,Kim, Youngmi,Jeong, Harim,Kang, Misook Elsevier 2008 Journal of industrial and engineering chemistry Vol.14 No.5

<P><B>Abstract</B></P><P>To enhance the evolution of hydrogen from the photocatalytic hydrolysis of sodium borohydride solution, three photocatalysts; 0.5mol% In, Sn, and Sb, incorporated into TiO2, were prepared using a hydrothermal method. The particle sizes of well-developed anatase were about 5.0nm, except those for Sb–TiO2, which were above 5.0nm, with closed to cubic form shapes, except for pure TiO2 that was spherical. In the X-ray photoelectron spectra (XPS) for Ti2p3/2, a band at 460.34eV, attributed to partial MTiO3, was seen in Sn–TiO2. Additionally, two bands of Sn3d5/2 at 484.5 and 486.8eV, assigned to the Sn and SnO or SnO2, respectively, appeared over Sn–TiO2, with the ratios of metal–OH/metal–O in the O1s bands, imply a hydrophilic property, also increased in this compared with the other catalysts. The hydrogen evolution from sodium borohydride (NaBH4) hydrolysis was enhanced in the Sn–TiO2 photocatalytic system compared to those for the other catalysts; hydrogen was evolved at 140ml/gh over Sn–TiO2, which was different from the 100ml/gh for the hydrolysis of pure NaBH4, and increased to 1350ml/g after 5h. In particular, the evolved hydrogen increased with increasing amounts of Sn–TiO2, and the production continued for 150h.</P>

        Lee, Yeji,Song, Hyerin,Ahn, Heesang,Choi, Jong-ryul,Kim, Kyujung Elsevier 2018 Sensors and actuators. B Chemical Vol.274 No.-

<P><B>Abstract</B></P> <P>A hypoalbuminemia diagnosis kit, based on an extraordinary optical transmission sensor, for accurate and fast measurements of albumin concentrations in human serum was demonstrated. First, using nanohole arrays, the sensitivity of the sensor was confirmed to be approximately 460 nm/RIU by theoretical simulations and using standard solutions with specific refractive indices. In the preliminary study of albumin measurements, non-labeled detection of a difference in albumin concentration of approximately 10 mg/ml was achieved with a spectral peak shift of 0.8972 nm and proved to be capable of diagnosing hypoalbuminemia without a surface modification process. Additionally, we successfully classified 100 samples with different concentrations into normal and hypoalbuminemia groups with a probability greater than 98%. According to the results, the hypoalbuminemia diagnosis kit has a potential to be practically employed for real-time, preclinical, label-free diagnosis with integration of advanced microfluidics and miniaturized optics.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A hypoalbuminemia diagnosis kit based on an EOT sensor was demonstrated. </LI> <LI> The sensitivity of the sensor with nanohole arrays is 463.6 nm/RIU. </LI> <LI> Label free detections for the difference of albumin concentrations about 10 mg/ml was achieved. </LI> <LI> Normal and hypoalbuminemia groups was distinguished with a probability of over 98%. </LI> </UL> </P>
