In the case of the Posterior Cruciate Ligament (PCL) reconstruction, the different fixations are used according to the tunnel method of the femur, the tibia and the type of the autotransplantation. Three popular ACL reconstructions have been regarded as the gold standard for more than 10 years, several hundred thousands of the surgeries have been accomplished. However, the secondary diseases such as the weakness of the quadriceps femoris muscle by donor-site morbidity of autotransplantation, the stiffness of the joint, the fracture of the patella, the pain in the vincent of the patellar tendon, can happen and the size of the transplanting tendon is restricted. Based on the autotransplantation, PCL reconstruction of Achilles tendon using the high tibia test of the fresh cadaver accumulated fatigue test, the advanced PCL, the collateral ligament, the meniscus reconstruction and its accumulated fatigue damage test using interference screw was carried out.

In this paper, we study an area-preserving piecewise linear map with the feature of dangerous border collision bifurcations. On the invariant set of the system, KAM-type structures, which is a typical characteristic of an area preserving map, is investigated. Specially, we show that certain KAM-curves on the invariant set are non smooth curves.

<P>We identified a p-cumate degrading gene cluster (designated cmt) with a novel organization in our genomic studies of the terpene-degrading Rhodococcus sp. strain T104. The mutant strain SN140, isolated for the inability to grow on p-cumate, accumulates 2,3-dihydroxy-p-cumate as identified by liquid chromatography-mass spectrometry and 300 MHz proton nuclear magnetic resonance spectroscopy. Reverse transcriptase polymerase chain reaction experiments showed that the cmt genes are operonic and induced specifically by growth on p-cumate. This report is the first example of identifying the genes for p-cumate degradation in a gram-positive organism based on functional data.</P>
Oxalic Acid Production in Submerged Culture of Aspegillus niger F22

Oxalic acid has a nematicidal activity against the root-knot nematode Meloidogyne incognita. High producer of oxalic acid was isolated, and then named as Aspergillus niger F22. Oxalic acid production was investigated under various temperatures from 20 – 33oC and rotational speeds in 5 L jar fermenters. Yield of oxalic acid increased with decreasing temperature. The highest yield was obtained at 23oC, showing the yield of oxalic acid of 8.7 g/L, whereas oxalic acid production was least at 33oC. At 20oC, the yield was lower than that of 23oC. At a rotational speed of 300 rpm, serious oxygen depletion was present from 48 - 72 h, resulting in low productivity of 26.2 mg /L·h. When a rotational speed was set at 600 rpm, dissolved oxygen tension was over 40% and oxalic acid production increased up to approximately 55%. Viscosity during the culture differed with temperatures. Viscosity increased with the increment of temperatures. When A. niger F22 was cultured at 23oC, viscosity was 810 cP, which was favorable for oxalic acid production.

A Phase 1 Study Using Autologous Natural Killer Cells in Patients with HAIC-Hepatocellular Carcinoma

Aims: Natural killer (NK) cell-based immunotherapy has recently been tried with advances of understanding the role of immune defense against hepatocellular carcinoma (HCC). To improve NK cells therapy, we focused to increasing delivery of NK cells and synergic effect combined with hepatic arterial infusion chemotherapy (HAIC). Methods: We did a prospective, open label, phase 1 trial of the safety and efficacy of autologous NK cells through hepatic arterial infusion as sequential therapy after HAIC in advanced HCC patients. Between March 2016 and July 2017, 11 patients were included who showed favorable response more than stable disease (SD) after 2 sessions of HAIC in advanced HCC patients with child A. A total 4 sessions of HAIC were performed the protocols of infusion of cisplatin (25/m2) and 5-fluorouracil (750/m<sup>2</sup>) for 4 days every 3-4 weeks interval. The peripheral blood mononuclear cells of patients by leukapheresis were ob-tained after 3<sup>rd</sup> HAIC and NK cells were expanded for 2 weeks under Current Good Manufacturing Practices (cGMP). Patients received planned dosage of NK cells through chemoport into hepatic artery for 5 days after 4<sup>th</sup> HAIC (3 patients; 2.5x108, 3 patients; 5x10<sup>8</sup>, 5 patients; injection of 10x10<sup>8</sup> NK cells). The primary end point was safety of NK cell injection; secondary endpoint included objective response rate (modified Response Evaluation Criteria In Solid Tumors), time to progression, dura-tion of response and immunologic efficacy. Results: Any adverse events of NK cells injection were none according to dosage. An objective response was observed in 7 patients (63.6%) included three complete responses and four partial responses. Stable disease was observed in 2 patients and progressive disease was in 2 patients and thus disease control rate was 81.8%. The mean duration of time to progression was 9.7±5.3 month and duration of response without chemotherapy was 6.1±5.2 month. The newly metastatic lesion was occurred in 3 patents (27.2%; lymph node 1 patients, Lung 2 patients). Two patients were died by tumor progression and others were still alive. The increasing immunologic response was observed in 5 patients (55 %) to evaluate cytotoxicity and NK cell proportion of peripheral mononuclear cells after NK cell injection. Conclusions: The HAIC and NK cells immunotherapy is safe and effective treatment in the advance HCC patient with favorable liver function. The additional studies are urgently required to establish the new novel treatment.

<P>Hydrogen sulfide (H<SUB>2</SUB>S) is a multifunctional signaling molecule that exerts neuroprotective effects in oxidative stress. In this article, we report a mitochondria-localized two-photon probe, SHS-M2, that can be excited by 750 nm femtosecond pulses and employed for ratiometric detection of H<SUB>2</SUB>S in live astrocytes and living brain slices using two-photon microscopy (TPM). SHS-M2 shows bright two-photon-excited fluorescence and a marked change in emission color from blue to yellow in response to H<SUB>2</SUB>S, low cytotoxicity, easy loading, and minimum interference from other biologically relevant species including reactive sulfur, oxygen, and nitrogen species, thereby allowing quantitative analysis of H<SUB>2</SUB>S levels. Molecular TPM imaging with SHS-M2 in astrocytes revealed that there is a correlation between the ratiometric analysis and expression levels of cystathionine β-synthase (CBS), the major enzyme that catalyzes H<SUB>2</SUB>S production. In studies involving DJ-1, a Parkinson’s disease (PD) gene, attenuated H<SUB>2</SUB>S production in comparison with wild-type controls was observed in DJ-1-knockout astrocytes and brain slices, where CBS expression was decreased. These findings demonstrate that reduced H<SUB>2</SUB>S levels in astrocytes may contribute to the development of PD and that SHS-M2 may be useful as a marker to detect a risk of neurodegenerative diseases, including PD.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/jacsat/2013/jacsat.2013.135.issue-26/ja404004v/production/images/medium/ja-2013-04004v_0009.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/ja404004v'>ACS Electronic Supporting Info</A></P>
