Attempts using in vitro and in vivo selection procedures have been made to search for hammerhead ribozymes that have higher activities than the wild-type ribozyme and also to determine whether other sequences might be possible in the catalytic core of the hammerhead ribozyme. Active sequences selected in the past conformed broadly to the consensus core sequence except at A9, and no sequences were associated with higher activity than that of the hammerhead with the consensus core, an indication that the consensus sequence derived from viruses and virusoids is probably the optimal sequence [Vaish et al. (1997) Biochemistry 36, 6495-6501]. Recently, during construction of ribozyme expression vectors, we isolated a mutant hammerhead ribozyme, with an insertion of G between A9 and 610.1, that appeared to show significant activity [Kawasaki et at (1996) Nucleic Acids Res. 24, 3010-3016; Kawasaki et al. (1998) Nature 393, 284-289]. We, therefore, characterized kinetic properties of the G-inserted mutant ribozymes in terms of the NUX rule. We demonstrate that the NUX rule is basically applicable to the G-inserted ribozymes and, more importantly, one type of G-inserted ribozyme was very active with k_(cat), value of 6.4 min^(-1) in 50 mM Tris-HCl (pH 8.0) and 10 m1VI MgCl₂ at 37℃.

Background/Aims: Infliximab (IFX), an antibody to tumor necrosis factor, (TNF)-α has effi cacy in treating Crohn`s disease (CD). However, knowledge of the potential effects of IFX on patients` immune profi les is lacking. The purpose of this study was to reveal the immunological effects of IFX. Methods: Twenty-two patients with a CD activity index (CDAI) of 194.2±92.9 and an average duration of disease of 3.26 months and 21 healthy controls were included. Patients were to have their fi rst IFX remission induction therapy with 3 infusions (5 mg/kg) at weeks 0, 2, and 6. Oral 5-aminosalicylic acid was the only ongoing medication in the patient population. Blood samples at baseline, 12 hours after the first infusion and at week 14 were labeled with anti-CD4/ CD25 antibodies for immunohistochemical measurement of regulatory T-cells (Treg). Serum cytokines and chemokines were measured by suspension array and ELISA. Results: CDAI signifi cantly decreased prior to the second IFX infusion (p<0.001). Clinical remission rates were 77.3% and 91% by the second and third infusions, respectively. At baseline, interleukin (IL)-6 (p<0.03), IL-8 (p<0.03), IL-10 (p=0.050), IL-13 (p<0.01), transforming growth factor-β1 (p<0.01), and ``regulated on activation, normal T cell expressed and secreted`` (RANTES) (p<0.01) were elevated in patients. After the initial IFX infusion, TNF-α (p<0.04), IL-6 (p<0.03), interferon (IFN)-γ (p<0.04), IFN-γ-inducible protein-10 (p<0.01), monocyte chemoattractant protein-1 (p<0.01), macrophage infl ammatory protein-1β (p<0.01), and RANTES (p<0.01) were decreased. IFX infusion was associated with an increase in Treg (p<0.01) and a decrease in the Th1 (IFN-γ)/Th2 (IL-4) ratio (p<0.03). Conclusions: IFX use was associated with restoration of the Th1/Th2 balance after a single infusion and seemed to promote induction of naive Th0 lymphocytes to Treg. LiNiO_2의 합성 최적조건 확립 및 과량의 리튬함량과 Al 도핑이 전기화학적 특성에 미치는 영향

졸-겔법을 이용하여 LiNiO_2, Li_(1+x)NiO_2(x=0.00-0.05) 및 LiAl_yNi_(1-y)O_2 (y=0.00-0.3)을 합성하였다. 우수한 전기화학적 성능을 보이는 LiNiO_2를 합성하기 위해 금속이온에 대한 아디프산의 몰비의 영향, 리튬의 함량, Al의 도핑의 영향을 실험하였다. 그 결과 합성된 LiNiO_2는 아디프산과 총 금속이온의 몰비가 1.0일때 우수한 결정 특성을 나타내었다. 겔 전구체가 분해되는 동안 발생하는 기체를 분석할 결과 LiNiO_2,의 결정성을 향상시키는데 산소가 매우 중요한 역할을 하는 것을 관찰하였다. 합성한 Li_(1+x)NiO_2는 모두 전형적인 층상구조를 나타내었으나 리튬함량이 증가하면 전기화학적 특성이 저하됨을 관찰할 수 있었다. Al을 도핑한 시료는 도핑되지 않은 LiNiO_2보다 초기용량은 낮지만 우수한 사이클 특성을 나타내었다. Al 도핑된 Li_Al_yNi_(1-y)O_2는 고온에서도 전기화학적 특성이 우수함을 관찰할 수 있었다. LiNiO_2, Li_(1+x)NiO_2(x=0.00-0.05) and LiAl_yNi_(1-y)O_2 (y=0.00-0.3) powers were synthesized using a sol-gel method. To synthesize LiNiO_2 with a good electrochemical performance, the experiments were carried out as function of the molar ratio of adipic acid to total metal ions, excess Li content, and Al doping. The synthesized LiNiO_2 powders showed a good crystal quality when the molar ratio of adipic acid to total metal ions was 1.0. The analysis of gas composition evolved during the decomposition of gel precursors revealed that oxygen played an important role in improving the crystal quality of LiNiO_2 powders. All the prepared Li_(1+x)NiO_2 powders had a typical LiNiO_2 layered structure, but the electrochemical performance was degraded with the increasing the lithium content. Al-doped LiAl_yNi_(1-y)O_2 powdwers were better than LiNiO_2 powders in cyclicality though it showed a low initial discharge capacity. The Al doped LiAl_yNi_(1-y)O_2 powders presented a good electrochemical performance even at higher temperatures.
